ABSTRACT

DNA methylation modifications are known to influence epigenetic phenomena and have been a focus of forensic science research for some time. Degraded DNA after bisulfite treatment is widely used in DNA methylation analysis. In this study, we analyzed methylation levels at 12 CpG sites of four selected genomic regions by pyrosequencing after bisulfite treatment. DNA was extracted from buccal swab samples collected from 102 Japanese individuals who were 21-77 years old. We also developed a simple method to quantify the degradation levels of bisulfite-converted DNA by real-time PCR, and evaluated the effect of DNA degradation on age estimation. We found that the methylation levels and chronological ages were highly correlated in the four selected regions, and the mean absolute deviation (MAD) between chronological and estimated ages was low at 3.88 years. These results indicated that pyrosequencing analysis at the 12 CpGs was useful for age estimation in the Japanese population. To develop a sensitive quantification method, we analyzed the amplification efficiency of short and long fragments from 10 regions by real-time PCR. The amplification efficiency was highest for CCDC102B, and the degradation levels of bisulfite-converted DNA for the 102 samples were categorized as moderately or heavily degraded. For the younger age groups (20-49 years), the MADs were lower for moderately degraded DNA than they were for heavily degraded DNA. This finding indicates that degradation levels affected the accuracy of age estimation in most of the samples; the exception was the samples from the 50-77 years age group.

ABSTRACT

In many industrial processes that include fluid flow, cavitation erosion of different engineering structures (pumps, turbines, water levels, valves, etc.) during their operation is expected. Metallic, ceramic, and composite materials are usual candidates considered for application in such extreme conditions. In this study, the idea is to synthesize refractory ceramic material based on talc with the addition of zeolite for utilization as protective coatings in cavitating conditions. Two talc-based refractories with zeolites from two Serbian deposits were produced. The behaviors of the samples in simulated cavitation conditions were examined by an advanced non-destructive methodology consisting of monitoring mass loss and surface degradation using image analysis compiled with principal component analysis (PCA), interior degradation by ultrasonic measurements, and the microstructure by a scanning electron microscope (SEM). Lower mass loss, surface degradation level, and modeled strength decrease indicated better cavitation resistance of the sample with Igros zeolite, whereby measured strength values validated the model. For the chosen critical strength, the critical cavitation period as well as critical morphological descriptors, Area and Diameter (max and min), were determined. A Young's elasticity modulus decrease indicated that surface damage influence progressed towards interior of the material. It can be concluded that the proposed methodology approach is efficient and reliable in predicting the materials' service life in extreme conditions.

ABSTRACT

Although grassland degradation simultaneously affects plant productivity and soil nutrient concentrations, the relationship between plant productivity and soil nutrient concentrations during the process of grassland degradation is not yet well documented. A 4-year survey in the Qinghai-Tibetan Plateau was conducted to simultaneously investigate the relationships between plant productivity and soil nutrient concentrations in an alpine grassland at an overall degradation level and individual degradation levels. Our results showed that the total plant, sedge, and forb biomasses decreased, whereas the grass and legume biomasses first increased and then decreased as the level of alpine grassland degradation increased. Soil organic carbon (C), total nitrogen (N), total phosphorus (P), available N, and available P concentrations also decreased with the increase in degradation level. Our results also showed that plant productivity was positively correlated with soil nutrient concentrations (soil organic C, total T, total P, available N, available P) at an overall degradation level, whereas plant productivity was positively correlated with only the soil organic C concentration at each degradation level. Our findings suggested that the alpine grassland degradation conditions had different effects on the plant productivity of four functional groups (sedges, grasses, legumes, forbs) and affected the relationship between plant productivity and soil nutrient concentrations.

A introdução de espécies florestais de rápido crescimento em áreas com solos arenizados e degradados pode ser uma alternativa tanto para a prevenção como para a minimização desse problema. O trabalho objetivou avaliar o crescimento em altura dominante do Pinus elliottii Engelm. e Pinus taeda L. em relação aos solos arenizados e degradados por ação antrópica, no oeste do estado do Rio Grande do Sul (RS). Para isso, foram selecionadas árvores dominantes em povoamentos com 29 anos de idade, em áreas com dois níveis de degradação, as quais foram abatidas e seccionadas pelo método de Smalian para a obtenção de discos de madeira para a análise dendrocronológica e determinação da altura dominante por idade, no período de 1982 a 2010. Os crescimentos em altura dominante dessas espécies, quando comparados entre si e por nível de degradação do solo, apresentaram tendências diferentes de desenvolvimento no tempo, indicando a presença de polimorfismo das curvas.


The introduction of fast-growing tree species in areas with arenized degraded soils can be an alternative to minimize this problem. This research was conducted to study the dominant height growth of Pinus elliottii Engelm. and Pinus taeda L. in relation of arenized soil and degraded by antropic action in the western state of Rio Grande do Sul (RS), Brazil. For this purpose, we selected dominant trees in stands with 29 years old, in areas with two levels of degradation, which were felled and sectioned by Smalian method to obtain wood sample for dendrochronological analysis and determination of dominant height for age in the period 1982-2010. The dominant height growth of these species, when compared by level of soil degradation, showed different trend of development in the time, indicating the presence of polymorphism curves.
