ABSTRACT

In the Valpolicella area (Verona, Italy) Vitis vinifera cv. Corvina is the main grape variety used to produce Amarone wine. Before starting the winemaking process, the Corvina grapes are stored in a withering (i.e., dehydrating) warehouse until about 30% of the berry weight is lost (WL). This practice is performed to concentrate the metabolites in the berry and enrich the Amarone wine in aroma and antioxidant compounds. In compliance with the guidelines and strict Amarone protocol set by the Consorzio of Amarone Valpolicella, withering must be carried out by setting the grapes in a suitable environment, either under controlled relative air humidity (RH) conditions and wind speed (WS)-no temperature modification is to be applied-or, following the traditional methods, in non-controlled environmental conditions. In general, the two processes have different dehydration kinetics due to the different conditions in terms of temperature, RH, and WS, which affect the accumulation of sugars and organic acids and the biosynthesis of secondary metabolites such as stilbenes and glycoside aroma precursors. For this study, the two grape-withering processes were carried out under controlled (C) and non-controlled (NC) conditions, and the final compositions of the Corvina dried grapes were compared also to evaluate the effects on the organoleptic characteristics of Amarone wine. The findings highlighted differences between the two processes mainly in terms of the secondary metabolites of the dried grapes, which affect the organoleptic characteristics of Amarone wine. Indeed, by the sensory evaluation, wines produced by adopting the NC process were found more harmonious, elegant, and balanced. Finally, we can state how using a traditional system, grapes were characterised by higher levels of VOCs (volatile compounds), whilst wines had a higher and appreciable complexity and finesse.

ABSTRACT

Collagens extracted from different tissues and fish species display different physicochemical properties, thus novel sources require characterization. Gulf corvina (Cynoscion othonopterus) is processed industrially for food. Of the by-products, the swim bladder is used for fish maw, but other tissues are treated as waste. In the present study, pepsin-soluble collagen from Gulf corvina skin and swim bladder was extracted and characterized. Skin produced a higher collagen yield (82 ± 1.53 %) than swim bladder (69 ± 1.60 %). Both collagens exhibited electrophoresis bands corresponding to ([α1(I)]2α2(I)) and ß chains, all characteristic of type I collagen. Spectra analysis showed the collagens to maintain their triple-helix structure. The skin collagen had a lower denaturation temperature (29.8 °C) than the swim bladder collagen (32.5 °C), due to its relatively low imino acid content (168 vs. 190 /1000 residues, respectively). Both collagens were highly soluble in acidic pH ranges; Zeta potential values were 5.5 for the skin collagen and 6.2 for the swim bladder collagen. Gulf corvina skin and swim bladder are excellent sources of type I collagen with similar physicochemical properties.

ABSTRACT

The present study has been conducted to find out new findings on maximum length and weight values of Umbrina cirrosa in the Bay of Saros (Northern Aegean Sea, Turkey). On September, 11, 2016, a single specimen of Umbrina cirrosa with 68.8 cm total length and 2600.00 g total weight was caught by handline at a depth of 20 m. The provable size is the second largest length ever reported in the all seas of the world.


O presente estudo foi realizado para descobrir novas descobertas sobre os valores máximos de comprimento e peso de Umbrina cirrosa na Baía de Saros (Mar Egeu do Norte, Turquia). Em 11 de setembro de 2016, um único espécime de Umbrina cirrosa com 68.8 cm de comprimento total e 2600.00 g de peso total foi capturado por linha de mão a uma profundidade de 20 m. O tamanho provável é o segundo maior comprimento já registrado em todos os mares do mundo.

ABSTRACT

Grapevine represents a particularly interesting species as concerns phenotypic plasticity, considering that the terroir, meaning the contribution of the geography, geology, and climate of a certain place, together with the agronomical practices utilized, may deeply influence the berry phenotype at the physiological, molecular, and biochemical levels. This phenomenon leads to the production of wines that, although produced from the same variety, exhibit different enological profiles and represents an issue of increasing interest from both a biological and an economic point of view. The main objective of the present study was to deepen the understanding of phenotypic plasticity in grapevine, trying to dissect the role of one its important components - the soil - by investigating the singular effect that different physico-chemical soil properties can produce in terms of berry plasticity at the phenological, physiological, and biochemical levels in a red and a white variety of great economic importance in Italy and overseas: Corvina and Glera. The results indicated a genotype-dependent response to the soil factor, with higher biochemical plasticity in Corvina with respect to Glera and suggested a key role of specific soil properties, including the skeleton, texture, and mineral composition, on the metabolite profile of berry skin.

ABSTRACT

BACKGROUND: A neutral, heat-sensitive serine protease (NHSSP) originating from the feather-degrading fungus Onygena corvina (O. corvina) was described and defined as an alkaline serine protease of the subtilisin type S8 family, exhibiting an enzymatic activity at neutral pH. Generally, broad specificity proteases, such as proteinase K or trypsin, have found numerous applications in research and biotechnology. RESULTS: We report the cloning and expression in the yeast PichiaPink™ system, as well as purification, and characterization of the NHSSP. Recombinant, His6-tagged NHSSP was efficiently expressed from an optimized, synthetic gene and purified using a simple protocol based on ammonium sulfate fractionation and hydrophobic interaction chromatography. The enzyme shows atypical C-terminal processing, the coded preproprotein undergoes signal peptide removal and maturation through the clipping of a propeptide section and 10 amino acids (aa) from the C-terminus, including the His6-tag. The deletion variant has been constructed, devoid of the C-terminal ORF segment, thus eliminating the need for C-terminal processing. Both NHSSP variants exhibit very similar enzymatic characteristics. The purified enzymes were characterized to determine the optimal proteolytic conditions. We revealed that the mature NHSSP is reproducibly active over a wide pH range from neutral to mild acidic (pH of 5.0 to 8.5), with an optimum at pH 6.8, and at temperatures of 15 to 50 °C with an optimum at 38-42 °C. Interestingly, we demonstrated that the protease can be fully deactivated by a moderate increase in temperature of about 15 °C from the optimum to over 50 °C. The protease was partially sensitive to serine protease inhibitors, and not inhibited by chelating or reducing agents and detergents. SDS induced autolysis of NHSSP, which points to a high stimulation of its proteolytic activity. CONCLUSIONS: The NHSSP was produced as a recombinant protein with high efficiency. Compared to proteinase K, the most common serine protease used, NHSSP shows an approx. twofold higher specific activity. Protein sequencing can be a valuable technical application for the protease. The protein coverage is significantly higher in comparison to trypsin and reaches about 84-100% for ß-lactoglobulin (BLG), antibody (mAb) light and heavy chains. Furthermore, the option to perform digestions at neutral to slightly acidic pH-values down to pH 5.0 avoids modification of peptides, e.g. due to deamidation.

ABSTRACT

Withering is a practice traditionally used in various regions to produce sweet or dry wines. During withering there is an increase in sugar content but also a modification in volatile compound profiles. Controlling metabolic changes through the dehydration process to obtain wines with desired characteristics is therefore a challenging opportunity. The effects of two different withering technologies, post-harvest or on-vine with blocked sap vessel flow, on the volatile profile of young and aged Corvina red wines was investigated. The results showed that modulation of wine aroma due to the withering process is associated with fermentative metabolites, such as esters, higher alcohols, and acids, as well as grape-related compounds such as C6 alcohols, terpenes and norisoprenoids. Significant differences were also found by comparing the two withering techniques. Post-harvest in a traditional "fruttaio" warehouse wines showed higher content of ethyl acetate, ethyl butanoate, ß-citronellol and 3-oxo-α-ionol, whereas post-harvest withering on-vine increased ß-damascenone in wines. The type of withering technique has an influence on the evolution of some aroma compounds during the aging of wine, among them linalool, (E)-1-(2,3,6-trimethylphenyl)buta-1,3-diene (TPB), n-hexyl acetate, ethyl acetate, ethyl 3-methylbutanoate, 3-oxo-α-ionol and ß-damascenone.

BACKGROUND: During the production of red wine, moderate uptake of oxygen in the post-fermentative phase helps the stabilization of colour and the decrease of astringent tannins. However, the influence of oxygen uptake during the fermentative phase in must has not been completely investigated. In this study we evaluated the effect of controlled oxygen supply during fermentation-maceration of Corvina grapes on colour characteristics, tannins, volatile compounds, acetaldehyde production and oxidative stability of the finished wine. RESULTS: Oxygen supply during fermentation improved the formations of stable pigments of Corvina wines due to the higher production of acetaldehyde. However, in wines treated with oxygen a lower production of fruity esters by yeasts was observed. Wines obtained from higher oxygen exposure during fermentation-maceration showed reduced ability to react with oxygen during storage. CONCLUSIONS: Fermentative and post-fermentative oxygenation should be considered as a technological approach for modifying colour composition and stability, as well as oxidative behaviour of wine during aging. © 2020 Society of Chemical Industry.

ABSTRACT

Estimating the growth of fishes is critical to understanding their life history and conducting fisheries assessments. It is imperative to sufficiently sample each size and age class of fishes to construct models that accurately reflect biological growth patterns, but this may be a challenging endeavor for highly-exploited species in which older fish are rare. Here, we use the Gulf Corvina (Cynoscion othonopterus), a vulnerable marine fish that has been persistently overfished for two decades, as a model species to compare the performance of several growth models. We fit the von Bertalanffy, Gompertz, logistic, Schnute, and Schnute-Richards growth models to length-at-age data by nonlinear least squares regression and used simple indicators to reveal biased data and ensure our results were biologically feasible. We then explored the consequences of selecting a biased growth model with a per-recruit model that estimated female spawning-stock-biomass-per-recruit and yield-per-recruit. Based on statistics alone, we found that the Schnute-Richards model described our data best. However, it was evident that our data were biased by a bimodal distribution of samples and underrepresentation of large, old individuals, and we found the Schnute-Richards model output to be biologically implausible. By simulating an equal distribution of samples across all age classes, we found that sample distribution distinctly influenced model output for all growth models tested. Consequently, we determined that the growth pattern of the Gulf Corvina was best described by the von Bertalanffy growth model, which was the most robust to biased data, comparable across studies, and statistically comparable to the Schnute-Richards model. Growth model selection had important consequences for assessment, as the per-recruit model employing the Schnute-Richards model fit to raw data predicted the stock to be in a much healthier state than per-recruit models employing other growth models. Our results serve as a reminder of the importance of complete sampling of all size and age classes when possible and transparent identification of biased data when complete sampling is not possible.

ABSTRACT

The genetic relationships of native or introduced Plagioscion squamosissimus in five Brazilian Neotropical basins were evaluated using the mitochondrial atpase6/8 genes. Results revealed that the population of the Tocantins River basin is more basal than the native populations of the Amazon and Parnaíba River basins. Moreover, the populations of P. squamosissimus that were introduced in the São Francisco and upper Paraná River basins originated from the population of the Parnaíba River.

Abstract This study focused on the characterization and analysis of communities and infra-communities of metazoan parasites of Plagioscion squamosissimus caught in Promissão Reservoir in the Tietê River in Borborema (21°39′58"S, 49°8′49"W), state of São Paulo, Brazil. Fifty adult specimens caught by professional fishermen in March 2015 were necropsied. The fish presented an average standard length of 25.2 ± 2.2 cm and average weight of 328.82 ± 89.03 g. A total of 5,227 specimens of metazoan parasites were collected: 2,880 (55.1%) adult Diplectanum piscinarius (Monogenoidea: Diplectanidae) and 2,347 (44.9%) Austrodiplostomum compactum metacercariae (Digenea, Diplostomidae), both with 100% prevalence and mean abundance of 57.6 and 46.9, respectively. Parasite diversity was low (species richness = 2), with a Simpson index (D) equal to 0.505, and low values of Brillouin (HB = 0.687) and Margalef diversity (I = 0.117) indices. Berger-Parker's index of dominance (p = 0.551) indicated a slight dominance of the monogenean parasite D. piscinarius. There was a positive correlation, assessed by Pearson coefficient between parasite abundance of D. piscinarius and standard length (r = 0.43) and weight (r = 0.51) of hosts.


Resumo O objetivo deste trabalho foi caracterizar e analisar as comunidades e infracomunidades de metazoários parasitos de corvinas capturadas no Reservatório de Promissão, Rio Tietê, município de Borborema (21° 67′S, 49° 14′O), Estado de São Paulo. Foram examinados 50 espécimes, capturados por pescadores profissionais no mês de março de 2015, e os parasitas coletados foram quantificados, preparados e montados para identificação taxonômica e análise das comunidades de parasitos. Os peixes analisados no estudo apresentaram comprimento padrão médio de 25,2 ± 2,2 cm e peso médio de 328,82 ± 89,03 g. Foram coletados 5227 espécimes de parasitas metazoários, sendo 2880 (55,1%) Diplectanum piscinarius (Monogenoidea: Diplectanidae) e 2347 (44,9%) metacercárias de Austrodiplostomum compactum (Digenea, Diplostomidae), ambos com prevalência de 100% e abundância parasitária de 57,6 e 46,9, respectivamente. Foi encontrada baixa diversidade parasitária (riqueza de espécies=2), com índice de Simpson (D) igual a 0,505 e baixos valores dos índices de Shannon (H'=0,688) e de diversidade de Margalef (I=0,177). O índice de dominância de Berger-Parker (d=0,551) indicou uma leve dominância do monogenético D. piscinarius. Houve correlação positiva intermediária, avaliada pelo coeficiente de Pearson, entre a abundância parasitária de D. piscinarius e comprimento padrão (r=0,43) e peso (r=0,51) dos hospedeiros.

ABSTRACT Austrodiplostomum compactum is a digenetic trematode whith metacercariae that occur in the eyes of a wide variety of fish species. In Brazil, A. compactum metacercariae have been reported in many fish species, but there are only a few studies in the Amazon region. Due to the lack of information in the Amazon, the aim of this study was to report the occurrence of A. compactum metacercariae in the eyes of 15 specimens of Plagioscion squamosissimus from the Lake Catalão, in Amazonas, Brazil. Fishes were collected using gill nets placed randomly in the lake, euthanized in a field laboratory, and had their eyes dissected and examined under a stereomicroscope. A total of 801 metacercariae were registered infecting the eyes of the hosts with a parasitic prevalence of 100%. There was a significant positive correlation between the number of metacercariae recorded and the standard body length of fishes. No significant correlation was found between the number of metacercariae and the weight and relative condition factor (Kn) of the hosts. Values of the variance-to-mean ratio and Green's index suggested an aggregate distribution of the parasites in the hosts, with a low degree of aggregation. This is the first report of occurrence of A. compactum parasitizing a fish in the Lake Catalão.


RESUMO Austrodiplostomum compactum é um trematódeo digenético cuja metacercária ocorre nos olhos de uma grande variedade de espécies de peixes. No Brasil, metacercárias deste parasita foram relatadas em diversas espécies de peixes, mas existem poucos estudos na região amazônica. Devido à falta de informação na região amazônica, o objetivo deste estudo foi relatar a ocorrência de metacercárias de A. compactum nos olhos de 15 espécimes de Plagioscion squamosissimus provenientes do Lago Catalão, Amazonas, Brasil. Os peixes foram coletados usando redes de pesca distribuídas aleatoriamente no lago e sacrificados em laboratório de campo, onde os olhos foram removidos e examinados sob microscópio estereoscópico. Um total de 801 metacercárias foram registradas infectando os olhos do hospedeiro, com uma prevalência parasitária de 100%. Houve uma correlação positiva significativa entre o número de metacercárias e o comprimento padrão dos peixes. Nenhuma correlação significativa entre o número de metacercárias e o peso e o fator de condição relativa (Kn) dos hospedeiros foi evidenciada. Valores do índice de dispersão e do índice de Green sugerem uma distribuição agregada dos parasitas com um baixo grau de agregação nos hospedeiros. Este é o primeiro relato da ocorrência de A. compactum parasitando um peixe no Lago Catalão.

The composition and changes of the fungal population and of the metabolites present in grapes and in ferments of Vitis vinifera L. cv. Corvina, one of the major components of the Amarone musts, were dissected aiming at the identification of constant characteristics possibly influenced by the productive process. The fungal populations and metabolomic profiles were analyzed in three different vintages. 454-pyrosequencing on the ribosomal ITS1 region has been used to identify the fungal population present in Corvina grapes and fresh must. Samples were also subjected to metabolomics analysis measuring both free volatile compounds and glycosylated aroma precursors through an untargeted approach with comprehensive two-dimensional gas chromatography time-of-flight mass spectrometry. Albeit strongly dependent on the climate, both the mycobiota and metabolome of Corvina grapes and fresh musts show some characteristics recursive in different vintages. Such persistent characteristics are likely determined by the method adopted to produce Amarone or other dry wines made from partially dried grapes. In particular, the harsh conditions imposed by the prolonged withering appear to contribute to the shaping of the fungal populations. The fungal genera and metabolites present in different vintages in V. vinifera L. cv. Corvina grapes and fresh musts represent core components of the peculiar technique of production of Amarone. Their identification allows the in-depth understanding and improved control of the process of production of this economically and culturally relevant wine.

ABSTRACT

ResumenLas corvinas son especies de interés comercial que han sido sobreexplotadas en el Golfo de Nicoya, Costa Rica. El objetivo de este estudio fue describir por primera vez, el desove espontáneo y la ontogenia de las larvas de la corvina aguada Cynoscion squamipinnis en cautiverio, con el fin de realizar repoblamiento y proyectos de maricultura. Reproductores silvestres (n= 6, 1-2 Kg), fueron capturados y mantenidos en la Estación de Biología Marina Juan Bertoglia Richards (Puntarenas, Costa Rica) por un periodo de dos años (octubre 2006-diciembre 2008). Durante este periodo, el estado de madurez de las hembras (n= 3) fue registrado mediante muestras de cánula y los machos (n= 3) mediante masaje abdominal. Todos los reproductores fueron colocados juntos en un tanque de 18 t, con aireación, a una salinidad entre 33-35 ups y expuestos a una temperatura constante (29 ± 1 °C). De enero a marzo 2009 ocurrió un periodo de desove espontáneo, obteniéndose 162 000 huevos en tres desoves. El porcentaje de fertilización fue de 50-60 %, y la supervivencia después de la eclosión fue entre 60-85 %. El diámetro de los huevos fue de 0.852 mm (Desviación Estándar= 0.039), con una gota de aceite de 0.269 mm (DE= 0.016). En el desarrollo embrionario, la primera división mitótica ocurrió una hora después del desove (hdd), la segunda división 1:30 hdd, la tercera división 2 hdd, la cuarta división a las 2:30 hdd, y la quinta 3:00 hdd. La mórula fue observada a las 3:30 hdd, la blástula a las 4:30 hdd, la gástrula a las 8:30 hdd, la forma C a las 10:00 hdd, y la forma S ocurrió de las 10-19 hdd. La eclosión ocurrió 19 hdd. La larva midió 2.234 mm (DE= 0.122) de longitud total (LT), y 2.179 mm (DE= 0.119) de longitud notocordial (LN). La preflexión inició 49 hdd, la flexión 11 días después del desove (ddd) (3.767 mm LT, DE= 0.209), y la postflexión a los 14 ddd (4.015 mm LT, DE= 0.302). A los 45 ddd, los juveniles pesaron 3.68 g (DE= 1.09). El tiempo de eclosión de las larvas de la corvina aguada fue menor al de otras especies de corvina. Los tiempos de formación en las etapas embrionarias difirieron poco con respecto a los tiempos observados para otras especies de corvinas. Las diferencias observadas con respecto a otras especies, probablemente responden a las características genéticas propias de cada especie y a la temperatura de incubación de los huevos. La obtención de desoves espontáneos sin aplicaciones hormonales a los reproductores, y la producción de juveniles en cautiverio demostraron que la corvina aguada puede ser considerada en programas de repoblamiento y proyectos de maricultura.


Abstract:The croakers or drums are commercial species, which have been overfished in the Nicoya Gulf, Costa Rica. This study aimed to describe, for the first time, the reproduction and the ontogeny of weakfish, Cynoscion squamipinnis in captivity, in order to perform restocking and mariculture projects. Wild fish (n= 6, 1-2 Kg) were captured and maintained in the Estación de Biología Marina Juan Bertoglia Richards (Puntarenas, Costa Rica) for a two years period (October 2006- December 2008). During this period, maturation stage was monitored periodically by cannula samples in the females (n= 3) and gentle massage in males (n= 3). All fish were stocked in an 18 t tank, with aeration, 33-35 ups of salinity, and a constant temperature (29 ± 1 °C). The spawning period occurred from January to March 2009, producing 162 000 eggs in three spontaneous spawns. The fertilization percentage was 50-60%, and survival after hatching was 60-85%. The egg diameter was 0.852 mm (Standard deviation (SD)= 0.039), and oil drop of 0.269 mm (SD= 0.016). In the embryonary development, the first mitotic division (MD) was observed one hour after spawning (has), the second MD was 1:30 has, the third MD was 2:00 has, the fourth MD was 2:30 has, and fifth MD at 3:00 has. Morule was observed 3:30 has, the blastule 4:30 has, the gastrule 8:30 has, C shape at 10:00 has, and C shape at 12:00 has. After 19 has hatching larvae occurred. The total length (TL) of the larvae was 2.234 mm (SD= 0.122), and the nothochordial length (NL) was 2.179 mm (SD= 0.119). Preflexion stage was observed 49 has, flexion stage was 11 days after spawn (das) (3.767 mm LT (SD= 0.209)), and postflexion stage was 14 das (4.015 mm LT (SD= 0.302)). After 45 das, the juvenile weights 3.68 g (SD= 1.09). Hatch time of the weakfish larvae was minor than of others croaker species. The stages times of embrionary development were a little different from others croaker species, and probably respond to genetic characteristics of each species and the eggs incubation temperature. The spontaneously spawning without broodstock hormonal applications, and the juveniles production in captivity showed that weakfish is a potential species for restocking programs and mariculture projects. ABSTRACT

This study represents the first investigation on ecology of endophytic bacteria isolated from 3 and 15 year-old vine stems of Vitis vinifera cv. Corvina. The analysis was performed by means of culture-dependent techniques. The obtained results showed that new grapevine endophytic genera are being discovered. Moreover, Bacilli and Actinobacteria are frequently isolated from 3 year-old plants, whereas Alpha- and Gamma- Proteobacteria classes are more prevalent in the 15 year-old plants. Shannon-Wiener (H) index and analysis of rarefaction curves revealed greater genus richness in young grapevine plants. Furthermore, results evidenced an increase of genotypic group number within specific genera (e.g., Rhizobium and Pantoea). Among isolated strains from 3 and 15 year-old stems, respectively, 34 and 39% produce siderophores; 22 and 15% secrete ammonia; 22 and 21% produce indole-3-acetic acid; 8.7 and 41% solubilize phosphate. Besides, two strains isolated from 15 year-old grapevines showed 1-aminocyclopropane-1-carboxylate deaminase activity. Antifungal activity analysis evidenced that two Bacillus strains possess growth antagonistic effect toward all the tested fungal strains. Therefore, the present study extends our knowledge of the diversity of the endophytic bacteria by providing new insights into the complexity of the grapevine microbiome.

ABSTRACT

The early life history stages are inadequately known for most fishes of the Neotropical region. Thus, larvae and juveniles of the species Pachyurus bonariensis and Plagioscion ternetzi, two corvina species found in the Pantanal of Mato Grosso, Brazil, were described, assessing ontogenetic changes in their external morphology, pigmentation, fin development, morphometry and meristics. Fish were collected in Chacororé Bay, on the Cuiabá River, between March 2000 and March 2004. Eighty individuals of each species were analyzed, comprising 60 larvae and 20 juveniles. Pachyurus bonariensis larvae exhibited a mouth in a terminal position that became sub-terminal; large, well-pigmented, spherical eyes that became small and elliptical in postflexion; preopercular spines (two internal and four external); 23 to 27 myomeres; initially sparse pigmentation that intensified, mainly in the ventral region; and the following fin formation sequence and total number of spines and rays: caudal, dorsal (XI+29-32), anal (II+six), pelvic (I+five) and pectoral (15-17). In contrast, the larvae of P. ternetzi exhibited a terminal mouth; large, well-pigmented, spherical eyes that decreased in size during development; preopercular spines (three internal and four external); 23 to 26 myomeres; pigmentation that was initially sparse, became evident only in late postflexion stage, with the presence of some chromatophores on the top of the head; and the following fin formation sequence and total number of spines and rays: caudal, dorsal (XI+30-36), anal (II+six), pelvic (I+five) and pectoral (16-18). In relationship the morphometric variables, only the snout-anal fin length differed between the two species, being initially larger in P. ternetzi, whereas it only became larger in juvenile P. bonariensis after 34 mm. Despite the difficulty of intraspecific identification among fish larvae collected in natural environments, the morphological and morphometric tools used in the present study were effective in separating the early stages of development of the two morphologically similar species that share the same environment for reproduction.


Las primeras fases del ciclo de vida no son conocidas lo suficiente para la mayoría de los peces de la región Neotropical. Así la descripción de larvas y juveniles de Pachyurus bonariensis y Plagioscion ternetzi, dos especies de corvinas que se encuentran en el Pantanal matogrossense, Brasil, fue realizada a través de cambios ontogénicos en la morfología externa, en la pigmentación y en el desarrollo de las aletas, al igual que en la morfometría y merística. El material fue recolectado en la bahía de Chacororé, río Cuiabá, entre marzo de 2000 y marzo 2004. Se analizaron 80 individuos de cada especie, siendo 60 larvas y 20 juveniles. Las larvas de P. bonariensis presentaron boca en posición terminal pasando a sub-terminal, ojos grandes, bien pigmentados y esféricos, tornándose pequeños y elípticos en post-flexión, espinas en el pre-opérculo (dos internas y cuatro externas); número total de miómeros variando entre 23 y 27, pigmentación inicialmente escasa intensificándose, principalmente, en la región ventral; secuencia de formación y número total de espinas y radios de las aletas: caudal, dorsal (XI+29-32), anal (II+6), pélvica (I+5) y pectoral (15-17). Por otro lado, las larvas de P. ternetzi presentan una boca terminal: ojos grandes, bien pigmentado y esféricos, disminuyendo a medida que se desarrolla, espinas en el pre-opérculo (tres internas y cuatro externas); número total de miómeros variando entre 23 y 26; pigmentación inicialmente escasa, volviéndose evidente solo al final de la post-flexión, con la presencia de algunos cromatóforos en la región superior de la cabeza; secuencia de formación y número total de espinas y radios de las aletas: caudal, dorsal (XI+30-36), anal (II+6), pélvica (I+5) y pectoral (16-18). En la relación de las variables morfométricas, sólo la longitud de la aleta anal-hocico diferían entre las dos especies, siendo inicial-mente mayor en P. ternetzi, mientras que sólo se hizo más grande en los jóvenes de P. bonariensis después de 34 mm. A pesar de la dificultad de identificación intraespecífica entre larvas de peces recolectadas en ambientes naturales, las herramientas morfológicas y morfométricas utilizadas en el presente estudio fueron eficaces para la separación de las primeras etapas de desarrollo de las dos especies morfológicamente similares que comparten el mismo ambiente para la reproducción.

ABSTRACT

Poultry processing plants and slaughterhouses produce huge quantities of feathers and hair/bristle waste annually. These keratinaceous wastes are highly resistant to degradation. Onygena corvina, a non-pathogenic fungus, grows specifically on feathers, hooves, horn, and hair in nature. Hence, the proteases secreted by O. corvina are interesting in view of their potential relevance for industrial decomposition of keratinaceous wastes. We sequenced and assembled the genome of O. corvina and used a method called peptide pattern recognition to identify 73 different proteases. Comparative genome analysis of proteases in keratin-degrading and non-keratin-degrading fungi indicated that 18 putative secreted proteases from four protease families (M36, M35, M43, and S8) may be responsible for keratin decomposition. Twelve of the 18 predicted protease genes could be amplified from O. corvina grown on keratinaceous materials and were transformed into Pichia pastoris. One of the recombinant proteases belonging to the S8 family showed high keratin-degrading activity. Furthermore, 29 different proteases were identified by mass spectrometry in the culture broth of O. corvina grown on feathers and bristle. The culture broth was fractionated by ion exchange chromatography to isolate active fractions with five novel proteases belonging to three protease families (S8, M28, and M3). Enzyme blends composed of three of these five proteases, one from each family, showed high degree of degradation of keratin in vitro. A blend of novel proteases, such as those we discovered, could possibly find a use for degrading keratinaceous wastes and provide proteins, peptides, and amino acids as valuable ingredients for animal feed.

ABSTRACT

Re-introduction is an important tool for recovering endangered species; however, the magnitude of genetic consequences for re-introduced populations remains largely unknown, in particular the relative impacts of historical population bottlenecks compared to those induced by conservation management. We characterize 14 microsatellite loci developed for the Seychelles paradise flycatcher and use them to quantify temporal and spatial measures of genetic variation across a 134-year time frame encompassing a historical bottleneck that reduced the species to ~28 individuals in the 1960s, through the initial stages of recovery and across a second contemporary conservation-introduction-induced bottleneck. We then evaluate the relative impacts of the two bottlenecks, and finally apply our findings to inform broader re-introduction strategy. We find a temporal trend of significant decrease in standard measures of genetic diversity across the historical bottleneck, but only a nonsignificant downward trend in number of alleles across the contemporary bottleneck. However, accounting for the different timescales of the two bottlenecks (~40 historical generations versus <1 contemporary generation), the loss of genetic diversity per generation is greater across the contemporary bottleneck. Historically, the flycatcher population was genetically structured; however, extinction on four of five islands has resulted in a homogeneous contemporary population. We conclude that severe historical bottlenecks can leave a large footprint in terms of sheer quantity of genetic diversity lost. However, severely depleted genetic diversity does not render a species immune to further genetic erosion upon re-introduction. In some cases, the loss of genetic diversity per generation can, initially at least, be greater across re-introduction-induced bottlenecks.

ABSTRACT

Plagioscion squamosissimus is a species from the Amazon basin that was introduced into the Tietê River system. The present study aimed to analyse the feeding habits of this species in Bariri Reservoir and to verify the possible occurrence of ontogenetic changes in its diet composition. The samples were gathered in four periods of the year: February, June, September and November 2003. The fish were gathered with different fishing net meshes in three different reservoir portions. The alimentary items found in the stomachs were identified to the lowest possible taxonomic level and had abundance, occurrence frequency, volume and biomass determined. The Alimentary Index (IAi) was calculated for each alimentary item consumed by "corvina" for each studied period. Comparisons among the diet of different size classes of P. squamosissimus were done using the similarity coefficient of Jaccard and the Cluster Analysis (UPGMA). The Friedman proof was performed to verify if there is a significant ontogenetic variation in the species diet and changes in the consumption of different alimentary categories by P. squamosissimus among the sampled periods. P. squamosissimus presented a piscivorous feeding habit, although other items were also consumed. The biggest values of IAi were obtained for the alimentary item fish in the months of June (0.47) and November (0.39). The item Ephemeroptera (Campsurinae) was the most representative in February (0.30) and June (0.45). Despite the fact that P. squamosissimus consumed an ample spectrum of alimentary items, the ontogenetic changes were evident through the exploration of aquatic insects by the younger classes and by a diet mainly composed of fish in adult individuals. The alimentary plasticity of P. squamosissimus evidenced in this study might have contributed to the success of this species in Bariri Reservoir.


Plagioscion squamosissimus é uma espécie da Bacia Amazônica que foi introduzida no sistema do Rio Tietê. O presente estudo teve como objetivo analisar o hábito alimentar da espécie no reservatório de Bariri e verificar possível ocorrência de mudanças ontogenéticas na composição da dieta. As amostragens foram realizadas em quatro períodos do ano: fevereiro, junho, setembro e novembro de 2003. Os peixes foram coletados com redes de diferentes malhagens em três porções do reservatório. Os itens alimentares encontrados nos estômagos foram identificados até o menor nível taxonômico possível e tiveram abundância, frequência de ocorrência, volume e biomassa determinados. O Índice de Importância Alimentar (IAi) foi calculado para cada item alimentar consumido pela corvina para cada período estudado. Comparações entre as dietas das diferentes classes de tamanho de P. squamosissimus foram feitas utilizando-se o coeficiente de similaridade de Jaccard e a Análise de Cluster. A Prova de Friedman foi realizada para verificar se há variação ontogenética significante na dieta da espécie e se existem mudanças no consumo das categorias alimentares nos períodos amostrados. P. squamosissimus apresentou um hábito alimentar piscívoro, embora outros itens alimentares também tenham sido consumidos. Os maiores valores do IAi foram obtidos para o item alimentar peixes nos meses de junho (0,47) e novembro (0,39). O item Ephemeroptera foi mais representativo em fevereiro (0,30) e junho (0,45). Apesar do fato de P. squamosissimus consumir um amplo espectro de itens alimentares, as mudanças ontogenéticas ficaram evidentes pela exploração de insetos aquáticos pelas classes mais jovens e por uma dieta principalmente composta por peixe em indivíduos adultos. A plasticidade alimentar de P. squamosissimus evidenciada neste estudo pode ter contribuído para o sucesso da espécie nesse reservatório.

ABSTRACT

Os métodos sensoriais são acurados e amplamente utilizados na avaliação do frescor de peixe. Esta pesquisa descreve o desenvolvimento de um protocolo sensorial para corvina (Micropogonias furnieri) baseado no Método de Índice de Qualidade (MIQ). O protocolo de Índice de Qualidade (IQ) pontuou atributos de aspecto geral, de olhos e de brânquias, além da cor do rim e da musculatura, cujo somatório variou de zero (máximo frescor) a 22 (limite de aceitabilidade). As contagens de bactérias atingiram o limite aceitável no 14o dia de estocagem a 0oC. O protocolo criado especificamente para a corvina eviscerada poderá ser empregado nos diversos segmentos como na comercialização, estocagem e processamento, reduzindo perdas econômicas e auxiliando na proteção da saúde do consumidor.


Sensory methods are the accurate and widely used methods to evaluate fish freshness. This paper describes the development of a sensorial scheme for whitemouth croaker (Micropogonias furnieri) based on Quality Index Method (QIM). The Quality Index protocol (QI) it punctuated attributes of general aspect, of eyes, of gills, besides the kidneys' and musculature's color, which sum varied from zero (maximum freshness) to 22 (acceptability limit). The counting of bacterial reached the accepted limit on the 14th day of storage at 0oC. The protocol specifically created for the gutted whitemouth croaker stored in ice can be used in several segments as commercialization, storage and processing, reducing economical losses and aiding in the protection of the consumer's health.
