ABSTRACT

The aim of this study was to evaluate different times for timed artificial insemination (TAI) in buffalo submitted to a P4/E2/eCG-based protocol. In this study, 204 buffaloes were distributed into one of two groups (TAI56, n=103 and TAI64, n=101). At a random stage of the oestrous cycle (Day 0 = D0), in the morning (TAI56, a.m.) or afternoon (TAI64, p.m.), buffaloes received an intravaginal progesterone device (P4; 1.0 g) plus EB (2.0 mg i.m.). On D9 a.m. (TAI56) or p.m. (TAI64), the P4 was removed and buffaloes received PGF2a (0.53 mg i.m. sodium cloprostenol) and eCG (400 IU i.m.). On D10 a.m. (TAI56) or p.m. (TAI64), 24 h after P4 removal, buffaloes were treated with EB (1.0 mg i.m.). Buffaloes from TAI56 and TAI64 were inseminated 56 and 64 h after P4 removal (D11, p.m. and D12, a.m., respectively). Ultrasound examinations were performed on D0 to ascertain ovarian follicular status, at TAI to measure the diameter of the dominant follicle (DF) and D42 for pregnancy diagnosis. The statistical analysis was performed using the GLIMMIX procedure of SAS®. There was no difference between TAI56 and TAI64 for the diameter of the DF at TAI and the pregnancy per TAI. It was concluded that TAI 56 or 64 h after P4 removal did not affect fertility in buffaloes submitted to the induction of ovulation with EB. The present research supports that is possible to perform TAI at any time throughout the day in buffalo synchronized during the non-breeding season.(AU)


O objetivo deste estudo foi avaliar diferentes momentos para a realização da IATF em búfalas submetidas a um protocolo à base de P4/E2/eCG. Neste estudo, 204 búfalas foram distribuídas em um de dois grupos (IATF56, n=103 e IATF64, n=101). Em estágio aleatório do ciclo estral (Dia 0 = D0), pela manhã (IATF56, manhã) ou pela tarde (IATF64, tarde), as búfalas receberam um dispositivo intravaginal de progesterona (P4; 1,0 g) e BE (2,0 mg i.m.). No D9 pela manhã (IATF56) ou pela tarde (IATF64), a P4 foi removida e as búfalas receberam PGF2a (0,53 mg i.m. cloprostenol sódico) e eCG (400 UI i.m.). No D10 pela manhã (IATF56) ou pela tarde (IATF64), 24 h após a remoção da P4, as búfalas foram tratadas com BE (1,0 mg i.m.). As búfalas dos grupos IATF56 e IATF64 foram inseminadas 56 e 64 h após a remoção da P4 (D11, pela tarde e D12, pela manhã, respectivamente). Avaliações ultrassonográficas foram realizadas no D0 para verificar o status folicular ovariano, na IATF para medir o diâmetro do folículo dominante (FD) e no D42 para o diagnóstico de gestação. A análise estatística foi realizada utilizando o procedimento GLIMMIX do SAS®. Não houve diferença entre os grupos IATF56 e IATF64 no diâmetro do FD na IATF e na prenhez por IATF. Conclui-se que a IATF 56 ou 64 h após a remoção da P4 não afeta a fertilidade de búfalas submetidas à indução da ovulação com BE. A presente pesquisa evidencia que é possível realizar a IATF durante todo o dia em búfalas sincronizadas durante a estação reprodutiva desfavorável.(AU)

ABSTRACT

The chemical constituents of essential oils obtained by hydrodistillation of the leaves of Atalantia roxburghiana Hook. f. and Tetradium trichotomum Lour., as well as the leaves and fruits of Macclurodendron oligophlebia (Merr.) Hartl. (Rutaceae) are being reported. The essential oils were analysed by using gas chromatography (GC) and gas chromatography coupled with mass spectrometry (GC-MS). Sabinene (36.9%) was the most singly abundant compound in the leaf of A. roxburghiana. The major constituents present in the leaf oil of T. trichotorum were (E)-ß-ocimene (24.8%), α-pinene (10.4%), (Z)-ß-ocimene (9.4%) and ß-caryophyllene (8.0%). On the other hand, while α-pinene (17.5%), ß-caryophyllene (15.5%) and caryophyllene oxide (10.6%) occurred in higher proportion in the leaf of M. oligophlebia, the fruit oil was dominated by benzyl benzoate (16.8%), (E, E)-farnesol (8.3%) and ß-caryophyllene (6.0%).


Se muestran los constituyentes químicos de los aceites esenciales obtenidos, por hidrodestilación, de las hojas de Atalantia roxburghiana Hook. f. y de Tetradium trichotomum Lour., así como de las hojas y frutos de Macclurodendron oligophlebia (Merr.) Hartl. (Rutaceae). Los aceites esenciales fueron analizados por Cromatografía de Gases (CG) y por Cromatografía de Gases acoplada a Espectrometría de Masas (CG-EM). El compuesto más abundante en las hojas de A. roxburghiana es el sabineno (36.9%); mientras que los mayoritarios en el aceite de las hojas de T. trichotomum fueron (E)-ß-ocimeno (24.8%), α-pineno (10.4%), (Z)-ß-ocimeno (9.4%) y ß- cariofileno (8.0%). En las hojas de M. oligophlebia los compuestos más abundantes fueron α-pineno (17.5%), ß-cariofileno (15.5%) y óxido de cariofileno (10.6%); sin embargo, en el aceite obtenido del fruto fueron benzoato de bencilo (16.8%), (E, E)-farnesol (8.3%) y ß- cariofileno (6.0%).

ABSTRACT

The aim of this study was the indication of an additive for the ensilage of sugarcane (Saccharum officinarum L.). In a laboratory trial, nine treatments were applied (g of additive/kg of fresh forage - FF) to the sugarcane (RB867515), before ensiling in minisilos (15 x 30 cm PVC tubes) during 78 days: untreated; urea (5) + sodium benzoate (0.5); urea (7.5) + benzoate (0.5); urea (5) + benzoate (0.75); urea (7.5) + benzoate (0.75); sodium propionate (1, 2 and 4); calcium hydroxide (10). Urea + benzoate in the lowest doses, propionate in the higher concentration and calcium hydroxide were selected, considering the ethanol content (26.5, 27.2 and 7.4 g/kg DM, respectively), total DM loss (88, 46 and 58 g/kg DM, respectively) and digestibility (541, 496 and 516 g/kg DM, respectively) of the silages. Silages treated with these doses of additives and the untreated silage (80 d of storage) were fed (nine + seven d) to 16 castrated male sheep (Santa Inês) housed in metabolic cages. The silage with calcium hydroxide presented coefficients of apparent digestibility of DM (0.44), of NDF (0.4) and DM intake (20 g/kg live weight) in the higher levels. Calcium hydroxide was superior to propionate and urea + benzoate, considering alcoholic fermentation control and reduction of losses in the silage and the forages nutritional value.

Em um ensaio de laboratório, nove tratamentos foram aplicados (g de aditivo/kgde forragem fresca - FF) à cana-de-açúcar (RB867515) antes da ensilagem em minisilos (tubos de PVC de 15 x 30 cm) durante 78 d: sem tratamento; uréia (5) + benzoato de sódio (0,5); uréia (7,5) + benzoato (0,5); uréia (5) + benzoato (0,75); uréia (7,5) + benzoato (0,75); propionato de sódio (1, 2 e 4); hidróxido de cálcio (10). A silagem com hidróxido de cálcio apresentou coeficientes de digestibilidade aparente da MS (0,44), da FDN (0,40) e de ingestão da MS (20 g/kg peso vivo) nos níveis mais altos. Hidróxido de cálcio foi superior ao propionato e à uréia + benzoato, considerando-se o controle da fermentação alcoólica, a redução de perdas e o valor nutritivo da silagem.

Two experiments were conducted aiming to evaluate the effects of two ovulatory inducers (Exp.1) and equine chorionic gonadotropin (eCG; Exp.2) on follicular and luteal dynamics in a fixed-time AI (FTAI) protocol in locally adapted Curraleiro Pé-Duro cows. In Exp. 1 multiparous cows (n=12) received an intravaginal device containing 1g of progesterone (P4) for 8 days and 2mg of estradiol benzoate (EB) intramuscularly (IM) at device insertion (Day 0). At device removal (Day 8) 0.150mg of Sodium D-Cloprostenol was administered IM and the cows were randomly assigned to receive 1mg of EB (EB8) or 1mg of estradiol cypionate (EC8) IM, or to not receive any ovulatory inducer (Control). All the animals participated in all treatments (crossover). The interval from P4 removal to ovulation was shorter and less variable in the EB8 treatment group (P≤0.05). In Exp. 2 (crossover), multiparous cows (n=12) received the same hormonal treatment as the EB8 group in Exp.1. At device removal (Day 8) cows were randomly assigned to receive 300UI of eCG IM or to not receive eCG (Control). No difference was ascertained on follicular and luteal parameters in Exp. 2 (P>0.05). We concluded that EB can be used as the ovulatory inducer (Exp. 1) in a FTAI protocol in Curraleiro Pé-Duro cows. However, eCG (Exp. 2) was not able to stimulate follicular and luteal development. This result is probably due to the adaptive capacity of Curraleiro Pé-Duro cows that maintained a satisfactory body condition score even in dry and hot environments.(AU)


Foram realizados dois experimentos com o objetivo de avaliar o efeito de dois indutores da ovulação e da gonadotrofina coriônica equina (eCG) na dinâmica folicular e luteal, em um protocolo de inseminação artificial em tempo fixo (IATF) em vacas localmente adaptadas da raça Curraleiro Pé-Duro. No experimento 1, vacas pluríparas receberam um dispositivo intravaginal contendo 1g de progesterona (P4) durante oito dias e 2mg de benzoato de estradiol (BE) intramuscular (IM) no momento da inserção do dispositivo (dia zero). Na retirada do dispositivo (dia oito), as vacas receberam 0,150mg de D-cloprostenol sódico IM e foram separadas aleatoriamente para receber 1mg de BE IM (BE8) ou 1mg de cipionato de estradiol IM (CE8), ou nenhum indutor da ovulação (controle). Todos os animais participaram de todos os tratamentos (crossover). O intervalo entre a retirada da P4 e a ovulação foi menor e menos variável no tratamento BE8 (P≤0,05). O momento da ovulação foi mais precoce e mais concentrado nos animais do grupo BE 8. No experimento 2 (crossover), vacas pluríparas receberam o mesmo tratamento hormonal do grupo BE8 do experimento1. Na retirada do dispositivo (dia 8), as vacas foram separadas aleatoriamente para receberem 300UI de eCG IM, enquanto o controle não. Não houve diferença nos parâmetros foliculares e luteais avaliados no experimento 2 (P>0,05). Em conclusão, o BE pode ser utilizado como indutor da ovulação (experimento 1) em protocolos de IATF em vacas Curraleiras Pé-Duro. Entretanto, o eCG (experimento 2) não foi capaz de estimular o desenvolvimento folicular e luteal. Esse resultado é devido provavelmente à capacidade adaptativa das vacas Curraleiras Pé-Duro em manter uma condição corporal satisfatória mesmo em condições de clima seco e quente.(AU)

ABSTRACT

SUMMARY This study aimed to evaluate the effect of using two estradiol esters, on the diameter of the ovulatory follicle (DFOL), and pregnancy rates of dairy cows submitted to FTAI program. Therefore, 94 females crossbred Dutch-Gir were subjected to the following hormone protocol started on a random day D0, by insertion of an intravaginal device of progesterone (P4) associated to 2.0 mg of estradiol benzoate (EB) intramuscular (im). In D8, the devices were removed and 500 µg of sodium cloprostenol prostaglandin F2 was administered and 300 IU of equine chorionic gonadotropin (eCG) i.m. At this time the animals were divided in two groups: 50 cows were allocated to EC group and received 1 mg of estradiol cypionate i.m., while the other 44 received 1.0 mg of EB i.m. 24 hours after removal of the devices (D9), being called the BE Group. ABSTRACT

La hiperamonemia se presenta en forma secundaria por aumento en la producción de amonio, como en la hemorragia gastrointestinal o disminución de la eliminación, como ocurre en errores innatos del metabolismo, principalmente en aquellos con defectos en el ciclo de la urea, insuficiencia hepática o fármacos. Clasificar la hiperamonemia y reportar las opciones terapéuticas en niños, su abordaje clínico y revisión de la literatura. Estudio prospectivo, descriptivo y transversal de niños con hiperamonemia. Variables: edad, género, etiología, niveles de amonio, clínica, tratamiento. 21 pacientes, 12 (57,12%) varones y 9 (42,88%) hembras. Edad promedio 3,91 años (rango:<1mes-14 años). Amonio promedio general 214,66 mmol/l (rango:110-980), clasificados según severidad: sin insuficiencia hepática 11/21 con promedio de amonio 99,44 y 201 mmol/l en hiperamonemia leve y moderada respectivamente. Clínica y laboratorio de insuficiencia hepática en 10/21 con promedio de amonio de 114,44, 287,51 y 756,66 en leve, moderada y severa hiperamonemia, con una diferencia significativa entre el nivel de amonio y la presencia o ausencia de insuficiencia hepática (p<0,0001); 5/10 con insuficiencia hepática ingresaron a terapia intensiva, 4 de ellos presentaron encefalopatía hepática, un paciente fallecido. Etiología: Error innato del metabolismo 33,33%, toxicidad por medicamentos 23,80%, hepatitis viral A fulminante 19,04% y otros virus 9,52%, hepatitis autoinmune 4,76% y urosepsis 4,76%. En los casos leves-moderados se administró lactulosa dosis respuesta vía oral 19/21 y por enema rectal 7/21 con L-carnitina en 15/21 y en Hiperamonemia severa adicionalmente Benzoato de sodio en 4/21 y hubo indicación de hemodiálisis en 3 pacientes. Restricción proteica en todos, vitaminoterapia y 6 niños tratados con ácido ursodeoxicólico. La hiperamonemia es multifactorial, requiere diagnóstico temprano, la clasificación de severidad permite el tratamiento oportuno para evitar complicaciones....

To classify the report hyperammonaemia and therapeutic options in children, its clinical approach and review of the literature. Prospective, descriptive and transversal children with hyperammonaemia. Variables: age, gender, etiology, ammonia levels, clinical treatment. 21 patients, 12 (57,12%) males and 9 (42,88%) females. Clinical and laboratory liver failure 10/21 with ammonium averaging 114,44, 287,51 and 756,66 as mild, moderate and severe hyperammonemia, with a significant difference between the level of ammonia and the presence or absence of liver failure (p < 0,0001), 5/10 with liver failure admitted to intensive care, 4 of them had hepatic encephalopathy, a patient died. Etiology: An inborn error of metabolism 33,33%, 23,80% drug toxicity, fulminant viral hepatitis and other viruses 19,04% 9,52% 4,76% autoimmune hepatitis and urosepsis 4,76%. In mild-moderate cases were given oral lactulose Dose 19/21 and by enema rectal 7/21 with L-carnitine in 15/21 and further severe Hyperammonemia sodium benzoate 4/21 and was indication of hemodialysis in 3 patients. Protein restriction at all, vitamin therapy and 6 children treated with ácidoursodeoxicólico. Hyperammonemia is multifactorial, requires early diagnosis, classification of severity allows early treatment to avoid complications and development of irreversible neurological sequelae

Background: Tissue texture, or tenderness, may be checked-out by analyzing the force necessary to cause muscle fibers rupture (shearing force) and the result is expressed in kilogram-force (kgf). The aim of this paper was to analyze the texture of broiler chicken breast muscles fi xed in a 10% formaldehyde solution and kept in a 0.5% sodium benzoate and 0.5% acetic acid solution up to 180 days so any alteration regarding resistance and the best moment for dissection could be described. Materials, Methods & Results: Forty chicken breasts were used and muscle texture was analyzed by the shearing force whose values were correspondent to the maximum force necessary to chisel the samples. Eight of 40 breasts were taken as a control group (fresh muscles). The remaining breast muscles were fi xed in a 4% formaldehyde solution and kept in plastic containers with this solution during seven days and, then, eight breasts went through texture analysis. The 24 remaining breasts were set in plastic containers with 0.5% sodium benzoate and 0.5% acetic acid solution. The texture analyses were taken out at 30, 90 and 180 days of conservation, and eight breasts were used in each time. Data were submitted to statistical analysis by the 5% Tukey's test. The average shearing force of the control, formaldehyde and 30-day sodium benzoate and acetic acid groups differed among them and from the 90-day and 180-day of sodium benzoate and acetic acid groups. The average shearing forces of the two last groups did not statistically differ among them. Discussion: The longer the muscles were kept in the sodium benzoate and acetic acid solutions, the more they became soft, and the shearing force decreased from 21.46 Kgf (seven days of conservation) to 19.20 Kgf (30 days), 17.23 Kgf (90 days) and 16.32 Kgf (180 days) not reaching, however, values from 8.97 to 15.31 Kgf, as described in chicken breasts conserved in 96oGL ethylic alcohol. The average shearing force of chicken breasts conserved in sodium benzoate and acetic acid progressively decreased during 180 days of conservation, when it reached a value of 16.32 Kgf, similar to the value of 15.32 Kgf of chicken breasts conserved for 45 days in formaldehyde. By the seventh day of conservation in 10% formaldehyde, samples presented an average shearing force of 21.46 Kgf, which was higher than the 15.33 Kgf of chicken breasts conserved during 45 days or the 14.74 to 17.37 Kgf of the same tissue kept up to a year in formaldehyde solution. It was concluded that conservation of breast broiler chickens fixed in formaldehyde and conserved in sodium benzoate and acetic acid made them softer through within 180 days, resulting in a better structure malleability and suggesting that dissection of anatomical muscle pieces may occur in an easier way in this moment. Although the sodium benzoate and acetic acid effect in texture of broiler chicken muscle tissue fixed in formaldehyde has been studied, it is believed that, due this great tissue homogeneity, these data might be taken or used as basis to similar studies in other species.

Background:  Tissue  texture,  or  tenderness,  may  be  checked-out  by  analyzing  the  force  necessary  to  cause  muscle  fibers rupture  (shearing  force)  and  the  result  is  expressed  in  kilogram-force  (kgf).  The  aim  of  this  paper  was  to  analyze  the  texture of  broiler  chicken  breast  muscles  fi  xed  in  a  10%  formaldehyde  solution  and  kept  in  a  0.5%  sodium  benzoate  and  0.5%  acetic acid  solution  up  to  180  days  so  any  alteration  regarding  resistance  and  the  best  moment  for  dissection  could  be  described.Materials,  Methods  &  Results:  Forty  chicken  breasts  were  used  and  muscle  texture  was  analyzed  by  the  shearing  force whose  values  were  correspondent  to  the  maximum  force  necessary  to  chisel  the  samples.  Eight  of  40  breasts  were  taken as  a  control  group  (fresh  muscles).  The  remaining  breast  muscles  were  fi  xed  in  a  4%  formaldehyde  solution  and  kept in  plastic  containers  with  this  solution  during  seven  days  and,  then,  eight  breasts  went  through  texture  analysis.  The  24 remaining  breasts  were  set  in  plastic  containers  with  0.5%  sodium  benzoate  and  0.5%  acetic  acid  solution.  The  texture analyses  were  taken  out  at  30,  90  and  180  days  of  conservation,  and  eight  breasts  were  used  in  each  time.  The  shearing force  was

ABSTRACT

The objective of this study was to evaluate the effect of medroxy-progesterone acetate (MAP) with or without estradiol benzoate (EB) on follicular growth during the estrous cycle in cattle. In the first experiment, Hereford cows were synchronized with a synthetic analogue of PGF2 alpha and were treated with two different doses of MAP (250 or 500 mg) with or without EB for 7 days starting on day 8 of the estrous cycle. Follicular growth was inhibited (P<0.05) in all cows except controls and those receiving 250mg MAP without EB. Seventy-five percent of the animals (15/20) showed estrus on days 21 and 22 of the cycle rather than at MAP withdrawal, demonstrating that these treatments did not induce estrus. To determine whether the EB treatment altered endometrial sensitivity to oxytocin and thus the luteolytic cascade, multiparous pre-synchronized cows received 5 mg of EB followed 6 hours later with 50 IU of oxytocin (OT; n=9). Eight hours after EB injection, endometrial fragments were collected from the cows on days 4, 13 and 17 of the estrous cycle and COX-2 gene expression was measured by PCR. EB increased COX-2 mRNA levels only on day 17 of the estrous cycle (P<0.05). In conclusion, MAP alone or associated with EB is able to suppress bovine follicular growth. However, EB in the presence of MAP is not efficient to induce luteolysis in cows when injected on day 8 of the estrous cycle.


Este estudo teve como objetivo avaliar o efeito do acetato de medroxi-progesterona (MAP) com ou sem benzoato de estradiol (BE) sobre o crescimento folicular durante o ciclo estral bovino. No primeiro experimento, vacas da raça Hereford foram sincronizadas com um análogo sintético de PGF2á e tratadas com duas doses diferentes de MAP (250 ou 500mg), com ou sem EB, durante 7 dias, iniciando-se no oitavo dia do ciclo estral. Observou-se uma inibição do crescimento folicular (P<0,05) em todas as vacas, exceto no grupo controle e no grupo que recebeu 250mg de MAP sem BE. Os 75 por cento dos animais não exibiu estro no momento da remoção do MAP, mas sim nos dias 21 e 22 do ciclo, demonstrando que os tratamentos não induziram cio. Para se determinar se o tratamento com BE alterou a sensibilidade endometrial à ocitocina e, assim, a cascata luteolítica, vacas multíparas pré-sincronizadas receberam 5mg de BE, seguidos, após 6 horas, de 50 UI de ocitocina (OT; n=9). Entretanto, o BE, na presença de MAP é ineficaz na indução da luteólise bovina, quando injetado no oitavo dia do ciclo estral.

ABSTRACT

Este trabalho foi realizado com o objetivo de estudar a eficácia do protocolo de curta duração de sincronização do estro. As ovelhas foram divididas em quatro grupos: Grupo Controle (esponjas de MAP por 12 dias, e eCG na retirada); Os Grupos I, II e III permaneceram com a esponja por quatro dias e na retirada foi aplicado 100 µg de PGF e adicionalmente: no Grupo I (0,1 mg de Benzoato de estradiol - BE na colocação da esponja e 48h após a retirada 50 µg de GnRH); Grupo II (35 mg de progesterona injetável e 0,1 mg de BE na colocação da esponja e na retirada 400 UI de eCG e 50 µg de GnRH 48h após); Grupo III (35 mg de progesterona injetável e 0,2 mg de BE na colocação da esponja e na retirada 400 UI de eCG e 50 µg de GnRH 56h após). Foram feitos exames de ultrassom, dosagens de progesterona, e observações do início do estro e da ovulação. A falta do eCG no Grupo I fez com que esse protocolo fosse menos eficaz na indução e sincronização do estro e ovulação. O Grupo Controle teve maior sincronia do estro e da ovulação.(AU)

Ewes were divided in four groups: Control Group (MAP sponges for 12 days, and eCG at withdrawal); Groups I, II and III used the sponge for four days, and 100 µg of PGF was applied at withdrawal; and additionally, Group I (0.1 mg of Estradiol benzoate - EB, in the sponge placement, and in the withdrawn 400 UI of eCG and 50 µg of GnRH 48h later); Group II (35 mg of injectable progesterone and 0.1 mg of EB in the sponge placement, and 400 UI of eCG at withdrawal, and 50 µg of GnRH 48h after); Group III (35 mg of injectable progesterone and 0.2 mg of EB in the sponge placement, and 400 UI of eCG at withdrawal, and 50 µg of GnRH 56h after). ABSTRACT

Os fungos termorresistentes dos gêneros Byssochlamys, Neosartorya e Talaromyces representam grave problema para a indústria processadora de alimentos, especialmente de frutas e hortaliças, em virtude da ocorrência de ascosporos extremamente resistentes e/ou de micotoxinas termo estáveis. Com o propósito de fornecer subsídios quanto ao uso de quantidades mais racionais de conservantes pela indústria de alimentos e, portanto, tornar seu consumo menos danoso à população, o presente estudo determinou in vitro a concentração mínima de metabissulfito de sódio, benzoato de sódio e sorbato de potássio necessária e suficiente para inibir o crescimento micelial e a germinação de ascósporos de B. fulva, N. fischeri e T.flavus em meio de cultivo acidificado (pH 3,5). Foi demonstrado que metabissulfito de sódio em baixas concentrações é tão eficiente na inibição dos fungos mencionados quanto aos demais conservantes em altas concentrações. Por comparação com os procedimentos utilizados pelas indústrias de alimentos, é possível inferir que estas podem reformular o uso de conservantes, considerando-se o tipo de matéria-prima, a temperatura de processamento, a combinação e concentração efetivamente necessária de aditivos, bem como os efeitos tóxicos desses compostos para a saúde humana.
