ABSTRACT

Protoceratium reticulatum is the main yessotoxin-producer along the Chilean coast. Thus far, the yessotoxin levels recorded in this region have not posed a serious threat to human health. However, a bloom of P. reticulatum during the austral summer of 2022 caused the first ban of shellfish collection, due to the high toxin levels. A bloom of P. reticulatum during the austral summer of 2020 allowed an evaluation of the fine-scale distribution of the dinoflagellate during a tidal cycle. High-resolution measurements of biophysical properties were carried out in mid-summer (February 18-19) at a fixed sampling station in Puyuhuapi Fjord, Chilean Patagonia, as part of an intensive 24-h biophysical experiment to monitor the circadian distributions of P. reticulatum vegetative cells and yessotoxins. High P. reticulatum cell densities (>20 × 103 cells L-1) were found in association with a warmer (14.5-15 °C) and estuarine (23.5-24.5 g kg-1) sub-surface water layer (6-8 m). P. reticulatum cell numbers and yessotoxins followed a synchronic distribution pattern consistent with the excursions of the pycnocline. Nevertheless, the surface aggregation of the cells was modulated by the light cycle, suggesting daily vertical migration. The yessotoxin content per P. reticulatum cell ranged from 9.4 to 52.2 pg. This study demonstrates both the value of fine-scale resolution measurements of biophysical properties in a highly stratified system and the potential ecosystem impact of P. reticulatum strains producing high levels of yessotoxins.

ABSTRACT

Fungal alcohol dehydrogenases (ADHs) participate in growth under aerobic or anaerobic conditions, morphogenetic processes, and pathogenesis of diverse fungal genera. These processes are associated with metabolic operation routes related to alcohol, aldehyde, and acid production. The number of ADH enzymes, their metabolic roles, and their functions vary within fungal species. The most studied ADHs are associated with ethanol metabolism, either as fermentative enzymes involved in the production of this alcohol or as oxidative enzymes necessary for the use of ethanol as a carbon source; other enzymes participate in survival under microaerobic conditions. The fast generation of data using genome sequencing provides an excellent opportunity to determine a correlation between the number of ADHs and fungal lifestyle. Therefore, this review aims to summarize the latest knowledge about the importance of ADH enzymes in the physiology and metabolism of fungal cells, as well as their structure, regulation, evolutionary relationships, and biotechnological potential.

ABSTRACT

The aggregation of alpha-synuclein (α-Syn) plays a critical role in the development of Parkinson's disease (PD) and other synucleinopathies. α-Syn, which is encoded by the SNCA gene, is a lysine-rich soluble amphipathic protein normally expressed in neurons. Located in the cytosolic domain, this protein has the ability to remodel itself in plasma membranes, where it assumes an alpha-helix conformation. However, the protein can also adopt another conformation rich in cross-beta sheets, undergoing mutations and post-translational modifications, then leading the protein to an unusual aggregation in the form of Lewy bodies (LB), which are cytoplasmic inclusions constituted predominantly by α-Syn. Pathogenic mechanisms affecting the structural and functional stability of α-Syn - such as endoplasmic reticulum stress, Golgi complex fragmentation, disfunctional protein degradation systems, aberrant interactions with mitochondrial membranes and nuclear DNA, altered cytoskeleton dynamics, disrupted neuronal plasmatic membrane, dysfunctional vesicular transport, and formation of extracellular toxic aggregates - contribute all to the pathogenic progression of PD and synucleinopathies. In this review, we describe the collective knowledge on this topic and provide an update on the critical role of α-Syn aggregates, both at the cellular and molecular levels, in the deregulation of organelles affecting the cellular homeostasis and leading to neuronal cell death in PD and other synucleinopathies.

ABSTRACT

ABSTRACT Acrostichum danaeifolium, a Neotropical fern, occurs preferentially in marshy areas or at the margins of lakes and mangroves. Microlepidoptera larvae burrow through the petioles of the fern, preferentially on the non-expanded leaves. The galleries in the petiole create a new microhabitat, harboring a rich fauna of arthropods. The aim of the present study was to assess the richness of ants associated with its petiole. The study was conducted in a population of A. danaefolium from the Atlantic Forest in Rio de Janeiro state, Southeastern Brazil. Six collections were carried out every two months (2009-2010), three in the dry and three in the rainy season. The leaves were divided into three development stages: non-expanded leaves (NEL), expanded leaves (EL) and senescent leaves (SL). Seven leaves from each phase were randomly collected from seven individuals. A total of fifteen ant species were recorded. The species with the highest frequency and density in fern petioles were Camponotus crassus and Crematogaster curvispinosa. The highest ant richness and abundance was found in senescent leaves. The high number of ants found in the petioles of Acrostichum danaefolium qualifies it as a potential key species in the marshes and flooded areas where it occurs.

ABSTRACT

Magnetic 45S5 bioactive glass (BG) based scaffolds covered with iron-loaded hydroxyapatite (Fe-HA-BG) nanoparticles were obtained and its cytotoxicity investigated. Fe-HA nanoparticles were synthesized by a wet chemical method involving the simultaneous addition of Fe2+/Fe3+ions. BG based scaffolds were prepared by the foam replica procedure and covered with Fe-HA by dip-coating. Fe-HA-BG magnetic saturation values of 0.049 emu g-1and a very low remanent magnetization of 0.01 emu g-1were observed. The mineralization assay in simulated body fluid following Kokubo's protocol indicated that Fe-HA-BG scaffolds exhibited improved hydroxyapatite formation in comparison to uncoated scaffolds at shorter immersion times. The biocompatibility of the materialin vitrowas assessed using human osteoblast-like MG-63 cell cultures and mouse bone marrow-derived stroma cell line ST-2. Overall, the results herein discussed suggest that magnetic Fe-HA coatings seem to enhance the biological performance of 45S5 BG based scaffolds. Thus, this magnetic Fe-HA coated scaffold is an interesting system for bone tissue engineering applications and warrant further investigation.

ABSTRACT

Macroalgae are photosynthetic, multicellular, sessile eukaryotic organisms that offer diverse habitats for the colonization of epiphytic bacteria, therefore establishing biological interactions of diverse complexity. This review focusses on the interactions between macroalgae and their Epiphytic Bacterial Community (EBC); the main aims are to ascertain whether (1) the epiphytic bacterial groups differ at the phylum and genus levels of the macroalgae; (2) the methodologies used so far to study these microorganisms are related in any way to eventual variations of the EBCs on macroalgae; and (3) the EBC of macroalgae has a functional means rather a simple taxonomic grouping. Results showed firstly the taxonomic grouping of macroalgae does not explain the composition and structure of the EBCs. Secondly, the methodology used is important for describing EBCs; and thirdly, multiple bacteria can have the same function and thus to describe the functionality of EBCs it is important to recognize host-specific and generalist bacteria. We recommend the incorporation of a complementary approach between the taxonomic composition and the functional composition analyzes of EBCs, as well as the use of methodological tools that allow analysis of interactions between the EBCs and their hosts, based on the "holobiont" concept.

Populations show spatial-temporal fluctuations in abundance, partly due to random processes and partly due to self-regulatory processes. We evaluated the effects of various external factors on the population numerical abundance of curimba Prochilodus lineatus in the upper Paraná River floodplain, Brazil, over a 19-year period. Panel data analysis was applied to examine the structure of temporal and spatial abundance while controlling auto-regressive processes and spatial non-homogeneity variances that often obscure relationships. As sources of population variation, we considered predation, competition, selected abiotic variables, construction of a dam upstream of the study area, water level and flood intensity during the spawning period. We found that biological interactions (predation and competition) were not significantly related to variations in curimba abundance; specific conductance was a space indicator of abundance, apparently linked to the biology of the species; intensity of floods determined inter-annual variation in abundances; Porto Primavera Dam negatively impacted the abundances at sites in the floodplain directly affected by discharges from the dam. Panel data analysis was a powerful tool that identified the need for intense flooding to maintain high abundances of curimba in the upper Paraná River. We believe our results apply to other species with similar life strategy.(AU)


As populações apresentam flutuações espaço-temporais em abundância devido a processos aleatórios e auto-regulatórios. Nesse trabalho avaliamos efeitos de vários fatores sobre a abundância da curimba, Prochilodus lineatus, em cinco ambientes da planície de inundação do alto rio Paraná, Brasil, durante 19 anos. A análise de dados em painel foi utilizada por examinar variações espaço-temporais na abundância considerando processos auto-regressivos e variâncias espacialmente heterogêneas, que por vezes dificultam a identificação das relações. Como fontes de variação na abundância da curimba foram consideradas: predação, competição, variáveis abióticas selecionadas, construção da barragem de Porto Primavera, nível da água do rio Paraná e intensidade de cheia no período reprodutivo. Foi identificado que predação e competição não foram significativamente associadas às abundâncias da curimba; condutividade específica foi indicadora espacial da abundância; intensidade das cheias foi determinante de variações interanuais na abundância; barragem de Porto Primavera influenciou, negativamente, a abundância em corpos d'água diretamente afetados. A análise de dados em painel mostrou ser uma poderosa ferramenta preditiva, que possibilitou identificar a necessidade de cheias intensas para manutenção de elevadas abundâncias de curimba no alto rio Paraná. The role of a dominant macrobenthic polychaete, Scolelepis squamata, in the colonisation of defaunated tropical sediments by sandy-beach nematodes was investigated and compared with a previous colonisation experiment carried out on a temperate sandy beach. Experimental cylinders, equipped with lateral windows allowing infaunal colonisation, were filled with defaunated sediment containing two treatments, with and without S. squamata. These cylinders were inserted into microcosms containing sediment with indigenous meiofauna collected from the field. The treatments were incubated in the laboratory at ambient temperature and salinity for 7, 14 and 21 days. The nematode assemblages in both treatments did not differ in composition between treatments and from the natural assemblages, suggesting that all the species were equally able to colonise the experimental cores. The presence of the polychaete did not affect the development of the nematode community composition, in contrast to the results from a previous temperate-beach experiment. However, our results did not indicate whether the difference in results was caused by the different behaviour of the polychaete specimens, or by the different composition and response of the present nematode community.


Esse estudo avaliou a influência da espécie Scolelepis squamata (Polychaeta) no processo de colonização de sedimentos defaunados obtidos em uma praia arenosa tropical, além de comparar esses resultados com um estudo similar realizado em uma praia temperada. Sedimentos, previamente defaunados, foram colocados em amostradores com aberturas laterais, que permitiram a colonização da meiofauna. Foram definidos dois tratamentos, um com a presença e outro com a ausência da espécie S. squamata, além do controle. Os amostradores desses tratamentos foram alocados em unidades experimentais do tipo microcosmo, as quais continham sedimento com a meiofauna residente. Os tratamentos foram incubados em laboratório por 7, 14 e 21 dias, com condições controladas de temperatura e salinidade. Os nematódeos não apresentaram diferenças significativas em termos de composição entre os tratamentos e nem em relação ao controle, sugerindo que todas as espécies desse grupo foram, igualmente, capazes de colonizar as unidades experimentais. Esses resultados indicaram que a presença do poliqueto não afetou a estrutura da comunidade de nematódeos, o que representou um resultado contrário ao obtido para a praia temperada. No entanto, não se pode concluir se essas diferenças entre as praias estariam relacionadas ao comportamento diferencial de S. squamata ou pela presença de comunidades de nematódeos distintas nas praias.
