ABSTRACT

Brain signalling pathways involved in subclinical anxiety and depressed mood can be modulated via the gut brain axis (GBA), providing the potential for diet and dietary components to affect mood. We investigated behavioural, physiological and gut microbiome responses to the Lacticaseibacillus rhamnosus strain HN001 (LactoB HN001™), which has been shown to reduce postpartum anxiety and depression, and a milk fat globule membrane-enriched product, Lipid 70 (SurestartTM MFGM Lipid 70), which has been implicated in memory in stress-susceptible Wistar Kyoto rats. We examined behaviour in the open field, elevated plus maze and novel object recognition tests in conjunction with the expression of host genes in neuro-signalling pathways, and we also assessed brain lipidomics. Treatment-induced alterations in the caecal microbiome and short-chain fatty acid (SCFA) profiles were also assessed. Neither ingredient induced behavioural changes or altered the brain lipidome (separately or when combined). However, with regard to brain gene expression, the L. rhamnosus HN001 + Lipid 70 combination produced a synergistic effect, reducing GABAA subunit expression in the amygdala (Gabre, Gat3, Gabrg1) and hippocampus (Gabrd). Treatment with L. rhamnosus HN001 alone altered expression of the metabotropic glutamate receptor (Grm4) in the amygdala but produced only minor changes in gut microbiota composition. In contrast, Lipid 70 alone did not alter brain gene expression but produced a significant shift in the gut microbiota profile. Under the conditions used, there was no observed effect on rat behaviour for the ingredient combination. However, the enhancement of brain gene expression by L. rhamnosus HN001 + Lipid 70 implicates synergistic actions on region-specific neural pathways associated with fear, anxiety, depression and memory. A significant shift in the gut microbiota profile also occurred that was mainly attributable to Lipid 70.

ABSTRACT

When looking to improve animal welfare, it is essential to understand the viewpoints of stakeholders in the industry. Previous research has engaged stakeholders such as farmers, veterinarians, and the public to better understand future dairy industry directions. However, the use of perspective-taking as a tool to overcome farmer's focus on current barriers to the industry has not been explored. Therefore, the aims of this study were to have farmers take the perspective of the cow, to elicit: (1) farm elements that are essential to create a cow-centric dairy system, and (2) how this system can be achieved in the next 50 years. To do this, we employed participatory methods (photo elicitation, timelining) to generate discussion with 12 New Zealand dairy farmers. Pasture access is frequently cited as an integral component for promoting good welfare in dairy cows, thus, this cohort of farmers was considered a good case study as they all had experience managing cows in grazing systems. Participants were asked to create a cow-centric farm design, and a pathway to implementation. Following thematic analysis of the multiple data sources, the results are presented under the following three themes: (1) cow-centric farm designs (environmental considerations, cow handling and care), (2) timeline to achieve and sequence of implementation ("low hanging fruit" and long-term investments), and (3) assumptions and resources needed (including financial considerations, technology, farmer buy-in, regulations and enforcement and other stakeholder involvement). By better understanding how current industry practice aligns, and also differs, from what the cow would want, we can work towards future management systems that incorporate the requirements of all stakeholders, including the cow.

ABSTRACT

Goats naturally browse different forages in various postures; this differs from typical farm practice, thus there are opportunities to improve goat welfare by understanding what and how they like to eat. We investigated if feeding preference was related to posture, feeder height relative to the ground, and type of feed. Sixteen adult, Saanen cross females participated in two experiments comparing a floor-level feeder (grazing posture; farm standard), with an elevated feeder (browsing posture; Exp1) and a platform-level feeder (raised, grazing posture; Exp2), when two forages (leaves, grass) were offered. Measurements included feed intake (g of DM/feeder), feeder switching frequency, first feeder visited, latency to visit first feeder and exploration and non-feeding activity time. Effects of posture (Exp1), height (Exp2) and feed type were analyzed. Type of feed affected preference for feeding posture and height. All goats consumed leaves over grass (Exp1: POP: 188 ± 6.52 g, GRA: 20.3 ± 7.19 g; Exp2: POP: 191 ± 6.15 g, GRA: 0.231 ± 6.91 g; P < 0.001), and the feeder containing leaves was often visited first (Exp 1: GRA/POP: 94% of visits, P < 0.001, POP/GRA: 53%, P = 0.724; Exp 2: GRA/POP: 91%, P < 0.001; POP/GRA: 69%, P = 0.041). When goats received only leaves, they consumed more from the floor-level (162 ± 22.2 g) vs. elevated level (102 ± 21.9 g) feeder (P = 0.039). When goats received only grass, there was no posture or height preference; however, they changed feeders more frequently (at least 4x (Exp1) and 2x (Exp2) more than other combinations; P > 0.01). Feed intake was negatively affected by exploring time (Exp1 only: r = -0.541; P < 0.001) and performing non-feeding activities (Exp1: r = -0.698; P < 0.001; Exp2: r = -0.673; P < 0.001). We did not identify a preference for elevated feeding posture; however, we suggest that our short test (compared to previous work) encouraged goats to make choices based on line-of-sight and also that the elevated feeder design (replicated from previous work) made leaf access harder. Nonetheless, we highlight that some goats actively used the elevated feeder; this coupled with the clear preference for leaves over grass, suggests that offering feed type and presentation diversity would allow individuals to express their natural feeding behavior more fully.

ABSTRACT

Individual variation in responses to commonly used tests of anxiety and spatial memory is often reported. While this variation is frequently considered to be 'noise', evidence suggests that it is, at least partially, related to consistent individual differences in behavioral responses (i.e., personality). The same tests used to assess anxiety are often used to profile personality traits, but personality differences are rarely considered when testing treatment differences in anxiety. Focusing on the rat literature, we describe fundamental principles involved in anxiety and spatial memory tests and we discuss how personality differences and housing conditions can influence behavioral responses in these tests. We propose that an opportunity exists to increase stress resiliency in environmentally sensitive individuals by providing environmental enrichment. We conclude by discussing different approaches to incorporating personality measures into the design and analysis of future studies; given the potential that variation masks research outcomes, we suggest that a strategy which considers the individual and its housing can contribute to improving research reproducibility.

ABSTRACT

There is growing evidence that animal personality is linked to a range of productivity traits in farm animals, including dairy cattle. To date, the methodology for assessing personality traits of dairy cattle is time consuming and often requires a test arena, which limits the opportunity for commercial farms to use personality traits of dairy cattle for individualized management. This study investigated whether personality traits of pastured dairy cattle, scored in short behavioral tests, are associated with daily behavioral patterns and milk production. Cows (n = 87) were exposed twice to each of 5 behavioral tests, where their responses to novel or putatively stressful situations were scored on an ordinal scale for investigative and reactive behavior toward a novel object and a novel human after exiting the milking parlor, response to restraint in a crush, avoidance distance from an approaching human in the paddock, and response to milking (step-kick behavior). Most behavior test scores were consistent over the 2 test repeats (using repeatability estimates, Mann-Whitney U test of difference between repeats, and Spearman rank correlation). Behavior test scores were subjected to a principal components analysis that revealed intertest relationships in 3 factors of correlated sets of test scores, interpreted as personality traits (fearful of humans, calm-investigative, and reactive to milking). Regression analyses determined how these traits were associated with daily grazing, ruminating, and lying behaviors, and milk production (after controlling for cow age, breed, lactation status, group, and climate variables). Cows that were more fearful of humans (high avoidance distance, reactive toward the novel human) had reduced lying time compared with cows that scored low on this trait. Cows that were more calm (during restraint) and investigative (toward the novel object) had greater grazing time, which likely contributed to their greater milk production compared with cows that scored low on this trait. Cows that were more reactive to milking produced less milk than cows that scored low on this trait. These results indicate that individual differences in daily behavior patterns and milk production of dairy cattle are associated with personality traits of cows, measured using several short behavioral tests. These methods may be useful for characterizing grazing dairy cattle on commercial farms, which could aid in understanding individual behavior patterns and provide opportunities for individualized management.

ABSTRACT

Separation of the cow and calf shortly after birth is a common practice on commercial dairy farms around the world, but there are emerging concerns about this practice among citizens and other stakeholders. Continuous improvement of on-farm management practices in collaboration with dairy sector stakeholders increases the likelihood that farming systems evolve in a way that is consistent with societal expectations. Few commercial dairy farms provide extended cow-calf contact, and there is little understanding of how dairy farmers view this practice. This study examined the views of New Zealand dairy farmers toward providing cow-calf contact, particularly the barriers to adopting such a system in a seasonal-calving pasture-based dairy system. Standard farm practice in New Zealand is to remove the calf from the cow around 24 h (but could be up to 48 h) after birth. These conventional farmers (n = 63) were randomly selected from the database of all dairy farmers in New Zealand and telephone-interviewed using a semistructured interview format. Their responses to questions about providing cow-calf contact (defined as contact beyond the standard practice of 48 h) were analyzed using thematic analysis. Three major themes of concern were identified by these farmers about providing cow-calf contact as follows: (1) poor animal welfare, especially the risk of mastitis in the dam, inadequate colostrum for the calf, increased stress from delayed separation, and lack of shelter for calves while outdoors with the cow; (2) increased labor and stress on staff; and (3) system-level changes required, including infrastructure and herd management. Many of these concerns stemmed from challenges related to the nature of large-scale seasonal-calving pasture-based dairy systems, where a large number of calves are born in a short period of time and may be exposed to inclement weather in late winter in some areas. Several small-scale farmers (n = 4) providing cow-calf contact for longer than standard practice of 48 h were also interviewed; all permitted contact for at least 4 wk. These farmers also felt that animal welfare and health were important, and that this was promoted in their cow-calf contact systems. Concerns about colostrum and mastitis, for example, were not raised by these farmers, but they did agree that additional infrastructure and shelter were important considerations for cow-calf contact systems. Some conventional farmers expressed cognitive dissonance in that they theoretically preferred cow-calf contact but could not see it being realistic or practical to implement. Farmers currently providing longer cow-calf contact may be a useful resource for better understanding of how practical and economical cow-calf contact systems could be adopted on commercial pastoral dairy farms.

ABSTRACT

Laboratory mice are commonly euthanised with carbon dioxide (CO2); however, there is ample evidence that this gas is aversive. Previous work suggests that sedation achieved via injection with benzodiazepines prior to CO2 administration could reduce aversive behaviours during euthanasia. We explored the potential of using a voluntarily ingested sedative (tiletamine-zolazepam, Zoletil®) prior to euthanasia. Male and female C57BL/6 mice were allocated into one of the five experimental groups, which differed in the dose of Zoletil: 0, 10, 20, 40, 80 or 100 mg/kg. A dose of 20 mg/kg was found to achieve mild sedation prior to euthanasia; mice which received this dose numerically reared and walked on the cage lid less, and showed ataxia, immobility and recumbency for longer than mice that received a lower dose. During euthanasia, mice that received 20 mg/kg showed fewer aversive responses to CO2. Doses of 40 to 100 mg/kg were associated with signs of moderate to severe sedation, but resulted in an incomplete intake of the sedative, which made the interpretation of the aversiveness to CO2 difficult. Voluntary oral administration of a sedative is an effective, affordable, and easy way to minimize the stress of mice to euthanasia with CO2.

ABSTRACT

This article considers the complexity and diversity of ethical concepts and beliefs held by Maori, the indigenous people of Aotearoa New Zealand (hereafter New Zealand), relating to animals. A combination of interviews and focus group discussions were conducted with individuals who identify as Maori and were working with wildlife, primarily in an eco-tourism and conservation context. Two main themes emerged from the data: ethical concepts relating to the environment, and concepts relating to the spiritual relationships between people, animals and the environment. These findings highlight that the connections between humans and animals through a Maori lens are nuanced in ways not typically accounted for in Western philosophy. This is of particular importance because of the extent to which standard Western thought is embodied in law and policy related to human treatment of animals and the environment. In New Zealand, relationships and partnerships are informed by Te Tiriti o Waitangi, one of New Zealand's founding documents. Where these partnerships include activities and environments involving human-animal interaction, policy and legislation should account for Maori knowledge, and diverse of thought among different hapu (tribal groups). We conclude by exploring ways of including Maori ethical concepts around animals in general, and wild animals in particular, in law and policy, providing a case study relevant to other bicultural or multicultural societies.

In pasture-based systems, cows may spend several hours away from the paddock and may also walk several kilometres to meet daily milking requirements; this could lead cows to experience time constraints for grazing, ruminating and lying time in the paddock. This study investigated how walking distance and time spent away from the paddock affected daily behavioural patterns (i.e., grazing, ruminating and lying time) and milk yield. Dairy cows were managed in three rotationally grazed groups (n = 29 cows each) on the same farm and milked twice daily. A triaxial ear tag accelerometer on each cow recorded daily duration of grazing and ruminating, and a leg-based accelerometer recorded the daily lying time, for 13 days. GPS collars on four cows per group recorded the daily walking distance and total time away from the paddock for the group. A mixed repeated measures model tested how time off-paddock and walking distance affected the daily behavioural patterns; age, breed, milk yield and maximum ambient temperature were used as covariates with group as the observational unit. A second similar model tested how these factors affected milk yield. Walking distance and time spent away from the paddock were not correlated. When daily walking distance increased (to a maximum of 4 km/d), cows spent more time grazing and less time ruminating, but lying time was not affected. This result may, in part, be related to the greater energy expenditure demands for walking longer distances and milk production. When time away from the paddock increased (to a maximum of 4 h/d), cows spent less time lying, but grazing and ruminating times were not affected. Milk yield was not affected by walking distance, but one of the groups experienced a lower milk yield when time away from the paddock was increased. This result suggests that, for some cows, lying times may be shorter when experiencing a longer time away from the paddock, which may also affect milk yield. Overall, this study indicates that paddock behaviours are associated with walking distance to the milking parlour and time spent away from the paddock. Efforts to reduce walking distance and time spent away from the paddock are likely to provide cows with greater opportunity to engage in daily behaviours in the paddock that meet their needs and maintain their milk yield.

Anticipatory behaviour occurs in the period before a reward or other positive event is presented and has been interpreted as an indicator of the welfare and emotional state of animals. The use of this indicator has received limited attention in dairy calves. Therefore, we investigated how anticipatory behaviour is affected by housing environment and reward quality, and if anticipatory behaviour changes when reward quality changes unexpectedly. Sixteen pairs of calves were assigned to treatments in a 2 x 2 factorial design (two housing environment and two reward quality combinations). Housing was either basic (2 m2/calf, river stone surface) or enriched (5 m2/calf, woodchip, and enrichment items), and the reward was access to either an additional basic or enriched pen. Calves were conditioned to anticipate reward pen access; anticipatory behaviour toward receiving the reward pen was measured. Signaling reward access increased the frequency of transitions between behaviours and duration of touching and looking at the signal and exit door. Basic-housed calves showed more anticipatory behaviour (increased frequency of transitions between behaviours) and decreased latency to access the reward compared to enriched-housed calves, but the reward pen quality had no effect on anticipatory behaviour. When the reward pen quality changed from enriched to basic unexpectedly, resulting in sudden reward loss, basic-housed calves decreased, while enriched-housed calves increased, anticipatory behaviour. However, there was no change in anticipatory behaviour during reward gain (change from basic to enriched reward pen). Our findings align with previous work showing that animals in basic housing show more anticipation for a reward, and demonstrate suppressed behavioural response when experiencing reward loss, suggesting greater sensitivity to reward. Sensitivity to reward has associations with mood state; thus, calves in basic environments may experience a more negative emotional state, and thus reduced welfare, compared to calves in enriched environments.

Isolation during calving is a common dairy cow behavior, however it has not been examined in large outdoor group settings. The provision of "hides" was monitored for its impact on calving location and cow-calf behavior. Stocking density and bedding management were either controlled (Phase 1) or managed according to farm practice (Phase 2). Hides were used for calving by 18% (Phase 1) and 22% (Phase 2) of the cows; a further 59% and 44% of cows moved into the hides after calving (Phase 1 and 2, respectively). When hides were not available, cows calved near the edges of the calving area. In Phase 2, as stocking density increased, cows tended to use the hides less. Older cows were less likely to isolate regardless of management. Cow-calf interaction with other cows and calves was lower when hides were available. There was no evidence that hides reduced incorrect matching of cows and calves by staff, however cases of "mismothering" (i.e., calves being taken by other dams) were observed. Since the majority of cows used the hides at some stage before or after calving, we suggest opportunities for seclusion should be provided in large calving groups.

ABSTRACT

We developed a simple, interactive poster design. Via brief infographics and simple numbered stickers, participants were able to provide input about their top three areas of importance regarding a specific topic (i.e., promoting natural behavior in goats). The tool was utilized in three scenarios-an international conference, a regional veterinary conference, and a regional school leadership day. After a short discussion with the presenter, participants ranked their top three areas of importance. Response rates ranged from 22% to 100%. The data collection performed was intended to demonstrate a proof-of-concept of the poster design; therefore, comparisons across tested cohorts were not made. However, we showed that when applied in different settings, the poster gathered some consistent opinions on which behaviors are the best exemplars of naturalness in goats. Response bias, from opting for socially desirable responses, as well as sampling bias from using the tool at specific conferences or with specific demographics, could be an issue. Nonetheless, these are not unique concerns, and we suggest that corrections used in focus groups could alleviate these biases. The flexibility of this interactive tool, and its capability to shift the audience from viewing to participating in research presents a novel alternative to traditional poster presentations.

The assessment of hoof conformation is important due to its recognized relationship with the biomechanical functionality of the hoof. Hoof conformation can be assessed using objective measures or subjective scores. However, to date, there are limited data using either method in dairy goats. Therefore, the aims were to (1) develop a reliable method of assessing hoof conformation in dairy goats, and (2) compare two aspects of a subjective assessment against corresponding objective measures as a means of validation. A total of 1035 goats contributed photographs across 16 commercial dairy goat farms. Photographs were taken of the left front and left hind hoof in the lateral and dorsal aspect at five assessments across the goats' first two lactations. Hoof conformation was assessed using five subjective scores (toe length, heel shape, fetlock shape, claw splay, and claw shape) and two objective measures (toe length ratio and claw splay distance). Following the training of two observers, high levels of inter and intra-reliability were achieved for both the subjective scores (>0.8 weighted kappa) and objective measures (>0.8 Lin's concordance correlation coefficient). Two aspects of the subjectively assessed ordinal scores were compared with the objective measures with high levels of accuracy (>0.8). This suggests that the subjective scores may be a suitable alternative to more time-consuming objective measures when assessment is completed using photographs.

ABSTRACT

Public interest is stimulating farming industries to improve animal welfare in production systems. Indoor housing of ruminants has received scrutiny because of perceived intensiveness and lack of naturalness. Animal welfare has traditionally focused on health benefits (e.g., bedding management and reducing disease) and reducing negative experiences (e.g., painful husbandry practices). Recent attention to animals having "a life worth living" extends expectations to provide increased care and opportunities for positive experiences and natural behaviors. Although not all natural behaviors necessarily contribute to improved welfare, we present evidence for why many are important, and for how they can be promoted in commercial systems. Worldwide, commercial dairy goats (Capra hircus) are frequently housed in large open barns with space to move and soft bedding for lying; however, this is not sufficient to promote the range of natural behaviors of goats, which in turn suggests that commercial housing could be improved. The basis for this thinking is from the range of behaviors expressed by the Capra genus. Collectively, these species have evolved cognitive and behavioral strategies to cope with harsh and changing environments, as well as variable and limited vegetation. The rocky and often steep terrain that goats inhabit allows for predator avoidance and access to shelter, so it is not surprising that domesticated goats also seek out elevation and hiding spaces; indeed, their hoof structure is designed for the movement and grip in such rugged environments. The browsing techniques and flexibility in diet selection of wild, feral and extensively managed goats, appears to be equally important to housed goats, highlighting the need for more complexity in how and what goats are fed. Goats naturally live in small, dynamic groups, governed by complex social structures in which horns play a strong role. Commercial housing systems should consider the benefits of more natural-sized social groups and revisit the rationale behind horn removal. We suggest that cognitive stimulation is a potential welfare improvement for goats in commercial settings. Goat cognitive abilities, which enabled success in complex and variable social and physical environments, are unchallenged in uniform environments, potentially leading to negative affective states. We make suggestions for housing improvements that could be readily adopted into current systems without compromising production efficiency.

ABSTRACT

Goats are browsers and select vegetation at various heights when foraging. On commercial farms, dairy goats are typically fed from low-level feed bunks. The objective of this study was to determine how feed intake and feeding behavior vary when goats are offered feed at variable heights, with the potential of evaluating the benefits of offering an elevated feeder to dairy goats. Thirteen Saanen X dairy goats were housed in a home pen with a lying area of wood shavings, where they were pre-exposed for 24 d to 3 feeder heights designed to result in differences in head height while feeding: floor level (head lowered relative to body), head level (head level relative to body), and elevated level (head and neck angled upward). Nine groups of 3 goats each were randomly selected and housed for 24 h in a test pen identical to the home pen except that it contained 1 of each of the 3 feeder heights. Each feeder contained ad libitum chopped alfalfa silage and a top-dressed corn-based supplement, refreshed twice daily. Refusals from inside and under each feeder were weighed to calculate intake. Feed intake increased with increasing feeder height (mean ± SE; 0.18, 0.29, and 0.34 ± 0.04 kg of DM/goat for floor-level, head-level, and elevated-level feeders, respectively). Total feeding time did not vary with feeder height, but feeding rate tended to be faster at the elevated-level feeder (14.5 ± 2.1 g of DM/min) compared with head-level (9.2 ± 2.3 g of DM/min) and floor-level (8.9 ± 2.1 g of DM/min) feeders. Goats visited the floor-level feeder (36.4 ± 8.4 visits/goat) less than the head-level (79.4 ± 8.4 visits/goat) and elevated-level (74.8 ± 8.4 visits/goat) feeders. The number of displacements per minute of feeding time (physical removal of another goat from the feeding place) was greater at the elevated-level feeder (0.46 ± 0.06 displacements/min) compared with the floor-level feeder (0.23 ± 0.06 displacements/min) and tended to differ from the head-level feeder (0.27 ± 0.06 displacements/min). We conclude that goats eat more from an elevated feeder and compete more to access this feeder.

ABSTRACT

Calf housing often only meets the basic needs of calves, but there is a growing interest in providing enrichments. This study described the behaviour of calves when they were given the opportunity to interact with two commonly available enrichment items. Female and male calves (approximately 11 days old) were pair-housed in 8 identical pens fitted with an automated brush and a hanging rope. Frequency and duration of behaviours were recorded on 3 separate days (from 12:00 until 08:00 the following day. Calves spent equal time using the brush and rope (27.1 min/day), but there was less variation in the use of the brush as opposed to the rope (coefficient of variation, CV: 23 vs. 78%, respectively). Calves had more frequent (94 bouts, CV: 24%) and shorter (17.8 s/bout, CV: 24%) brush use bouts compared to fewer (38 bouts, CV: 43%) and longer (38.3 s/bout, CV: 53%) rope use bouts. There was a diurnal pattern of use for both items. Frequency of play was similar to rope use, but total time playing was 8% of rope and brush use. Variability among calves suggested that individual preference existed; however, the social dynamics of the pair-housed environment were not measured and therefore could have influenced brush and rope use. Multiple enrichment items should be considered when designing improvements to calf housing.
