ABSTRACT

NARROW LEAF1 (NAL1) exerts a multifaceted influence on leaf morphology and crop yield. Recent crystal study proposed that histidine 233 (H233) is part of the catalytic triad. Here we report that unlike suggested previously, H234 instead of H233 is a component of the catalytic triad alongside residues D291 and S385 in NAL1. Remarkably, residue 233 unexpectedly plays a pivotal role in regulating NAL1's proteolytic activity. These findings establish a strong foundation for utilizing NAL1 in breeding programs aimed at improving crop yield.

ABSTRACT

Maternal obesity is associated with disturbance of lipid metabolism and obesity in offspring; however, the pathogenesis is still unclear. The present study elucidated the role of potential lipid metabolism-associated long non-coding RNA (lncRNA) and identified the pathways involved in mice born to obese dams. In the present study, maternal obesity was induced by feeding a high-fat diet for 10 weeks in female C57/BL6 mice, whereas control mice were fed a standard diet. All female mice mated with healthy male mice and were allowed to deliver spontaneously. The results demonstrated that female offspring from obese dams presented a tendency to become overweight in the first 8 weeks after birth; however, maternal obesity did not significantly alter the body weight of male offspring. RNA-sequencing analysis was performed on female offspring liver at 3 weeks old. Significantly dysregulated lncRNAs and downstream targets in female offspring liver were identified using bioinformatics analysis. lncRNA, microRNA (miRNA or miR) and mRNA expression levels in liver and AML12 cells were assessed using reverse transcription-quantitative PCR. A total of 8 upregulated and 17 downregulated lncRNAs were demonstrated in offspring from obese dams and lncRNA Lockd was indicated to be a key dysregulated lncRNA. Competing endogenous RNA (ceRNA) models suggested that the lncRNA Lockd/miR-582-5p/Elovl5 pathway was key for lipid metabolism in the liver of offspring from obese dams. Finally, small interfering RNA and miRNA inhibitor transfection was used to evaluate the ceRNA models in AML12 cells. Taken together, the results of the present study indicated that lncRNA Lockd-miR-582-5p-Elovl5 network may be disrupted in lipid metabolism and lead to obesity in the offspring of obese dams. This research will provide new insights into the molecular mechanism of obesity and lipid metabolism disorder.

ABSTRACT

OBJECTIVE: Elemene is widely used to treat malignant pleural effusion in China. This meta-analysis aimed to evaluate the efficacy and safety of elemene in treating malignant pleural effusion. METHODS: Electronic databases including Pubmed, the Cochrane Library, Embase and Chinese biomedical literature database were searched until March 2017. Clinical controlled trials (CCTs) assessing the efficacy and safety of elemene in the treatment of malignant pleural effusion were included. The quality of the included studies was evaluated using the quality evaluation criteria of the Cochrane Handbook version 5.1.0. RESULTS: A total of 46 CCTs were included, with 2992 patients. Results of meta-analysis showed that elemene significantly improved the overall response rate (ORR) in controlling malignant pleural effusion (risk ratio [RR] = 1.16; 95% CI: 1.08-1.23; P < .05). Subgroup results showed that the ORR of elemene in the treatment of lung cancer patients with malignant pleural effusion (RR = 1.20, 95% CI: 1.07-1.34; P < .05) was higher than that of other cancers (RR = 1.14, 95% CI: 1.05-1.23; P < .05). Meanwhile, elemene did not significantly increase the incidences of chest pain and fever (P > .05). CONCLUSION: Elemene is suggested to have the ability of improving the treatment outcome of malignant pleural effusion with acceptable safety.

ABSTRACT

By analyzing HyJet V phosphate ester hydraulic oil environmental impacts (oil, etc.) and confounding factors (pH, temperature, etc.), the feasibility was studied for the fluorescence detection of aircraft hydraulic oil leaks. By using the fluorescence spectrophotometer at various acidities and temperatures, the fluorescence properties of HyJet V phosphate ester hydraulic oil, Jet Oil II lubricant and 2197 lubricant were gained. The experimental results are shown as following: The fluorescence peaks of HyJet V phosphate ester hydraulic oil, Jet Oil II lubricant and 2197 lubricant are at 362, 405 and 456 nm, respectively. The impact of temperature on HyJet V phosphate ester hydraulic oil is less effective; Jet Oil II lubricant and 2197 lubricant fluorescence intensity decreases with increasing temperature. When acidity increases, the fluorescence peak of HyJet V phosphate ester hydraulic oil gradient shifts from 370 to 362 nm, and the fluorescence intensity decreases; the fluorescence peak of Jet Oil II lubricant is always 405 nm, while the fluorescence intensity decreases; the fluorescence peak of 2197 lubricant at 456 nm red shifts to 523 nm, and double fluorescence peaks appeare. The results are shown as following: under the influence of the environment and interference factors, the fluorescence characteristics of HyJet V phosphate ester hydraulic oil remain unchanged, and distinguish from Jet Oil II lubricant and 2197 lubricant. Therefore, the experiments indicate that the detection of HyJet V phosphate ester hydraulic oil leak is feasible by using fluorescence method.
