ABSTRACT

OBJECTIVE: Non-alcoholic fatty liver disease (NAFLD) is strongly associated with hyperlipidemia, which is closely related to high levels of sugar and fat. ß-sitosterol is a natural product with significant hypolipidemic and cholesterol-lowering effects. However, the underlying mechanism of its action on aquatic products is not completely understood. METHODS: A high-fat diet (HFD)-induced NAFLD zebrafish model was successfully established, and the anti-hyperlipidemic effect and potential mechanism of ß-sitosterol were studied using oil red O staining, filipin staining, and lipid metabolomics. RESULTS: ß-sitosterol significantly reduced the accumulation of triglyceride, glucose, and cholesterol in the zebrafish model. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed that differential lipid molecules in ß-sitosterol mainly regulated the lipid metabolism and signal transduction function of the zebrafish model. ß-sitosterol mainly affected steroid biosynthesis and steroid hormone biosynthesis in the zebrafish model. Compared with the HFD group, the addition of 500 mg/100 g of ß-sitosterol significantly inhibited the expression of Ppar-γ and Rxr-α in the zebrafish model by at least 50% and 25%, respectively. CONCLUSIONS: ß-sitosterol can reduce lipid accumulation in the zebrafish model of NAFLD by regulating lipid metabolism and signal transduction and inhibiting adipogenesis and lipid storage.

ABSTRACT

In recent years, the potent influence of tocotrienol (T3) on diminishing blood glucose and lipid concentrations in both Mus musculus (rats) and Homo sapiens (humans) has been established. However, the comprehensive exploration of tocotrienol's hypolipidemic impact and the corresponding mechanisms in aquatic species remains inadequate. In this study, we established a zebrafish model of a type 2 diabetes mellitus (T2DM) model through high-fat diet administration to zebrafish. In the T2DM zebrafish, the thickness of ocular vascular walls significantly increased compared to the control group, which was mitigated after treatment with T3. Additionally, our findings demonstrate the regulatory effect of T3 on lipid metabolism, leading to the reduced synthesis and storage of adipose tissue in zebrafish. We validated the expression patterns of genes relevant to these processes using RT-qPCR. In the T2DM model, there was an almost two-fold upregulation in pparγ and cyp7a1 mRNA levels, coupled with a significant downregulation in cpt1a mRNA (p < 0.01) compared to the control group. The ELISA revealed that the protein expression levels of Pparγ and Rxrα exhibited a two-fold elevation in the T2DM group relative to the control. In the T3-treated group, Pparγ and Rxrα protein expression levels consistently exhibited a two-fold decrease compared to the model group. Lipid metabolomics showed that T3 could affect the metabolic pathways of zebrafish lipid regulation, including lipid synthesis and decomposition. We provided experimental evidence that T3 could mitigate lipid accumulation in our zebrafish T2DM model. Elucidating the lipid-lowering effects of T3 could help to minimize the detrimental impacts of overfeeding in aquaculture.

ABSTRACT

This study aims to explore the hypoglycemic effect of lyophilized broccoli microgreens on type 2 diabetes (T2D) in mice. The experiment lasted 18 weeks, including 1 week of adaptation (normal diet) and 17-week experimental period (high-fat diet). After ingestion of broccoli microgreens, the body weight and glucose homeostasis were improved. Meanwhile, the blood lipid status, antioxidant indexes, and inflammatory factors level were improved. Moreover, the insulin resistance and the pathological changes in mice organs were reversed. In addition, the composition of gut microbiota and the production of propionic acid in intestinal content were improved. Our experiment proved that broccoli microgreens have the ability to regulate T2D and improve symptoms of mice T2D induced by high-fat diet and streptozotocin (STZ). PRACTICAL APPLICATIONS: For years, the functionality of broccoli microgreens has attracted much attention. This article will prove the therapeutic effect of broccoli microgreens on T2D and explain its principle of action in the management of T2D.

ABSTRACT

This study was founded for the purpose investigate the differences in effects of combined medication of pioglitazone and melbine and single-use of pioglitazone on the levels of hba1c, blood fat and insulin sensitivity of elder patients with type II diabetes mellitus (T2DM), to provide clinical reference and guidance for the treatment of T2DM in elder patients. For this purpose, we selected a total of 120 elder patients with T2DM who visited the clinic or were admitted to this hospital between July 2016 and July 2017 and divided them into the observation group and the control group (n=60 for each group). For the control group, they only took pioglitazone for treatment, while those in the observation additionally took melbine for treatment. Then, we observed the levels of FPG, 2hPG, HbA1c, blood fat, FINS, 2hINS and HOMA-IR. Results showed that after treatment, significant decreases were seen in levels of FPG, 2hPG and HbA1c in patients of two groups compared to the levels before treatment, and the levels in the observation group decreased more evidently than those in the control group (p<0.05); besides, the levels of total glyceride, total cholesterol and LDL-C were all significantly lower than those before treatment, with an elevated HDL-C, and those levels of TG, TC and LDL-C in the observation group were significantly lower than those in the control group; the level of HDL-C was higher than the control group (p<0.05). Similar decreases were identified in the levels of FINS, 2hFINS and HOMA-IR after treatment (p<0.05), and the levels in the observation group were also significantly lower than those in the control group (p<0.05). In the observation group, the incidence rate of adverse reactions was 10% (6/60), while in the control group was 8.33% (5/60), but the difference between the two groups showed no statistical significance (p>0.05). It is concluded that combined medication of pioglitazone and melbine can effectively reduce the levels of plasma glucose, blood fat and the HOMA-IR, with an elevated sensitivity to insulin, but no severe adverse reactions, manifesting a promising safety in long-term administration. It is worthy of being promoted in clinical practice.

ABSTRACT

The knowledge of frequency and temperature dependent dielectric properties of tissue is essential to develop ultra-wideband diagnostic technologies, such as a non-invasive temperature monitoring system during hyperthermia treatment. To this end, we characterized the dielectric properties of animal liver, muscle, fat and blood in the microwave frequency range from 0.5 GHz to 7 GHz and in the temperature range between 30 °C and 50 °C. The measured data were modeled to a two-pole Cole-Cole model and a second-order polynomial was introduced to fit the Cole-Cole parameters as a function of temperature. The parametric model provides access to the dielectric properties of tissue at any frequency and temperature in the specified range.

ABSTRACT

The present study was designed to analyze variations in blood fat, blood glucose and insulin-resistance in elder patients following surgery for hepatic carcinoma. It also investigated the correlation of insulin with the level of serum leptin and blood fat. A total of 80 patients with primary hepatic cancer who were admitted to The First Hospital of Lanzhou University for treatment between October 2014 and June 2016 were enrolled in the study. At the 1-year follow-up, the patients were divided into two groups based on their recurrence of hepatic cancer after surgery. The levels of serum leptin were detected prior to, one month and one year after surgery; the changes in blood fat, body mass index (BMI), waistline and hipline were measured at one year after surgery; alterations in the fasting blood glucose and blood glucose were measured at 2 h after meal. The fasting insulin (FINS) level and homeostasis model assessment-insulin resistance (HOMA-IR) index were also measured. Correlations between serum leptin and total cholesterol, FINS and fasting blood glucose were analyzed. In the recurrence group, the levels of serum leptin and FINS level were significantly reduced, while waistline and hipline were increased, compared with the non-recurrence group (P<0.05). The BMI and fasting blood glucose in the recurrence group was significantly elevated in comparison with the non-recurrence group (P<0.05). The HOMA-IR index was significantly increased in the recurrence group compared with the non-recurrence group (P<0.05). These results indicated that following surgery for hepatic cancer, the level of serum leptin in patients with recurrence was decreased with an increase in susceptibility to abnormal metabolism of blood fat and glucose. In addition, the serum leptin was negatively correlated with the total cholesterol level and fasting blood glucose and positively correlated with the FINS level in patients. It was concluded that leptin levels decreased in patients with postoperative recurrence, as well as the accumulation of visceral adipose tissue and the development of abnormal blood glucose metabolism was observed.

ABSTRACT

OBJECTIVE: To investigate the levels of blood fat, C-reactive protein (CRP) and hemorheological indicators in the elder patients with coronary heart disease (CHD), so as to provide evidence for prospective study and treatment of elder CHD. METHODS: We collected the clinical data of 127 elder CHD patients who admitted to this hospital between July 2016 and December 2017 to detect the levels of blood fat, CRP and hemorheological indicators. RESULTS: In elder CHD patients, levels of the total cholesterol (TC), triglyceride (TG) and low density lipoprotein cholesterin (LDL-C) were significantly higher than the normal reference, and comparison with the control group also showed significant increases (p < 0.01); average levels of the high-density lipoprotein cholesterin (HDL-C), phospholipid (PL), lipoprotein a [LP (a)] and free fatty acid were in the range of normal reference. Abnormal levels of TC, TG, LDL-C and HDL-C were identified in 59.06%, 58.27%, 51.18% and 18.11% of the elder CHD patients, most of which were concomitant with obesity or hypertension, and levels of these indicators were significantly higher than those in the control group with statistically significant differences (p < 0.01). Comparisons of the age, gender distribution, hypotension, exercise and sleep showed that differences had no statistical significance (p > 0.05). In comparison with the control group, the levels of CRP, the whole blood viscosities at high and low shears, plasma viscosity, hematocrit value, aggregation index and rigidity index of red blood cells (RBC) were all higher than those in the control group, and the differences had statistical significance (p < 0.01). However, the erythrocyte sedimentation rate (ESR), deformity index of RBC, blood flow rates in the bilateral middle cerebral arteries (MCA), anterior cerebral arteries (ACA), terminal internal carotid artery (TICA), posterior cerebral arteries (PCA), vertebral arteries (VA) and basilar artery (BA) were significantly lower than those in the control group, and the differences had statistical significance (p < 0.05 or 0.01). CONCLUSION: In elder CHD patients, anomaly is mainly seen in levels of TC, TG and LDL-C with concentrated, adhesive and aggregating blood.

ABSTRACT

The purpose of this study is to verify the effects of aquatic exercise on the health-related physical fitness, blood fat, and immune functions of children with disabilities. To achieve the aforementioned purpose, the researchers studied 10 children with grade 1 or grade 2 disabilities who do not exercise regularly. The researchers used SPSS 21.0 to calculate the averages and standard deviations of the data and performed a paired t-test to verify the differences in averages before and after an exercise. The study showed significant differences in lean body weight, muscular strength, cardiovascular endurance, flexibility, and muscular endurance. The researchers found statistically significant differences in triglyceride as well as in immunoglobulin G. The findings suggest that aquatic exercise affects the health-related physical fitness, blood fat, and immune functions of children with disabilities.

ABSTRACT

Diabetic nephropathy (DN) is currently a major public health problem worldwide. The objective of the present study was to evaluate the clinical effect of breviscapine injections in patients with DN. A meta-analysis was performed using the following databases to obtain published reports in any language: PubMed/MEDLINE, Embase, China National Knowledge Infrastructure, Chinese Evidence-Based Medicine, Wanfang Digital Periodicals, Chinese Academic Journals Full-text Database, Chinese Biological and Medical Database, China Doctoral and Masters Dissertations Full-text Database and the Chinese Proceedings of Conference Full-text Database. Two assessors independently reviewed each trial. A total of 35 randomized controlled trials, which performed studies on a total of 2,320 patients (1,188 in treatment groups and 1,132 in control groups), were included in the present meta-analysis. Data were analyzed using Stata version 11.0 for Windows. The results from the analysis demonstrated that breviscapine injections have greater therapeutic effects in patients with DN in comparison with the control group, including renal protective effects (reducing urine protein, serum creatinine and blood urea nitrogen) and adjustment for dyslipidemia (affecting levels of cholesterol, triglycerides and high density lipoproteins). These effects indicate that breviscapine injections are beneficial to patients with DN. Further studies are required to determine the mechanisms underlying the therapeutic effects of breviscapine.

Methods: Totally 80 cases of outpatients with hypertension complicated with CHD were randomly divided into the observation group (40 cases) and the control group (40 cases). The control group was given amlodipine 10 mg,po,qn, and the observation group was given amlodipine and atorvastatin tablets 20 mg,po,gn, and the treatment course was 6 weeks. The changes of blood pressure and blood lipid in the two groups before and after the treatment were observed, and the clinical curative effect and adverse reactions were also studied. Results:Compared with those before the treatment, systolic blood pressure, diastolic blood pressure and blood fat in the two groups were all improved after the treatment, and the difference was statisti-cally significant (P0. 05). Conclusion: Amlo-dipine atorvastatin tablets can significantly improve blood pressure and blood lipid in the patients with hypertension complicated with CHD with promising clinical curative effect and few side effects, which is worthy of wide use in clinics.

ABSTRACT

Objective:To discuss the curative effect of atorvastatin intensive treatment on lipid-lowering in the patients with type 2 diabetes and its influence on vascular endothelial function. Methods: Totally 74 cases of patients with type 2 diabetes were selected and divided into the intensive group ( n=37 cases) and the ordinary group ( n=37 cases) at random. The patients in the two groups were given routine medical treatment, such like diet adjustment, moderate exercise and drug control etc, and the patients in the inten-sive group were additionally given 40 mg atorvastatin orally, once a day, while the patients in the ordinary group were additionally given 20mg atorvastatin orally, once a day for 12 weeks. The changes of blood fat, von willebrand factor (VWF) of serum vascular endothe-lial function, plasma thrombomodulin ( PTM) and vascular endothelial cadherin ( VEC) levels of the patients before the treatment and in the 12th week after the medical treatment were observed and compared, and the adverse drug reactions ( ADR) were evaluated as well. Results:After the 12-week medical treatment, TC, TG and LDL-C levels of the patients in the two groups were obviously de-clined than those before the treatment, while HDL-C levels were obviously raised than those before the treatment ( P0. 05). Conclusion:Compared with atorvastatin, atorvastatin intensive treatment in the patients with type 2 diabetes has more favorable lipid-lowering effect, which can effectively reduce serum vWF, PTM and VEC levels to improve and rem-edy vascular endothelial function with high security.
