ABSTRACT

Objective To explore the expression and correlation of Fascin-1 and epidermal growth factor receptor (EGFR) in hormone receptor-positive breast cancer.Methods The immunohistochemical technique,EnVision method,was used to evaluate the expression of Fascin-1 and EGFR in 294 cases of hormone receptor-positive breast cancer,which contains 290 cases of estrogen receptor (ER) positive and 244 cases of progestrone receptor (PR) positive.According to ER,PR,Epidermal growth factor receptor 2 (HER2),and Ki-67 status,all cases of hormone receptor-positive breast cancer were categorized into 2 subtypes:160 cases of luminal A and 134 cases of luminal B.Results Fascin-1 and EGFR protein positive rates in hormone receptor-positive breast cancer was 13.9% (41/294) and 30.6% (90/294),respectively.Fascin-1 positive rate was significantly higher in EGFR positive cases (30.0%,27/90) than in EGFR negative cases (6.9%,14/204) (x2 =27.857,P =0.000).In the ER positive and PR positive cases,Fascin-1 positive rates were both significantly higher in EGFR positive cases than in EGFR negative cases (x2 =29.23,P =0.000;x2 =27.596,P =0.000,respectively).In the Luminal A and Luminal B subtype,Fascin-1 positive rates were also both significantly higher in EGFR positive cases than in EGFR negative cases (x2 =23.247,P=0.000;x2 =5.325,P=0.021,respectively).Conclusions EGFR signal pathway may positive regulate Fascin-1 expression in hormone receptor-positive breast cancer.

ABSTRACT

Objective To research the expression change and the sense of histone acetylation and p21WAF1 protein in breast cancer. Methods Pathological morphologic changes of breast cancer were identified by H. E. staining. Immunohistochemical study for p21WAF1 protein expression was performed on 80 breast cancers and 80 normal breast tissues. The expressions of acetylated histone H3, H4 and p21WAF1 protein in 80 breast cancers and 80 normal breast tissues were analyzed by Western blot. Results H.E. staining discovered that the tissue structure and cell morphous of breast cancer had visible atypia, compared with normal breast tissues. The immunohistochemical results displayed that the positive expression of p21 WAF1 in 80 breast cancers and 80 normal breast tissues was 49 cases(61.25 %) or 3 cases (3.75%), respectively. There was significantly difference in the expression level between the 80 breast cancers and 80 normal breast tissues (P < 0. 05). The expression levels of p21WAF1 protein in breast cancer tissues were higher than that in normal breast tissues, 0. 78 ± 0. 095 or 0. 65 ± 0. 055, respectively. The expression levels of acetylated histone H3, H4 protein in normal breast tissues were higher than those in breast cancer tissues. H3 was 2. 35 ± 0. 340 or 1.07 ± 0. 067, respectively, H4 was 3.44 ± 0. 202 or 1.11 ± 0. 086, respectively. There was significantly difference in the expression levels between the 80 breast cancers and 80 normal breast tissues (P <0. 01). Conclusions The occurrence and development of breast cancers may be related to the expression change of histone acetylation and p21WAF1 protein.

ABSTRACT

Objective To study the expression of VEGF, MMP-2 and MMP-9 in benign and malignant lesions of breast and their effects on breast carcinoma's invasiveness and metastasis. Methods VEGF,MMP-2 and MMP-9 expression was detected by SP method in 20 breast fibroadenoma and 12 normal breast specimens taken from non-cancerous regions adjacent to breast cancer tissue and 38 breast carcinomas. Results The expression rate of VEGF, MMP-2 and MMP-9 in breast carcinoma was remarkably higher than those in normal breast tissue and breast fibrogdenoma (63.2% vs 25.0% and 30. 0% ,78. 95% vs 33.3% and 35.0% and 71.1% vs 41.7% and 45.0% ,respectively; P <0.05). There was a correlation between VEGF, MMP-2 and MMP-9 expression and nodal metastasis ( P <0.05). High expression of VEGF, MMP-2 and MMP-9 was correlated with pathological grade ( P < 0.05, respectively). The overexpression of VEGF was related with those of MMP-2 and MMP-9 in breast cancer( VEGF and MMP-2:rs =0.541, P <0.01 ;VEGF and M M P-9: rs = 0.11, P < 0.01; MMP-2and M MP-9: rs= 0P < 0.01 ). Conclusion VEGF,MMP-2 and MMP-9 overexpression is strongly related to the invasiveness and metastasis of breast carcinoma, which might be valuable in evaluating tumor invasion and metastasis.

ABSTRACT

Objective To investigate the'expression of Survivin in breast cancer tissue microarray, and its implication to clinic pathology and prognosis. Methods The expression of Survivin was studied in 80 cases of breast disease tissues ( 18 cases of hyperplasia of mammary glands and 62 cases of breast canc-er) and 20 cases of adjacent normal tissues by S-P immunohistochemisty to analysis the relationships be-tween its expression and history grade, clinic stage and lymphaden metastasis. Results The positive rates of Survivin were 0(0/20) in normal mammary glands, 33. 3% (6/18) in hyperplasia of mammary glands and 72. 5% (45/62) in breast cancer. There were significant difference between them (X~2= 34. 62, P <0. 01 ). The expression of Survivin was closely correlated to the stage and lymphaden metatasis and survival time ( P <0.05 or P <0.01), and it was negatively correlated to the histological grade ( P >0.05).Conclusions The expression of Survivin shows an ascending tendency in the carcinogenesis and progres-sion of breast cancer. The Survivin can promote the invasion and the metastasis of the cancer cell, and it is closely correlated to the prognosis.
