ABSTRACT

The development of high-throughput sequencing techniques enabled a deeper and more comprehensive understanding of environmental microbiology. Specifically, the third-generation sequencing techniques represented by nanopore sequencing have greatly promoted the development of environmental microbiology research due to its advantages such as long sequencing reads, fast sequencing speed, real-time monitoring of sequencing data, and convenient machine carrying, as well as no GC bias and no PCR amplification requirement. This review briefly summarized the technical principle and characteristics of nanopore sequencing, followed by discussing the application of nanopore sequencing techniques in the amplicon sequencing, metagenome sequencing and whole genome sequencing of environmental microorganisms. The advantages and challenges of nanopore sequencing in the application of environmental microbiology research were also analyzed.

ABSTRACT

Objective To observe the changes of short-chain acyl-CoA dehydrogenase (SCAD) expression on human umbilical vein endothelial cell (HUVEC) apoptosis and investigate its relationship with apoptosis. Methods The HUVEC was cultured normally for 2-3 days. The apoptotic model of HUVEC was established by tert-butyl hydrogen peroxide (tBHP). The HUVEC was treated by different concentrations of tBHP (0, 10, 20, 30, 40, 50 μmol/L) for 12 hours and different time (0, 3, 6, 9, 12 hours) with 50 μmol/L tBHP to establish the apoptotic model of HUVEC. The cell viability was detected by methyl thiazolyl tetrazolium (MTT), the mRNA expression of SCAD was determined by real-time polymerase chain reaction (PCR), the protein expression of SCAD was achieved by Western Blot. The best concentrate and time were determined to interfere the HUVEC to achieve the apoptotic model of HUVEC. The SCAD gene of HUVEC was knocked down by RNA interference sequence (siRNA274, siRNA414, siRNA679). The mRNA expression of SCAD, the protein expression of SCAD and the activity of SCAD enzyme were detected to achieve the best RNA interference sequence. The HUVEC was intervened by the best RNA interference sequence and tBHP. The cell activity and apoptosis rate, the enzyme activity of SCAD, the mRNA and protein expression of SCAD, the contents of reactive oxygen species (ROS), aderosine triphosphate (ATP) and free fatty acid (FFA) were detected to observe the effect of SCAD on apoptosis of HUVEC. Results ① The cell viability, the mRNA expression and the protein expression of SCAD were decreased gradually in a concentration and time dependent manner with the increase of tBHP concentration and the prolongation of intervention time. The decline was most significant in the group of the 50 μmol/L tBHP to interfere HUVEC for 12 hours. ② The siRNA679 transfection was the most significant in reducing SCAD mRNA and protein expressions among the three interference sequences (siRNA274, siRNA414, siRNA679). ③ Compare with blank control group, the cell viability was significantly decreased in the siRNA679 group (A value: 0.48±0.09 vs. 1.00±0.09, P < 0.01), the apoptotic rate of HUVEC was significantly increased [(29.96±2.09)% vs. (2.90±1.90)%, P < 0.01], the expression of SCAD mRNA and SCAD protein, the activity of SCAD enzyme and the content of ATP were significantly decreased [SCAD mRNA (2-ΔΔCt): 0.50±0.16 vs. 1.34±0.12, SCAD/α-Tubulin: 0.67±0.11 vs. 1.00±0.06, the activity of SCAD enzyme (kU/g): 0.38±0.04 vs. 0.53±0.04, the content of ATP (μmol/g): 0.14±0.02 vs. 0.19±0.01, all P < 0.05], the contents of FFA and ROS were significantly increased [FFA (nmol/g): 0.84±0.07 vs. 0.47±0.04, ROS (average fluorescence intensity): 647.5±23.7 vs. 434.2±46.5, both P < 0.01]. Meanwhile, SCAD siRNA treatment triggered the same apoptosis as HUVEC treated with tBHP. Conclusions Down-regulation of SCAD may play an important role in HUVEC apoptosis. Increase in the expression of SCAD may become an important part in intervening HUVEC apoptosis.

ABSTRACT

Objective To explore the status,developing demands and directions of the education and training of military medical equipment.Methods The main problems of the PLA's medical equipment education and training were analyzed via being compared with those of foreign armies.According to the requirements on the campaign mode changing in the future war,the innovating mode under information-based military training,and the research and exploration of novel equipment and new technology,the developing directions in the future were ascertained.Results The interface was enhanced between new medical equipment development and educational training,and specifications were prepared for medical equipment application and maintenance.Conclutsion Exploring the pivot choke points and the developing directions will provide strategy guidance on the rapid and highly efficient elevation of military medical equipment supporting capacity.

ABSTRACT

AIM:To observe the effect of metformin on the expression of GSTM2 in spontaneously hypertensive rats,and to investigate the mechanism of the reversal of myocardial hypertrophy by metformin.METHODS:Sixteen weeks old WistarKyoto (WKY) and spontaneously hypertensive rats (SHR) were used as the experimental control.Eight weeks old WKY and SHR were administered to metformin (Met) continuously for 8 weeks as the experimental group.Systolic pressure of rats was regularly determined by NIBP instrument of heart rate and blood pressure.The hemodynamic parameters were tested by BL-420 biological experimental system and 4 track physiological recorders.The weights of left ventricle and rats were collected to calculate the left ventricular mass index.The activity of GSTM2 enzyme in left ventricle was detected by Enzyme-linked immunosorbent assay (ELISA).The protein expression of GSTM2 and p47phox and Nox4 in left ventricle were investigated by Western blot.The O2·-levels were measured by staining with dihydroethidium (DHE).RESULTS:Compared with the experimental control,the blood pressure and left ventricular mass index and left ventricular end-diastolic pressure (LVEDP) increased in SHR;the maximal rate of increase and decrease of left-ventricle pressure development (± dp/dt max) decreased significantly in SHR;the protein expression and enzyme activity of left ventricular GSTM2 were significantly decreased;the expression of p47phox,Nox4 and the generation of O2·-were significantly increased in the left ventricles of SHR;the value of P was less than 0.01 and the difference was statistically significant.Compared with the SHR group,the SHR were administered with metformin (Met) continuously for 8 weeks.The left ventricular mass index and left ventricular end-diastolic pressure(LVEDP) decreased observably in SHR administration group;the maximal rate of increase and decrease of left-ventricle pressure development(± dp/dt max) increased in SHR administration group;the protein expression and enzyme activity of left ventricular GSTM2 were significantly increased in SHR administration group.the expression of p47phox,Nox4 and the generation of O2·-was significantly decreased in the left ventricles of SHR administration group;the value of P was less than 0.01 and the difference was statistically significant.CONCLUSION:Metformin can significantly reverse the myocardial hypertrophy in SHR,which might be associated with the up-regulation of GSTM2 expression,decrease the expression of p47phox,Nox4 and O2·-generation,elimination of oxidative stress.

ABSTRACT

Objective:To analyze the related substances in the domestic products of minoxidil and minoxidil gel for the purpose of standard improvement. Methods:An HPLC-Q-TOF-MS method was developed. The test was performed on a Thermo Scientific Accu-core C18 column (100 mm × 2. 1 mm, 2. 6 μm) with a mobile phase of acetonitrile-water (7:93) (containing 0. 1% formic acid) at a flow rate of 0. 2 ml·min-1 , the detection wavelength was 230 nm. The full scan and two stage scanning was employed for detection with ESI source positive ion mode. The mass scan range was m/z 50-500. Results:14 related substances were separated and detected under the established HPLC-Q-TOF-MS condition. The structures and resource of 8 related substances were analyzed preliminarily. Conclusion:The HPLC-Q-TOF-MS technology has been proved to be effective in separation and identification of the related substances in minoxidil and minoxidil gel. The results were useful for quality control.

Objective To design a vehicle-mounted X-ray diagnosis simulation training system based on the field X-ray diagnostic vehicle to solve the problems during its training and utilization.Methods The field X-ray diagnostic vehicle had its internal and external structure analyzed,and was divided into several functional modules.The technologies of computer programming and database were used for the simulation of the modules.An example was given to simulate the operating process of the vehicle by establishing traumatic condition model,war injury image library and etc.esults The system developed realized the simulation training of the vehicle,and the trainee gained feedback after simulation as references.Conclusion The system implements informatized training of the field X-ray diagnostic vehicle,shortens the training period,decreases the training cost and increases the training efficacy.

AIM:To investigate the effect of short-chain acyl-CoA dehydrogenase ( SCAD) on collagen expres-sion and proliferation of rat cardiac fibroblasts and to explore the relationship between SCAD and cardiac fibrosis . METHODS:The model of proliferation and collagen expression of rat cardiac fibroblasts induced by angiotensin II was es -tablished.After treatment with siRNA-1186, the expression of SCAD at mRNA and protein levels , fatty acids beta oxida-tion rate, ATP, the enzyme activity of SCAD and free fatty acids in the rat cardiac fibroblasts were determined . RESULTS:The mRNA and protein expression of SCAD was decreased in the rat cardiac fibroblasts induced by angiotensin II compared with the control cells , and the expression of collagen I and collagen III was significantly upregulated .Com-pared with negative control group , SCAD expression and activity , fatty acid beta-oxidation rate and ATP significantly de-creased in siRNA-1186 group, but the content of free fatty acids were obviously increased in the rat cardiac fibroblasts , and the expression of collagen I and collagen III was significantly up-regulated.CONCLUSION:The expression and synthesis disorder of collagen may be triggered by down-regulation of SCAD .SCAD may be a promising therapeutic target for myocar-dial fibrosis .

ABSTRACT

Objective:To establish a method for the determination of omeprazole delayed release capsules and investigate the be-havior of reference preparations to provide experimental basis for generic drugs quality consistency evaluation. Methods:According to the first dissolution method (basket method) stated in 0. 931 of Chinese Pharmacopeia (2015 edition), the type of release media, solu-bility and stability of omeprazole in different media, effect of different treatment methods on the drug adsorption in the solution, differ-ent rotation speed and the methodology of ultraviolet spectrophotometry were investigated. Results:The rotation speed was 75 r·min-1 with the dissolution medium volume of 900 ml. The dissolution profiles of omeprazole delayed release capsules in three different media ( pH 6. 0 phosphate buffer, pH 6. 8 phosphate buffer and water containing 3% Tween-80) were determined with online filteration. The solubility of omeprazole in the different media was 0. 123, 0. 078 and 0. 275 mg·ml-1 , respectively. The results showed that ome-prazole was degraded 44%, 8% and 14% in 2 h in the above three release media, and degraded up to 43% in 6 h in water containing 3% Tween-80. The linear of omeprazole was 0. 209 4-20. 94, 0. 204 8-20. 48 and 0. 2016-20. 16 μg·ml-1 with the average recovery of 99.3% (RSD=0.7%,n=12), 99.7% (RSD=0.9%,n=12) and 99.5% (RSD=0.6%,n=12) respectively in the three media. Conclusion:The method is accurate and reliable, which can be used to study the quality consistency of omeprazole delayed re-lease capsules.

ABSTRACT

Objective:To evaluate the quality of carbazochrome sodium sulfonate Injection from different manufacturers, analyze the existing problems and provide the reference for the improvement of clinical medication safety. Methods:Statutory testing methods and the exploratory research methods were used to examine the samples, and the quality status of carbazochrome sodium sulfonate Injec-tion was evaluated according to the results. Results:Totally 89 batches of the samples were tested by the current standard. The quali-fied rate was 97. 8%, and the two batches of unqualified samples were carbazochrome sodium sulfonate and sodium chloride Injection with unqualified insoluble particles. Overall, the quality risk of carbazochrome sodium sulfonate for injection was low, and the safety of carbazochrome sodium sulfonate and sodium chloride Injection needed further research. Conclusion:The quality of carbazochrome so-dium sulfonate Injection basically meets the current standard, and the exploratory research suggests that the current standard is imper-fect. It is urgent to improve the related substance detection in the standard and determine the reasonable limits for the degradation im-purities and the other impurities. The clinical medication safety of carbazochrome sodium sulfonate and sodium chloride Injection needs further research.

ABSTRACT

ObjectiveTo comprehend the similarities and differences of postoperative quality of life between mastectomie avec dissection axillaire and modified radical mastectomy among patients with breast cancer,and in order to supply references for establishing corresponding nursing measures. Methods160patients with early breast cancer were selected,40 patients accepted the mastectomie avec dissection axillaire (the conservative group)and 120 patients accepted modified radical mastectomy (the radical group).The quality of life of patients was surveyed with questionnaires. ResultsThe EORTC QLQ-C30 score of the conservative group was not significant than the radical group.The EORTC QLQ-BR 23 score showed that the conservative group had a better self-evaluation of their configuration. ConclusionsThe patients who accepted the mastectomie avec dissection axillaire will live a better life than those who accepted modified radical mastectomy.

ABSTRACT

The recent fast development in biotechnology has resulted in a large number of therapeutic biologicals coming in the foreground for clinical application. Transdermal drug delivery (TDD) is designed for patching the drug on the skin, and then the drug molecules permeate through the skin into the subcutaneous capillary vessels. However,very few drugs can be administered transdermally at the therapeutic levels to pass through the barrier of stratum corneum. Several physical and chemical methods are applied to improve the permeability of skin. Research result shows that the low-frequency ultrasonic technique can extraordinarily increase the rate of permeation. As a result, it is becoming one of the potential methods that serve as the substitutes for traditional methods. In this paper is presented a type of equipment based on MSP430. The principle of design, the structure of hardware, and the applied function in this area are described.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the pharmacokinetics profile of fangchinoline and tetrandrine in rats after administration of single compound and mixture with other intergradient in traditional prescription.</p><p><b>METHOD</b>A method for determination of fangchinoline and tetrandrine in rat plasma by using HPLC-MS has been developed and validated. The pharmacokinetics of two compounds and two compounds in the effective component group (ECG) of Xiaoxuming decoction were compared.</p><p><b>RESULT</b>Compared with the single dose of compound experiment results, the t(max) of fangchinoline and tetrandrine were longer than those in the single dose of ECG experiment. At the meanwhile the rest parameter showed no significant difference.</p><p><b>CONCLUSION</b>Other components in the ECG of Xiaoxuming decoction delayed the absorption rate of fangchinoline and tetrandrine, the bioavailability of two compounds were the same as that of the single dose of compound experiment.</p>

ABSTRACT

Objective To investigate the reasonable perioperative management for elderly patients with esophageal cancer undergon esophagectomy.Methods 92 elderly patients with esophageal neoplasms were treated by esophagectomy.According to the specific pathogenetic condition of the patients,various perioperative managements were provided.After surgery,the operative complications,mortality and 3,5-year survival rate were observed.Results Esophagectomy was carried out for all cases.Cardiovascular and respiratory complications were found in 19,10 respectively.Both of anastomotic stricture and fistulae were found in 4 cases.1 and 2 complicated with chylopleural and disturbance gastric emptying respectively.Mortality was 4.35%,and the 3,5-year survival rate was 80.43%(74/92),34.79%(32/92) respectively.Conclusion Satisfactory perioperative managements are the key to successful esophagectomy.

ABSTRACT

@# ObjectiveTo observe the effect of telmisartan on plasma levels of inflammatory cytokine in coronary artery disease (CAD) patients complicated with diabetes mellitus and hypertension after percutaneous coronary intervention.MethodsFifty CAD patients who just had undertook angioplasty and implanted stents were randomly divided into two groups, the test group (telmisartan group, n=25) and control group (perindopril group, n=25). After treatment, patients were followed-up for 6 months; plasma samples were collected from each patient before and after percutaneous coronary intervention. Then plasma levels of C-reactive protein (CRP), monocyte chemoattractant protein-1 (MCP-1) were measured by enzyme-linked immunoassay. The changes of cholesterol, fasting plasma glucose (FPG), insulin and insulin resistance index (IRI) were observed.ResultsAt the end of 6 months, plasma levels of CRP and MCP-1 of patients in the test group significantly declined (P<0.01), and showing a inversely correlation with FPG (P<0.01), and FPG, insulin and IRI also declined. In the control group, only CRP and MCP-1 declined (P<0.05). Meanwhile, the frequency of cardiovascular events in test group was significantly lower than that in the control group.ConclusionTelmisartan can decline plasma levels of CRP, MCP-1 and frequency of cardiovascular events as well as increasing insulin sensitivity and improving glucose metabolism to unstable angina patients.

ABSTRACT

Objective To provide high-quality health care services to persons whose access is restricted by geography or environment and disaster or battle situation. Methods X-ray equipment was based on embedded Linux operating system, and used OMAP1510 as its hardware developing platform. Telemedicine image workstation was based on Windows operating system, and used VC++6.0 as its software developing platform. Results Telemedicine image workstation could achieve and process images from X -ray machine. Conclusion The system can be applied to real-time acquisition of medical images and telemedicine.

ABSTRACT

Objective To evalute the feasibility and safety of sirolimus (rapamycin)-eluting stent for the treatment of in-stent restenosis (ISR). Methods Eleven patients with ISR were treated with one or more Cypher~ TM sirolimus-eluting stents. All these patients received angiographic follow-up at 6 months. Results The success rate of the procedure was 100%. All patients were free of angina after 6 months. There were no repeat revascularizations, stent thrombosis or major adverse clinical events during the follow up period. No ISR was noted in the follow-up coronary angiography. Conclusion This study demonstrates the safety and potential of sirolimus-eluting stents for the treatment of ISR.

ABSTRACT

To investigate the relationship of between p21wAF1 and PCNA expression and the patients' prognosis in breast carcinoma. MethodExpression of p21wAF1 and PCNA was determined by S-P immunohistochemical technique in 152 patients with breast carcinoma. ResultsPositive immunostaining of p21WAf1was detected in 61(40.1% ) primary breast carcinoma, in which 43(70.5% ) were low expression, and 18(29.5% ) over expression of 152 PCNA positive cases, 69(45% ) expressed with low index and 83(55% )expressed with high index. p21wAf1 expression level was correlated with tubule formation (γs= 0.341, P<0.01 ),nuclear atypia(γs=0.516, P<0.01), mitosis(γs=0.351, P<0.01)and histological grade(γs=0.415, P<0.01)in breast cancer tissue. Univariate analysis demonstrated less disease-free survival in patients with higher p21WAF1 (x2 = 6.92, P< 0.01 ) and PCNA (x2 = 5.14, P<0.05)expression than in those with lower expression. The survival term of the patients with p21WAF1 -/PCNA low index, p21wAF1 +/PCNA low index, p21WAF1 +/PCNA high index and p21WAF1 -/PCNA high index groups was successively shortened. Conclusions( 1 ) Combined detection of p21wAF1/PCNA is helpful to judge the proliferating activity of the tumor cells and the patients prognosis. (2) Different expression level of p21WAF1in breast carcinoma is probably related the different regulating directions.

ABSTRACT

Objective To study the quality of Pinellia ternata grown in Funan. Methods The contents of free and hydrolyzed amino acids and ephedrine in the samples from four habitats were determined. The components of methanol and acidic ethanol extracts of the four samples were compared by HPLC. The components of petroleum ether extracts of the four samples were compared by TLC. Results The content of amino acids and ephedrine and the kinds of amino acids of P. ternata grown in Funan were almost the same as those from the other three, the main components of methanol and petroleum ether extracts of P. ternata grown in Funan were the same as those from other three. Some components of acidic ethanol extracts of the four samples were different. Conclusion The quality of P. ternata grown in Funan is as good as that of the other three.
