ABSTRACT

The sheep nasal bots Oestrus ovis is parasite of the nasal cavities and sinuses of small ruminants causing oestrosis, one of the most frequent parasitic diseases in sheep and goats. The widely use of ivermectin and closantel by the sheep breeders in the treatment and prophylaxis of gastrointestinal nematodes resulted in widespread cases of anthelmintic resistance. However, there is no report about cases of O. ovis with drug-resistance. In this study, we evaluated the prophylactics and therapeutic effects of both antiparasitics in sheep with O. ovis natural infestation. The trial was carried out from early December 2019 to March 2020, with 30 crossbred males lambs allocated into three groups of 10 animals each: control (without treatment), treated with ivermectin (0.2 mg/kg subcutaneously) and treated with closantel (10 mg/kg orally). The animals were kept together grazing the same pasture area. The treatment groups were drenched in two occasions 70 days apart: on 5th December 2019 and on 13th February 2020. On 19th March 2020, all lambs were slaughtered. The lamb heads were removed and sectioned along their longitudinal and sagittal axis to search for larvae. Recovered O. ovis larvae were counted and identified according to their developmental stage (L1, L2, and L3). Seven of the control lambs were infested with O. ovis larvae ranging from six to 17 larvae (11.6 mean infestation intensity). All recovered larvae from control group were intact and active. Three animals treated with ivermectin had O. ovis larvae (1-3 larvae), however they were dead and in degeneration. The animals treated with closantel did not have any larvae. The clinical suggestive signs of oestrosis were scarce over the experimental period. The averages of daily weight gain were similar (p > 0.05) among groups. Closantel and ivermectin had high efficacy against oestrosis and O. ovis parasitism did not hinder the performance of lambs.

ABSTRACT

Mammalian lice are obligate hematophagous ectoparasites that remain on the host throughout the life cycle. This study showed an endangered wild howler species (Alouatta belzebul) parasitized by Pediculus mjobergi. Twenty-seven primates were rescued during wildlife rescue activities in the process of vegetation suppression at the Belo Monte Hydroelectric Power Plant (UHE), located on the lower Xingu River, in the Brazilian Amazon basin. Among the 27 primates examined from two vegetal formations (alluvial rainforest and liana-infested forest) areas, 13 (48.15%) were parasitized by lice with all positive primates being from alluvial rainforest. Optical and scanning electron microscopy allowed the visualization of the structures and the diagnostic characters of the lice species in greater detail expanding the current description. These results highlight the importance of considering ecological and regional features to provide a better understanding of the factors that promote parasitism.

ABSTRACT

BACKGROUND: Haemonchus contortus, a gastrointestinal nematode parasite of sheep, is mainly controlled by anthelmintics; the occurrence of anthelmintic resistance leads to treatment failures and increases economic burden. Because molecular mechanisms involved in drug resistance can be elucidated by genomic studies, an extreme quantitative trait locus (X-QTL) mapping approach was used to identify co-segregation of the resistance phenotype with genetic markers to detect the genome-wide variants associated with monepantel resistance in H. contortus. METHODS: A cross between H. contortus isolates using parental susceptible (Par-S) males and monepantel resistant (Par-R) females resulted in SR progeny, while reciprocal cross resulted in RS progeny. Pools (n = 30,000) of infective larvae (L3) recovered from Par-R, and from SR and RS populations in the F3 generation, collected both before (unselected group) and 7 days after (selected group) selection with monepantel treatment in sheep hosts, were subjected to genome sequencing (Pool-Seq). Pairwise comparisons of allele frequencies between unselected and selected groups were performed for each population by Fisher's exact test (FET) and for both populations combined by a Cochran-Mantel-Haenszel (CMH) test. RESULTS: Mapping rates varied from 80.29 to 81.77% at a 90.4X mean coverage of aligned reads. After correction for multiple testing, significant (P < 0.05) changes in allele frequencies were detected by FET for 6 and 57 single nucleotide polymorphisms (SNPs) in the SR and RS populations, respectively, and by the CMH test for 124 SNPs in both populations. The significant variants located on chromosome 2 generated a selection signal in a genomic region harboring the mptl-1, deg-3 and des-2 genes, previously reported as candidates for monepantel resistance. In addition, three new variants were identified in the mptl-1 gene. CONCLUSIONS: This study expands knowledge on genome-wide molecular events underlying H. contortus resistance to monepantel. The identification of a genome region harboring major genes previously associated with monepantel resistance supports the results of the employed X-QTL approach. In addition, a deletion in exon 11 of the mptl-1 gene should be further investigated as the putative causal mutation leading to monepantel resistance.

ABSTRACT

Four species of Mammomonogamus are known from large African herbivores. A recent study demonstrated that a single Mammomonogamus species was shared by both western lowland gorillas (Gorilla gorilla gorilla) and African forest elephants (Loxodonta cyclotis) in Central African Republic, suggesting lower species diversity than previously described in literature. We examined more than 500 fecal samples collected from sympatric African forest elephants, western lowland gorillas, and African forest buffaloes (Syncerus caffer nanus) at four study sites across Central Africa and examined them by coproscopic methods to detect Mammomonogamus eggs, which were found at three of the study sites. Subsequently, sequences of 18S rDNA, 28S rDNA, and cox1 amplified from individual eggs were analyzed. Phylogenetic analyses of both nuclear and mitochondrial DNA revealed two clades: one formed by sequences originating from Gabonese buffaloes and the other comprising gorillas and elephants. The gorilla-elephant clade was further differentiated depending on the locality. We show the existence of at least two distinct species of Mammomonogamus, M. loxodontis in elephants and gorillas and M. nasicola in buffaloes. The available information on Mammomonogamus in African herbivores is reviewed.

ABSTRACT

This study examined the development of resistance to anthelmintics in Haemonchus contortus in lambs under suppressive or selective treatment regimens that included monepantel. Twenty Ile de France and 20 Santa Ines lambs were allocated to two anthelmintic treatment regimens, based on body weight and nematode faecal egg counts (FEC): targeted selective treatment (TST) or suppressive treatment, both with monepantel. Lambs of the TST group were treated individually when they presented with a packed cell volume (PCV) ≤20%. On 7 October 2016, the lambs were allocated to clean pastures, where they grazed in separated paddocks by group until late February 2017. The experimental area was contaminated with nematodes that were introduced with the experimental Ile de France and Santa Ines lambs, naturally infected with gastrointestinal nematodes. To maintain the grazing lambs in the suppressive treatment group and their pasture as free of worms as possible, these lambs were treated with anthelmintics before being allocated to their paddock and then were periodically treated with monepantel. However, the use of a suppressive treatment regimen that included monepantel over a period of 3 months resulted in the emergence of a population of resistant H. contortus. In the TST group, there was a rapid and progressive reduction in the efficacy of monepantel, which at the end of the experiment was only 76%. The Ile de France lambs were all treated one or more times during the experiment, whereas only two Santa Ines lambs in the TST required treatment. In conclusion, a population of H. contortus resistant to monepantel emerged quickly during the rainy season, even when sheep were submitted to selective treatment.

ABSTRACT

Iguaçu National Park, which contains the largest remnant of Atlantic Forest in southern Brazil and Iguaçu Falls as one of its main visiting points, is a year-round attraction for thousands of domestic and international tourists. Ring-tailed coatis are generalist, opportunistic carnivores that benefit from human association. These animals are the most abundant carnivores in this park, where they come into close contact with tourists and the resident population around the park. Moreover, as the park is surrounded by human dwellings, free roaming domestic dogs are frequently present, favoring the exchange of pathogens with wild animals. Wild carnivores are known to be infected with several pathogens, from viruses to arthropods, some of them passed on by domestic carnivores. Among the nematodes that infect wild carnivores, those of the Onchocercidae family are of concern due to their zoonotic potential. The objective of this research was to assess the prevalence of filarial nematodes in coatis and dogs in Iguaçu National Park, as well as the potential impact of infection on host health and body condition. To this end, 75 coatis and 50 adult dogs were captured in the aforementioned area. Seven species of filarioids, Dirofilaria immitis, Dirofilaria repens, Acanthocheilonema reconditum, Brugia sp., Mansonella sp. and two undetermined species, confirmed by histochemical tests, were diagnosed in the area. Knott's concentration test in coatis and dogs showed a prevalence of 81.6% and 16.0%, respectively. Seven microfilarial morphotypes were diagnosed in the coatis and two morphotypes were found in the dogs. A specific immunoassay test for Dirofilaria immitis revealed the presence of D. immitis infection in 1.33% of the coatis and 22% of the domestic dogs. The parasitic infection had negligible effects on the body condition of both dog and coati hosts, but an increase was found in eosinophil counts in coatis with filarial infection, as well as decreases in hematocrit and hemoglobin in the infected domestic dogs. These findings represent new locality and host records for all the filarioids diagnosed in the area of this study, with some of the diagnosed parasites, given their zoonotic potential, representing a health risk for park visitors and the local population.

ABSTRACT

Amazonia, the largest Brazilian biome, is one of the most diverse biomes around the world. Considering the Brazilian chiropteran species, 120 out of known 167 species are registered in Pará state, with 10 endemic species. Despite the high diversity of bats in Amazonia, studies on their parasites, especially on helminths, are scarce. Therefore, the present study aims to study the helminth fauna of different bats from the Pará state, Amazon biome, determine the descriptors of infection, and evaluate the host-parasite interactions, as well as evaluate differences in ecological indexes in accordance with the feeding guilds. The study was developed on 67 bats of 21 species captured in several areas of the Pará state. The animals were identified, divided into feeding guilds, and necropsied. The parasites obtained were identified and quantified. A total of 182 parasites were found in 20.89 % of the studied bats, representing nine species, as follows: Anenterotrema eduardocaballeroi, Anenterotrema liliputianum, Ochoterenatrema caballeroi, Tricholeiperia sp., Parahistiostrongylus octacanthus, Litomosoides guiterasi, Litomosoides brasiliensis, Capillariinae gen. sp., and Hymenolepididae gen. sp. Also, the results indicated that there was no impact of parasitism on host body condition and no relationship between sex and parasite intensity. In relation to the feeding guilds, the omnivores showed higher prevalence and mean intensity. Animals from regions closer to the equator tend to have greater richness in parasite species, but the present study revealed low diversity and richness in species. In conclusion, the ecological pattern observed for other animal groups, in which higher parasitic diversity are registered in lower latitudes, is not applicable to chiropterans from the study area.
