ABSTRACT

Salinity is one of the causes that limit crop production. Plant Growth Promoting Rhizobacteria (PGPR) are beneficial soil bacteria that play a significant role in promoting plant growth. These microorganisms can produce their effect through the emission of Volatile Organic Compounds (VOCs). Most of the research to study the effects of microbial VOCs on plant growth has been carried out under controlled conditions using partitioned Petri dishes. In this article, we describe an alternative method that has the advantage of allowing long-term trials, being able to let the plant have a greater development in growth and height, without space limitation. In the proposed method, M. piperita were planted in glass jars containing Murashige and Skoog solid media, with a small glass vial containing Hoagland media inserted into the jar. This small vial was inoculated with the specified bacterium and served as the source of bacterial volatiles. This way plants were exposed to mVOCs without having any physical contact with the rhizobacteria.•The procedure allows studying the effect of microbial VOCs on plant growth.•It also allows longer trials, being able to let the plant develop more without space limitation.

ABSTRACT

The objective of this work was to assess the performance of two aeration types, splash and blower, used on a commercial biofloc technology (BFT) farm of red tilapia, Oreochromis sp., and their effect on growth performance and water quality parameters. For this, red tilapia juveniles were randomly distributed into twelve tanks. Each tank had an independent aeration system, and two experimental groups were characterized: six tanks used splash aerators (SPL group; one per tank, 1 hp), and the remaining tanks used a blower aerator (BLW group) with the same potency (1 hp). Water quality parameters were registered daily, and after 90 days of commercial culture, we obtained final growth parameters for each batch (tank). We observed no statistical differences on growth parameters, while water quality parameters showed that dissolved oxygen and oxygen saturation were significantly higher for the SPL group. Therefore, this field study corroborated indications from prior research at laboratory conditions that, at a commercial scale, splash aerators are more adequate for the production of red tilapia in BFT conditions.(AU)

ABSTRACT

The applications of nanoparticles (Nps) as food additives, health enhancers, and antimicrobials in animal production are increasing. The aim of this study was to evaluate the effect of selenium (Se) nanoparticles (Se0Nps) stabilized with L-cysteine (Se0Nps/L-Cys), as a nutritional supplement, on immunological, oxidative status, and productive parameters in O. mykiss. TEM and SEM-EDS showed the accumulation of spherical Se0Nps entirely composed by elemental selenium (Se0) as intracellular and extracellular deposits in Pantoea agglomerans UC-32 strain. The in vitro antioxidant capacity of Se0Nps/L-Cys was significant more efficient ROS scavengers than Se0Nps and Na2SeO3. We also evaluate the effect of Se0Nps/L-Cys on cell viability and oxidative stress in RTgill-W1, RTS-11, or T-PHKM Oncorhynchus mykiss cell lines. Se0Nps/L-Cys showed less toxic and high antioxidant activity than Se0Nps and Na2SeO3. Finally, the dietary Se0Nps/L-Cys had a significant better effect on both plasma lysozyme and respiratory burst activity (innate immune response), on tissular Gpx activity (oxidative status), and on well-being (productive parameter) of O. mykiss when it is compared to Se0Nps and Na2SeO3. Se0Nps/L-Cys is a promising alternative for nutritional supplement for O. mykiss with better performance than Na2SeO3 and Se0Nps, ease to implementation, and reduced environmental impact.

ABSTRACT

Fall armyworm (FAW), Spodoptera frugiperda (Smith), is one of the major pests targeted by transgenic crops expressing insecticidal proteins from Bacillus thuringiensis (Bt) Berliner. However, FAW presents a high capacity to develop resistance to Bt protein-expressing crop lines, as reported in Brazil, Argentina, Puerto Rico and the southeastern U.S. Here, FAW genotypes resistant to pyramided maize events expressing Cry1F/Cry1A.105/Cry2Ab2 (P-R genotype) and Cry1A.105/Cry2Ab2 (Y-R genotype) from Brazil were used to investigate the interactions between non-Bt hosts (non-Bt maize, non-Bt cotton, millet and sorghum) and fitness costs. We also tested a FAW genotype susceptible to Bt maize and F1 hybrids of the resistant and susceptible genotypes (heterozygotes). Recessive fitness costs (i.e., costs affecting the resistant insects) were observed for pupal and neonate to adult survival of the P-R genotype on non-Bt cotton; larval developmental time of the P-R genotype on millet and sorghum; larval and neonate-to-adult developmental time of the Y-R genotype on non-Bt cotton and sorghum; the fecundity of the Y-R genotype on non-Bt cotton; and mean generation time of both resistant genotypes. However, on non-Bt cotton and non-Bt maize, the P-R genotype had a higher fitness (i.e., fitness benefits), displaying greater fecundity and rates of population increases than the Sus genotype. Non-recessive fitness costs (i.e., costs affecting heterozygotes) were found for fecundity and population increases on millet and sorghum. These findings suggest that, regardless of the disadvantages of the resistant genotypes in some hosts, the resistance of FAW to Cry1 and Cry2 Bt proteins is not linked with substantial fitness costs, and may persist in field conditions once present.

We search for positive utility impact on the invasive species Pterygoplichthys spp. The optimal level of replacement of sardine meal (SM) by Pterygoplichthys spp. meal (PLM) in practical feeds for fry of Nile tilapia (Oreochromis niloticus) was evaluated. We evaluated six experimental diets: 50% PLM-50% SM, 60% PLM-40% SM, 70% PLM-30% SM, 80% PLM-20% SM, 90% PLM-10% SM and 100% PLM-0% SM. In a recirculation system, 270 sex-reversed tilapia fry were used (0.7 ± 0.1 g). Diets were administered in triplicate and the experiment lasted 56 days with sampling every 14 days. There were no statistical differences in growth. At the end of the experiment, the SGR, ADG, CF, WG, DFI, FCR and PER were determined without significant differences (P > 0.05) between treatments, but there was a tendency that could indicated higher WG and PER in the 90% PLM-10% SM treatment (93.56 ± 0.43% and 8.44 ± 1.18 g, respectively). All survival rates were higher than 90% with no significant differences. Additionally, there were no statistical differences in the chemical composition of the whole fish, while apparent digestibility coefficients of protein (81-90%) and lipids (87-93%) also showed no significant differences. We concluded that 100% PLM can replace 100% SM in feeding fry of O. niloticus without affecting their survival, growth or chemical composition or the apparent digestibility of nutrients.


Buscamos por um impacto positivo de utilidade nas espécies invasoras como Pterygoplichthys spp. Foi avaliado o nível ótimo de substituição da farinha de sardinha (SM) pela farinha de Pterygoplichthys spp. (PML) em dietas práticas para tilápia do Nilo (Oreochromis niloticus). Foram utilizadas seis dietas experimentais; 50% PLM-50% SM, 60% PLM-40% SM, 70% PLM-30% SM, 80% PLM-20% SM, 90% PLM-10% SM e 100% PLM-0% SM. Em sistema de recirculação foram colocados 270 filhotes de tilápia machos (0.7 ± 0.1 g). As dietas foram triplicadas, o experimento durou 56 dias com amostragem a cada 14 dias, não houve diferença estatística para o crescimento. A sobrevida foi superior a 90%, sem diferenças significativas. Além disso, não houve diferenças significativas na composição proximal de todo o peixe, enquanto o aparente coeficiente de digestibilidade de proteínas (81-90%) e lipídios (87-93%) também não mostrou diferenças significativas. Podemos concluir que o uso de 100% de farinha de Pterygoplichthys spp. pode substituir 100% de farinha de sardinha na alimentação de jovens O. niloticus sem afetar a sobrevivência, o crescimento, a composição química e a digestibilidade aparente dos nutrientes.

In recent years, veterinary probiotic formulations constitute an interesting alternative to the use of antibiotics in animals for human consumption, but beneficial microorganisms must meet certain requirements to be included in these products. The objective of this work was to evaluate the safety and innocuity of beneficial autochthonous lactic bacteria (BALB) as well as to determine their beneficial, environmental, and technological characterization. Antibiotic resistance was assayed using phenotypic and genotypic methodology. A bovine vaginal fluid simulated medium (MSBVF) was designed where growth, pH changes, and expression of beneficial characteristics of lactic bacteria were evaluated; additionally, the optimal culture conditions in commercial media were determined in order to obtain the highest biomass production of the strains. Finally, the best strains were lyophilized and administered intravaginally to pregnant cows and their permanence in the vagina and adverse effects were evaluated. The results show that most of the strains were resistant to vancomycin, tetracycline, and streptomycin, with a high sensitivity to ampicillin, gentamicin, and clindamycin. The strains evaluated did not show gelatinase or hyaluronidase activity; however, 11 strains produced α-type hemolysis. The optimal growth of the microorganism was obtained in MRS broth, under slight agitation and without pH control. The strains grown in the MSBVF grew well and maintained the probiotic properties. Animals treated with probiotics bacteria did not show systemic or local inflammation. These strains can be included in a probiotic veterinary product to be applied to different bovine mucosa.

ABSTRACT

Colombia is the main producer of cape gooseberry (Physalis peruviana L.), a plant known for its various consumption practices and medicinal properties. This plant is generally grown in eroded soils and is considered moderately tolerant to unfavorable conditions, such as nutrient-poor soils or high salt concentrations. Most studies conducted on this plant focus on fruit production and composition because it is the target product, but a small number of studies have been conducted to describe the effect of abiotic stress, e.g., salt stress, on growth and biochemical responses. In order to better understand the mechanism of inherent tolerance of this plant facing salt stress, the present study was conducted to determine the metabolic and growth differences of P. peruviana plants at three different BBCH-based growth substages, varying salt conditions. Hence, plants were independently treated with two NaCl solutions, and growth parameters and LC-ESI-MS-derived semi-quantitative levels of metabolites were then measured and compared between salt treatments per growth substage. A 90 mM NaCl treatment caused the greatest effect on plants, provoking low growth and particular metabolite variations. The treatment discrimination-driving feature classification suggested that glycosylated flavonols increased under 30 mM NaCl at 209 substages, withanolides decreased under 90 mM NaCl at 603 and 703 substages, and up-regulation of a free flavonol at all selected stages can be considered a salt stress response. Findings locate such response into a metabolic context and afford some insights into the plant response associated with antioxidant compound up-regulation.

Chlorpyrifos is one of the most used insecticides in agro-ecosystems and is repeatedly applied due to the increase in pest resistance, which leads to environmental accumulation. The aim of this work was to evaluate the effect of chlorpyrifos on growth and aflatoxin B1 (AFB1) production by four Aspergillus section Flavi strains, under different water conditions-aW (0.93, 0.95 and 0.98)-on maize-based medium (MMEA) and maize grains supplied with 0.06 to 1.4 mmol/L of chlorpyrifos. MMEA plates were incubated at 18, 28, and 37 °C and plates with maize grains at 25 °C during 21 days. Chlorpyrifos stimulated the growth and AFB1 production of non-target organisms, such as Aspergillus section Flavi strains, both at low (0.06 mmol/L) and at high concentrations (1.4 mmol/L) on MMEA and maize grains. Stimulation occurred over a wide range of temperature and aw conditions. The toxin concentration produced by the two strains on MMEA at 18 °C increased when the concentration of chlorpyrifos also increased, being most significant at 0.6 mmol/L. In conclusion, the presence of chlorpyrifos should be considered as a factor, together with environmental conditions, for the development of effective production practices of maize grains, in order to avoid fungal growth and AFB1 production, to prevent both economic losses and risks to human and animal health.

ABSTRACT

Abstract The current agricultural system has led to the development of glyphosate (GP)-resistant weeds, causing an increase in GP doses and applications. Native mycota of pesticide-contaminated sites are the major source of pesticide-degrading microorganisms. The aims of the present study were to isolate the GP-tolerant culturable mycota in two soils with different pesticide exposure from Córdoba, Argentina, and to evaluate the growth parameters in native fungal isolates in the presence of GP and the effective dose that caused 50% growth reduction. The results showed that the genera Fusarium, Aspergillus, Mucor, Penicil-lium and Sterilia were the prevalent fungi isolated from soils both exposed and not-exposed to pesticides. The highest value (>100 mM) of effective concentration of herbicide that caused 50% growth inhibition (EC50), was found for Trichoderma isolates. Sterilia spp. had EC50 val-ues of 100mM, while Aspergillus spp. and Mucor had EC50 values between 50 and 100mM. The growth rate evaluation varied according to the isolates and GP concentrations. The data showed that all Aspergillus spp., Trichoderma spp., Mucor and three Sterilia spp. had the best growth performance in media supplied with GP after a variable acclimation period. This study provides valuable data for further studies that would allow to know the metabolic capacity of these fungal species that can be potential candidates for GP removal from contaminated environments.


Resumen El sistema agrícola actual condujo ai desarrollo de malezas resistentes a glifos-ato (GP), lo que deriva en el aumento de las dosis y el número de aplicaciones requeridas. La microbiota nativa de sitios contaminados con plaguicidas constituye la fuente principal de microorganismos capaces de degradar este herbicida. Los objetivos del presente estudio fueron aislar la microbiota cultivable tolerante a GP a partir de dos suelos agrícolas con diferente grado de exposición a plaguicidas, ambos de la provincia de Córdoba, Argentina, y evaluar en los hongos nativos aislados los parámetros de crecimiento en presencia de GP y la dosis efectiva que produce un 50% de reducción del crecimiento (EC50). Los géneros Fusarium, Aspergillus, Mucor, Penicillium y Sterilia fueron los de mayor prevalencia, tanto en suelos expuestos como no expuestos a plaguicidas. La mayor EC50 hallada (> 100 mM) correspondió a dos aislamientos de Trichoderma. En dos aislamientos de Sterilia, los valores de EC50 fueron de 100 mM, mientras que en los dos de Aspergillus y en uno de Mucor fueron de entre 50 y 100 mM. La velocidad de crecimiento varió según la concentración de GP empleada y el aislamiento considerado. Todos los aislamientos de Aspergillus, Trichoderma y Mucor, y tres aislamientos de Sterilia mostraron el mejor rendimiento en cuanto al crecimiento en los medios con GP después de un período de aclimatación variable. Este trabajo proporciona datos preliminares valiosos referidos a la capacidad metabólica de estos aislados y su potencial para remover GP en ambientes contaminados.

Previously, our group reported the establishment of a white callus cell line of Buddleja cordata Kunth that is a high producer of the secondary metabolite, verbascoside (VB, also named acteoside), under suspension culture conditions. Here, we present experimental evidence of the sustained ability of that cellular line to grow and produce high amounts of VB for 5 years of continuous culture. Cellular line profiles were determined at the early (at the beginning) and late stages (at the end of 5 years of continuous subculturing) by analyzing relevant parameters of culture growth, i.e., specific growth rate [µ], doubling time [dt], and growth index [GI], as well as VB production. Late-stage cultures exhibited a 61% faster growth rate than early-stage subcultures, and 25 and 3% lower doubling time and growth index. The extents of growth phases were found to be different. Similar amounts of biomass were found (9.5 g and 9.4 g L-1). Verbascoside production increased parallel to cell growth; maximal yield level occurred in the mid-exponential phase and lasted until the end of the stationary phase (i.e., from the 15th to the 25th day and from the 9th to the 21st day for the early and late stages, correspondingly). The content of VB was higher in the late-stage culture (1.43 ± 0945 g L-1) than in the early-stage culture (1.21 ± 0.0286 g L-1). Productivity values point out the potential use of B. cordata cell line in the biotechnological production of VB and for research focused on the biochemistry of secondary metabolism.

ABSTRACT

Animal growth is a complex and dynamic process that involves physiological and morphological changes from hatching to maturity. It is defined as the increase in body size per time unit. Mathematical functions, called growth models, have been used to explain growth patterns. The aim of this study was to compare the Gompertz-Laird, logistic, Richards, and Von Bertalanffy growth models to determine which best fits the data of the Creole chickens (CC). Three hundred forty-seven CC were individually weighed from hatching until 177 D of age. Birds were fed a starter diet (0-18 D of age; 19% crude protein (CP) and 3,000 kcal of ME/kg) and grower diet (19-177 D of age; 18% CP and 2,800 kcal of ME/kg). Data were analyzed using PROC NLIN to fit the nonlinear growth curve. The coefficient of determination (R2), Akaike information criteria (AIC), and Bayesian information criteria (BIC) were used to compare the goodness of fit of the models. The Von Bertalanffy (R2: 0.9382, 0.9415; AIC: 2,224.1, 2,424.8; BIC: 2,233.5, 2,434.3, for females and males, respectively) was the model that best explained growth of the birds. On the other hand, both the Gompertz-Laird and logistic models overestimated hatching BW and underestimated the final BW of CC. Females reached age of maximum growth faster than males. The asymptotic weight was higher in males (3,011 g) than in females (2,011 g). Body weight at inflection point was 892 g at 64 D of age for males and 596 g at 54 D for females. In conclusion, the best fit of the data was obtained with the Von Bertalanffy growth model; the information is intended to serve as the basis for utilizing CC.

ABSTRACT

The current agricultural system has led to the development of glyphosate (GP)-resistant weeds, causing an increase in GP doses and applications. Native mycota of pesticide-contaminated sites are the major source of pesticide-degrading microorganisms. The aims of the present study were to isolate the GP-tolerant culturable mycota in two soils with different pesticide exposure from Córdoba, Argentina, and to evaluate the growth parameters in native fungal isolates in the presence of GP and the effective dose that caused 50% growth reduction. The results showed that the genera Fusarium, Aspergillus, Mucor, Penicillium and Sterilia were the prevalent fungi isolated from soils both exposed and not-exposed to pesticides. The highest value (>100mM) of effective concentration of herbicide that caused 50% growth inhibition (EC50), was found for Trichoderma isolates. Sterilia spp. had EC50 values of 100mM, while Aspergillus spp. and Mucor had EC50 values between 50 and 100mM. The growth rate evaluation varied according to the isolates and GP concentrations. The data showed that all Aspergillus spp., Trichoderma spp., Mucor and three Sterilia spp. had the best growth performance in media supplied with GP after a variable acclimation period. This study provides valuable data for further studies that would allow to know the metabolic capacity of these fungal species that can be potential candidates for GP removal from contaminated environments.

The in vitro seed germination which results in the production of disease-free seedlings and greenhouse germination of the seeds of Mansonia altissima was investigated in order to establish a better way of germination of the timber species. Five levels of GA3 treatment were used in in vitro germination with three replicate and two seeds were inoculated in each of the jam bottle. Whereas, in greenhouse germination, five levels of different treatments were used, replicated three times and each Petri plate contained 15 seeds. The experiment was repeated twice and the data from each experiment was put together and used for the statistical analysis. The results showed that seeds germination occurred eight days after inoculation in in vitro but in the case of greenhouse germination, it took only five days. For in vitro rapid germination of Mansonia altissima, the MS medium should be supplemented with 1.0 μm of GA3. Equally, in greenhouse germination, the seeds need to be soaked in 1.0 mM of GA3 for 24 hours. Alternatively, in the absence of GA3, the seeds can be soaked in water for 24 hours before broadcasting the seeds on the seedbed for germination, as this will help to identify nonviable seeds.(AU)

BACKGROUND: The use of spineless cactus in finishing diets for lambs improves the carcass quality. However, data about its influence on meat traits is very scarce. The effects of spineless cactus inclusion in the finishing diet of lambs at four levels (0, 150, 300 and 450 g kg-1 of dry matter, DM) in replacement of Tifton hay on the physicochemical and sensory properties of meat were studied. RESULTS: The spineless cactus modified the chemical composition increasing the protein (P < 0.001) and the fat (P < 0.01) contents of meat, presenting the greatest values in lambs fed diets with 150 and 300 g kg-1 (DM) replacing levels, respectively. However, the spineless cactus did not influence the growth performance from 23.7 to 37.3 kg of body weight (on average). In addition, colour parameters of the meat were not affected by the inclusion of spineless cactus in the finishing diet of lambs, resulting in mean values of 37.55, 14.96 and 8.49 for lightness, redness and yellowness, respectively. There were no statistical differences among treatments for cooking loss and shear force, with mean values of 34.8% and 14.8 N, respectively. Finally, the sensorial properties were not affected by the inclusion of spineless cactus in the finishing diet of lambs. CONCLUSION: Spineless cactus could be included up to the level of 300 g kg-1 DM in the finishing diets of lambs to increase the fat content of meat without compromising its sensorial properties. However, further studies about the fatty acid composition of meat from lambs fed diets containing spineless cactus are necessary. © 2019 Society of Chemical Industry.

ABSTRACT

Entomopathogenic fungi are widely recognized as agents of biological control worldwide. Their use in agriculture for the regulation of pest populations is a promising alternative to conventional insecticides. Furthermore, recent studies have shown that entomopathogenic fungi fulfill an additional role in plants as growth promoters. The purpose of this investigation was to assess the growth and yield of corn plants colonized with Beauveria bassiana and its effect on the lepidopteran pest Rachiplusia nu. Effects of the fungus on plant growth, crop yield, and vertical transmission were evaluated in the field. Feeding preferences of R. nu larvae were assessed in the laboratory using a "choice test". Corn plants inoculated with B. bassiana showed an increase in height, number of leaves, grain weight, yield, and percentage of seed germination compared to control plants. Consumption of B. bassiana-colonized corn plants by R. nu larvae was reduced compared to feeding levels observed on non-inoculated plants. This study showed that endophytic B. bassiana can provide multiple benefits to Zea mays and can play an important role in future integrated pest management programs.

BACKGROUND: Glyphosate (GP) is one of the main pesticides used for maize production. Fusarium sp. is a fungal genus with several phytopathogenic species and toxigenic features. In this study, the culturable soil mycota was evaluated using the surface-spray method. The effect of GP on the growth parameters (growth rate and lag phase) of Fusarium spp. was also tested on solid media conditioned with different water activities. Finally, the GP effect on disease severity caused by Fusarium sp. in maize seedlings was studied. RESULTS: The results showed that Fusarium species are frequently isolated from GP-exposed soils. The GP concentrations tested had a significant effect on F. graminearum, F. verticillioides and F. oxysporum growth parameters on solid media. The pathogenicity tests showed that the disease severity of the maize seedlings significantly increased with increasing GP concentrations. CONCLUSIONS: This study showed that Fusarium species are frequently isolated from pesticide-exposed soils and the GP concentrations tested had a significant effect both on growth parameters and disease severity in maize. This study provides an approach to the effect of GP on Fusarium sp. growth and pathogenicity that reinforces the importance of evaluating all the factors that could affect feed and food production. © 2019 Society of Chemical Industry.

ABSTRACT

Juvenile growth rates are thought to be restricted by available food resources. In animals that grow throughout the year, such as tropical lizards, growth is therefore predicted to be faster during the rainy season. We test this prediction using a population of Anolis nebulosusby describing the growth trajectories of both sexes using nonlinear regression models, and we then correlate the growth rates of individuals with food available in the environment, precipitation, and temperature. The Von Bertalanffy model fits the growth rates of the females better, while the logistic-by-length model fits the males better. According to both models, the males grew faster than females, reaching slightly smaller sizes at adulthood. Males reached sexual maturity when 35 mm long, at an age of seven months, and females matured at 37 mm (SVL), taking nine months to reach this size. In 1989, juvenile males and females grew more in both seasons (rainy and dry) than adults; for 1990, there were no differences by season or between age classes. These results are interesting since in the 1989 and 1990 rainy seasons, practically the same orders of prey and the greatest abundance of prey available in the environment were registered. A possible explanation could be that predation was more intense in 1990 than in 1989. There is little evidence that food, temperature, and humidity affect growth rates of A. nebulosus, refuting our predictions. This is mainly due to the low variation in growth observed in 1990. Therefore we think that the growth of this species reflects a complex combination of ecological and genetic factors.
