ABSTRACT

Background: Osteosarcoma (OS) is a malignancy originating from mesenchymal tissue. Microfibril-associated protein 2 (MFAP2) plays a crucial role in cancer, notably promoting epithelial-mesenchymal transition (EMT). However, its involvement in OS remains unexplored. Methods: MFAP2 was silenced in U2OS cells using shRNA targeting MFAP2 (sh-MFAP2) and validated by quantitative real-time polymerase chain reaction (qRT-PCR). We extracted gene chip data of MFAP2 from multiple databases (GSE28424, GSE42572, and GSE126209). Correlation analyses between MFAP2 and the Notch1 pathway identified through the gene set variation analysis (GSVA) enrichment analysis were conducted using the Pearson correlation method. Cellular behaviors (viability, migration, and invasion) were assessed via the Cell Counting Kit-8 (CCK-8), wound healing, and Transwell assays. EMT markers (N-cadherin, vimentin, and ß-catenin) and Notch1 levels were examined by western blotting and qRT-PCR. Cell morphology was observed microscopically to evaluate EMT. Finally, the role of MFAP2 in OS was validated through a xenograft tumor model. Results: OS cell lines exhibited higher MFAP2 mRNA expression than normal osteoblasts. MFAP2 knockdown in U2OS cells significantly reduced viability, migration, and invasion, along with downregulation of N-cadherin and vimentin, as well as upregulation of ß-catenin. MFAP2 significantly correlated with the Notch1 pathway in OS and its knockdown inhibited Notch1 protein expression. Furthermore, Notch1 activation reversed the inhibitory effects of MFAP2 knockdown on the malignant characteristic of U2OS cells. Additionally, MFAP2 knockdown inhibited tumor growth, expression levels of EMT markers, and Notch1 expression in OS tumor tissues. Conclusions: Our study revealed that MFAP2 was an upstream regulator of the Notch1 signaling pathway to promote EMT in OS. These findings suggested MFAP2 as a potential OS therapy target.

ABSTRACT

Food system transformation has been a widely discussed topic in international society over time. For the last few decades, China has made remarkable achievements in food production and has contributed greatly to the reduction in global hunger and poverty. Examining experiences and lessons from China's food security practices over the years is helpful to promote a national food system transformation for China, as well as other developing countries. This study systematically reviews the literature on Chinese food security studies, with the aim of assessing China's food security achievements and examining the remaining and emerging issues in the pursuit of food system transformation. The results show that China has continuously promoted food system transformation in land consolidation, agri-food production technologies, management and organization modes, food reserves, trade governance, and food consumption. These transformations ensure not only food availability, timeliness, and nutrition, but also in terms of the ecological, social, and economic sustainability, feasibility, and justice of food security. However, China is also confronting new challenges in food security, for example, malnutrition, environmental unsustainability, and reductions in diversified agri-food. In the future, China is expected to be committed to promoting healthy diets, sustainable agricultural production, climate change mitigation, and the reduction of food waste and loss to enhance its agri-food system's resilience.

ABSTRACT

Small-scale agriculture (SA) is regarded as unsustainability because of its low benefit. To protect traditional agricultural landscapes like terraces through SA will be difficult. However, in China, terraces are still maintained well by smallholders now. This study takes the family as a basic unit and SA in Hani terraced region as an object to explore its sustainability from the perspective of labor productivity (LP) through the questionnaire method. The findings are that peasant households work on both farm and non-farm jobs. They get a low income (3854.5 yuan RMB) from hybrid-rice-cropping and a high income (44,665.8 yuan RMB) from non-farm jobs but spend a small part of labors (34.23 person•days) in growing hybrid-rice and expended lots of labor (522 person•days) for non-farm jobs. In conclusion, LP of hybrid-rice-cropping is 1.32 times that of non-farm jobs for a household. The result shows SA in Hani terraced region has a higher return of labor investment than non-farm jobs. It is different from the common impression of the low benefit of SA and also explains why SA still continues to exist in the mountainous area in China now. However, SA in Hani terraced region also faces challenges with salary level rise of non-farm jobs and part-time farmers' requirement for living quality improvement. In the future, promoting industrial integration development in this region to add local employment for improving farmers' income is a feasible approach to protect terraced landscapes.
