ABSTRACT

The intestinal extracellular matrix (ECM) helps maintain appropriate tissue barrier function and regulate host-microbial interactions. Chondroitin sulfate- and dermatan sulfate-glycosaminoglycans (CS/DS-GAGs) are integral components of the intestinal ECM, and alterations in CS/DS-GAGs have been shown to significantly influence biological functions. Although pathologic ECM remodeling is implicated in inflammatory bowel disease (IBD), it is unknown whether changes in the intestinal CS/DS-GAG composition are also linked to IBD in humans. Our aim was to characterize changes in the intestinal ECM CS/DS-GAG composition in intestinal biopsy samples from patients with IBD using mass spectrometry. We characterized intestinal CS/DS-GAGs in 69 pediatric and young adult patients (n = 13 control, n = 32 active IBD, n = 24 IBD in remission) and 6 adult patients. Here, we report that patients with active IBD exhibit a significant decrease in the relative abundance of CS/DS isomers associated with matrix stability (CS-A and DS) compared to controls, while isomers implicated in matrix instability and inflammation (CS-C and CS-E) were significantly increased. This imbalance of intestinal CS/DS isomers was restored among patients in clinical remission. Moreover, the abundance of pro-stabilizing CS/DS isomers negatively correlated with clinical disease activity scores, whereas both pro-inflammatory CS-C and CS-E content positively correlated with disease activity scores. Thus, pediatric patients with active IBD exhibited increased pro-inflammatory and decreased pro-stabilizing CS/DS isomer composition, and future studies are needed to determine whether changes in the CS/DS-GAG composition play a pathogenic role in IBD.

ABSTRACT

With the popularization and application of artificial intelligence and medical image big data in the field of medical image, the universality of modes and the rapid development of deep learning have endowed multi-mode fusion technology with great development potential. Technologies of 5G and artificial intelligence have rapidly promoted the innovation of online hospitals. To assist doctors in the remote diagnosis of cancer lesions, this article proposes a cancer localization and recognition model based on magnetic resonance images. We combine a convolution neural network with Transformer to achieve local features and global context information, which can suppress the interference of noise and background regions in magnetic resonance imaging. We design a module combining convolutional neural networks and Transformer architecture, which interactively fuses the extracted features to increase the cancer localization accuracy of magnetic resonance imaging (MRI) images. We extract tumor regions and perform feature fusion to further improve the interactive ability of features and achieve cancer recognition. Our model can achieve an accuracy of 88.65%, which means our model can locate cancer regions in MRI images and effectively identify them. Furthermore, our model can be embedded into the online hospital system by 5G technology to provide technical support for the construction of network hospitals.

ABSTRACT

As uncontrolled urban growth has increasingly challenged the sustainable use of urban land, it is critically important to model urban growth from different perspectives. Using the SLEUTH (Slope, Land use, Exclusion, Urban, Transportation, and Hill-shade) model, the historical data of Hefei in 2000, 2005, 2010, and 2015 were collected and input to simulate urban growth from 2015 to 2040. Three different urban growth scenarios were considered, namely a historical growth scenario, an urban planning growth scenario, and a land suitability growth scenario. Prediction results show that by 2040 urban built-up land would increase to 1434 km2 in the historical growth scenario, to 1190 km2 in the urban planning growth scenario, and to 1217 km2 in the land suitability growth scenario. We conclude that (1) exclusion layers without effective limits might result in unreasonable prediction of future built-up land; (2) based on the general land use map, the urban growth prediction took the governmental policies into account and could reveal the development hotspots in urban planning; and (3) the land suitability scenario prediction was the result of the trade-off between ecological land and built-up land as it used the MCR -based (minimum cumulative resistance model) land suitability assessment result. It would help to form a compact urban space and avoid excessive protection of farmland and ecological land. Findings derived from this study may provide urban planners with interesting insights on formulating urban planning strategies.

ABSTRACT

OBJECTIVE: To study the characteristics of local cytokines profile in human gastric cancer lesions and offer guides for immunotherapy of gastric cancer. METHODS: The cytokines profile in isolated CD4+, CD8+ T subsets and epithelial cells from cancer tissue, contrasted with benign gastric mucosa of 15 gastric cancer patients were analyzed by a highly sensitive radioactivity labeled semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) technique. RESULTS: The predominance of increased Th2 and reduced Th1 immune response in both CD4+ and CD8+ T subsets from cancer tissue, compared with benign gastric mucosa, were observed. IL-6, IL-8 and TNF-alpha in CD8+ T cells from cancer tissue showed statistical higher levels than those from benign gastric mucosa (P = 0.029, P = 0.022, P = 0.002), so did IL-10, TGF-beta, TNF-alpha in T cells and epithelial cells. CONCLUSION: A shift of Th1/Th2 cytokines to Th2 is found in T cells from local tumor lesion of gastric cancer, which suggests an immune suppressive status in local tumor lesion. The obvious change of cytokine profile in CD8+ T cell suggests they play a main role in immune suppression. To study the cytokine expression of different cell subsets, we are able to offer guides for immunotherapy for patients with gastric cancer.
