ABSTRACT

A rapid validated ultra-fast liquid chromatography-photodiode array detector (UFLC-PDA) method was developed to identify and quantify ayapanin (AY) and umbelliferone (UM) simultaneously in Ayapana triplinervis Vahl methanolic extract. The method was validated for linearity, limit of detection (LOD; 3:1σ/S), limit of quantification (LOQ; 10:1σ/S), precision, accuracy, specificity and robustness. The response was linear with a good correlation between concentration and mean peak area through a correlation coefficient of 0.9996, y = 7025.7x - 2269.8 and 0.9997, y = y = 16,262x - 946 with LOD of 6.256 ± 0.52 and 3.325 ± 0.36, and LOQ of 18.838 ± 0.18 and 8.870 ± 0.85 for AY (0.67% w/w) and UM (0.18% w/w), respectively. The relative standard deviation (%) of precision and recovery of AY and UM was <2.0%. The proposed method was simple, accurate, specific, precise and reproducible.

ABSTRACT

This study investigated the gastroprotective effect of Ayapana triplinervis leaves against indomethacin-induced gastric ulcer in male albino rat. Gastric ulceration was developed by single oral dose of indomethacin (30 mg/kg). Experimental rats were pretreated with omeprazole (positive control 20 mg/kg), hydromethanolic extract of A. triplinervis (200 mg/kg) for 28 days just before the indomethacin treatment. Free acidity, total acidity, pepsin activity and gastric volume, gastric pH, tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6) in stomach and serum prostaglandin E2 levels were assessed in control, ulcerated group and A. triplinervis pretreated groups. Oxidative stress biomarkers, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in serum also evaluated. All the parameters were deviated from control in indomethacin-treated group but significantly protected in A. triplinervis-pretreated group. The active ingredient(s) present in the A. triplinervis have antioxidant and gastroprotective properties that prevent the indomethacin-induced gastric ulcer. PRACTICAL APPLICATIONS: A. triplinervis has been widely consumed from earlier time as traditional medicine for the treatment of gastric problem and gastric ulcer in India. This is the first report that hydromethanolic extract of A. triplinervis has potent therapeutic properties against gastric ulcer. This work will provide a clue to the pharmaceutical industry to develop an effective gastroprotective agent.

ABSTRACT

Zika virus (ZIKV) is an emerging mosquito-borne virus of medical concern. ZIKV infection may represent a serious disease, causing neonatal microcephaly and neurological disorders. Nowadays, there is no approved antiviral against ZIKV. Several indigenous or endemic medicinal plants from Mascarene archipelago in Indian Ocean have been found able to inhibit ZIKV infection. The purpose of our study was to determine whether essential oil (EO) from Reunion Island medicinal plant Ayapana triplinervis, whose thymohydroquinone dimethyl ether (THQ) is the main component has the potential to prevent ZIKV infection in human cells. Virological assays were performed on human epithelial A549 cells infected with either GFP reporter ZIKV or epidemic viral strain. Zebrafish assay was employed to evaluate the acute toxicity of THQ in vivo. We showed that both EO and THQ inhibit ZIKV infection in human cells with IC50 values of 38 and 45 µg/mL, respectively. At the noncytotoxic concentrations, EO and THQ reduced virus progeny production by 3-log. Time-of-drug-addition assays revealed that THQ could act as viral entry inhibitor. At the antiviral effective concentration, THQ injection in zebrafish does not lead to any signs of stress and does not impact fish survival, demonstrating the absence of acute toxicity for THQ. From our data, we propose that THQ is a new potent antiviral phytocompound against ZIKV, supporting the potential use of medicinal plants from Reunion Island as a source of natural and safe antiviral substances against medically important mosquito-borne viruses.

ABSTRACT

Ayapana triplinervis (Asteraceae) é uma erva medicinal da Amazônia conhecida como "Japana branca" e "Japana roxa" utilizada como tônico, digestivo, antidiarréico dentre outros. Objetivou-se caracterizar neste estudo a morfoanatomia caulinar e foliar de A. triplinervis, para corroborar se esta espécie apresenta morfotipos, além de fornecer dados para sua identificação. Amostras foram coletadas no Horto de Plantas Medicinais da Universidade Federal Rural da Amazônia e Embrapa Amazônia Oriental, analisadas morfologicamente e segundo técnicas usuais em anatomia vegetal. O caule é cilíndrico branco ou roxo, com epiderme unisseriada persistente e endoderme com bainha amilífera e estrias de Caspary. As folhas são simples, opostas, triplinervadas, com base atenuada ou decorrente. Possuem estômatos anomocíticos, cera em placas ou granular, glândulas peroladas, mesofilo dorsiventral com feixes colaterais e ductos secretores. A. triplinervis apresentou caracteres morfoanatômicos diferentes entre os tipos branca e roxa, o que possibilita a distinção dos dois morfotipos.


Ayapanatriplinervis (Asteraceae), also known as "Japana branca" and "Japana roxa" in Brazil, is a medicinal herb from the Amazon used as tonic, digestive, antidiarrheal, among others. This study aimed to characterize the morphoanatomy of the aerial vegetative axis of A. triplinervis to corroborate that this species has morphotypes, in order to provide data for their identification. Samples were collected in the Garden of Medicinal Plants of the Federal Rural University of Amazônia and Embrapa Amazônia Oriental, and they were analyzed morphologically and by the standardtechniques of plant anatomy. The stem is cylindrical, white or purple, with uniseriate epidermis, trichomes and endoderm with starch sheath and Casparian strips. The leaves are simple, opposite, with attenuate or arising basis; they have anomocytic stomata, epicuticular wax plates or granules, pearl glands, dorsiventral mesophyll with collateral vascular bundles and secretory ducts. Ayapana triplinervis showed different morphoanatomical features betweenthe white and purple types, which enablesthe distinction between the two morphotypes.
