ABSTRACT

Narrative care for families suffering from perinatal loss is rarely provided by medical institutions in China Mainland. However, with the advancement of the Chinese narrative medicine theory and practice, the clinical significance of narrative care has been increasingly recognized. Based on the principles of Chinese narrative medicine, this narrative case study described traumatic narrative foreclosures occuring in a family suffering from stillbirth, and highlighted the multidisciplinary collaboration for practising narrative care in the process of supporting the bereaved in our hospital. Meanwhile, we advocate the establishment of a narrative care ecology by training more obsteticians and nurses with good narrative competence in purpose of helping the family experiencing perinatal losses to overcome their tramatic narrative foreclosures, increasing the chances of another successful pregnancy and childbirth as well as enhancing their quality of life.

ABSTRACT

The application of Nb microalloying to high-carbon pearlite bridge cable wire rod steel has always been controversial, especially in the actual production process, which will be affected by the cooling rate, holding temperature and final bonding temperature. In this paper, the experimental characterization, finite element simulation and phase diagram calculation of the test steel were carried out, then the microstructure and properties of different parts of Nb microalloying of bridge cable wire rods were compared and analyzed. The phase transition interval of pearlite during the water-cooling process of bridge cable wire rods is increased due to the refinement of austenite grains, and the significant increase in the end temperature of the phase transition makes the average interlamellar spacing of pearlite increase. The cooling rate of different parts of bridge cable wire rods simulated by Abaqus has little difference. At the same time, Nb microalloying effectively increases the proportion of low-angle grain boundaries, so that the overall average misorientation representing the surface defects is reduced. This helps to reduce the surface energy and increase the stability of the microstructure. Combined with the mechanical properties of microtensile rods, it is found that the grain refinement effect of Nb is greater than that of coarsening interlamellar spacing during hot rolling deformation in actual production, which makes the tensile strength at the 1/4 section increase significantly. The overall tensile strength and area shrinkage of the steel wire have also been effectively improved.

ABSTRACT

PURPOSE: E-cadherin, a transmembrane glycoprotein mediating Ca2+-independent homotypic cell-cell adhesion in epithelial cell, plays an essential role in metastasis. It has been postulated that E-cadherin downregulation is a crucial mechanism in the pathogenesis of endometriosis. To evaluate the effect on the cell behavior after knockdown of E-cadherin gene (CDH1) in cultured human endometrial epithelial cells (EECs) isolated from normal endometrium. METHODS: EECs were isolated from the endometrial tissues of fertile woman who underwent total hysterectomy due to cervical intraepithelial neoplasia III. CDH1 expression was knocked down by small hairpin RNA. The EECs transfected with empty vector served as control. Transwell assay was used to test EECs migration or invasion. qRT-PCR and western blot were used to detect mRNA and protein levels. RESULTS: The results showed that knockdown of E-cadherin expression can increase cell migration and invasion, and up-regulate mRNA and protein levels of ß-catenin, cyclinD1, and c-myc. CONCLUSIONS: Down-regulation of E-cadherin expression may activate the Wnt/ß-catenin pathway in endometrial cells, which may together participate in the occurrence of endometriosis.

ABSTRACT

A uterine fibroid is a leiomyoma that originates from the smooth muscle layer of the uterus. A variety of endometrial abnormalities are associated with uterine fibroids. This study aims to investigate the methylation status of the E-cadherin gene (CDH1) promoter region in the endometrium of patients with uterine fibroids. The methylation of CDH1 was studied using methylation-specific polymerase chain reaction in the endometrial tissue of 102 patients with uterine fibroids and 50 control patients. The E-cadherin expression was examined by flow cytometry. The methylation rate of CDH1 promoter region was 33.3% in the endometrium of patients with uterine fibroids and 8% in the endometrium of women without fibroids. The frequency of CDH1 promoter methylation in the endometrium of patients with fibroids was significantly higher than that in the endometrium of women without fibroids (P = .001). Furthermore, the E-cadherin expression level in methylation-positive tissues was significantly lower than that in methylation-negative tissues (P = .017). These results suggest that epigenetic aberration of CDH1 may occur in the endometrium of patients with fibroids, which may be associated with E-cadherin protein expression in endometrial tissue.
