ABSTRACT

Edible, plant-derived foodstuffs are recognized as precious sources of polyphenol compounds, whose consumption has proven to have multiple beneficial effects on human health. However, the awareness that cooking processes are able to induce quali-quantitatively changes in their native occurrence and that their bioavailability after food ingestion is poor led the research to move toward the preparation of nutraceutical supplements aimed at maximizing their content by effective extractive techniques and protecting them from degradation. The present work fits into this context, proposing a green, ready-to-use formulation of capitula, stems, and leaves of Algerian artichokes, in which natural deep eutectic solvents were exploited as extracting solvents but not removed at the end of the process. MTT test on the Caco-2 cell line highlighted that mitochondrial redox activity inhibition was absent below the 50 µg/mL tested dose. Simulated in vitro digestion was used as a predictive model for formulation bioaccessibility, where the joint approach with UHPLC-HRMS techniques allowed to define the release of each polyphenol from the investigated matrices. The capitula-based sample was the richest one in flavonoids, especially luteolin and apigenin glycosides, which survived in the intestinal digesta. On the contrary, simple phenols characterized the stem sample, whose release was mainly in the gastric chyme.

ABSTRACT

Increasing interest has been shown in globe artichoke for pharmaceutical and food applications as a significant source of polyphenol compounds. With the aim to improve the polyphenol profile of globe artichoke, an open-field experiment is set up to study the effect of gibberellic acid (GA3) application on the cynaropicrin, caffeoylquinic acid and flavone levels of three genotypes ('Apollo', 'Tema 2000' and 'Violet de Provence'), along with crop earliness, yield and bio-morphological plant response. The results indicate that GA3 treatment is more effective in terms of total polyphenol and caffeoylquinic acid accumulation in 'Apollo', regardless of the plant parts analyzed. In particular, the leaves of 'Apollo' were the richest source of luteolin derivatives of 5-O- and 1,5-di-O-caffeoylquinic acids. On average across the genotypes, GA3 treatment favored the accumulation of apigenin derivatives in the floral stem, and caffeoylquinic acid and cynaropicrin in the leaves. From the perspective of yield, GA3 treatment allowed us to anticipate the first harvest in each genotype, while either maintaining ('Apollo', 'Violet de Provence') or enhancing ('Tema 2000') the fresh weight yield. In conclusion, this study could be interesting for promoting GA3 usage to ameliorate the globe artichoke nutraceutical profile and to anticipate the first harvest for fresh marketing without significant yield losses.

ABSTRACT

The knowledge of the organization of the domesticated gene pool of crop species is an essential requirement to understand crop evolution, to rationalize conservation programs, and to support practical decisions in plant breeding. Here, we integrate simple sequence repeat (SSR) analysis and phenotypic characterization to investigate a globe artichoke collection that comprises most of the varieties cultivated worldwide. We show that the cultivated gene pool of globe artichoke includes five distinct genetic groups associated with the major phenotypic typologies: Catanesi (which based on our analysis corresponds to Violetti di Provenza), Spinosi, Violetti di Toscana, Romaneschi, and Macau. We observed that 17 and 11% of the molecular and phenotypic variance, respectively, is between these groups, while within groups, strong linkage disequilibrium and heterozygote excess are evident. The divergence between groups for quantitative traits correlates with the average broad-sense heritability within the groups. The phenotypic divergence between groups for both qualitative and quantitative traits is strongly and positively correlated with SSR divergence (FST) between groups. All this implies a low population size and strong bottleneck effects, and indicates a long history of clonal propagation and selection during the evolution of the domesticated gene pool of globe artichoke. Moreover, the comparison between molecular and phenotypic population structures suggests that harvest time, plant architecture (i.e., plant height, stem length), leaf spininess, head morphology (i.e., head shape, bract shape, spininess) together with the number of heads per plant were the main targets of selection during the evolution of the cultivated germplasm. We emphasize our findings in light of the potential exploitation of this collection for association mapping studies.

ABSTRACT

Abstract The aim of this work is to study three cultivars of artichoke (Cynara cardunculus var. scolymus): Gauchito, Guri and Oro Verde in terms of their in vitro chemoprevention and anti-inflammatory properties. These cultivars show good productive performance. The phenolic composition of their fresh leaves and edible bracts was analyzed by high performance liquid chromatography and high resolution mass spectrometry (HPLC-HRMS), showing mainly caffeoylquinic acids and flavonoids. Caffeoylquinic acids were quantified and the highest content was found in Gauchito cultivar. In this cultivar, the content of dicaffeoylquinic acids in fresh bracts was six times higher than that in fresh leaves (10064.5 ± 378.3 mg/kg versus 1451.0 ± 209.3 mg/kg respectively). Luteolin flavonoids were detected in leaves. The extracts from fresh bracts and leaves were assessed in their in vitro bioactivity against human neuroblastoma cells (SH-SY5Y). Inhibition of SH-SY5Y cells proliferation by Gauchito and Guri leaf extracts (8 µg/mL) was higher than 50 %. The leaf extracts of the same cultivars showed an inhibitory effect on human interferon IFN-I, decreasing its activity 50% at 40 µg/mL. Interestingly, the bract extracts did not show in vitro bioactivity at these concentrations, nor did the pure compounds chlorogenic acid, cynarin, apigenin and luteolin (at 2 µg/mL). These results suggest that Gauchito and Guri leaf extracts have potential for human neuroblastoma chemoprevention and treatment of inflammatory processes.

ABSTRACT

Phenolic compounds are well-known bioactive compounds in plants that can have a protective role against cancers, cardiovascular diseases and many other diseases. To promote local food development, a comprehensive overview of the phenolic compounds' composition and their impact on human health from typical Mediterranean plants such as Punica granatum L., Ziziphus jujuba Mill., Arbutus unedo L., Celtis australis L., Ficus carica L., Cynara cardunculus var. Scolymus L. is provided. Moreover, the potential use of these data for authenticity determination is discussed. Some of the plants' phenolic compounds and their impact to human health are very well determined, while for others, the data are scarce. However, in all cases, more data should be available about the content, profile and health impacts due to a high variation of phenolic compounds depending on genetic and environmental factors. Quantifying variation in phenolic compounds in plants relative to genetic and environmental factors could be a useful tool in food authentication control. More comprehensive studies should be conducted to better understand the importance of phenolic compounds on human health and their variation in certain plants.

BACKGROUND: Traditionally, globe artichoke cultivation in the Mediterranean basin is based on monoculture and on use of high amounts of nitrogen fertiliser. This raises issues regarding its compatibility with sustainable agriculture. We studied the effect of one typical conventional (CONV) and two alternative cropping systems [globe artichoke in sequence with French bean (NCV1), or in biannual rotation (NCV2) with cauliflower and with a leguminous cover crop in inter-row spaces] on yield, polyphenol and mineral content of globe artichoke heads over two consecutive growing seasons. RESULTS: NCV2 showed statistical differences in terms of fresh product yield with respect to the monoculture systems. In addition, the dihydroxycinnamic acids and dicaffeoylquinic acids of non-conventional samples were one-fold significantly higher than the conventional one. All the samples reported good mineral content, although NCV2 achieved a higher Fe content than conventional throughout the two seasons. After two and three dates of sampling, the CONV samples showed the highest levels of K content. CONCLUSION: In our study, an acceptable commercial yield and quality of 'Spinoso sardo' were achieved by shifting the common conventional agronomic management to more sustainable ones, by means of an accurate choice of cover crop species and rotations introduced in the systems. © 2017 Society of Chemical Industry.

The existence of variability is a prerequisite for genetic improvement in plants. Globe artichoke is a high nutritious vegetable with medical value, representing a profitable alternative for rural producers. This research was conducted with the aim of evaluating the phenotypic variability in a commercial cultivar of artichoke (Cynara cardunculus var. scolymus L) established from seeds. Field plants were assessed when primary head reached commercial stage. An amount of 21 quantitative and 5 multicategoric characters were assessed. The quantitative data were submitted to multivariate analysis. For quantitative characters the distance between individuals varied from 3.0 to 50.9, revealing high intrapopulation variability. The greater relative contribution characters for genetic divergence were the primary head fresh mass (79.88%) and bottom fresh mass (8.43%). This indicates the possibility of plant selection for head quality in this population. The clustering analysis through UPGMA method, based on quantitative characters, allowed the formation of five groups. For multicategoric traits, the similarity among individuals varied from 1.22% to 100%. Within the existing population variability, it was possible to select plants with superior quantitative traits desirable for in natura consumption, as primary head fresh weight and length, bottom fresh mass, bract basis length and width, as well as non-quantitative traits as round head shape, absence of thorn and presence of violet color in the head.


A existência de variabilidade é um pré-requisito para o melhoramento genético em plantas. A alcachofra é uma hortícola de alto valor nutritivo e medicinal, representando uma alternativa rentável para os produtores rurais. Este trabalho objetivou avaliar a variabilidade fenotípica de uma cultivar comercial de alcachofra (Cynara cardunculus var. scolymus L) estabelecida por sementes. As plantas foram cultivadas no campo e avaliadas quando o capítulo primário atingiu o estádio comercial. Foram avaliados 21 caracteres quantitativos e cinco multicategóricos, com posterior análise multivariada. Para os caracteres quantitativos, a distância entre os indivíduos variou de 3 a 50,9, revelando elevada variabilidade intrapopulacional. Os caracteres com maior contribuição relativa para divergência genética foram a massa fresca do capítulo primário (79,88%) e massa fresca do fundo (8,43), indicando a possibilidade de seleção de plantas com qualidade de capítulo nesta população. A análise de agrupamento pelo método UPGMA, com base nos caracteres quantitativos, permitiu a formação de cinco grupos. Para as variáveis multicategóricas, similaridade entre os indivíduos variou de 1.22% a 100%. Foi possível selecionar indivíduos com características superiores quanto aos principais caracteres quantitativos desejáveis para o consumo in natura, como: massa fresca e comprimento do capítulo primário, massa fresca de fundo, comprimento e largura da base das brácteas, comprimento das brácteas externas, bem como características não quantitativas como capítulo de formato circular, ausência de espinhos e presença de cor violeta no capítulo.
