ABSTRACT

In the present study, we explored the effects of displacement directionality in mating behavior (i.e., lateralized and non-lateralized movements) on mating success (i.e., copulation occurs) and efficiency (i.e., time length at which copulation is achieved), and its association with sex and sexual experience in A. diaperinus. To do so, we carried out mating experiments and recorded the behavior of the mating pair during the whole mating sequence (i.e., precopulatory and copulatory phases). During the precopulatory phase, independently of sex and sexual experience, all beetles performed non-lateralized (i.e., backside or frontside) approaches; however, only sexually experienced beetles showed lateralized approaches (i.e., right-side and left-side). Notably, experienced males exhibited greater mating success than virgin males. After the approach, both virgin and experienced males displayed lateralized and non-lateralized mounts on the females with distinct mating success. Regardless of their sexual experience, 100% of successful mating attempts were achieved when males mounted from the females' right side. Furthermore, the development of lateralized approaches and mounts reduces the time of mating sequence span compared with non-lateralized behaviors. We highlight the importance of lateralization in mating behavior and sexual experience to achieve higher mating success, addressing a potential learning ability of beetles based on experience.

A total of 648 one-day-old chicks (Cobb®) which were allocated to one of three treatments (no application of the commercial product - control, and two concentrations of the product 2.00 and 3.33g/m2) in eight replicates with 27 birds each. The commercial product, which was based on cypermethrin was applied in the chambers with an atomizer. The lesser mealworm population was sampled weekly (1, 7, 14, 21, 28, 35 and 42 days of the experiment) using traps, at three points (front, middle and back) inside the pen. The performance, carcass and cut yields were evaluated. At 21, 28, 35 and 42 days of the experiment, the number of lesser mealworm adults and larvae was lower in the treatment groups involving 2.00 and 3.33g/m2 of the product than in the control group. On the first and 14th days of the study, the number of lesser mealworms adults was lower in the group treated with 3.33g/m2 of the product compared to the control group, although the results did not differ statistically from those obtained with 2.00g/m2 of the product. Um total de 648 pintos de um dia de idade (Cobb®) foram distribuídos em três câmaras com os tratamentos (sem aplicação do produto comercial - controle, e duas concentrações do produto comercial - 2,00 e 3,33g/m2) em oito repetições com 27 aves cada. O produto comercial a base de cipermetrina foi aplicado nas câmaras com um atomizador. A população de cascudinho foi amostrada semanalmente (1, 7, 14, 21, 28, 35 e 42 dias do experimento) usando armadilhas, em três pontos (frente, meio e fundo) de cada parcela. ABSTRACT

The lesser mealworms (Alphitobius diaperinus) constitute a common cosmopolitan pest in poultry flocks and may colonize the litter in adult and larval forms. Previous studies have documented their potential as carriers of enteric pathogens. In this context, S. enterica constitutes a prioritized zoonotic agent in the poultry industry due to the sanitary risks and economic losses associated with its presence. The aim of this study is to describe the presence of S. enterica strains in larval and adult forms of A. diaperinus collected from poultry litter belonging to industrial farms located in the central zone of Chile. A total of 403 specimens (203 adults and 200 larvae) were sampled from three farms and 25 flocks. For bacteriological isolation, beetles were processed to differentiate external and internal contamination. Then, isolates were serotyped according to the Kauffman-White scheme and antimicrobial resistance phenotypes were determined using the disk diffusion method. Gene sequences from the megaplasmid pESI were identified through a PCR based test. These procedures led to the detection of 15 S. enterica isolates, belonging to serotypes Infantis (14) and Livingstone (1), from both adults (6) and larval (9) specimens, with a similar external (7) and internal (8) distribution. Furthermore, all S. Infantis isolates showed antimicrobial resistance and evidence of megaplasmid pESI carriage, with all possessing multidrug-resistant phenotypes. Our results confirm that A. diaperinus constitutes a potential reservoir of zoonotic Salmonella strains of sanitary and economic concern for the industry and for public health.

ABSTRACT

Poultry litter reuse in Brazil is a common practice to reduce broiler production costs. Quicklime and shallow fermentation treatments are methods used to reduce microbial contamination and infestation of insects such as Alphitobius diaperinus (Panzer). The aim of this study was to evaluate the physicochemical parameters of reused poultry litter to better characterize the effects of quicklime and shallow fermentation on Salmonella and A. diaperinus control. Ammonia and humidity concentrations significantly increased on the litter treated with shallow fermentation and pH when treated with virgin and hydrated quicklime. For A. diaperinus control, shallow fermentation with 2 and 3 L of water and 3 L plus 600g of quicklime/m2 eliminated 100% of the insects. Results of assessed physicochemical parameters indicated that the treatments with quicklime and shallow fermentation are inefficient to control Salmonella spp. because they do not reach the indexes required for this pathogen elimination, mainly ammonia and pH. Ammonia index produced by microbial fermentation in shallow fermentation treatment eliminates A. diaperinus.

ABSTRACT

The lesser mealworm control is mostly made by insecticides, as the association of chlorpyrifos, cypermethrin and citronellal. The traditional insecticide application method (TRADM) depends on many liters of water and, with the objective of reducing water consumption, the aim was to compare TRADM with atomizer-equipment method (ATOMM). Four and three houses from farm A and B were used, respectively. Houses 1, 3 and 7 were treated with TRADM (1 liter of insecticide diluted in 500 liters of water) and houses 2, 4, 5 and 6 with ATOMM (1 liter of insecticide diluted in 9 liters of water). Twenty-four hours before and 30 (for farm A) and 45 days (for farm B) after the insecticide application, the number of insects was counted in each house (using 18 mapped traps/house). Farms were not considered different (p>0.05) and comparison was made between the houses. Houses were different (p<0.001) in the pre-treatment count, but they were equal in the second count (p=0.97). The insecticide application increased the number of traps without any insect (p<0.0001). In conclusion, the application method of chlorpyrifos, cypermethrin, and citronellal insecticide using atomizer equipment presented similar results than using the traditional application method. Both methods were effective in reducing the infestation of all houses, not making any difference if the dilution of the product was in 500 liters or in 9 liters of water; thus, it is possible to spend less water to run this management.


O controle do cascudinho é realizado principalmente por inseticidas, como a associação entre clorpirifos, cipermetrina e citronelal. O método tradicional de aplicação do inseticida (TRADM) depende de grande volume de água para a diluição e, visando à redução do uso de água, o estudo objetivou comparar TRADM com o método utilizando atomizador (ATOMM). Quatro galpões da propriedade A e três da B foram utilizados. Os galpões 1, 3 e 7 foram tratados com TRADM (1 litro inseticida:500 litros de água) e os galpões 2, 4, 5 e 6 tratados utilizando ATOMM (1 litro de inseticida:9 litros de água). Vinte e quatro horas antes e 30 (para A) e 45 dias (para B) após a aplicação foi realizada a contagem de cascudinhos em cada galpão (18 iscas mapeadas/galpão). Não houve diferença (p>0,05) entre as propriedades e os galpões foram considerados nas comparações. Estes diferiram quanto à contagem pré-tratamento (p<0,001), mas não diferiram na contagem pós-tratamento (p=0,97). A aplicação do inseticida aumentou o número de iscas sem insetos (p<0,0001). Concluindo, a aplicação do inseticida a base de clorpirifos, cipermetrina e citronelal usando atomizador apresentou resultado semelhante ao do método tradicional. Ambos os métodos são efetivos para reduzir a infestação de todos os galpões, não fazendo diferença se a diluição do produto foi realizada em 500 ou 9 litros de água; dessa forma, é possível gastar menor quantidade de água para realizar esse manejo.

ABSTRACT

The lesser mealworm, Alphitobius diaperinus Panzer 1797 (Coleoptera: Tenebrionidae), is a cosmopolitan insect pest affecting poultry production. Due to its cryptic behavior, insecticide control is usually not efficient. Thus, sustainable and effective methods would have an enormous and positive impact in poultry production. The aim of this study was to confirm the identity of the male-produced aggregation pheromone for a Brazilian population of A. diaperinus and to evaluate its biological activity in behavioral assays. Six male-specific compounds were identified: (R)-limonene (1), (E)-ocimene (2), 2-nonanone (3), (S)-linalool (4), (R)-daucene (5), all described before in an American population, and a sixth component, (E,E)-α-farnesene (6), which is apparently exclusive to a Brazilian population. Y-Tube bioassays confirmed the presence of a male-produced aggregation pheromone and showed that all components need to be present in a similar ratio and concentration as emitted by male A. diaperinus to produce a positive chemotactic response.

ABSTRACT

This present study was developed with the objective of detect Salmonella sp. by conventional bacteriology and qPCR techniques in samples of flooring material from transport crates (meconium); raising environment (swab of cages and drinking fountains); cloacal swab; food and insects from growing, rearing and production phases in a commercial group of laying hens. A total of 864 samples were collected, among whom 248 originated from growing, 392 from rearing and 224 from production phase. Among the 864 samples, 2,8% where positives in bacteriologic technique and 15.3% in qPCR. Contamination was higher in growing and rearing phases and declined in production phase. Twenty four isolations of Salmonella where typified as Salmonella Agona (41.7%), Salmonella Livingstone (33.3%), Salmonella Cerro (16.7%), Salmonella Senftenberg (4.2%) and Salmonella Schwarzengrund (4.2%). During growing phase Salmonella Livingstone was identified. These findings suggest vertical contamination in the group. During rearing and production phases, isolated materials belong to serovars Agona, Cerro, Senftenberg and Schwarzengrund, pointing to horizontal contamination. It is possible to conclude that both vertical and horizontal contaminations are important during the cycle of commercial egg production and contamination in rearing phase is higher than in growing and production phases.(AU)


O presente estudo foi desenvolvido com objetivo de detectar Salmonella sp. pelas técnicas de bacteriologia convencional e PCR em tempo real em amostras de forros de caixas de transporte (mecônio), de ambientes de criação (suabes de gaiola e de bebedouro), suabes de cloaca, ração e insetos oriundos das fases de cria, recria e produção de um lote de poedeiras comerciais. Foram coletadas 864 amostras das quais 248 foram originadas da cria, 392 da recria e 224 da produção. Das 864 amostras 2,8% foram positivas na técnica bacteriológica e 15,3% no PCR em tempo real. A contaminação aumentou da fase de cria para a recria e declinou na fase de produção. Vinte e quatro isolados de Salmonella foram tipificados como Salmonella Agona (41,7%), Salmonella Livingstone (33,3%), Salmonella Cerro (16,7%), Salmonella Senftenberg (4,2%) e Salmonella Schwarzengrund (4,2%). Na fase de cria identificou-se Salmonella Livingstone. Esses achados sugerem a contaminação vertical do lote. Nas fases de recria e produção os isolados pertenceram aos sorovares Agona, Cerro, Senftenberg e Schwarzengrund apontando para a contaminação horizontal. Pode-se concluir com este estudo que tanto a contaminação vertical como a horizontal são importantes no ciclo de produção de ovos comerciais e que a contaminação na fase de recria é maior que na fase de cria e de produção.(AU)

Among the 864 samples, 2,8% where positives in bacteriologic technique and 15.3% in qPCR. Contamination was higher in growing and rearing phases and declined in production phase. Twenty four isolations of Salmonella where typified as Salmonella Agona (41.7%), Salmonella Livingstone (33.3%), Salmonella Cerro (16.7%), Salmonella Senftenberg (4.2%) and Salmonella Schwarzengrund (4.2%). During growing phase Salmonella Livingstone was identified. These findings suggest vertical contamination in the group. During rearing and production phases, isolated materials belong to serovars Agona, Cerro, Senftenberg and Schwarzengrund, pointing to horizontal contamination. It is possible to conclude that both vertical and horizontal contaminations are important during the cycle of commercial egg production and contamination in rearing phase is higher than in growing and production phases.(AU)


O presente estudo foi desenvolvido com objetivo de detectar Salmonella sp. pelas técnicas de bacteriologia convencional e PCR em tempo real em amostras de forros de caixas de transporte (mecônio), de ambientes de criação (suabes de gaiola e de bebedouro), suabes de cloaca, ração e insetos oriundos das fases de cria, recria e produção de um lote de poedeiras comerciais. Foram coletadas 864 amostras das quais 248 foram originadas da cria, 392 da recria e 224 da produção. Das 864 amostras 2,8% foram positivas na técnica bacteriológica e 15,3% no PCR em tempo real. A contaminação aumentou da fase de cria para a recria e declinou na fase de produção. Vinte e quatro isolados de Salmonella foram tipificados como Salmonella Agona (41,7%), Salmonella Livingstone (33,3%), Salmonella Cerro (16,7%), Salmonella Senftenberg (4,2%) e Salmonella Schwarzengrund (4,2%). Na fase de cria identificou-se Salmonella Livingstone. Esses achados sugerem a contaminação vertical do lote. Nas fases de recria e produção os isolados pertenceram aos sorovares Agona, Cerro, Senftenberg e Schwarzengrund apontando para a contaminação horizontal. Pode-se concluir com este estudo que tanto a contaminação vertical como a horizontal são importantes no ciclo de produção de ovos comerciais e que a contaminação na fase de recria é maior que na fase de cria e de produção.(AU)

ABSTRACT

The aim of this study was to evaluate the in vitro bioactivity of tea tree (Melaleuca alternifolia) essential oil against larvae and adult forms of lesser mealworms (Alphitobius diaperinus) and its influence on the soil fauna. Tests were performed in triplicate using pure tea tree oil (TTO; 1, 5, 10, 25, 50, and 100%), TTO nanoparticles (1, 3, and 7.5%), or terpinen-4-ol, the main compound of the tea tree oil, at the same concentrations of TTO. Larvae and adult mortality occurred at concentrations up to 10 and 50% of TTO, respectively. No larvicidal or insecticidal effect of TTO nanoparticles was observed. Terpinen-4-ol showed insecticidal and larvicidal effect at concentrations higher than 25%. The evaluation of TTO effect on soil organisms was performed by standard ecotoxicological tests (ISO) with the springtail species Folsomia candida. Only TTO was used for ecotoxicological tests in doses of 1, 5, 10, 25, 50, and 100 mg kg-1 of soil. TTO had no negative effects on F. candida survival or reproduction. Therefore, it was concluded that M. alternifolia oil may be a new alternative for control of the lesser mealworm.(AU)

Complementarmente, determinou-se o perfil de suscetibilidade aos antimicrobianos: amoxicilina (10 mcg), ampicilina (10 mcg), ciprofloxacina (5 mcg), enrofloxacina (5 mcg), florfenicol (30 mcg), neomicina (30 mcg), sulfonamida (300 mcg), tetraciclina (30 mcg) e trimetoprim-sulfametoxazol (25 mcg) dos serovares tipificados isolados. As amostras foram submetidas às análises microbiológicas pelos métodos bacteriológicos convencionais. Salmonella sp. foi isolada em 6,2% (4/64) do fígado, 4,7% (3/64) do coração, 3,1% (2/64) dos sacos da gema e 4,7% (3/64) do mecônio, num total de 4,7% (12/256) (pinto de um dia); em 10,2% (13/128) das amostras ambientais, sendo 9,4% (9/96) de suabes de arrasto 12,5%, (4/32) de larvas e adultos Alphitobius diaperinus e em 4,4% (28/640) das amostras em abatedouros, sendo 6,5% (21/320) dos inglúvios e 2,2% (7/320) dos conteúdos cecais de abatedouro. Salmonella enterica ser. Enteritidis foi identificada em suabes de arrasto e em amostras de Alphitobius diaperinus, enquanto Salmonella enterica ser. Typhimurium foi encontrada nos inglúvios e cecos. Salmonella enterica ser. Enteritidis apresentaram 75% (6/8) de resistência às sulfonamidas e Salmonella enterica ser. Typhimurium 100% (3/3). A amoxicilina foi outro antimicrobiano com elevada frequência de resistência. Adicionalmente, 20,7% (11/53) dos serovares apresentaram resistência simultânea a pelo menos dois princípios ativos. Conclui-se que Salmonella encontra-se amplamente distribuída no fluxo de produção de frangos de corte, e a via vertical continua sendo uma fonte de introdução de Salmonella sp. à cadeia de produção; cama e insetos podem perpetuar e veicular Salmonella de interesse zoonótico no ambiente avícola; a existência de cepas resistentes aos antimicrobianos, bem como a resistência múltipla, constituem ameaça à saúde pública.(AU)

It also aimed at determining the susceptibility profile to amoxicillin (10 mcg), ampicillin (10 mcg), ciprofloxacin (5 mcg), enrofloxacin (5 mcg), florfenicol (30 mcg), neomycin (30 mcg), sulfonamide (300 mcg), tetracycline (30 mcg) and trimethoprim-sulfamethoxazole (25 mcg) of typed serovars isolated. The samples were submitted to microbiological analyses by the conventional method. Salmonella sp. was isolated in 4.7% (12/256) of samples of one-day old chicks, in 6.2% (4/64) of the liver, 4.7% (3/64) of the hearts, 3.1% (2/64) of the yolk sacs, 4.7% (3/64) of meconium, and in10.2% (13/128) of the environment samples, being 9.4% (9/96) from drag swabs, 12.5% (4/32) from larvae and adult Alphitobius diaperinus and 4.4% (28/640) of the slaughterhouses samples, being 6.5% (21/320) of crops and 2.2% (7/320) of cecum samples. Salmonella enterica ser. Enteritidis was identified only in drag swabs and Alphitobius diaperinus. Salmonella enterica ser. Typhimurium was observed in the crops and cecum, which have shown high frequency of resistance to sulfonamides, 75% (6/8) and100% (3/30), respectively. Additionally, 20.7% (11/53) of the serovars have shown multiresistance to at least two of the tested drugs. In conclusion, Salmonella can be widely spread in broiler production flow, and the vertical pathway is still a major source of Salmonella insertion in the poultry production chain. The litter and bugs can perpetuate and disseminate Salmonella sp. as well as both the existence of strains resistant to antimicrobial and the occurrence of multiresistance are a threat to public health.(AU)

ABSTRACT

Global poultry production is plagued by a wide variety of arthropods. The problems associated with their chemical control have led to an increasing search for control alternatives, and entomopathogenic fungi seem to be a promising strategy. Despite the large number of insects and mites considered as important pests in animal production, studies on the use of entomopathogenic fungi for their control are still scarce compared with agricultural pests, particularly in Brazil. This article reviews some damages and control aspects of the main arthropod pests that affect Brazilian poultry production, including house flies, lesser mealworms, and feather mites, by the use of the entomopathogenic fungi Beauveria bassiana and Metarhizium anisopliae. Studies published in the last 20 years were reviewed, and the main problems and limitations of that pest-control strategy are discussed.(AU)

A estabilidade de ambiência na avicultura moderna tem favorecido a proliferação de insetos praga. Nesse cenário, Alphitobius diaperinus (Panzer) (Coleoptera: Tenebrionidae), comumente denominado cascudinho, surge como principal inseto cosmopolita infestando camas de aviários. Neste sentido o objetivo deste trabalho foi avaliar os fatores cal hidratada, temperatura e umidade na cama de aviários sobre a mortalidade de A. diaperinus em condições de laboratório. Foram realizados bioensaios laboratoriais simulando o ambiente de criação em recipientes com a adição de cama de aviário, ração e adultos e larvas de A. diaperinus. A variável resposta foi a mortalidade aos sete e 10 dias após a instalação dos experimentos. A adição de cal hidratada na cama de aviário na dosagem de 400g m-² promoveu mortalidade parcial de adultos (35,62%±2,74) e larvas (45,62%±1,75) de A. diaperinus. O fator umidade na cama de aviário, independente do percentual, não provocou mortalidade significativa de adultos de A. diaperinus. A temperatura da cama de aviário em 45°C proveu mortalidade total de adultos e larvas de A. diaperinus nas condições propostas.


The stability of in the modern poultry industry has been favored the proliferation of insect pests. The response variable was mortality at seven and 10 days after installation of the experiments. The addition of hydrated lime in the poultry litter at a dosage of 400g m-² promoted partial mortality of the adult population (35.62%±2,74) and larvae (45.62%±1,75) of A. diaperinus. The moisture in the litter, regardless of percentage, did not cause significant mortality of adults of A. diaperinus. The temperature in the litter at 45°C caused total mortality of adults and larvae of A. diaperinus at the conditions studied.
