ABSTRACT

The interleukin-23 (IL-23)/IL-17 immune axis has been linked to the pathology of psoriasis, but how this axis contributes to skin inflammation in this disease remains unclear. We measured inflammatory cytokines associated with the IL-23/IL-17 immune axis in the serum of patients with psoriasis using enzyme-linked immunosorbent assays. Psoriasis was induced in male C57BL/6J mice using imiquimod (IMQ) cream, and animals received intraperitoneal injections of recombinant mouse anti-IL-23A or anti-IL-17A antibodies for 7 days. The potential effects of the IL-23/IL-17 immune axis on skin inflammation were assessed based on pathology scoring, hematoxylin-eosin staining of skin samples, and quantitation of inflammatory cytokines. Western blotting was used to evaluate levels of the following factors in skin: ACT1, TRAF6, TAK1, NF-κB, and pNF-κB. The serum of psoriasis patients showed elevated levels of several cytokines involved in the IL-23/IL-17 immune axis: IL-2, IL-4, IL-8, IL-12, IL-17, IL-22, IL-23, and interferon-γ. Levels of IL-23p19 and IL-17 were increased in serum and skin of IMQ-treated mice, while ACT1, TRAF6, TAK1, NF-κB, and pNF-κB were upregulated in the skin. A large proportion of NF-κB p65 localized in nucleus of involucrin+ cells in the epidermis and in F4/80+ cells of the dermis of psoriatic lesional skin. Treating these animals with anti-IL-23 or anti-IL-17 antibodies improved pathological score and immune imbalance, mitigated skin inflammation and downregulated ACT1, TRAF6, TAK1, NF-κB, and pNF-κB in skin. Our results suggest that skin inflammation mediated by the IL-23/IL-17 immune axis in psoriasis involves activation of the ACT1/TRAF6/TAK1/NF-κB pathway in keratinocytes and macrophage.

ABSTRACT

Cutaneous squamous cell carcinoma (CSCC) is a common cancer in humans and is the second major type of skin cancer that causes death in humans. In this article, we investigated the effects of alkannin on CSCC progression. We revealed that alkannin curbed CSCC cell viability in a dose-dependent manner and accelerated CSCC cell apoptosis. In addition, alkannin expedited macrophage M1 polarization while curbing M2 polarization. Moreover, alkannin elevated phosphatase and tensin homolog (PTEN) abundance in CSCC cells. The results of bioinformatics analysis revealed that alkannin might modulate CSCC via PTEN. Downregulation of PTEN reversed the effects of alkannin on apoptosis of CSCC cells and M1/M2 polarization of macrophages. Alkannin reduced CSCC tumor growth in a mouse xenograft model. In conclusion, alkannin curbed the advancement of CSCC by expediting apoptosis and facilitating M1 polarization of macrophages by upregulating PTEN. These data may offer a therapeutic approach against CSCC.

ABSTRACT

BACKGROUND: Long non-coding RNAs (lncRNAs) play important roles in the progression of tumors. However, the function and expression of SCARNA2 in cutaneous squamous cell carcinoma (cSCC) is still unreported. METHODS: A quantitative polymerase chain reaction was applied to study the expression of SCARNA2 and miR-342-3p. Cell counting kit-8, flow cytometry and transwell assays were performed to study cell growth, cycle and cell invasion. RESULTS: We found that SCARNA2 expression is up-regulated in cSCC cell lines and SCARNA2 expression is higher in cSCC tissues than in adjacent non-tumor specimens. Ectopic expression of SCARNA2 promoted cell growth, cell cycle and invasion in SCC13 cells. In addition, the data indicate that miR-342-3p expression is down-regulated in cSCC cell lines and miR-342-3p is down-regulated in cSCC tissues compared to adjacent non-tumor specimens. We showed that the SCARNA2 expression is negatively associated with miR-342-3p in cSCC. Moreover, we noted that SCARNA2 sponges miR-342-3p expression in cSCC cells. Overexpression of SCARNA2 suppressed the miR-342-3p expressed in SCC13 cells. We found that elevated expression of SCARNA2 promotes cell growth, cell cycle and invasion via regulating miR-342-3p expression in SCC13 cells. CONCLUSIONS: These data suggest that SCARNA2 acts in an oncogenic role and may be a potential target for cSCC.

ABSTRACT

Stable rotor levitation is a challenge for rotational gyroscopes (magnetically suspended gyroscopes (MSG) and electrostatically suspended gyroscopes (ESG)) with a ring- or disk-shaped rotor, which restricts further improvement of gyroscope performance. In addition, complicated pick-up circuits and feedback control electronics propose high requirement on fabrication technology. In the proposed gyroscope, a ball-disk shaped rotor is supported by a water-film bearing, formed by centrifugal force to deionized water at the cavity of the lower supporting pillar. Water-film bearing provides stable mechanical support, without the need for complicated electronics and control system for rotor suspension. To decrease sliding friction between the rotor ball and the water-film bearing, a supherhydrophobic surface (SHS) with nano-structures is fabricated on the rotor ball, resulting in a rated spinning speed increase of 12.4% (under the same driving current). Rotor is actuated by the driving scheme of brushless direct current motor (BLDCM). Interaction between the magnetized rotor and the magnetic-conducted stator produces a sinusoidal rotor restoring torque, amplitude of which is proportional to the rotor deflection angle inherently. Utilization of this magnetic restoring effect avoids adding of a high amplitude voltage for electrostatic feedback, which may cause air breakdown. Two differential capacitance pairs are utilized to measure input angular speeds at perpendicular directions of the rotor plane. The bias stability of the fabricated gyroscope is as low as 0.5°/h.

ABSTRACT

Friction between contacting surfaces of metal materials restricts the application of mechanical support in the high-precision inertial device of a rotational gyroscope. Instead, a disk- or ring-shaped rotor is electrostatically or magnetically suspended. However, stability of the rotor suspension restricts further improvement of the measurement precision. In the developed rotational gyroscope, a stable mechanical rotor supporting scheme with low friction is achieved by fabrication of a superhydrophobic surface with similar nanostructures of the lotus leaf on the carbon steel ball of the ball-disk-shaped rotor and the addition of a water film between the rotor ball and bronze hemispherical supporting bowl, which forms a water film bearing. The special design of the ball-disk-shaped rotor makes it possible for the application of a low-friction water bearing in the gyroscope, with rotor tilting motion. With a superhydrophobic surface, friction is further decreased and the rated spinning speed increases 12.4%, resulting in approximately the same proportion of increase in the scale factor. Moreover, superhydrophobic surface reduces mechanical damping torque for precessional motion to one order smaller than electrostatic feedback torque. Thus, through close-loop control, stable damping characteristics for precessional motion are obtained. The gyroscope exhibits excellent performance with the parameters of the measurement range, scale factor, nonlinearity, resolution, bias stability, and dynamic setting time tested to be -30°/s to 30°/s, -0.0985 V/(°/s), 0.43%, 0.1°/s, 0.5°/h, 0.1 s, respectively.

BACKGROUND: Dermatological disease significantly affects patient's health-related quality of life (HrQoL). Skindex is one of the most frequently used dermatology-specific HrQoL measures. Currently no Chinese version of Skindex is available. The aim of this study was to translate and culturally adapt Skindex-29 and Skindex-16 into Chinese, and to evaluate their reliability and validity. METHODS: Translation and cultural adaption were performed following guidelines for cross-cultural adaption of health-related quality of life measures. Subsequently, a cross-sectional study was conducted in which patients with dermatological disease (n = 225) were enrolled. The Chinese version of Skindex-29 and Skindex-16 and Dermatology Life Quality Index (DLQI) were completed. Reliability was evaluated with internal consistency using Cronbach's alpha. Validity was evaluated using known-groups validity, convergent validity and factor structure validity. RESULTS: There were both seven items of Skindex-29 and Skindex-16 requiring a second forward- and backward- translation to achieve the final satisfactory Chinese version. The internal consistency reliability was high (range of Cronbach's alpha for the scales of Skindex-29 0.85-0.97, Skindex-16 0.86-0.96). Known-group validity was demonstrated by higher scores from patients with inflammatory dermatosis than from patients with isolated skin lesions (P < 0.05). Evidence of factor structure validity of the Skindex-29 and Skindex-16 was demonstrated by both exploratory factor analysis that accounted for 68.66% and 77.78% of the total variance, respectively, and confirmatory factor analysis with acceptable fitness into the expected three-factor structure. CONCLUSION: This study has developed semantically equivalent translations of Skindex-29 and Skindex-16 into Chinese. The evaluation of the instruments' psychometric properties shows they have substantial evidence of reliability and validity for use as HrQoL instruments in Chinese patients with dermatological disease.

ABSTRACT

Spectrum mobility as an essential issue has not been fully investigated in mobile cognitive radio networks (CRNs). In this paper, a novel support vector machine based spectrum mobility prediction (SVM-SMP) scheme is presented considering time-varying and space-varying characteristics simultaneously in mobile CRNs. The mobility of cognitive users (CUs) and the working activities of primary users (PUs) are analyzed in theory. And a joint feature vector extraction (JFVE) method is proposed based on the theoretical analysis. Then spectrum mobility prediction is executed through the classification of SVM with a fast convergence speed. Numerical results validate that SVM-SMP gains better short-time prediction accuracy rate and miss prediction rate performance than the two algorithms just depending on the location and speed information. Additionally, a rational parameter design can remedy the prediction performance degradation caused by high speed SUs with strong randomness movements.

ABSTRACT

Indoor positioning systems based on the fingerprint method are widely used due to the large number of existing devices with a wide range of coverage. However, extensive positioning regions with a massive fingerprint database may cause high computational complexity and error margins, therefore clustering methods are widely applied as a solution. However, traditional clustering methods in positioning systems can only measure the similarity of the Received Signal Strength without being concerned with the continuity of physical coordinates. Besides, outage of access points could result in asymmetric matching problems which severely affect the fine positioning procedure. To solve these issues, in this paper we propose a positioning system based on the Spatial Division Clustering (SDC) method for clustering the fingerprint dataset subject to physical distance constraints. With the Genetic Algorithm and Support Vector Machine techniques, SDC can achieve higher coarse positioning accuracy than traditional clustering algorithms. In terms of fine localization, based on the Kernel Principal Component Analysis method, the proposed positioning system outperforms its counterparts based on other feature extraction methods in low dimensionality. Apart from balancing online matching computational burden, the new positioning system exhibits advantageous performance on radio map clustering, and also shows better robustness and adaptability in the asymmetric matching problem aspect.

BACKGROUND: The Psoriasis Disability Index (PDI) is a widely used instrument to measure the impact of psoriasis on patients. There has not been psychometric evaluation of the Chinese version of PDI. The aim of this study was to evaluate its reliability and validity among Chinese patients with psoriasis. METHODS: A multi-center, cross-sectional study was conducted at 9 hospitals including patients aged 18 years and over. Reliability was determined by internal consistency using Cronbach's alpha. Validity was assessed through convergent validity and known groups validity. Dimensionality of the PDI was examined by exploratory factor analysis in working patients and nonworking patients respectively. RESULTS: In all, 831 patients were studied. Internal consistency of the PDI was satisfactory. Cronbach's alpha coefficient was 0.91 for the total score and over 0.70 for each subscale of the PDI. Evidence of convergent validity of the PDI was proved by excellent and moderate to good correlations with the Dermatology Life Quality Index (DLQI) and four subscales of the Short Form-36 (SF-36) (role-physical, bodily pain, social functioning, and role-emotional): r = 0.51-0.78. Known groups validity was confirmed that the PDI score discriminated well among patients with different severity of psoriasis. The dimensionality of the PDI was determined by the presence of two-factor structure for working patients and three-factor structure for nonworking patients which accounted for 57.3% and 62.3% of the variance respectively. CONCLUSION: The Chinese version of the PDI is a reliable and valid instrument to assess the impact of psoriasis on patients' lives and could be used in future quality of life assessment of Chinese patients with psoriasis.
