ABSTRACT

Until now, there has not been organized consensus for standardization in bariatric surgery In Russia. We present the results of the first Bariatric Surgery Consensus Conference conducted in Barnaul (March, 2023). A list of questions was proposed within 6 blocks: 1) general issues of bariatric surgery, 2) sleeve gastrectomy, 3) one-anastomosis gastric bypass («mini-gastric bypass¼), 4) Roux-en-Y Gastric Bypass, 5) Single Anastomosis Duodenal Switch and other options for biliopancreatic bypass, 6) rare procedures. Consensus (>70% agreement) was reached for 51 out of 96 statements. Stratification by the level of expertise was carried out, and responses of the expert group were compared with responses of all participants.

ABSTRACT

Bacteriophages-viruses that infect bacterial cells - are the most abundant biological entities on Earth. The use of phages in fundamental research and industry requires tools for precise manipulation of their genomes. Yet, compared to bacterial genome engineering, modification of phage genomes is challenging because of the lack of selective markers and thus requires laborious screenings of recombinant/mutated phage variants. The development of the CRISPR-Cas technologies allowed to solve this issue by the implementation of negative selection that eliminates the parental phage genomes. In this manuscript, we summarize current methods of phage genome engineering and their coupling with CRISPR-Cas technologies. We also provide examples of our successful application of these methods for introduction of specific insertions, deletions, and point mutations in the genomes of model Escherichia coli lytic phages T7, T5, and T3.

ABSTRACT

AIM: To present the results of work of National Medical Research Center of Treatment and Rehabilitation, reassigned for COVID-19 patients treatment during pandemic. Run-up methodology, procedures and working process organization are detailed. MATERIALS AND METHODS: 354 COVID-19 patients were treated from 13.04.2020 to 10.06.2020 [age 59 (470) years, 56% women, body mass index 28.5 (24.932.2) kg/m2]. Patients were admitted at 8 (611) day of sickness. In-hospital stay was 16 (1420) days. RESULTS: NEWS scale at the day of admittance was 2 (14); 2 (13) in patients discharged alive and 6 (47) in died patients, p=0.0001. So prognostic accuracy of NEWS scale was confirmed as very well (area under ROC-curve = 0.819). 69 patients (19.5%) were treated at intensive care department for 7 (413) days. 13 patients died, 11 of them had COVID-19 as direct or indirect cause of death. Total in-hospital mortality was 3.67%, in-hospital mortality of COVID-19 patients 3.1%. 17 healthcare workers (HCW), contacted with COVID-19 patients were infected (2.67%). 4 HCW, who had no direct contact with patients were also infected and 7 HCW were infected before the first patient was admitted. No one of them died. CONCLUSION: Complex tasks of healthcare organization during COVID-19 pandemic can be solved quickly with acceptable quality, characterized by low levels of patients; mortality and HCW infection.

ABSTRACT

INTRODUCTION: The term textiloma is used to describe foreign body (swab) forgotten during surgery. We don't know any case of the description of endoscopic ultrasonography (EUS) for intra-abdominal textiloma. OBJECTIVES: To determine the EUS capabilities in the topical diagnosis of intra-abdominal textiloma. MATERIALS AND METHODS: Patient, female, 57-year-old was admitted to our hospital with post-operative hernia. 10 months before the patient underwent surgery (laparotomy, splenectomy) in another hospital. For ethical reasons, we do not name that hospital. Intra-abdominal lesion has been accidentally revealed by computed tomography during the pre-operative examination prior to hernioplasty. Ultrasound unclear detected hyperechoic lesion with a dense acoustic shadow. The patient was sent to the EUS to clarify the diagnosis and to determine the topography of the lesion, fine-needle aspiration - if required. RESULTS: EUS revealed a hyperechoic corrugated border of the lesion with a dense acoustic shadow - between the gastric wall and the lower surface of the liver. Taking into account preceding surgery EUS suspected textiloma. The patient underwent hernioplasty and revision of the abdominal cavity. Textiloma has been revealed between the stomach and liver. CONCLUSION: EUS correctly diagnosed textiloma and accurately determined its location.

ABSTRACT

Type EFPA-30 apparatus for electrophoresis on cellulose acetate films was developed at the Specialized Design Office for Biophysical Equipment of the Moscow Research and Production Association BIOFIZPRIBOR. Three autonomic separation cameras are united in one block, that permits separation of 12, 24, and 36 samples; the apparatus is supplied with time transducer, acoustic and visual indication. Clinical trials of the apparatus were carried out at the Institute of Biological and Medical Chemistry of the USSR Academy of Medical Sciences, its commercial production is to be started. The apparatus is intended for protein separation and may be used for studies of hemoglobins, serum proteins, lipoproteins, enzymes, and other biologic compounds. Methods for hemoglobin and serum protein separation are presented.

ABSTRACT

Conductometric detectors can be employed for analysis of the blood serum proteins. Conductometric curves of various sera show at least 12 peaks corresponding to certain protein fractions and inorganic components. Isotachophoresis of individual protein preparations detects impurities in them. The type of conductometric curves of various blood sera helps diagnose abnormal shifts. Use of a spacer is not conducive to a more accurate detection of the blood serum protein compounds.

ABSTRACT

The conditions were optimized for freezing storage, restoring and further cultivation of hybridoma cells producing antibodies to viral antigens. The effect of density of cellular suspension frozen,concentration of calf embryo serum in cryoprotected medium and mild conditions of the restoring of hybridoma were studied. To restore deeply frozen hybridoma 24 hole plastic panels with a layer of feeding cells of the HEPES and insulin containing medium were used. The fulfilling of these requirements makes possible restoration of intact antibody-producing hybridoma from 10(2)-10(3) frozen cells.
