ABSTRACT

The potato/tomato psyllid Bactericera cockerelli is the Candidatus Liberibacter solanacearum bacterium vector that causes diseases in Solanaceae crops. Pest control is based on synthetic chemical insecticides, plant extracts, and natural enemies such as parasitoids. Tamarixia triozae feeds on nymphs of B. cockerelli, reaching up to 95% parasitism. This work aimed to evaluate the parasitic performance of T. triozae on tomato leaves with B. cockerelli N3 nymphs, using two domesticated (Floradade and Micro-Tom) and one Wild tomato variety. Several assays were completed to identify the parasitoid attraction toward un-infested plants (healthy) and infested plants (damaged) of three varieties. Parasitism preference and "Y" tube olfactometer tests were performed, respectively. The parasitism of Tamarixia triozae showed a preference toward plants of the Floradade variety by 44% compared with the other two varieties (p = 0.0003). T. triozae was more attracted to damaged plants of the Wild variety (p = 0.0523). Healthy plants of Floradade and Micro-Tom varieties attracted a higher proportion of parasitoids, except in the Wild variety, where T. triozae was more attracted to damaged plants. Taken together, the results of this study show that the domestication degree in tomato plants positively influenced the interactions between tomato plants and the parasitoid, T. triozae.

ABSTRACT

BACKGROUND: Chagas disease (American trypanosomiasis) is an important neglected tropical illness, which has the flagellate protozoan Trypanosoma cruzi as etiological agent and blood-feeding insects of the Triatominae subfamily as vectors. Despite its importance for disease epidemiology, field studies targeting microgeographic dispersal of triatomines in endemic areas are rare. The ability wingless nymphs have to move (crawl) within peridomestic settings is a key component regarding the design and development of rational control strategies. METHODS: We double-marked Triatoma sordida fourth-instar nymphs (N4) with a reliable fluorescent dye and a trace element. This new methodology allowed us to simultaneously evaluate (i) nymph dispersal and (ii) the effectiveness of copper (Cu), chromium (Cr), and cadmium (Cd) trace elements as potential new markers. In the mark-release-recapture (MRR) experiment, 390 T. sordida N4 were released in the peridomicile of a single rural household, 130 individuals at each of three release points, at distances of 2, 5, and 10 m from the chicken coop (CC) and 27, 32, and 35 m away from the horse corral (HC). All specimens were double marked (Cu/blue, Cr/orange, Cd/green). Recaptures occurred in two intervals: 1-3 days and 15-17 days after release. RESULTS: Specimens were successfully recaptured at all distances up to 10 m. A total of 19, 23, and 10 specimens were able to disperse 2, 5, and 10 m, respectively, to reach the CC. No insects were recaptured at the HC. Of the three analyte/paint combinations tested, Cr/orange gave the most promising results; Cu/blue marker and Cd/green marker performed very poorly with only 4/19 and 0/10 analyte/paint ratios, respectively. CONCLUSIONS: Triatoma sordida N4 could cover a distance of 10 m in 17 days. This indicates that nymphs seem to have a reduced dispersal capability compared to adults. Ninety-one percent of the 22 recaptured orange-marked nymphs were still Cr positive after the 17-day period evaluated. This makes this analyte a good candidate for future investigations that will apply this marking method in MRR studies.

ABSTRACT

The identification of Blood Meal Source (BMS) in hematophagous vectors contributes to a better understanding of the ecology of hemoparasite transmission. Those insects can endure long periods without feeding, waiting for a favorable setting. Although this represents an important behavior observed in those groups, such as triatomines, little is known about how time can affect BMS detection, especially considering extended periods. To comprehend to which extent this behavioral phenomenon can impact molecular detection, we submitted two groups of Rhodnius robustus to increasing periods of starvation under experimental conditions. It was possible to recover the BMS until the 12th week of the starvation process. Nymphs were more resistant to prolonged periods of starvation (up to more than 189 days) than adults (maximum of 137 days), with no significant difference between their weights after being fed. The study brought new insights to the understanding of Trypanosoma cruzi transmission by R. robustus in the nature, with a temporal perspective.

ABSTRACT

Rickettsia rickettsii infection causes the highest human fatality rate among all Rickettsia species of the world and is endemic in Southeast Brazil. In this part of the country most human spotted fever cases are related to unnaturally high populations of ticks, usually a mix of two species, Amblyomma dubitatum Neumann and Amblyomma sculptum Berlese and their local host, capybara (Hydrochoerus hydrochaeris). At the same time, an increase in the number of SF notifications as well as its urbanization was observed, and a better characterization of disease epidemiology is mandatory for control measures and to halt its expansion. It was recently noticed in southeast Brazil that SF endemic areas were characterized by overgrowth populations of A. sculptum and decreased populations of A. dubitatum. Hence, characterization of areas with potential to endemicity, eco-epidemiological studies and control measures may rely on the evaluation of A. sculptum/A. dubitatum rate. However, in potentially endemic scenarios, discriminating the two tick species, particularly the nymphs considered the main vector stage, is a challenge in face of several hundreds to thousands of ticks that should be examined for quantitative studies. We herein present additional morphological features to an existing identification key for Amblyomma nymphs that considerably diminishes the labor to distinguish nymphs of these two tick species. Specifically, the oval-shaped scutum of A. dubitatum and a conspicuous scapula of A. sculptum are prominent discriminating features.

ABSTRACT

The study of intestinal microbiota in vector insects like triatomines is paramount in parasitology because many parasitic species inhabit the vector's gut. Although knowledge on the gut microbiota in various vectors of the parasitic flagellate Trypanosoma cruzi has grown, research efforts have focused on South American triatomines. This study reports the isolation of bacterial microbiota in the anterior and posterior gut of Meccus pallidipennis (a triatomine species endemic to Mexico) by culture, as well as its identification by phenotypic and biochemical tests and its quantification by counting colony-forming units. The study was performed on fifth-instar nymph and adult specimens of M. pallidipennis, either laboratory-bred or collected in the field and either infected or not with T. cruzi. Overall, 17 bacterial species were identified, with the genera Bacillus and Staphylococcus being the most prevalent regardless of the origin of the insects. No differences were observed in the number of bacterial species in the gut of laboratory-bred and field-collected insects, neither with respect to life stage or infection status. In general, the Shannon-Weaver diversity index was higher in non-infected insects than in infected ones. Further studies using non-culture methods are required to determine whether bacterial species diversity is modified by laboratory breeding.

The genera Macrogynoplax Enderlein, 1909 and Kempnyia Klapálek, 1914 are reported for the first time from Tocantins State, a new locality record for Brazil. The nymphs of M. matogrossensis Bispo & Neves (2005) and K. goiana Bispo & Froehlich (2004) are described. Additionally, we also describe the female of K. goiana.

Consequentemente, o aumento da transparência e da condutividade devido à barragem de Porto Primavera no rio Paraná pode estar favorecendo essas espécies de Ephemeroptera. Nossos resultados demonstram a importância da preservação do Parque nacional das várzeas do rio Ivinhema, principalmente para o gênero Guajirolus sp., o qual apenas foi registrado nessa região. ABSTRACT

The objective of this study was to assess, for the first time, the in vitro acaricidal activity of two topical formulations containing thymol, on immature stages of Rhipicephalus sanguineus sensu lato. For this purpose, two base formulations were prepared: an oil-in-water (O/W) emulsion and a hydroalcoholic solution, containing different thymol concentrations (0.5 to 20 mg/mL). We used the larval packet test for non-engorged larvae and nymphs, and the immersion test for engorged larvae and nymphs. For emulsion, a mortality rate of 94.2% was achieved at 0.75 mg/mL in non-engorged larvae. For engorged larvae, there was 95.0% mortality at 5.0 mg/mL. Non-engorged nymphs showed 83.3% mortality at 2.5 mg/mL, and for engorged nymphs, 86.0% mortality was verified at 5.0 mg/mL. For the hydroalcoholic solution, the mortality found for non-engorged larvae was 88.1% at 2.5 mg/mL. For engorged larvae, the highest mortality was 25.0% at 20 mg/mL; non-engorged nymphs had 91.0% mortality at 1.0 mg/mL and for engorged nymphs; the maximum value verified was 18.3% mortality at 20 mg/mL. Preliminary stability tests were carried, and the hydroalcoholic solution remained stable under all the conditions analyzed. The O/W emulsion showed signs of early instability at the concentration of 5.0 mg/mL. The results obtained indicate that the acaricidal activity of thymol, when included in the proposed formulations, was enhanced against non-engorged larvae with topical treatment in comparison with data in the literature. Although there were variations in toxicity between the different stages, these formulations are promising for future therapeutic use.

ResumenEn México se han registrado 29 especies del género Amnestus, de las cuales, seis se encuentran en el estado de Veracruz. Se describen e ilustran las características morfológicas de los estadios inmaduros de A. rugosus, incluyendo notas sobre su biología y comportamiento. Las ninfas y adultos fueron recolectados en los remanentes de la vegetación riparia del bosque mesófilo en la cuenca del río La Antigua, Veracruz. El muestreo se realizó en la zona montañosa del centro del estado y en la parte media de la cuenca. Se seleccionaron 12 sitios ubicados en cinco municipios, y se realizaron muestreos en época de lluvias (julio) y en secas (abril); las muestras fueron procesadas en embudos de Berlese-Tullgren y Winkler. Se recolectaron y revisaron 419 ejemplares, 320 adultos y 99 ninfas de A. rugosus. El tamaño y coloración del cuerpo y los patrones de puntuación son las principales diferencias entre los estados inmaduros de A. rugosus, A. ficus y A. morelensis.A. rugosus representa un nuevo registro para Veracruz.


Abstract:In Mexico there are 29 species of Amnestus, of which six species are from Veracruz. The morphological features and illustrations of immature stages of A. rugosus are presented. Notes about the biology and behavior were included. The nymphal and adult stages of A. rugosus were found in remnants of riparian vegetation of the mesophilous forest in La Antigua river basin, Veracruz, Mexico. Twelve sites in five localities were selected and the specimens were sampled in rainy (July) and dry (April) seasons of 2012. The samples were processed in Berlese-Tullgren funnel and Winkler. A total of 419 individuals, 320 adults and 99 nymphs of A. rugosus were revised. The size and coloration of the body and punctuation patterns the immature stages of A. rugosus, A. ficus, and A. morelensis are the main differences among these species. Abstract Brazilian floodplains have suffered great changes in their natural characteristics in recent decades, mainly in the flood pulse. The Upper Paraná River floodplain is one of the few places where are found remained areas in which such peculiar characteristics keep reflecting on its high biodiversity. Ephemeroptera nymphs are one of the higher density groups among benthic community, occurring in many water bodies like large rivers and secondary channels. We sought to understand which factors are needed for the species establishment and how much important is the species colonization, especially in environments with anthropogenic changes. The marginal areas, which are more structured with presence of macrophytes, showed the highest density and richness even in the Paraná River that has great human impact. We verified dominance of Americabaetis alphus, Tricorythopsis araponga, Tricorythopsis artigas on the Parana River, correlated with transparency, depth and electric conductivity, while the dominance of Traverella sp. was correlated with water temperature, especially in marginal areas. Consequently, the increasing transparency and electric conductivity due to the Porto Primavera dam in Parana River can be favoring those Ephemeroptera species. We demonstrated the importance of preserving the wetlands of Ivinhema River State Park mainly for Guajirolus sp., which was only registered in this region. Therefore, our study provides support for understanding gaps from previously studies using artificial substrates in three large rivers which are of great importance to the upper Paraná River floodplain.


Resumo Muitas planícies de inundação brasileiras vêm sofrendo enormes alterações em suas características naturais nas últimas décadas, principalmente no pulso de inundação. A planície de inundação do alto rio Paraná é um dos poucos lugares onde ainda há áreas nas quais essas características peculiares se mantêm, o que reflete em sua alta biodiversidade. Dentre a comunidade zoobentônica, Ephemeroptera é um dos grupos com maior densidade, ocorrendo em diversos corpos aquáticos como rios de grande porte e canais secundários. Nós buscamos compreender quais fatores são necessários para o estabelecimento das espécies e o quão importante é a colonização dessas espécies, principalmente em ambientes com modificações antrópicas. As áreas marginais mais estruturadas com presença de macrófitas apresentaram altas densidade e riqueza mesmo no rio Paraná que apresenta grande impacto antrópico. Verificamos que as dominâncias de Americabaetis alphus, Tricorythopsis araponga e Tricorythopsis artigas no rio Paraná foram correlacionadas com maiores valores de transparência, profundidade e condutividade, enquanto que a dominância de Traverella sp. foi correlacionada com temperatura, principalmente em áreas marginais. Consequentemente, o aumento da transparência e da condutividade devido à barragem de Porto Primavera no rio Paraná pode estar favorecendo essas espécies de Ephemeroptera. Nossos resultados demonstram a importância da preservação do Parque nacional das várzeas do rio Ivinhema, principalmente para o gênero Guajirolus sp., o qual apenas foi registrado nessa região. Portanto, nosso estudo vem trazer subsídios para compreensão de lacunas verificadas em estudos anteriores utilizando substratos artificiais em três rios de enorme influência na planície de inundação do alto rio Paraná.

ABSTRACT

Plant extract have been recognized as important natural source of insecticide. Herein, we assessed the toxicity of leaves-extract and seeds-extract of Annona mucosa (Annonaceae) against brown stink bug, Euschistus heros (F.) (Hemiptera: Pentatomidae), a soybean pest in Neotropical America. For this, bioassays with nymphs and adults were conducted using the concentrations of 5, 10, 20, 40 and 80 mg.mL-1 of leaves and seeds extract of A. mucosa and DMSO (10%), as control. Ten replicates were conducted by treatment, with five nymphs (3rd instar) by replicate (bioassay with nymphs) and one couple of adults (< 24h of old) by replicate (bioassay with adults). The number of nymphs death (mortality) and survival of adults were assessed. For adults the fecundity (number of eggs) and fertility (number of nymphs hatched) of females also were assessed. The nymphs bioassay has promising results in seeds extract, when compared to leaves extract. The mortality of nymphs treated with seeds extract was dependent-concentration, with increased in mortality in concentrations > 5.0mg.mL-1, while nymphs treated with leaves extract do not exhibit difference between treatments. In adults the survival was reduced, when were treated with seed extract of A. mucosa at concentrations higher than 20mg.mL-1, with reduction up to 92 days in contrast to control. A drastic reduction in the fecundity and fertility was observed, which highlight deleterious action of seed extract against reproductive potential of E. heros. This is the first report about toxicity A. mucosa (leaves and seed) on E. heros, and our results suggest that seeds extract of A. mucosa is promising for control of nymphs and adults of brown stink bug.


Extratos de plantas tem sido reconhecidos como um importante recurso natural de inseticidas. Nesta pesquisa avaliou-se a toxicidade do extrato de folhas e de sementes de Annona mucosa (Annonaceae) sobre o percevejo marrom, Euschistus heros (F.) (Hemiptera: Pentatomidae), uma praga-chave na cultura da soja na America Neotropical. Para tanto, bioensaios com ninfas e adultos foram conduzidos usando as concentrações de 5, 10, 20, 40 e 80 mg.mL-1 do extrato de folhas e sementes de A. mucosa e DMSO (10%), como controle. Dez repetições foram conduzidas por tratamento, com cinco ninfas (3º instar) por repetição (bioensaio com ninfas) e um casal de adultos (< 24h de idade) por repetição (bioensaio com adultos). Foram avaliados o número de ninfas mortas (mortalidade) e a sobrevivência dos adultos. Para fêmeas também foi avaliada a fecundidade (número de ovos) e fertilidade (número de ninfas eclodidas). O extrato de semente de A. mucosa apresentou resultados promissores na mortalidade de ninfas de E. heros, quando comparado ao extrato de folhas. A mortalidade de ninfas tratadas com extrato de semente foi dependente da concentração, com aumento da mortalidade em concentrações > 5,0mg.mL-1, enquanto as ninfas tratadas com extrato de folhas não apresentaram diferença entre os tratamentos. A sobrevivência dos adultos foi reduzida, quando estes foram tratados com extrato de semente de A. mucosa em concentrações maiores que 20mg.mL-1, com redução de até 92 dias, em contraste ao controle. Foi observada uma drástica redução na fecundidade e fertilidade, o que destaca a ação deletéria do extrato de sementes sobre o potencial reprodutivo de E. heros. Este é o primeiro registro sobre a toxicidade de A. mucosa (folhas e sementes) sobre E. heros e os resultados sugerem que o extrato de semente de A. mucosa é promissor para o controle de ninfas e adultos do percevejo marrom.

In the present study, we provide morphological descriptions of the nymph of Amblyomma parvitarsum, A. tonelliae, and redescriptions of A. argentinae and A. sculptum. A taxonomic key, with relevant morphological characters illustrated by scanning electron micrographs, is provided for nymphs of the 24 species of the genus Amblyomma occurring in Argentina. Species included are A. argentinae, A. aureolatum, A. auricularium, A. boeroi, A. brasiliense, A. calcaratum, A. coelebs, A. dissimile, A. dubitatum, A. incisum, A. longirostre, A. neumanni, A. nodosum, A. ovale, A. parvitarsum, A. parvum, A. pseudoconcolor, A. pseudoparvum, A. rotundatum, A. sculptum, A. tigrinum, A. tonelliae, A. triste and A. varium. Principal morphological characters used for discrimination among species are presence/absence of auriculae, cornua and festoons with tubercles, size and shape of spurs of coxa I, margin and punctations of scutum, shape of basis capituli and length of cervical grooves. The geographical distribution of each tick species included in this work is presented and the importance of an accurate determination to species level of the Amblyomma nymphs to make epidemiological inferences is also discussed.

Rhipicephalus sanguineus is a species of tick which is widely distributed in America, Africa and Australia and is probably the most prevalent among all the other ixodid tick species. The present study demonstrated the effects of the arthropod growth regulator fluazuron (Acatak(®)), in the formation of the integument and the digestive processes of R. sanguineus nymphs fed on rabbits treated with different doses of this chemical acaricide. For this, three different doses of fluazuron (20mg/kg, 40mg/kg and 80mg/kg) were applied "pour on" to the hosts divided into three different treated-groups (II, III, IV) of three animals each. A fourth group (I) of rabbits (n=3) was given distilled water as control. On the first day after treatment (24h), the hosts were artificially infested with R. sanguineus nymphs. After full engorgement (7 days), the nymphs were removed and placed on labeled Petri dishes and kept in BOD incubator for 7 days. The engorged nymphs were then taken to ultra-structural analysis. Results revealed the following main ultra-structural changes in the nymphs integument and midgut of the different treated groups (II, III, IV): cuticle disorganization and the absence of subdivisions, damages in the integument epithelial cells, size of digestive cells, amount of endosomes, autophagic and digestive vacuoles, accumulated digestive residues, lipid droplets and organelles found in the digestive cells' cytoplasm, as well as the presence of microvilli in their plasma membranes. It is concluded that fluazuron may act on the integument and midgut cells of R. sanguineus engorged nymphs by impairing the synthesis of the new cuticle and the digestive processes (absorption of the blood ingested from the host, digestion - hemolysis, formation of digestive residues and release of nutrients to be converted into lipid, as well as for the synthesis of structural protein), which interfere in the development of nymphs, being able to prevent the emergence of adults after periodical ecdysis. These data indicate the possibility to use this arthropod growth regulator (AGR) in the control of R. sanguineus, at least in the nymphal stage of its biological cycle.

ABSTRACT

Chinavia Orian (1965) is one of the most diverse genera of Pentatomidae, distributed in the Afrotropical, Neotropical and Nearctic Regions. Thirty-two species are recorded for Brazil, some of them having potential economic impact because they are found on crops and referred to as pests. The morphology of the five nymphal instars of Chinavia armigera (Stål, 1859), Chinavia aseada (Rolston, 1983), Chinavia brasicola (Rolston, 1983) and Chinavia runaspis (Dallas, 1851) are described here. Through a comparative study, identification keys were developed to allow an early identification of the 12 Chinavia species of Rio Grande do Sul.

ABSTRACT

Com o objetivo de apresentar um levantamento da fauna de Ephemeroptera no Estado do Mato Grosso, investigamos as ninfas de Ephemeroptera, principalmente de duas bacias hidrográficas do leste do estado. Sessenta e sete espécies/morfoespécies em 41 gêneros e oito famílias foram encontradas. Quatro espécies (Cloeodes redactus Waltz & McCafferty, 1987 e Waltzoyphius roberti Thomas & Peru, 2002 [Baetidae], Tricorytopsis chiriguano Molineri, 2001 [Leptohyphidae] e Microphlebia surinamensis Savage & Peters, 1983 [Leptophlebiidae]) foram registradas pela primeira vez no Brasil. As famílias Euthyplociidae, Polymitarcyidae e Coryphoridae foram registradas pela primeira vez para o Mato Grosso, assim como 19 gêneros e 18 espécies. Vinte e cinco espécies não foram identificadas devido à falta de informação envolvendo o estágio ninfal de seus respectivos gêneros. Assim como demonstrado em outros trabalhos com o grupo, a ordem se mostrou mais diversa do que se conhece atualmente, especialmente se considerarmos as várias fisionomias e bacias hidrográficas do estado que se mantêm desconhecidas.


Aiming to present a survey of Ephemeroptera fauna of Ephemeroptera from Mato Grosso State we investigated mayfly nymphs mainly from two hydrographic basins located in the eastern area of the state. Sixty-seven species/morphospecies in 41 genera and eight families were encountered. Four species (Cloeodes redactus Waltz & McCafferty, 1987 and Waltzoyphius roberti Thomas & Peru, 2002 [Baetidae], Tricorytopsis chiriguano Molineri, 2001 [Leptohyphidae] and Microphlebia surinamensis Savage & Peters, 1983 [Leptophlebiidae]) are recorded for the first time from Brazil. The families Euthyplociidae, Polymitarcyidae and Coryphoridae are recorded for the first time from Mato Grosso, as well as 19 genera and 18 species. Twenty-five species could not be identified due to the lack of knowledge regarding the nymphal stage of their respective genera. As demonstrated in others surveys of mayflies in Brazil, the order is much more diverse than currently recognized, especially if we consider that several physiognomies and hydrographic basins of the state remain undocumented.

ABSTRACT

Together with the larval stage, the nymphal stage of ticks of the genus Amblyomma are the most aggressiveticks for humans entering areas inhabited by wildlife and some domestic animals in Brazil. However, due to the absence of morphological descriptions of the nymphal stage of most Brazilian Amblyomma species, plus the lack of an identification key, little or nothing is known about the life history of Amblyomma spp. nymphs in the country. In the present study, morphological description of the nymphal stage,illustrating important external characters through scanning electron microscopy, is provided for nymphsof 15 Amblyomma species that occur in Brazil, for which the nymphal stage had never been described: A. aureolatum, A. auricularium, A. calcaratum, A. coelebs, A. fuscum, A. humerale, A. incisum, A. latepunctatum, A. naponense, A. nodosum, A. ovale, A. pacae, A. pseudoconcolor, A. scalpturatum, A. varium. In addition, the nymphal stage of 12 Amblyomma species, which had been previously described, are redescribed: A. brasiliense, A. cajennense, A. dissimile, A. dubitatum, A. longirostre, A. oblongoguttatum, A. parkeri, A. parvum, A. romitii, A. rotundatum, A. tigrinum, A. triste. The descriptions and redescriptions totalized 27 species. Only 2 species (A. geayi, A. goeldii) out of the 29 Amblyomma species established in Brazil are not included in the present study. A dichotomous identification key is included to support taxonomic identification of the nymphal stage of 27 Amblyomma species established in Brazil.
