ABSTRACT

2,4-dinitroaniline (2,4DNBA), a significant hazardous chemical, is extensively used in industry and agriculture. The chemical accumulates in the environment for a long time, causing irreversible damage to the ecosystem. Currently, it is quite challenging to identify it by common analysis and detection techniques. Herein, a luminescent organic cocrystal (TCNB-8HQ) was prepared using 1,2,4,5-tetracyanobenzene (TCNB) as the electron acceptor and 8-hydroxyquinoline (8HQ) as the electron donor. The prepared TCNB-8HQ was used as a fluorescent probe with a fast and specific response to 2,4DNBA. This detection method possessed a linear range of 0.5-200 µmol/L with a detection limit as low as 0.085 µmol/L to detect 2,4DNBA in real samples with satisfactory spiking recovery. As revealed by fluorescence spectrum and UV-vis absorption spectrum, the detection mechanism involved competitive absorption between cocrystal material and 2,4DNBA. Moreover, the feasibility of the system was explored by preparing portable indicator strips for 2,4DNBA from organic cocrystal (TCNB-8HQ). This study not only provided an environmentally friendly gram-level preparation strategy to synthesize the fluorescent material but also investigated their application in chemical detection.

ABSTRACT

Lanthanide metal-organic frameworks (Ln-MOFs) with exceptional optical performance and structural diversity offer a unique platform for the development of luminescent materials. However, Ln-MOFs often suffer from luminescence quenching by high-vibrating oscillators, especially in aqueous solution. Thus, multiple strategies have been adopted to improve the luminescence of Ln3+. Anomalous research about water-induced lanthanide luminescence enhancement of Ln-MOFs is in the primary stage. Here, two Eu-based metal-organic framework (Eu-MOF) isomers named QXBA-Eu-1 and QXBA-Eu-2 were constructed by using the same ligand under different solvent thermal conditions, which exhibited distinctive water- and methanol-boosting emission behaviors. As for QXBA-Eu-1, water and methanol molecules replaced the free N,N-dimethylacetamide (DMA) molecules in the framework, repressed the rotation or libration suppression of the QXBA linker, and formed hydrogen bonds with the coordinated water molecules, which suppressed the O-H high-energy vibrations, reduced nonradiative transitions, stabilized the T1 state, and facilitated the intersystem crossing (ISC) process. For QXBA-Eu-2, water molecules tended to replace the coordinated DMA ligands, which altered the S1 and T1 energy levels of the ligand and facilitated the ligand-to-metal energy transfer (LMET) process and strengthened the luminescence of Eu3+. Importantly, free solvent molecules and the hydroxylation of Eu3+ centers also restrained the rotation or libration of the QXBA linker, by which the nonradiative transition was further inhibited and the lanthanide luminescence enhanced. Thus, this work not only opened an unprecedented path to enhance lanthanide luminescence in aqueous solution but also expanded its application scope.

BACKGROUND: Painless gastroenteroscopy is a widely developed diagnostic and treatment technology in clinical practice. It is of great significance in the clinical diagnosis, treatment, follow-up review and other aspects of gastric cancer patients. The application of anesthesia techniques during manipulation can be effective in reducing patient fear and discomfort. In clinical work, the adverse drug reactions of anesthesia regimens and the risk of serious adverse drug reactions are increased with the increase in propofol application dose application dose; the application of opioid drugs often causes gastrointestinal reactions, such as nausea, vomiting and delayed gastrointestinal function recovery, after examination. These adverse effects can seriously affect the quality of life of patients. AIM: To observe the effect of modified ShengYangYiwei decoction on gastrointestinal function, related complications and immune function in patients with gastric cancer during and after painless gastroscopy. METHODS: A total of 106 patients with gastric cancer, who were selected from January 2022 to September 2022 in Xiamen Traditional Chinese Medicine Hospital for painless gastroscopy, were randomly divided into a treatment group (n = 56) and a control group (n = 50). Before the examination, all patients fasted for 8 h, provided their health education, and confirmed if there were contraindications to anesthesia and gastroscopy. During the examination, the patients were placed in the left decubitus position, the patients were given oxygen through a nasal catheter (6 L/min), the welling needle was opened for the venous channel, and a multifunction detector was connected for monitoring electrocardiogram, oxygen saturation, blood pressure, etc. Naporphl and propofol propofol protocols were used for routine anesthesia. Before anesthesia administration, the patients underwent several deep breathing exercises, received intravenous nalbuphine [0.nalbuphine (0.025 mg/kg)], followed by intravenous propofol [1.propofol (1.5 mg/kg)] until the palpebral reflex disappeared, and after no response, gastroscopy was performed. If palpebral reflex disappeared, and after no response, gastroscopy was performed. If any patient developed movement, frowning, or hemodynamic changes during the operation (heart rate changes during the operation (heart rate increased to > 20 beats/min, systolic blood pressure increased to > 20% of the base value), additional propofol [0.propofol (0.5 mg/kg)] was added until the patient was sedated again. The patients in the treatment group began to take the preventive intervention of Modified ShengYangYiwei decoction one week before the examination, while the patients in the control group received routine gastrointestinal endoscopy. The patients in the two groups were examined by conventional painless gastroscopy, and the characteristics of the painless gastroscopies of the patients in the two groups were recorded and compared. These characteristics included the total dosage of propofol during the examination, the incidence of complications during the operation, the time of patients' awakening, the time of independent activities, and the gastrointestinal function of the patients after examination, such as the incidence of reactions such as malignant vomiting, abdominal distension and abdominal pain, as well as the differences in the levels of various immunological indicators and inflammatory factors before anesthesia induction (T0), after conscious extubation (T1) and 24 h after surgery (T2). RESULTS: There was no difference in the patients' general information, American Society of Anesthesiologist classification or operation time between the two groups before treatment. In terms of painless gastroscopy, the total dosage of propofol in the treatment group was lower than that in the control group (P < 0.05), and the time of awakening and autonomous activity was significantly faster than that in the control group (P < 0.05). During the examination, the incidence of hypoxemia, hypotension and hiccups in the treatment group was significantly lower than that in the control group (P < 0.01). In terms of gastrointestinal function, the incidences of nausea, vomiting, abdominal distension and abdominal pain in the treatment group after examination were significantly lower than those in the control group (P < 0.01). In terms of immune function, in both groups, the number of CD4+ and CD8+ cells decreased significantly (P < 0.05), and the number of natural killer cells increased significantly (P < 0.05) at T1 and T2, compared with T0. The number of CD4+ and CD8+ cells in the treatment group at the T1 and T2 time points was higher than that in the control group (P < 0.05), while the number of natural killer cells was lower than that in the control group (P < 0.05). In terms of inflammatory factors, compared with T0, the levels of interleukin (IL) -6 and tumor necrosis factor-alpha in patients in the two groups at T1 and T2 increased significantly and then decreased (P < 0.05). The level of IL-6 at T1 and T2 in the treatment group was lower than that in the control group (P < 0.05). CONCLUSION: The preoperative use of modified ShengYangYiwei decoction can optimize the anesthesia program during painless gastroscopy, improve the gastrointestinal function of patients after the operation, reduce the occurrence of examination-related complications.

ABSTRACT

The concentration of vanillymandelic acid (VMA) in urine is closely related with pheochromocytoma diagnosis. Thus, it is essential to develop more accurate and convenient fluorescence sensing strategies toward VMA. Until now, the design of double ratiometric detection methods for VMA was still in the unexplored stage. In this work, novel Ln3+-based metal-organic frameworks (QBA-Eu and QBA-Gd0.875Eu0.125) possessing dual emission peaks was fabricated successfully, which served as isomers of YNU-1 and exhibited more excellent water stability in fluorescence and structure than the ones of YNU-1. The formation of the complex between QBA ligands and VMA molecules via hydrogen bonds within QBA-Eu frameworks produced a new emission band centered at 450 nm and resulted in the decline of monomer emission intensity for QBA at 390 nm. Owing to the reduced energy gap [ΔE (S1 - T1)], the antenna effect was hampered and luminescence of Eu3+ ions also decreased. The developed double ratiometric (I615nm/I475nm, I390nm/I475nm) fluorescence sensors based on QBA-Eu and QBA-Gd0.875Eu0.125 possessed the advantages of fast response (4 min), low detection limits (0.58 and 0.51; 0.22 and 0.31 µM), and wide linear ranges (2-100 and 2-80 µM), which met the requirements of pheochromocytoma diagnosis. We also applied them to determine VMA in an artificial urine sample and diluted human urine sample and obtained satisfactory results. They will become prospective fluorescence sensing platforms for VMA.

BACKGROUND: Painless gastroscopy is a widely used diagnostic and therapeutic technology in clinical practice. Propofol combined with opioids is a common drug for painless endoscopic sedation and anaesthesia. In clinical work, adverse drug reactions of anaesthesia schemes are often one of the important areas of concern for doctors and patients. With the increase in propofol dosage, the risk of serious adverse drug reactions, such as respiratory depression and hypotension, increases significantly; the use of opioids often causes gastrointestinal reactions in patients after examination, such as nausea, vomiting, delayed recovery of gastrointestinal function and other complications, which seriously affect their quality of life. AIM: To observe the effect of wrist-ankle acupuncture therapy on the anaesthesia regimen and anaesthesia-related complications during and after painless gastroscopy examination. METHODS: Two hundred patients were selected and randomly divided into a treatment group (n = 100) and a control group (n = 100). Both groups were routinely anaesthetized with the nalbuphine and propofol regimen, gastroscopy began after the patient lost consciousness, and given supportive treatment and vital sign monitoring. If the patient interrupted the surgery due to intraoperative torsion, intravenous propofol was used to relieve his or her discomfort. The treatment group received wrist-ankle acupuncture on this basis. RESULTS: The general data before treatment, American Society of Anesthesiologist (ASA) grade and operation time between the two groups was no significant difference. The Wakeup time, and the Self-ambulation time in the treatment group was significantly faster than that in the control group (P < 0.05). The total dose of propofol in the treatment group was 109 ± 8.17 mg, significantly lower than that in the control group (P < 0.05). The incidence of respiratory depression and hypotension was not significantly different, but the incidence of hiccups was significantly lower than that in the control group (P < 0.05). After the examination, the incidence of nausea, vomiting, abdominal distension, and abdominal pain was 11%, 8%, 6%, and 5%, respectively, which was significantly lower than that in the control group (P < 0.05). In addition, both the operators and the patients were more satisfied with this examination, with no significant difference between the groups (P > 0.05). CONCLUSION: Wrist-ankle acupuncture treatment can optimize the painless gastroscopy and anaesthesia scheme, reduces propofol total dose; shortens patient Wakeup time and Self-ambulation time, improves patient compliance and tolerance, is beneficial to clinical application.

ABSTRACT

Background: Pancreatic ductal adenocarcinoma (PDAC) is one of the deadliest malignancies worldwide. Immune escape is considered to be a reason for immunotherapy failure in PDAC. In this study, we explored the correlation between immune escape-related genes and the prognosis of PDAC patients. Methods: 1163 PDAC patients from four public databases, including The Cancer Genome Atlas (TCGA), International Cancer Genome Consortium (ICGC), Array-express, and Gene Expression Omnibus (GEO), were included in our study. Cox regression analysis was used to identify the 182 immune genes which were significantly associated with overall survival (OS). And then we established an immune escape-related gene prognosis index (IEGPI) score using several datasets as the training cohort and validated it using the validation cohort. Kaplan-Meier (KM) and Cox regression analysis were used to detect the relationship of IEGPI score with OS. We further explored the relationship between the IEGPI and immune indexes. And the prediction value of response for immunotherapy in Tumor Immune Dysfunction and Exclusion (TIDE) dataset. Results: We establish an IEGPI score based on 27 immune escape genes which were significantly related to the prognosis of OS in PDAC patients. Patients in the high-IEGPI group had a significantly worse overall survival rate compared with that in the low-IEGPI groups by KM curves and cox-regression. 5 of the 32 cancer types in TCGA could be significantly distinguished in survival rates through the low- and high-IEGPI groups. Moreover, the correlation between the IEGPI score was negatively correlated with an immune score in several datasets. And higher IEGPI better recurrence-free survival (RFS) and OS in the patients after patients were treated with both PD-1 and CTLA4 in the public datasets (P<0.05). Intriguingly, by using RT-PCR, we verified that the gene of PTPN2, CEP55, and JAK2 were all higher in the BxPC-3 and PANC-1 than HPDE5 cells. Lastly, we found that the IEGPI score was higher in K-rasLSL.G12D/+, p53LSL.R172H/+, Pdx1Cre (KPC) mice model with anti-PD-L1 than that without anti-PD-L1. Conclusion: Using the immune escape-related genes, our study established and validated an IEGPI score in PDAC patients from the public dataset. IEGPI score has the potential to serve as a prognostic marker and as a tool for selecting tumor patients suitable for immunotherapy in clinical practice.

ABSTRACT

Photochromic materials are constructed with molecules accompanied by structural change after triggering by light, which are of great importance and necessity for various applications. However, because of space-confinement effects, molecule stacking of these photoresponsive chromophores within coordination polymers (CPs) always results in an efficiency decrement and a response delay, and this phenomenon will lead to a poor photochromic property. Herein, a CP (named CIT-E) with a 3-fold-interpenetrating network structure, which was prepared with (Z)-1,2-diphenyl-1,2-bis[4-(pyridin-3-ylmethoxy)phenyl]ethene (1Z) and a CuI cluster, showed fast reversible photochromic behavior. Under UV-light illumination, the color of CIT-Z changed from pale yellow to reddish brown. With the illumination of green light, the polymer could return to its initial color within 10 s. To reveal the mechanism of reversible photochromic behavior of CIT-Z, single-crystal structures of each color state were fully studied, and other scientific study methods were also used, such as time-dependent density functional theory calculation and control experiments. It was found that, with light illumination, this behavior of CIT-Z was the result of a ligand-to-metal charge-transfer process, and this process was triggered by subtle molecular conformation variation of tetraphenylethylene. It should be noted that CIT-Z has high thermal and chemical stability, which are excellent advantages as smart photoresponsive materials. As a proof of concept, a uniform thin film with such a fascinating photochromic property allows applications in invisible anticounterfeiting and dynamic optical data storage. Overall, the present study opens up a new avenue toward reversible photochromic materials.

ABSTRACT

Porous coordination polymers or metal-organic frameworks with reversible phase-transition behavior possess some attractive properties, and can respond to external stimuli, including physical and chemical stimuli, in a dynamic fashion. Their phase transitions can be triggered by adsorption/desorption of guest molecules, temperature changes, high pressure, light irradiation, and electric fields; these mainly include two types of transitions: crystal-amorphous and crystal-crystal transitions. These types of porous coordination polymers have received much attention because of their interesting properties and potential applications. Herein, reversible phase transition porous coordination polymers are summarized and classified based on different stimuli sources. Corresponding typical examples are then introduced. Finally, examples of their applications in gas separation, chemical sensors, guest molecule encapsulation, and energy storage are also presented.

ABSTRACT

A series of silver coordination polymers (CPs) have been synthesized through self-assembly of three pyridinecarboxylic acid hydrazide (p-, m-, o-position) ligands with silver clusters (named Ag1-iah, Ag2-iah, and Ag3-iah). These silver CPs show different one- and two-dimensional topologies including cross-helical chains, planar network, and parallel helical chains for Ag1-iah, Ag2-iah, and Ag3-iah, respectively. The combination of experimental and computational results reveals the critical role in the space distribution of the coordination site of silver clusters and ligands in controlling the silver CPs' dimensionality and packing arrangement and modulating the optical properties and stability. Luminescent investigations reveal that Ag3-iah can selectively detect dichloromethane or trichloromethane in tetrachloromethane. These silver CPs provide a good model to study the influence of the space distribution of the coordination site of ligands on their packing arrangement and properties.

ABSTRACT

Over the last decade, the controllable reversible phase transition of functional materials has received growing interest as it shows unique suitability for various technological applications. Although many metal-organic frameworks (MOFs) possess a lamellar structure, the reversible structural transformation of MOFs between their three-dimensional (3D) phase and two-dimensional (2D) phase remains a largely unexplored area. Herein, we report for the first time a europium MOF with unprecedented reversible morphology in different solvents at room temperature. This europium MOF displayed a 3D nanorod morphology in organic solvent and a 2D nanobelt architecture in water. As a proof of concept for potential applications of this reversible-phase-transition MOF, we were able to use a delamination recovery method to load dye molecules that previously could not be loaded into europium MOFs.

ABSTRACT

Six petroleum-degrading strains were isolated from oil-contaminated soil at Dagang oil field and oil sewage on Bohai offshore drilling platform in Tianjin using enrichment culture and isolation method. The physiological biochemical test together with 16S rDNA sequencing analysis indicated that they belonged to Bacillus (S1, S2, S3, S4), Pseudomonas (W1) and Ochrobactrum (W2), respectively. The strain S3 had the maximum degradation rate of alkane (41.3%) and aromatic hydrocarbon (30.9%) among all isolated strains showing the better degradation efficiency by endogenous bacteria when compared to that by the exogenous bacteria. The four Bacillus strains were used to construct microbiome, thereafter subjected to petroleum degradation efficiency test and analyzed. The results showed that microbiome F3 consisting of S1 and S4 had the maximum degradation rates of alkane (50.5%) and aromatic hydrocarbon (54.0%), which were 69.9% and 156.1% higher than those by single bacterium, respectively. Furthermore, they were 22.1% and 74.6% respectively higher than those by the most optimal degradation bacterium S3. Microbiome F4 consisting of S2 and S3 had the minimum degradation rates of alkane (18.5%) and aromatic hydrocarbon (18.9%) which were 55.3% and 39.0% lower than the degradation rates of single bacterium, respectively. The results also demonstrated that there were both microbial synergy promotion and antagonism inhibition among bacteria of the same genus in the petroleum degradation period. Bacteria with close affinity in Bacillus genus displayed mainly promoted petroleum degradation effect.

ABSTRACT

OBJECTIVES: To investigate the effect of bilateral arcuate artery suture hemostasis of corpus uteri (haemostasia) for postpartum hemorrhage due to uterine inertia during caesarean section, and to explore the change of blood vessels and blood flow of the uterus after surgery. METHODS: From May 2009 to Dec. 2011, the 212 patients in No. 202 People's Liberation Army Hospital received bilateral arcuate artery suture hemostasis of corpus uteri for postpartum hemorrhage due to uterine inertia during caesarean section. Among them, 127 patients who failed to respond to conservative management and received haemostasia were defined as the 'haemostasia' group. 23 patients who received the suture after they failed to respond to conservative management and other conventional surgical hemostasis were defined as the 'other + haemostasia' group. 62 patients who received the suture simultaneously with conservative management were defined as the 'drug + haemostasia' group. The suture was done by the following steps: (1) The uterus should be exteriorised, and the fundus of uterus should be towards the head. (2) Transfix the anterior and posterior wall of corpus uteri with big blunt round needle and absorbable suture. The entry point was 2 cm above the uterine incision and 2 cm to lateral border of corpus uteri. The suture spanned the fundus of uterus, and was stretched tightly in front of the fundus, then tied knots were made. Bleeding volume, prompt hemostatic rate, effect rate, total effect rate and operation time were recorded. The resistance index (RI) of uterine artery, systolic/diastolic blood pressure (S/D), the visualization ratio of uterine artery and the mean value of artery diameter were obtained through color Doppler ultrasonography and enhancement CT 6 - 12 months after the surgery. RESULTS: (1) In the 'drug + haemostasia' group, the bleeding volume was (532 ± 28) ml. The operation time was (34 ± 3) min, and the prompt hemostatic rate was 97%. While the 'haemostasia' group had more bleeding volume, longer operation time and lower prompt hemostatic rate than the 'drug + haemostasia' group, with no statistically significant difference (P > 0.05). In 'other + haemostasia' group, the bleeding volume was (1379 ± 95) ml. The operation time was (79 ± 15) min, and the prompt hemostatic rate was 78%. The differences were significant when compared to the other groups (P < 0.01). There was no statistically significant difference on total effect rate among the three groups (P > 0.05). (2) There was no statistically significant difference on the RI and S/D of bilateral uterine artery among all the groups 6-12 months after the surgery. (3) The visualization ratio of left uterine artery of the 'other + haemostasia' group was lower (87%) than the 'haemostasia' group (97%) and the 'drug + haemostasia' group (95%, P < 0.05). There was no statistically significant difference between the 'haemostasia' group and the 'drug + haemostasia' group on the visualization ratio of bilateral uterine artery and the mean value of bilateral uterine artery diameter (P > 0.05). CONCLUSIONS: The bilateral arcuate artery suture hemostasis of corpus uteri is a simple, rapid, effective and safe method to control postpartum hemorrhage due to uterine inertia during caesarean section. The ovary and uterine blood flow are not affected after the surgery.

ABSTRACT

To explore one-step method for the preparation of Ganoderma lucidum multicomponent microemulsion, according to the dissolution characteristics of triterpenes and polysaccharides in Ganoderma lucidum, formulation of the microemulsion was optimized. The optimal blank microemulsion was used as a solvent to sonicate the Ganoderma lucidum powder to prepare the multicomponent microemulsion, besides, its physicochemical properties were compared with the microemulsion made by conventional method. The results showed that the multicomponent microemulsion was characterized as (43.32 +/- 6.82) nm in size, 0.173 +/- 0.025 in polydispersity index (PDI) and -(3.98 +/- 0.82) mV in zeta potential. The contents of Ganoderma lucidum triterpenes and polysaccharides were (5.95 +/- 0.32) and (7.58 +/- 0.44) mg x mL(-1), respectively. Sonicating Ganoderma lucidum powder by blank microemulsion could prepare the multicomponent microemulsion. Compared with the conventional method, this method is simple and low cost, which is suitable for industrial production.

ABSTRACT

OBJECTIVE: To observe the effect of catgut embedding (CE) at "Shimen" (CV 5) on the endometrial receptivity, pinopodes expression of the endometrial surface and leukaemia inhibitory factor (LIF) immunoactivity during periimplantation period. METHODS: Thirty Kunming mice were randomly divided into control, CE of "Shimen" (CV 5, CE-CV 5) and medication groups (n = 10/group). For mice of CE-CV 5 group, a piece of catgut was embedded into the subcutaneous tissue of "Shimen" (CV 5) on the day when the vagina-discharged epistom was found (pregnancy day 1, Pd 1). On Pd 4, mifepristone (0.1 mL, diluted in propylene glycol) was subcutaneously injected into CV 5 in rats of medication group. The expression of endometrial pinopodes (sampled on Pd 5) was observed by scanning electron microscopy. The immunoactivity of LIF of the endometrium was detected by immunohistochemistry. RESULTS: Compared with control group, the mean optical density (OD) values of LIF in CE-CV 5 and medication groups decreased significantly (P < 0.05), suggesting an downregulation of the endometrial LIF expression. No significant difference was found between CE-CV 5 and medication group in LIF expression (P > 0.05). Electronic microscopic observation showed (1) abundant mature pinopodes on the endometrial surface, with a large number of well-stacked ecphyma in control group; (2) fewer pinopodes and well-arranged smooth epithelial cells on the endometrial surface in catgut embedding group; and (3) no pinopodes found in the endometrium of medication group. CONCLUSION: Catgut embedding at "Shimen" (CV 5) can suppress the establishment of the mouse endometrial receptivity, which is possibly related to its effect in inhibiting the expression of endometrial pinopodes and LIF.

ABSTRACT

In order to comprehensively understand current situation about studies on channels and to open train of thought for the study, the relative literatures are analyzed and summarized from morphology, biophysics, biochemistry and so on. Probe into existing models of channel lines from nerves, blood vessels and nerves, gap junction, connective tissue; summarize biophysical characte-ristics of channels from temperature, loose connective tissue, biopressure effect, coherence electromagnetic field, etc.; and summarize biochemical characteristics of channels from ion concentration, exhalant of CO2, extracellular matrix and enzymology on the channel lines. Although the studies have not revealed essence of channel lines, these results reflect survey of studies on tissue structures and biological characteristics of channel lines at present.

ABSTRACT

OBJECTIVE: A novel recombinant interleukin6-Pseudomonas exotoxin fusion protein, having targeted cytotoxicity for leukemic cells highly expressing IL6R has been molecularly designed and constructed in this study. METHODS: Firstly, REDLK at C-terminus of Pseudomonas exotoxin PE40 was replaced with KDEL using point mutagenesis technology. Secondly, a cDNA encoding interleukin-6 devoid of N-terminal 24 amino acids [IL6D24] was fused to 5' terminus of PE40KDEL DNA by the method of gene splicing by overlap extension, which could generate recombinant IL6D24-PE40KDEL and IL6D24-Linker-PE40KDEL fusion genes. Thirdly, recombinant fusion genes IL6D24-PE40KDEL and IL6D24-Linker-PE40KDEL were ligated into the EcoR I and Sma I cloning sites in the pBV220 plasmids respectively and then transformed into E.coli HB101 cells. The expressed recombinant exotoxin fusion proteins were purified to electrophoresis purity by Mono Q column chromatography. Its selectively killing was tested by the MTS colorimetric method using both U937 and CEM cells lines. RESULTS: Recombinant exotoxin fusion proteins IL6D24-PE40KDEL was expressed as a form of inclusion bodies at higher level of 40% approximately of total proteins in bacterial cells. Western blot showed that the purified products could react specifically with IL6 monoclonal antibody and PEA antiserum, respectively. IL6D24-PE40KDEL was selectively cytotoxic to U937 cells expressing IL6R-positive with ID50 of 250 ng/ml, and but not CEM cells expressing IL6R-negative. CONCLUSIONS: Two novel recombinant interleukin6-Pseudomonas exotoxin fusion proteins IL6D24-PE40KDEL and IL6D24-Linker-PE40KDEL have been successfully constructed and they can selectively kill the leukemic cells expressing highly IL6R.
