ABSTRACT

Objective To investigate the expression levels and significance of hypoxia inducible factor-1 α (HIF-1 α),vascular endothelial growth factor (VEGF) and matrix metalloproteinase-2 (MMP-2) in pathological scar.Methods 73 patients with pathological scar from March 2015 to February 2017 in our hospital were selected,including 42 cases in hypertrophic scar group and 31 cases in keloid group.20 cases of non pathological scar patients and 20 cases of normal plastic surgery were selected as control group.mRNA and protein expression levels of HIF-1,VEGF and MMP-2 in skin tissue of each group were detected by real-time fluorescence quantitative and Western blot,and the clinical significance was analyzed.Results Compared with non pathological scar tissue and normal skin tissue,the mRNA and protein expression levels of HIF-1α,VEGF and MMP-2 in pathological scar tissue were significantly increased,and the differences were statistically significant (P < 0.05).The mRNA and protein expression levels of HIF-1α in keloid group were significantly higher than those in hypertrophic scar group (P < 0.05);there was no significant difference in the expressions of mRNA and protein of VEGF between keloid group and hypertrophic scar group (P > 0.05);the mRNA and protein expression levels of MMP-2 in keloid group were significantly lower than those in hypertrophic scar group (P < 0.05).Pearson correlation analysis showed that there was a significant positive correlation between HIF-1α and VEGF,MMP-2 in pathological scar tissues (r =0.623,0.507,P < 0.05).Conclusions The expressions of HIF-1α,VEGF and MMP-2 in pathological scar tissue is obviously increased,which is closely related to the formation of pathological scar.

ABSTRACT

Objective To investigate the expression of matrix metalloproteinase 2 (MMP2) and MMP9 in the breast cancer and its relationship with tumor invasive potentiality,node metastasis and prognostic significance.Methods 130 cases of breast cancer and 10 cases of breast benign disease were studied by immunohistochemical staining with MMP2,MMP9 monoclonal antibodies.Results The positive expression rate of MMP2 and MMP9 were 83.08% (108/130) and 71.54% (93/130) in the breast cancers,which was significantly higher than those in breast benign disease ( P <0.01 ).The expressions of MMP2 and MMP9 obviously increased with tumor sizes and clinical staging( P <0.05,P < 0.01 ).Over expression of MMP2 and MMP9 in the breast cancers with axillary lymph node metastases were significantly higher than those without lymph node metastases ( P < 0.01 ).Conclusions The expression of MMP2 and MMP9was closely correlated to lymph node metastasis,and may act as markers for lymph node metastasis and prognostic in breast cancer.

ABSTRACT

Objective Serum indicators of oxidative protein damage (OPD) were analyzed to explore the effect on renal MMP/TIMP system of OPD in diabetic nephropathy. Methods AOPP levels and PCO levels were measured by modified Witko-Sarsat's method and 2, 4-dinitrophenylhydrazine spectrophotometric method, expression of serum MMP-2 were determined by ELISA, and MMP-2 activity were detected by zymography . Results AOPP levels of group DN1, group DN2 were higher than those of group DM(78.23±19.30 vs 61.25±12.13,101.59±30.22 vs 61.25±12.13,F=41.988,P<0.01). PCO levels of group DN1 and group DN2 were higher than those of group DM (0.84±0.03 vs 0.66±0.02,1.05±0.05 vs 0.66±0.02,F=205.763,P<0.01). Pearson correlation analysis indicated AOPP and PCO were positively correlated with the expression (r=0.460,0.480,P<0.05) and activity (r=0.385,0.560,P<0.05) of serum MMP-2. Multiple stepwise regression analysis showed that AOPP and PCO were major influential factors of serum MMP-2 expression (P<0.05,P<0.01) and activity(P<0.01,P<0.05). Conclusions OPD might be involved in the imbalance of renal matrix metabolism, which was correlated with the development of diabetic nephropathy.

ABSTRACT

Objective To study the expression of VEGF, MMP-2 and MMP-9 in benign and malignant lesions of breast and their effects on breast carcinoma's invasiveness and metastasis. Methods VEGF,MMP-2 and MMP-9 expression was detected by SP method in 20 breast fibroadenoma and 12 normal breast specimens taken from non-cancerous regions adjacent to breast cancer tissue and 38 breast carcinomas. Results The expression rate of VEGF, MMP-2 and MMP-9 in breast carcinoma was remarkably higher than those in normal breast tissue and breast fibrogdenoma (63.2% vs 25.0% and 30. 0% ,78. 95% vs 33.3% and 35.0% and 71.1% vs 41.7% and 45.0% ,respectively; P <0.05). There was a correlation between VEGF, MMP-2 and MMP-9 expression and nodal metastasis ( P <0.05). High expression of VEGF, MMP-2 and MMP-9 was correlated with pathological grade ( P < 0.05, respectively). The overexpression of VEGF was related with those of MMP-2 and MMP-9 in breast cancer( VEGF and MMP-2:rs =0.541, P <0.01 ;VEGF and M M P-9: rs = 0.11, P < 0.01; MMP-2and M MP-9: rs= 0P < 0.01 ). Conclusion VEGF,MMP-2 and MMP-9 overexpression is strongly related to the invasiveness and metastasis of breast carcinoma, which might be valuable in evaluating tumor invasion and metastasis.

ABSTRACT

Objective To observe the effect of high-dose vitamin C on MMP2 expression and invasive ability in PANC-1. Methods Transwell invasion assay was used to compare the invasive ability of PANC-1 cells in different concentrations of vitamin C treated groups. RT-PCR and Western blot were used to detect and compare the levels of MMP2 mRNA and protein expression in each group. Results Compared with the 0mM vitamin C treated group, the mRNA expression of MMP2 was significantly decreased in 1.0,5.0,10. 0mM group(0. 510 ±0. 004 vs 0. 792 ±0. 006, 0. 391 ±0. 007 vs 0. 792 ±0. 006, 0. 282 ±0. 008vs 0. 792 ± 0. 006, P < 0. 05 ). Compared with the 0mM vitamin C treated group, the protein expression of MMP2 was significantly decreased in 1.0,5.0,10. 0mM group(0. 519 ±0. 004 vs 0. 761 ±0. 014,0. 310 ±0. 007 vs 0. 761 ±0. 014,0. 297 ±0. 008 vs 0. 761 ±0. 014, P <0. 05). Compared with the 0mM vitamin C treated group, the invaded cell number was significantly decreased in 1.0,5.0,10. 0mM groups ( 452 ± 22 vs653 ± 28,340 ± 32 vs 653 ± 28,409 ± 33 vs 653 ± 28, P < 0. 05 ). Conclusion High-dose vitamin C can decrease the expression of MMP2 in PANC-1 cells, and weaken its invasive ability.
