ABSTRACT

This study investigated the transcriptomic responses of subcutaneous adipose tissue (SAT) and liver in newborn Hanwoo calves subjected to maternal overnutrition during mid- to late gestation. Eight Hanwoo cows were randomly assigned to control and treatment groups. The treatment group received a diet of 4.5 kg of concentrate and 6.5 kg of rice straw daily, resulting in intake levels of 8.42 kg DMI, 5.69 kg TDN, and 0.93 kg CP-higher than the control group (6.07 kg DMI, 4.07 kg TDN, and 0.65 kg CP), with respective NEm values of 9.56 Mcal and 6.68 Mcal. Following birth, newly born calves were euthanized humanely as per ethical guidelines, and SAT and liver samples from newborn calves were collected for RNA extraction and analysis. RNA sequencing identified 192 genes that were differentially expressed in the SAT (17 downregulated and 175 upregulated); notably, HSPA6 emerged as the most significantly upregulated gene in the SAT and as the singular upregulated gene in the liver (adj-p value < 0.05). Additionally, differential gene expression analysis highlighted extensive changes across genes associated with adipogenesis, fibrogenesis, and stress response. The functional enrichment pathway and protein-protein interaction (PPI) unraveled the intricate networks and biological processes impacted by overnutrition, including extracellular matrix organization, cell surface receptor signaling, and the PI3K-Akt signaling pathway. These findings underscore maternal overnutrition's substantial influence on developmental pathways, suggesting profound cellular modifications with potential lasting effects on health and productivity. Despite the robust insights that are provided, the study's limitations (sample size) underscore the necessity for further research.

ABSTRACT

This study aimed to determine the persistent duration of maternal immunity against lumpy skin disease virus (LSDV) in dairy calves born from vaccinated cows using a virus neutralization test (VNT). The performance of the VNT and an in-house-ELISA test was also determined. Thirty-seven pregnant cows from 12 LSD-free dairy farms in Lamphun province, Thailand were immunized with a homologous Neethling strain-based attenuated vaccine and calved from December 2021 to April 2022. Blood samples from dam-calve pairs were collected within the first week after calving. Subsequently, blood samples were taken from the calves at monthly intervals over a period of 4 months and tested for the humoral immune response using a VNT. The calf sera were also tested with an in-house ELISA test to estimate the accuracy of both tests using a Bayesian approach. For the results, antibodies against LSDV can persist in cows for 4-9 months post-vaccination. Moreover, neutralizing antibodies and LSDV-specific antibodies against LSDV were detected in the majority of calves (75.68%) during the first week after colostrum intake. However, the percentage of seropositive calves declined to zero by day 120, with seropositivity dropping below 50% after day 60. Only a small number of seropositive calves (approximately 13.51%) were observed on day 90. These findings indicated that passive immunity against LSDV can last up to 3 months. The median of posterior estimates for sensitivity (Se) and specificity (Sp) of the VNT were 87.3% [95% posterior probability interval (PPI) = 81.1-92.2%] and 94.5% (95% PPI = 87.7-98.3%), respectively. The estimated Se and Sp for the ELISA test were 83.1% (95% PPI = 73.6-92.6%) and 94.7% (95% PPI = 88.4-98.5%), respectively. In conclusion, this study illustrates the transfer and persistence of maternal passive immunity against LSDV to calves under field conditions. This highlights a potential three-month vaccination gap in calves born from vaccinated cows, while an in-house ELISA test can be used as an ancillary test for LSDV immune response detection. However, further research is required to assess the vaccination protocols for calves as young as 2 months old to precisely determine the duration of maternal immunity.

ABSTRACT

Septic arthritis is common in newborn calves due to poor birth and housing hygiene. This study investigated the pathological deformities caused by arthritis in the carpal bones of calves using geometric morphometry. The changes in the carpal joint bones of newborn calves with septic arthritis were examined through shape analysis. The study included 20 healthy Simmental calves and 30 Simmental calves with septic arthritis. Dorso-palmar x-ray images of the carpal joint were taken, and geometric morphometry was performed on these images using 25 landmarks. The first principal components (PC1) represented 26.92% of the total variation, while PC2 represented 13.84%. One of the most significant shape changes with increasing PC1 occurred in the os carpi intermedium. The study found that it was statistically possible to discriminate between radiometric carpal joint images of Simmental calves in the control and arthritis groups using geometric morphometry. In newborn calves with septic arthritis, the trochlea radi was located more proximally. There was an enlargement of the os carpi intermedium and a tendency towards the os carpi ulnare in female calves with septic arthritis. These results indicate significant bone deformation due to septic arthritis. Geometric morphometric methods can be clinically useful, as demonstrated in this study. Researchers can statistically explore these shape analyses, opening new avenues for research in this field. This method not only enhances our understanding of morphological changes but also provides a framework for clinical investigations and discoveries in related areas.

ABSTRACT

OBJECTIVE: Compare immune responses induced by 2 commercial intranasal (IN) modified-live viral (MLV) vaccines given individually or coadministered and evaluate prevention of infection and lung pathology following bovine herpesvirus-1 (BHV-1) challenge. ANIMALS: 36 male Holstein calves (ages, 5 to 12 days). METHODS: In a randomized complete block design, each calf received an IN injection of either vaccine diluent (Placebo), an MLV vaccine containing bovine herpesvirus-1 (BHV-1; N3), bovine coronavirus vaccine (BC), or both N3 and BC (BC + N3) with a booster 4 weeks later. Nasal secretions and blood were collected weekly. Three weeks after the booster, the calves were challenged with BHV-1, sampled for virus shedding, and euthanized 10 days later to quantify lung pathology. The study period was September 7, 2020, to April 6, 2021. RESULTS: Calves were seropositive for BHV-1 and BC before vaccination. No significant difference in BC-specific serum immunoglobin G and nasal immunoglobin A antibody responses in the BC versus BC + N3 group or BHV-1-specific serum immunoglobin G and nasal immunoglobin A antibody responses in the N3 versus BC + N3 group. Cytokine responses to BHV-1 and BC did not differ among groups. BHV-1 shedding after challenge was significantly reduced in N3 groups versus Placebo and BC. There was a significant reduction in lung pathology in the N3 + BC group versus Placebo. CLINICAL RELEVANCE: This study provides evidence an MLV vaccine containing BHV-1 and an MLV BC vaccine can be coadministered to neonatal calves without significantly altering immune responses to the 2 viruses or compromising the prevention of BHV-1 respiratory disease. Calves receiving the BC + N3 vaccine had a significant reduction in lung pathology after BHV-1 aerosol challenge.

ABSTRACT

The aim of this study was to evaluate the influence of in acid whey (AW) multiplied Lactiplantibacillus plantarum LUHS135 (L.pl135), Lacticaseibacillus paracasei LUHS244 (L.pc244), and their biomass combination on newborn calves' feces and blood parameters. Additionally, the antimicrobial and mycotoxin-reducing properties and the resistance to antibiotics of the tested lactic acid bacteria (LAB) strains were analyzed. In order to ensure effective biomass growth in AW, technological parameters for the supplement preparation were selected. Control calves were fed with a standard milk replacer (SMR) and treated groups (from the 2nd day of life until the 14th day) were supplemented with 50 mL of AWL.pl135, AWL.pc244, and AWL.pl135×L.pc244 (25 mL AWL.pl135 + 25 mL AWL.pc244) in addition to SMR. It was established that L.pl135 and L.pc244 possess broad antimicrobial activities, are non-resistant to the tested antibiotics, and reduce mycotoxin concentrations in vitro. The optimal duration established for biomass growth was 48 h (LAB count higher than 7.00 log10 CFU mL-1 was found after 48 h of AW fermentation). It was established that additional feeding of newborn calves with AWL.pl135, AWL.pc244, and AWL.pl135×L.pc244 increased lactobacilli (on average by 7.4%), and AWL.pl135 and AWL.pc244 reduced the numbers of Enterobacteriaceae in calves' feces. The tested supplements also reduced the lactate concentration (on average, by 42.5%) in calves' blood. Finally, the tested supplements had a positive influence on certain health parameters of newborn calves; however, further research is needed to validate the mechanisms of the beneficial effects.

The objective was to estimate the genetic parameters of serum total protein concentration in newborn calves (calfSTP) and post parturient dairy cows (cowSTP). The study included 1013 calves and 989 cows from 10 dairy farms. Calf blood samples were collected 24-48 h after parturition while cow blood and colostrum samples were collected in the first 24 h after calving. Blood serum total protein and colostrum total solids content were determined using a Brix refractometer. Chemical analysis of colostrum was performed with Milkoscan. Univariate mixed linear models were used to estimate the heritability of calfSTP and cowSTP and their genetic and phenotypic correlations with colostrum traits. The heritability estimates of calfSTP and cowSTP were 0.21 and 0.20 (p < 0.05), respectively. Strong genetic correlations (r > 0.90) were detected between calfSTP and colostrum total solids and protein content (p < 0.05). Corresponding phenotypic correlations were 0.31-0.33 (p < 0.05). No genetic or phenotypic correlations were detected with colostrum fat content while the respective correlations with lactose were negative (-0.82 and -0.19, p < 0.05). No genetic correlations were detected between cowSTP and colostrum traits and only a low negative phenotypic one with lactose was detected. The results confirm that genetic selection aiming to improve the passive transfer of immunity in newborn calves and general fresh cow health would be feasible.

ABSTRACT

Housing confinement, adaptation to different light/dark conditions, and social deprivation could modify the amount of total locomotor behavior of calves recommended for their psychophysical health. Total locomotor behavior was recorded by means of an activity data logger every 5 min for 6 consecutive days. To do that eight clinically healthy 30-day-old Holstein calves living in calf boxes under natural photoperiod and environmental conditions were enrolled. ANOVA (analysis of variance) showed a statistical effect of the day of monitoring and animal. In the temporal distribution of the resting-activity frequency, it was observed that the calves presented periods of total locomotor behavior with the existence of two peaks, one between 06:00-07:00 and another between 17:00-18:00, which corresponds to time of food intake. In all animals, a diurnal daily rhythm of total locomotor behavior was observed during time of monitoring. Intrasubject and intersubject variabilities were statistically different in mesor, amplitude, and robustness of rhythm. In conclusion, the total locomotor behavior showed a diurnal daily rhythmicity in 30-day-old calves. The characteristics of rhythm were different from individual to individual and from day to day. The recorded intersubject variability must be taken in consideration during the monitoring of farm animals and justifies the application of the device to each animal, as precision livestock farming suggests.

ABSTRACT

Bovine viral diarrhea virus (BVDV), serving as an important pathogen for newborn calves, poses threat to reproductive and economic losses in the cattle industry. To survey the infection rate and genetic diversity of BVDV in newborn calves in northern China, a total of 676 sera samples of newborn calves were collected from four provinces between 2021 and 2022. All sera samples were individually detected for BVDV infection by RT-PCR and ELISA. Our results showed that the overall serological rate was 9.76% (66/676) and the average positive rate of BVDV RNA was 8.14% (55/676) in the newborn calves. Eight BVDV strains were successfully isolated from RT-PCR positive sera samples, and four isolates displayed the cytopathic effect (CPE). Based on phylogenetic tree at the genome level, the eight strains were classified into subgenotype 1c. Moreover, the BVDV isolates had a close genetic relationship with the GSTZ strain at either nucleotide or codon usage level. Interestingly, in comparison of synonymous codon usage patterns between the BVDV isolates with CPE and ones without CPE, there were four synonymous codons (UCG, CCC, GCA, and AAC) which displayed the significant differences (p < 0.05) at codon usage pattern, suggesting that synonymous codon usage bias might play a role in BVDV-1c biotypes. In addition, the usage of synonymous codons containing CpG dinucleotides was suppressed by the BVDV-1c isolates, reflecting one of strategies of immune evasion of BVDV to its host. Taken together, our study provided data for monitoring and vaccination strategies of BVDV for newborn calves in northern China.

ABSTRACT

We evaluated the effects of probiotic compounds on the composition of the gut microbiota. Forty newborn calves were random allocated to the lactic acid bacteria + yeast group (LS group), lactic acid bacteria group (L group), yeast group (S group), and control group (D group). Probiotics containing Lactococcus lactis subsp. lactis, Pediococcus pentosaceus, Lactobacillus plantarum, Saccharomyces cerevisiae, and Kluyveromyces marxianus were fed to calves in the three treatment groups for 15 days. The feeding process lasted 15 days. Fecal samples were collected from all calves at the end of the trial and analyzed using high-throughput 16S rRNA sequencing. Totals of 1,029,260 high-quality reads and 420,010,128 bp of sequences were obtained. Among the four groups, the alpha diversity of gut microbes was significantly higher in newborn cattle in the LS group than in those in the L, S, and D groups. Overall, the dominant phyla were Firmicutes, Actinobacteria, and Bacteroidetes, whereas Bifidobacterium was the most abundant phylum in the gut of cattle in the LS group. Newborn calves from the compound probiotic groups had closely clustered gut bacterial communities and had lower rates of diarrhea. Overall, compound probiotics regulated the intestinal microbiota community structure of newborn calves and improved intestinal health. New information relevant to the prevention of diarrhea is provided by our research in newborn calves.

ABSTRACT

The aim of this study was to evaluate the effects of dietary vitamin C supplementation on blood parameters of pre-parturient (PP) dairy cows and growth performance and immune system of their newborn calves. Forty PP cows (at approximately 21 days before calving and an average weight 791 ± 50 kg) were allocated into two experimental treatments: (1) basal diet without vitamin C supplementation (CO) and (2) basal diet with 20 g of vitamin C supplementation from 21 days before calving to parturition (VC). After parturition, the experiment continued by grouping the calves into four dietary treatments with 8 calves in each treatment. The experimental treatments were (1) control calves with no vitamin C supplementation and from cows that received no vitamin C supplement (CON), (2) calves supplemented with 600 mg of vitamin C per day and from cows that received no vitamin C supplement (CVC), (3) calves supplemented with no vitamin C and from cows that received 20 g of vitamin C per day (MVC), and (4) calves supplemented with 600 mg of vitamin C per day and from cows that received 20 g of vitamin C per day (CMVC). Serum concentrations of glucose, HDL and LDL, cholesterol, triglycerides, total protein, and albumin of cows were not affected by vitamin C supplementation during pre-parturient period. However, cows that received VC diet had lower (P < 0.05) malondialdehyde (MDA) and aspartate aminotransferase (AST) concentrations, higher total antioxidant capacity (TAC), and vitamin C concentration in their blood compared to CO cows. Vitamin C supplementation had no effect on plasma iron, copper, and zinc concentrations of PP cows. Similarly, vitamin C supplementation had no effect on total feed intake and feed conversion ratio (FCR) of suckling calves. However, calves in the CMVC group had higher (P < 0.05) overall daily weight gain compared to the other groups. Calves in the CVC and CMVC groups had lower (P < 0.05) blood MDA concentration on days 7 and 21. The highest (P < 0.05) blood TAC level was recorded in CMVC calves. Control group calves had lower (P < 0.05) blood superoxide dismutase activity compared to the other calves. Blood levels of alanine aminotransferase on days 7 and 21 and aspartate aminotransferase on day 7 were higher (P < 0.05) for calves in the CON and MVC groups. Based on the results, vitamin C supplementation had positive health effects on the oxidative parameters of PP dairy cows and also improved the performance and health status of the calves.

Calf diarrhea is one of the most concerning challenges facing both the dairy and beef cattle industry. Maintaining healthy gut microbiota is essential for preventing gastrointestinal disorders. Here, we observed significantly less bacterial richness in the abnormal feces with watery or hemorrhagic morphology compared to the normal solid feces. The normal solid feces showed high relative abundances of Osllospiraceae, Christensenellaceae, Barnesiella, and Lactobacillus, while the abnormal feces contained more bacterial taxa of Negativicutes, Tyzzerella, Parasutterella, Veillonella, Fusobacterium, and Campylobacter. Healthy calves had extensive bacterial-bacterial correlations, with negative correlation between Lactobacillus and potential diarrheagenic Escherichia coli-Shigella, but not in the abnormal feces. We isolated Lactobacillus species (L. reuteri, L. johnsonii, L. amylovorus, and L. animalis), with L. reuteri being the most abundant, from the healthy gut microbiota. Isolated Lactobacillus strains inhibited pathogenic strains including E. coli K88 and Salmonella Typhimurium. These findings indicate the importance of a diverse gut microbiota in newborn calf's health and provide multiple potential probiotics that suppress pathogen colonization in the gastrointestinal tract to prevent calf diarrhea.

In this study, we investigated the effect of dietary supplementation with acetate and L-tryptophan-conjugated bypass amino acid (ACT) during late pregnancy on the production performance of cows pre- and postpartum and their offspring. Eight multiparous Holstein cows (calving date ±15 d, 2nd parity; n = 4) were supplied with diets without ACT supplementation (Control) or with 15 g/day ACT supplementation (ACT). The results showed that ACT improved the feed intake (FI) in dry cows. No differences in blood hematological parameters were found between the two groups of prepartum cows. The serum glutamic-oxaloacetic transaminase activity increased and the triglyceride concentration decreased in the ACT-treated group compared to the control group. In the postpartum cows, milk compositions were not affected by ACT supplementation. Saturated fatty acid (SFA) content in the colostrum was significantly lower in the ACT-treated group than in the control group. Serum glucose (GLC) level was significantly higher in the ACT-treated group than in the control group. Monocyte and GLC levels were lower in calves of groups where their dams had received ACT. Overall, we found higher FI in the dry cows, lower colostrum SFA levels, and heavier calf birth weight (5.5 kg) when the dams were supplemented with ACT, suggesting a positive nutrient compensation by ACT supplementation to dry cows.

A 2-yr study was conducted at the Range Cattle Research and Education Center, University of Florida - Institute of Food and Agricultural Sciences (IFAS) (Ona, FL), to evaluate differences in the metabolism of Cu and Se of Angus (Bos taurus) and Brahman (Bos indicus) cattle. Thirty-two pregnant beef cows (n = 8 Brahman and 8 Angus/yr) were enrolled in the study in the first trimester of gestation. This study consisted of three phases: 1) restriction (day 0 to 90), 2) supplementation (day 91 to 150), and 3) calving. During all three phases, cows were individually fed and housed in partially covered drylot pens. During the restriction and supplementation phases, cows were provided a 1.5 kg/d of a grain-based concentrate supplement, which was fortified with flowers of S (50 g of supplemental S/cow daily; restriction phase) or Cu and Se (100 and 3 mg/d of Cu and Se, respectively; supplementation phase). Blood and liver samples were collected from all cows at 30 d intervals and from both cows and calves within 24 h of calving. Colostrum and milk samples were collected at calving and 7 d after birth. All data were analyzed using the MIXED procedure of SAS, where cow and calf were the experimental unit. During the restriction phase, a breed × day effect (P = 0.03) was observed where Brahman had greater liver Cu concentration than Angus cows in all sampling days. For liver Se concentration, a tendency (P = 0.07) for a breed effect was observed where Angus cows tended to have greater liver Se concentration than Brahman. During the supplementation phase, breed (P < 0.001) and day (P < 0.01) effects were observed, where Brahman cows had greater liver Cu concentration than Angus. For liver Se concentration, a day effect (P < 0.001) was observed, where liver Se concentration increased (P < 0.001) from day 90 to 120 and remained unchanged (P = 0.86) until day 150. At calving, no effects of breed (P = 0.34) were observed for liver Cu concentration of cows; however, Brahman calves tended (P = 0.09) to have greater liver Cu concentration than Angus calves. For Se liver concentration at calving, Angus cows tended (P = 0.07) to have greater liver Se concentration than Brahman cows; however, no breed differences (P = 0.70) were observed for liver Se concentration of calves at birth. In summary, substantial differences in multiple indicators of Cu and Se status were observed between Angus and Brahman cattle, implying that Angus and Brahman cattle possibly have different mechanisms to maintain adequate Cu and Se status.

ABSTRACT

The aim of this study was to assess the effects of feeding immunized egg proteins (IEP) on the health and performance of newborn dairy calves. Sixty-four Holstein calves, both male and female, were divided over two treatments. Calves either received IEP or a placebo (PCB) in their colostrum and calf milk replacer (CMR) for the first 14 days of their life. Until day 49, CMR was offered at 15% of birth weight (BW), at 10% on days 49-57 and at 5% on days 57-63. In addition, calves received starter concentrate, chopped straw and water from 3 days old until 70 days old at the end of study. Individual CMR and concentrate intake were measured daily whilst BW was recorded weekly. Visual faecal scoring and health observations were conducted daily. Faecal samples were collected weekly up to 4 weeks and during the first 4 days of scouring to screen for presence of Cryptosporidium parvum, rotavirus, coronavirus, E. coli and Salmonella. Results indicated that feeding IEP increased BW (p < .05) at 42 and 56 days old, and BW also tended (p = .06) to be higher after weaning at 63-70 days old compared to the PCB group. When analysed using a repeated measures model, compared to feeding PCB, feeding IEP increased total concentrate consumption (p = .001) by 3.6kg/calf. Over the entire study, daily water intake was higher (p = .002) for the IEP group when compared with the PCB group. In the IEP group, 12 calves were scored as scouring whereas there were 14 calves in the PCB group. There were no significant differences between treatments in faecal pathogen load of neither healthy nor scouring calves. In conclusion, supplementing IEP during the first 14 days of calf life improved the performance of newborn calves. Further work is warranted to understand the mode of action of IEP in calves.

ABSTRACT

In this study, arabinogalactan (ARB) and Lactobacillus uvarum LUHS245 antimicrobial properties against pathogenic bacteria (Klebsiella pneumoniae, Pseudomonas aeruginosa 17-331, Acinetobacter baumanni 17-380, Proteus mirabilis, MRSA M87fox, Enterococcus faecalis 86, Enterococcus faecium 103, Bacillus cereus 18 01, and Streptococcus mutans) and resistance to antibiotics were evaluated and the role of their supplementation on the main metabolic and fecal variables of newborn calves was established. The animal trial involved 48 Holstein female calves randomly allocated in four homogeneous groups of 12 animals each, on the basis of body weight in the second day of life. Calves were fed with a standard milk replacer diet from the second day of life until 14th day, either unsupplemented or supplemented with 50 mL of LUHS245 (≥7.5 log10 CFU mL-1), 30 g of ARB, or with both (50 mL of LUHS245 and 30 g ARB). In vitro data showed that the LUHS245 inhibited the growth of Salmonella enterica and Bacillus cereus (inhibition zones 13.0 and 21.3 mm, respectively). Supplementation of LUHS245 and ARB either alone or together, lowered total bacterial count in the feces and reduced lactate and serum alanine aminotransferase concentrations in blood. This study showed that LUHS245 supplementation alone or together with ARB seemed to have some positive influence on certain health parameters in newborn calves. Further research with larger cohorts of animals is warranted to validate the beneficial effects of the tested supplements.

The objective of this study was to evaluate the effects of rumen-protected betaine supplementation on performance of postpartum dairy cows and immunity of newborn calves. Twenty-four multiparous Holstein dairy cows were randomly divided into the control (CON, n = 12) and rumen-protected betaine (BET, n = 12) groups after blocking by parity and milk yield during the previous lactation cycle. The cows were fed a basal total mixed ration diet without BET (CON) or with BET at 20 g/d per cow (BET) from four weeks before expected calving to six weeks postpartum. The results showed that betaine supplementation had no effect on dry matter intake and milk yield of the cows. The BET cows tended to increase feed efficiency (energy-corrected milk/dry matter intake) and body weight loss postpartum compared to the CON cows. The plasma ß-hydroxybutyrate concentrations of the BET cows were greater at d seven after calving than those of the CON cows. Moreover, compared to the CON calves, the BET calves had greater plasma total protein and globulin concentrations. The plasma glucose concentrations of the BET calves tended to decrease relative to CON cows. In conclusion, rumen-protected betaine supplementation from four weeks before expected calving tended to increase fat mobilization of postpartum dairy cows, and might improve the immunity of newborn calves.

A deficiência de ferro em bezerros neonatos está associada ao desenvolvimento de anemia, que favorece o aparecimento de outras enfermidades como pneumonia e diarreia. Avaliou-se o efeito da suplementação de ferro sobre o eritrograma, teores séricos de ferro, ceruloplasmina e transferrina, bem como o potencial para toxicidade do protocolo utilizado por meio da avaliação dos teores de ureia, creatinina e enzimas hepáticas. Para tal avaliação foram utilizados 40 bezerros neonatos da raça Holandesa, alocados em cinco grupos experimentais com oito animais em cada grupo, que foram submetidos aos seguintes protocolos: administração intramuscular de 5mL de solução fisiológica estéril no 5º dia de idade (grupo controle G1), e administração intramuscular de 5mL de ferro dextrano 10% nos seguintes momentos: no 5º dia de idade (G2); no 5o e no 20º dias de idade (G3); no 5o e no 30º dias de idade (G4) e no 5o, 20o e 45º dias de idade (G5). Foram coletadas amostras de sangue até 8 horas após o nascimento e aos 5, 10, 20, 30, 60 e 90 dias de idade para realização do eritrograma, avaliação dos teores séricos de ferro, ceruloplasmina, transferrina, ureia, creatinina, bilirrubina total e direta, e das atividades das enzimas aspartato aminotransferase (AST), fosfatase alcalina (ALP) e gamaglutamiltransferase (GGT). Os animais que receberam ferro suplementar apresentaram menor oscilação nos parâmetros eritrocitários, embora os animais do grupo controle não tenham desenvolvido anemia. Notou-se também aumento, embora não significativo, nos teores séricos de ferro e das proteínas de fase aguda ceruloplasmina e transferrina, cuja atividade está relacionada ao metabolismo desse mineral. Os teores séricos de ureia, creatinina, bilirrubina total e direta e as atividades das enzimas GGT, AST e ALP não foram influenciados pelos protocolos de administração de ferro suplementar. Os protocolos de tratamento empregados não ocasionaram hepatoxidade ou nefrotoxidade aos animais. Concluiu-se que a suplementação com ferro dextrano por via parenteral em bezerros que recebem outras dietas que não apenas leite não traz benefícios que justifiquem sua indicação, embora sejam necessários mais estudos que avaliem a influência da suplementação com ferro sobre o tempo necessário para a recuperação, custos com o tratamento e impacto sobre a vida produtiva dos animais na idade adulta.(AU)


Iron deficiency in newborn calves is associated with the development of anemia, which favors the development of other infirmities such as pneumonia and diarrhea. The present study evaluated the effect of iron supplementation on erythrogram, serum levels of iron, ceruloplasmin and tranferrin, as well as potential toxicity of the protocol used by means of evaluation of urea, creatinine and hepatic enzyme activities. 40 newborn Holstein calves were allocated into 5 experimental groups comprising 8 calves each, which were subjected to the following treatment protocols: intramuscular administration of 5mL of sterile saline on the 5th day of age (control group G1), intramuscular administration of 5mL of 10% dextran iron in the following moments: on the 5th day of age (G2); on the 5th and in the 20th day of age (G3); on the 5th and 30th day of age (G4); on the 5th, 20th and 45th days of age (G5). Blood samples were taken until 8 hours after birth and with 5, 10, 20, 30, 60, and 90 days of age, and subjected to hemogram, evaluation of serum levels of iron, ceruloplasmin, transferrin, urea, creatinine, total and direct bilirrubin, and serum activities of aspartate aminotransferase (AST), alcaline phosphatase (ALP), and gamma-glutamyltransferase (GGT). Calves that received iron supplementation at any time presented less variation in the erythrocyte parameters, although calves in the control group did not develop anemia. Serum concentration of iron and acute phase protein ceruloplasmin and transferrin, which activities are related to iron metabolism, also increased, although not significantly. Serum levels of urea, creatinine, bilirubins and activities of AST, ALP, and GGT were not influenced by the administration protocols used in this experiment. The results of the experiment led to the conclusion that the supplementation with parenteral dextran iron in calves that receive diets other than exclusive milk does not bring sufficient advantages to be indicated, although more studies are necessary to evaluate the influence of iron supplementation on the outcome of infections in newborn calves, especially its influence on cost of treatment, time necessary for discharge and impact on its productive life.(AU)

ABSTRACT

The objective was to verify if thermography is able to detect inflammatory signs on the skin surface by comparing the umbilical region of healthy calves and calves presenting omphalitis. Twenty healthy calves (control group) had their lateral umbilical region and abdominal region examined with a thermal imaging camera in order to obtain a regional thermograph. The thermographic examination was then performed on 27 calves (Omphalitis group) presenting omphalitis diagnosed by physical examination. The maximum temperature of the lateral umbilical region in calves (aged < 30 days) was 35.7°C ± 1.8 for the control group and 37.0°C ± 1.1 for the omphalitis group and was significantly different (p = 0.002). No difference was found on the temperature of the abdominal umbilical region. In the abdominal and lateral region the highest temperature site was differently positioned between the groups. In conclusion, thermography is able to detect inflammatory signs on the skin of newborn calves and has advantages as a non-invasive, fast and safe method of supporting veterinary diagnosis.

ABSTRACT

As afecções umbilicais merecem destaque entre as enfermidades que acometem bezerros no primeiro mês de vida, pela alta incidência durante essa fase da vida dos neonatos e pelas relevantes perdas econômicas, decorrentes da mortalidade, custos de tratamento e atendimento veterinário, além de sequelas que poderão determinar menores ganho de peso e produção leiteira. O diagnóstico dessas enfermidades muitas vezes é prejudicado pela dificuldade em se detectar os acometimentos das estruturas intra-abdominais do umbigo, o que afetará negativamente a escolha do tratamento mais adequado e o estabelecimento do real prognóstico para cada animal. No presente estudo, realizou-se a análise ultrassonográfica dos animais com inflamação/infecção dos componentes umbilicais sendo possível observar algumas características em relação a mensurações e ecogenicidade, presentes nos componentes umbilicais acometidos. A partir dessas avaliações concluiu-se que a medida mais adequada para identificar animais com possíveis processos inflamatórios nos componentes umbilicais é a espessura da parede dos vasos umbilicais quando comparada ao diâmetro dos mesmos. Ainda, o estudo possibilitou verificar algumas particularidades do comportamento dos componentes do cordão umbilical em bezerros sadios da raça Holandesa, bem como determinar que os vasos umbilicais intra-abdominais iniciam uma involução em porção mais interna e progridem para as extremidades próximas ao anel umbilical com o evoluir da idade. Destaca-se que o confronto desses resultados com aqueles poucos descritos na literatura disponível mostrou-se com notáveis diferenças.(AU)


Umbilical disorders deserve mention among the illnesses affecting calves in the first month of life. The high incidence during this phase of life of newborns and relevant economic losses arising from mortality, cost of treatment and veterinary care, beyond sequelae may determine lower weight gain and milk production. The diagnosis of these diseases is often hampered by the difficulty to detect the involvement of intra-abdominal umbilical structures, which negatively affect the choice of the most appropriate treatment and the actual outcome for each calf. In the present study, we performed ultrasound examinations of calves with inflammation/infection in the umbilical components and observed some characteristics related with measurements and echogenicity present in umbilical affected components. From these evaluation it was concluded that the wall thickness of the umbilical vessels is the more reliable standard to determine changes in these components, as compared with that of the diameter of the umbilical vessels. Furthermore, from this study we observed some peculiarities of involution of the intra-abdominal umbilical cord components in healthy Holstein calves used for the control group to characterize the behavior of the umbilical structures during the progress of the age in the newborns. It is noteworthy that the comparison of these results with those found in the rare literature available showed notable differences.(AU)

ABSTRACT

The objective of this study was to determine the gamma-glutamyl transferase (GGT) levels and its relationship with diarrhea and passive transfer of immunity in Holstein calves within 24 hours and 30 days of life from Leopolis municipality, the north Parana region. Colostrum is rich in immunoglobulins and vital for immunity to newborn calves, since bovine placenta does not allow the passage of immunoglobulin to the fetus. Calves undergo various challenges that can lead to disease and death in the first month of life, including diarrhea. Diarrhea has a multifactorial etiology, and the passive immunity transferred through ingestion of colostrum is able to protect the calf against many of these etiologic agents. GGT measurements indirectly infer the amount of immunoglobulin ingested by the calf. Higher serum GGT levels (381.72 IU / L) were found at 24 hours, and a significant reduction was observed at 30 days (66.22 IU / L). When the presence or absence of diarrhea was associated with GGT levels above and below 200 IU / L, no statistical significance (P> 0.05) was observed, since 80% of animals with diarrhea had serum GGT levels higher than 200 IU / L. Under the conditions of this study, there was no relationship between the GGT concentration and the occurrence of diarrhea, and no mortality was observed despite some animals presented diarrhea.


O colostro é rico em imunoglobulinas e vital para a imunidade de bezerros recém-nascidos, uma vez que placenta bovina não permite a passagem de imunoglobulinas para o feto. Os bezerros são submetidos a diversos desafios que podem levar à doença e morte no primeiro mês de vida, dentre as quais a diarreia. A etiologia da diarreia é multifatorial e a imunidade passiva transferida através da ingestão de colostro é capaz de proteger o bezerro contra muitos desses agentes etiológicos. O objetivo deste estudo foi determinar a dosagem de gama-glutamil transferase (GGT) e sua relação com diarreia e a transferência de imunidade passiva em novilhas da raça Holandesa nos tempos de 24 horas e 30 dias de vida, no município de Leópolis, na região norte do Paraná. As mensurações de GGT infere indiretamente a quantidade de imunoglobulinas ingeridas pelo bezerro. Os níveis séricos de GGT demonstraram valores médios maiores 24 horas após o nascimento (381,72 UI / L) e uma redução significativa foi observada aos 30 dias de vida (66,22 UI/L). Quando a presença ou ausência de diarreia foi associada com níveis de GGT acima e abaixo de 200UI/L, não foi observado significância estatística (p>0,05), uma vez que 80% dos animais com diarreia tinham níveis séricos de GGT superior a 200 UI/L. Sob as condições que foi realizado o presente estudo, não houve relação entre a concentração de GGT e a ocorrência de diarréia, e nenhuma mortalidade foi observada apesar de alguns animais apresentarem diarreia.
