ABSTRACT

Inflammation can regulate hepatic drug metabolism enzymes and transporters. The impact of inflammation on renal drug transporters remains to be elucidated. We aimed to quantify the effect of inflammation (caused by acute pyelonephritis) on the in vivo activity of renal OAT1/3, using the probe drug furosemide. Pregnant women (second or third trimester) received a single oral dose of furosemide 40 mg during acute pyelonephritis (Phase 1; n = 7) and after its resolution (Phase 2; n = 7; by treatment with intravenous cefuroxime 750 mg TID for 3-7 days), separated by 10 to 14 days. The IL-6, IFN-γ, TNF-α, MCP-1, and C-reactive protein plasma concentrations were higher in Phase I vs. Phase II. The pregnant women had a lower geometric mean [CV%] furosemide CLsecretion (3.9 [43.4] vs. 6.7 [43.8] L/h) and formation clearance to the glucuronide (1.1 [85.9] vs. 2.3 [64.1] L/h) in Phase 1 vs. Phase 2. Inflammation reduced the in vivo activity of renal OAT1/3 (mediating furosemide CLsecretion) and UGT1A9/1A1 (mediating the formation of furosemide glucuronide) by approximately 40% and 54%, respectively, presumably by elevating the plasma cytokine concentrations. The dosing regimens of narrow therapeutic window OAT drug substrates may need to be adjusted during inflammatory conditions.

ABSTRACT

The objective of this study was to produce cellulose acetate (CA) from oat (OH) and soybean (SH) hulls through an eco-friendly solvent-and catalyst-free hydrothermal process, and also to compare the acetylation of cellulose by the conventional synthesis process, employing sulfuric acid as the catalyst, and acetic acid as solvent. Cellulose was extracted from OH and SH using a one-step chlorine-free process, resulting in materials with 86 and 81 % cellulose, respectively. CA samples obtained by hydrothermal process had degrees of substitution ranging from 0.95 to 1.47, and from 1.10 to 1.50, for OH and SH samples, respectively, being classified as monoacetates, while the conventional acetylation resulted in cellulose di- and triacetates. The acetylation by the hydrothermal process did not affect cellulose fibers morphology or crystallinity. CA samples obtained by the conventional process showed changes in their surface morphology, with decrease in crystallinity indexes. Viscosimetric average molar mass increased for all modified samples, with mass gain ranging from 16.26 to 519.70 %. The hydrothermal treatment proved to be a promising process for obtaining cellulose monoacetates with some advantages, including short reaction times, it is a one-step process, and lower effluents generation when compared to the conventional processes.

ABSTRACT

Probenecid is an old uricosuric agent used in clinics to treat gout and reduce the renal excretion of antibiotics. In recent years, probenecid has gained attention due to its ability to interact with membrane proteins such as TRPV2 channels, organic anion transporters, and pannexin 1 hemichannels, which suggests new potential therapeutic utilities in medicine. Some current functions of probenecid include their use as an adjuvant to increase the bioavailability of several drugs in the Central Nervous System (CNS). Numerous studies also suggest that this drug has important neuroprotective, antiepileptic, and anti-inflammatory properties, as evidenced by their effect against neurological and neurodegenerative diseases. In these studies, the use of probenecid as a Panx1 hemichannel blocker to reduce neuroinflammation is highlighted since neuroinflammation is a major trigger for diverse CNS alterations. Although the clinical use of probenecid has declined over the years, advances in its use in preclinical research indicate that it may be useful to improve conventional therapies in the psychiatric field where the drugs used have a low bioavailability, either because of a deficient passage through the blood-brain barrier or a high efflux from the CNS or also a high urinary clearance. This review summarizes the history, pharmacological properties, and recent research uses of probenecid and discusses its future projections as a potential pharmacological strategy to intervene in neurodegeneration as an outcome of neuroinflammation.

ABSTRACT

This study aimed to evaluate the effects of dietary fibers (apple, oat, pea, and inulin) in meat loaves treated with papain enzyme. In the first step, dietary fibers were added to the products at the level of 6%. All dietary fibers decreased the cooking loss and improved the water retention capacity throughout the shelf life of the meat loaves. Besides, the dietary fibers increased the compression force of meat loaves treated with papain, mainly oat fiber. The dietary fibers decreased the pH, especially the treatment with apple fiber. In the same way, the color was changed mainly by the apple fiber addition, resulting in a darker color in both raw and cooked samples. TBARS index increased in meat loaves added with both pea and apple fibers, mostly for the last one. In the next step, the combination of inulin, oat, and pea fibers was evaluated in the meat loaves treated with papain, combining fibers up to 6% total content likewise decreased cooking and cooling loss and increased the texture of the papain-treated meat loaf. The addition of fibers improved the acceptability of the texture-related samples, except for the three-fiber mixture (inulin, oat, and pea), which was related to a dry, hard-to-swallow texture. The mix of pea and oat fibers conferred the best descriptive attributes, possibly related to improved texture and water retention in the meat loaf, and comparing the use of isolated oat and pea, the perception of negative sensory attributes was not mentioned, such as soy and other off-flavors. Considering these results, this study showed that dietary fibers combined with papain improved the yielding and functional properties with potential technological use and consistent nutritional claims for elderly.

ABSTRACT

Oats are known for their nutritional value and also for their beneficial properties on human health, such as the reduction of cholesterol levels and risk of coronary heart disease; they are an important export product for Chile. During the last decade (2010-2022) over 90% of the oat cultivated area in Chile has been covered with Avena sativa L. cv. Supernova INIA. This lack of genetic diversity in a context of climate change could limit the long-term possibility of growing oats in Chile. The present study is a phenotypic and genetic analysis of 132 oat cultivars and pure lines of diverse origin that can be considered as potential breeding material. The germplasm was evaluated for 28 traits and analyzed with 14 SSR markers. The effects of genotypes on phenotype were significant over all traits (P ≤ 0.05). Most traits exhibited moderate to high broad-sense heritability with exceptions such as yield (H2 = 0.27) and hulls staining (H2 = 0.32). Significant undesirable correlations between traits were generally of small biological importance, which is auspicious for achieving breeding objectives. Some of the heritability data and correlations provided here have not been previously reported. The overall phenotypic diversity was high (H' = 0.68 ± 0.18). The germplasm was grouped into three phenotypic clusters, differing in their qualities for breeding. Twenty-six genotypes outperforming Supernova INIA were identified for breeding of conventional food-oats. The genetic diversity of the germplasm was moderate on average (He = 0.58 ± 0.03), varying between 0.32 (AM22) and 0.77 (AME178). Two genetic subpopulations supported by the Structure algorithm exhibited a genetic distance of 0.24, showing low divergence of the germplasm. The diversity and phenotypic values found in this collection of oat genotypes are promising with respect to obtaining genetic gain in the short term in breeding programs. However, the similar genetic diversity, higher phenotypic diversity, and better phenotypic performance of the germplasm created in Chile compared to foreign germplasm suggest that germplasm harboring new genetic diversity will be key to favor yield and quality in new oat cultivars in the long term.

ABSTRACT

The purpose of this study was to evaluate the performance of beef cattle with two levels of energy-protein supplementation of low-consumption on African Bermudagrass pasture, overseeded in winter with oat and ryegrass, with or without irrigation. Twenty-four castrated Angus steers (11 months old and had an initial average of 220 kg body weight (BW)) were used in experimental area of 3.6 ha. The experiment was completely randomized in a 2×2 factorial design, with three replicates. The evaluation period was 249 days (July/2019 to March/2020). The treatments were: irrigated pasture with supplementation of 1 g/kg BW or 2.7 g/kg BW and non-irrigated pasture with 1 g/kg BW of supplement or 2.7 g/kg BW. The grazing method was continuous with a variable stocking rate. Irrigation provided pastures with better chemical composition in winter and spring. Irrigation increased the daily accumulation rate in winter (84.6 vs. 45.9 kg DM/ha/day), providing a greater stocking rate (1,702 vs. 1,385 kg/ha) and, consequently, body weight gain per hectare. Supplementation of 2.7 g/kg BW provided a greater stocking rate in winter (1,652 vs. 1,435 kg/ha) and spring (3,096 vs. 2,811 kg/ha), not changing in summer. The association of irrigation and supplementation of 2.7 vs. 1.0 g/kg BW improves the intake pattern by the animal in summer without changing productivity parameters. Irrigation increases productivity and the nutritional value of pasture with higher livestock production per area in periods of water deficit. The supply of 2.7 vs. 1.0 g/kg BW provides a greater stocking rate and body weight gain per hectare.(AU)

ABSTRACT

The objective was to evaluate the ruminal disappearance rate of dry matter and NDF of black oat haylage subjected to mechanical and chemical dehydration and six wilting periods in the field, as follows: 0, 3, 6, 9, 12 and 15 days. Forage haylageswere ensiled and stored in PVC mini-silos for 60 days until opening for in situ evaluations. The pre-set ruminal incubation times were: 0, 6, 12, 24, 36 and 48 hours. Regardless of the dehydration time, forage subjected to mechanical dehydration showed ruminal disappearance rates of dry matter and NDF, thus DISDM-48h and DISNDF-48h were superior for this method compared to the chemical method. There was a reduction in DISDM-48h and DISNDF-48h with advancing time of wilting in the field for haylage dehydrated by the chemical method. In general, the chemical dehydration method seems to be more related to better haylage making practices than nutritional factors, where the haylage obtained by mechanical dehydration is superior.(AU)

Introdução: Mueslis de aveia contêm betaglucanas, uma fibra solúvel com propriedades hipocolesterolêmicas e hipoglicêmicas. Os benefícios do muesli de aveia podem ser alegados em produtos para atrair potenciais consumidores. Objetivos: Este estudo avaliou a influência de uma alegação funcional na aceitação de um muesli de aveia. Além disso, avaliou-se o efeito da aveia do muesli sobre as percepções de saciedade e humor. Métodos: Formularam-se dois produtos: muesli de aveia e muesli de flocos de arroz (controle), para a realização do estudo. Para os testes sensoriais, o muesli de aveia foi apresentado com duas codificações: uma com alegação de alimento funcional e outra sem alegação. Aos consumidores (n=160) foram abordadas questões sobre gosto, interesse de consumo e benefício antes e depois da degustação do mueslis, utilizando escalas hedônicas. Para entender os efeitos do muesli na saciedade e humor, foi realizado um ensaio clínico com 9 mulheres adultas saudáveis (aproximadamente 22 anos). Um desenho cruzado foi usado, com um período de washout de uma semana entre as intervenções. Os indivíduos preencheram um questionário de humor e saciedade usando escalas analógicas visuais antes e após consumir o mueslis no café da manhã. Resultados: O muesli de aveia com alegação foi apontado como o produto mais benéfico entre as amostras (p<0,05), apresentando maiores escores para gosto e interesse de uso. O consumo de muesli de aveia não influenciou o humor e a saciedade dos participantes pois não foram encontradas diferenças significativas entre as intervenções (p>0,05). Conclusão: A alegação funcional influenciou a aceitação dos benefícios do muesli de aveia para a saúde dos consumidores.


Background: Oats-based mueslis contain beta-glucans, a type of soluble fiber known for its hypocholesterolemic and hypoglycemic properties. The claim of these health benefits, oat muesli products may attract potential consumers. Objectives: This study aimed to assess the impact of a functional claim on consumers' acceptance and attitudes toward oat muesli. Additionally, the study evaluated the effects of muesli containing oats on satiety and mood perceptions. Methods: Two muesli products were formulated: one with oats and another with rice flakes (control) for sensory analysis. The oat muesli was presented with two codings (n=160): one with a functional food claim and another without the claim. Consumers were asked about their liking, interest in usage, and perceived benefits before and after tasting the mueslis using hedonic scales. Furthermore, a clinical trial was conducted with 9 healthy adult women subjects (average age 22 years) in a crossover design with a one-week washout period between interventions. The subjects completed a self-perceived mood and satiety questionnaire using visual analog scales (VAS) at Results: The oat muesli with the functional claim was perceived as the most beneficial product among the samples (p<0.05), receiving high scores for liking and interest in use. However, oat muesli consumption did not significantly influence participants' average mood and satiety during the trial (p>0.05), as no significant differences between interventions were found. Conclusion: The inclusion of a functional claim positively affected the acceptance of the health benefits associated with oat muesli.

ABSTRACT

The aim of the present study was to evaluate the effect of two types of nutritional supplementation during late gestation on the chemical composition, energy value, and IgG concentration in the colostrum and the IgG concentration in the blood serum of lambs. Pregnant Merino Precoz ewes (n = 36) carrying single fetuses were used. Animals were kept grazing on the Mediterranean annual grassland. From day ~90 of pregnancy, animals were allocated into three groups: daily supplementation with oat grain or lupine grain and a control group without supplementation. Immediately after parturition, colostrum was collected from each ewe, and a blood sample was taken from the lambs 24 h after birth. For the evaluation of the chemical composition of the colostrum, an EKOMILK® milk analyzer was used. The energy value of the colostrum was calorimetrically evaluated. IgG concentrations were measured by simple radial immunodiffusion. Data were analyzed by analysis of variance. Colostrum content of protein and non-fat solids was higher in the group supplemented with oat grain than in the lupine grain supplemented and control groups (p ≤ 0.05). In contrast, ewes supplemented with lupine grain had the highest concentration of fat in their colostrum (p ≤ 0.05). Oat grain supplementation resulted in higher concentrations of IgG, both in sheep colostrum and in the blood serum of their lambs (p ≤ 0.05), being higher than those observed in the lupine grain and control groups. Ewes that gave birth to male lambs had significantly higher concentrations of IgG in their colostrum compared to ewes that gave birth to females (p ≤ 0.05). The colostral IgG concentration positively correlated with the serum IgG concentration of the lambs (r = 0.32; p ≤ 0.05). The results indicate that the quality of colostrum and the immunological status of the newborn lambs can be improved by supplementation with oat grain.

BACKGROUND: The simultaneous administration of drugs with food can compromise the bioaccessibility and absorption of nutrients. The objective of this study was to evaluate the influence of the use of losartan potassium (LP), metformin hydrochloride (MH), and simvastatin (S) on the in vitro bioaccessibility of micronutrients (Cu, Fe, Mn, and Zn) in oat flour from Bahia, Brazil. METHODS: The experimental procedure consisted of the in vitro extraction of the bioaccessible fraction of Cu, Fe, Mn, and Zn in oat flour-with and without LP (50 mg), MH (500 mg), and S (20 mg)-using the unified bioaccessibility method (UBM), simulating the conditions of the gastrointestinal tract. For decomposition of the samples (oat flour and residue), a digester block with a closed system was used. To determine the total content (flour and residual fraction) and bioaccessible micronutrients, inductively coupled plasma optical emission spectrometry (ICP OES) was used. RESULTS: The bioaccessible contents (µg g-1) without the addition of drugs were: Cu 5.86 ± 0.21, Fe 32.80 ± 1.32, Mn 87.90 ± 1.90, and Zn 30.33 ± 2.05, with bioaccessibility ranging from 31.5 % for Fe to 99 % for Mn. The in vitro extraction method was validated by mass balance with recovery values from 89.78 % for Cu to 101.94 % for Mn. The range of bioaccessible contents (µg g-1) were: Cu (<4.14), Fe (32.10 ± 0.20-54.10 ± 2.03), Mn (81.40 ± 0.93-93.22 ± 0.80), and Zn (<10.80-29.11 ± 2.20). The estimation of the bioaccessibility of Cu, Mn, and Zn in oat flour were compromised in the presence of LP, MH, and S (p < 0.05). CONCLUSION: Chemical interactions can occur between drugs and micronutrients. Taken together, our results highlight that LP, MH, and S can interfere with the bioaccessibility of Cu, Fe, Mn, and Zn in oat flour in patients who use these drugs, suggesting its rational use in further investigations.

ABSTRACT

Current efforts to prevent dyslipidemia are focused on the development of functional products as an alternative for hypertriglyceridemia management. This study assessed the metabolic effect of the daily consumption of a bean and oats snack bar (BOSB) on hypertriglyceridemia biomarkers among Mexican women. An 8-weeks randomized parallel clinical trial (ID: NCT0496694, https://clinicaltrials.gov/ct2/show/NCT04966494) was conducted with 26 hypertriglyceridemic women allocated to BOSB group (TG = 208.18 ± 56.97 mg/dL) and control group (TG = 182.28 ± 51.39 mg/dL). Only the BOSB group consumed 50 g of the product per day. Fasting blood samples were taken from women with an adherence ≥ 90%. A targeted proteomic analysis with plasma samples of control and BOSB groups were conducted using a human obesity antibody array kit and bioinformatic tools provided by the Ingenuity Pathways Analysis (IPA) software. Serum TG levels in the BOSB group decreased by 37.80% (132.04 ± 27.83 mg/dL) compared with the control group (178.87 ± 32.01 mg/dL); glucose levels decreased by 5.69% in the BOSB group (87.55 ± 3.36 mg/dL). A modest body weight (5%) reduction was also found. Forty proteins were differentially modulated by the BOSB consumption (fold change > 1.2). The proteomic analysis revealed the involvement of BOSB bioactives in prevention of monocytes recruitment and localized inflammatory response, inhibition of pre-adipocyte maturation and adipogenesis, inhibition of hepatic b-oxidation, and potential satiety regulation. These results are promising since the mere intervention with the BOSB reduced serum TG without diet restriction, giving insights for further research in prevention of hypertriglyceridemia.

This study aimed to evaluate the effect of high-intensity ultrasound as pre-treatment in the development of fermented whey and oat beverages. Oat malt was produced, incorporated into a whey formulation (35, 50 and 65% v/v of whey) and ultrasonicated (at 40 kHz and 11 W/cm2) for 0, 3 or 10 min, prior to fermentation with L. casei 431. The treatments were identified as 35/65/0, 50/50/0, 65/35/0, 35/65/3, 50/50/3, 65/35/3, 35/65/10, 50/50/10 and 65/35/10, referring to the whey percentage, oat percentage, and the ultrasound time (min), respectively. The beverages 50/50/0 and 50/50/3 registered the highest (P < 0.05) growth with 1.96 and 2.00 log CFU ml, respectively. In general, the final average population of L. casei 431 was 7 to 8.86 log CFU/ml, being this adequate for a probiotic beverage. The highest antioxidant activity was found in the 35/65/3, 35/65/10, 50/50/3 and 50/50/10 beverages without difference (P < 0.05) among them. There was no effect of gender on the acceptance of the probiotic beverages. The best accepted beverage by women was 50/50/3 and both genders disliked the beverage 35/65/10. There was no relationship between the acceptance of the beverages and the consumers' habit by fermented milk beverages. No difference in the preference between the 50/50/0 and the 50/50/3 beverage was found. It is concluded that the probiotic beverage containing 50% whey and 50% oat and ultrasonicated for 3 min generated the highest levels of L. casei 431 growth, high antioxidant activity and good consumer acceptance and preference.

Bread is a highly consumed food whose nutritional value can be improved by adding an oat flour (Avena sativa L.-variety Bonaerense INTA Calen-Argentina) to a high-industrial quality wheat flour (Triticum aestivum L.). This cultivar of oat contains high amounts of ß-glucans, which act as a prebiotic fiber. Wheat flour was complemented with different amounts of oat flour (5, 15, and 25%). A contribution of hydrophilic components from oat flour was evident in the oat-wheat mixtures. At the same time, the high content of total dietary fiber led to changes in the rheological properties of the dough. Mixtures with a higher proportion of oats showed an increase in alveographic tenacity (stiffer dough), higher stability, and a lower softening degree in farinographic assays. The dough showed significant increases in hardness and gumminess, without significant changes in cohesiveness, i.e., no disruption to the gluten network was observed. Relaxation tests showed that the blends with a higher oat content yielded 10 times higher stress values compared to wheat dough. Analysis of the oat-wheat breads showed improvements in nutritional parameters, with slight decreases in the volume and crust color. The crumb showed significant increases in firmness and chewing strength as the amount of oats added increased. Nutritional parameters showed that lipids, dietary fiber, and ß-glucans were significantly increased by the addition of oats. Sensory analysis achieved high response rates with good-to-very good ratings on the hedonic scale set. Thus, the addition of oats did not generate rejection by the consumer and could be accepted by them. Breads with wheat and oats showed nutritional improvements with respect to wheat bread, since they have higher dietary fiber content, especially in ß-glucans, so they could be considered functional breads.

BACKGROUND: The effects of growth stage (GS) and ensiling were assessed on whole-crop oat's (Avena sativa L. cv. Cantara) chemical composition, silage fermentation quality and in situ ruminal degradability. Oat was harvested and ensiled at six GS: boot, heading, water ripe, early milk, early dough and grain ripe (144, 151, 178, 234, 362 and 512 g kg-1 of dry matter (DM) of whole-crop forage, respectively). RESULTS: GS influenced chemical composition, silage fermentation quality and ruminal degradability of whole-crop oat. Lower DM and higher water-soluble carbohydrates (WSC) contents lead to adequate forage compaction and fermentation at early GS; however, effluent was produced until the dough stage (L and Q; P ≤ 0.003). Advancing in maturity increased (P < 0.001) crop yield (4.5 to 9.4 t DM ha-1 ), DM (144 to 512 g kg-1 ), neutral detergent fibre (NDF; 537 to 571 g kg-1 DM), lignin (44.6 to 71.3 g kg-1 DM) and starch contents (26.4 to 201 g kg-1 DM), and reduced (P < 0.001) crude protein (107 to 60 g kg-1 DM) and WSC (115 to 17.5 g kg-1 DM). DM and NDF ruminal degradability declined with maturity for fresh and ensiled forages (L and Q; P < 0.05). Density and buffering capacity decreased with GS (L and Q; P < 0.001), whereas pH and soluble protein increased (L and Q; P ≤ 0.004). CONCLUSION: The growth stage of oat influenced the nutritive value and ruminal degradation to a greater extent than ensiling, and thus it can play a paramount role in whole-crop oat silage quality. © 2021 Society of Chemical Industry.

ABSTRACT

The hydrothermal carbonization (HTC) optimization of oat husk was performed using a response surface methodology. Furthermore, anaerobic digestion (AD) of spent liquor and hydrochar addition were evaluated in the biomethane potential (BMP) test. Results found that temperature influences the most in the studied responses (i.e., mass yield (MY) and higher heating value (HHV)). Optimal hydrochar MY (53.8%) and HHV (21.5 MJ/kg) were obtained for 219.2 °C, 30 min, and 0.08 of biomass/water ratio. A successful prediction capability of the optimization approach was observed, archiving an error < 1% between predicted and validated responses. The BMP experiment showed the feasibility of spent liquor as a potential substrate to be treated by AD (144 NmLCH4/gCOD). Hydrochar boosted the methane production of spent liquor increasing up to 17% compared to digestion with no hydrochar addition. These findings provide new insights regarding oat husk valorization by integrating HTC and AD for energy production.

RESUMEN El objetivo de este trabajo fue estudiar la fermentación láctica como bioestrategia para potenciar la actividad antioxidante de la avena. Se evaluó la capacidad de 31 cepas de bacterias lácticas (BL) para adaptarse a un sistema de avena/agua (SAA) mediante la determinación del crecimiento, la acidificación y la actividad fermentativa (método impedimétrico; tiempo de detección [DT], velocidad máxima de cambio de conductabilidad [VMCC] y porcentaje de cambio de conductabilidad [PCC]). Además, se determinó el contenido de compuestos fenólicos (CF) mediante el método de Folin-Ciocalteu (equivalentes de ácido gálico [EAG]), así como de péptidos, de aminoácidos libres y de actividad antirradicalaria (métodos DPPH•y ABTS•+) de los extractos metanólicos y acuosos obtenidos a partir de los SAA fermentados (SAAf). Seis cepas mostraron la mejor adaptabilidad al SAA y produjeron altos valores de VMCC (0,34-0,47 μS/min) y PCC (53,6-66,6%), con bajos valores de DT (≤ 3 h). Estos cambios fueron acompañados por modificaciones en la concentración de CF, péptidos y aminoácidos libres, los cuales fueron dependientes de la cepa. Se evidenció un incremento de la concentración de CF en los SAAf de las seis cepas (29,1-39,85 μg EAG/ml) con respecto al SAA control (17,1 μg EAG/ml). Además, hubo un incremento (9-25,5%) en la actividad antioxidante de los extractos metanólicos de SAAf medida por ambos métodos. Respecto del contenido en péptidos/aminoácidos libres de los SAAf y de su actividad antioxidante, se encontraron modificaciones de menor magnitud. Los resultados muestran que las BL son capaces de adaptarse a la avena como sustrato de fermentación e incrementar su contenido de compuestos antioxidantes.

Moreover, the content of phenolic compounds (PC) was determined using the Folin-Ciocalteu method (gallic acid equivalents [GAE]), free peptides and amino acids and free radical scavenging activity (DPPH•and ABTS•+methods) of methanolic and aqueous extracts obtained from fermented OWS/SAAs (fOWS/SAAf) were determined. Six strains have shown the best adaptability to SAA, with high values of VMCC (0.34-0.47 μS/min) and PCC (53.6-66.6%), and low values of DT (≤ 3 h). In these f/OWS/SAAf the chemical composition was also modified (PC concentration, peptides and free amino acids) with strain-dependent behavior. The PC content in f/OWS/SAAf using these six strains (29.1-36.9 μg GAE/ml) was higher than the control content in OWS/SAA control (17.1 ± 1.9 μg GAE/ml). An increase (9-25.5%) in antioxidant activity in f/OWS/SAAf methanolic extracts was detected using both methods. Minor modifications were observed in the peptide and free amino acid content of SAA and their antioxidant activity. Our results show LB ability to adapt to oat as fermentation substrate and increase the content of its antioxidant compounds.

ABSTRACT

Functional clinical nutrition is an integrative science; it uses dietary strategies, functional foods and medicinal plants, as well as combinations thereof. Both functional foods and medicinal plants, whether associated or not, form nutraceuticals, which can bring benefits to health, in addition to being included in the prevention and treatment of diseases. Some functional food effects from Avena sativa L. (oats), Linum usitatissimum L. (brown flaxseed), Glycine max L. (soya) and Moringa oleifera have been proposed for nutritional disorders through in vitro and in vivo tests. A formulation called a bioactive food compound (BFC) showed efficiency in the association of oats, flaxseed and soy for dyslipidemia and obesity. In this review, we discuss the effects of BFC in other nutritional disorders, as well as the beneficial effects of M. oleifera in obesity, cardiovascular disease, diabetes mellitus type 2, metabolic syndrome, intestinal inflammatory diseases/colorectal carcinogenesis and malnutrition. In addition, we hypothesized that a BFC enriched with M. oleifera could present a synergistic effect and play a potential benefit in nutritional disorders. The traditional consumption of M. oleifera preparations can allow associations with other formulations, such as BFC. These nutraceutical formulations can be easily accepted and can be used in sweet preparations (fruit and/or vegetable juices, fruit and/or vegetable vitamins, porridges, yogurt, cream, mousses or fruit salads, cakes and cookies) or savory (vegetable purees, soups, broths and various sauces), cooked or not. These formulations can be low-cost and easy-to-use. The association of bioactive food substances in dietary formulations can facilitate adherence to consumption and, thus, contribute to the planning of future nutritional interventions for the prevention and adjuvant treatment of the clinical conditions presented in this study. This can be extended to the general population. However, an investigation through clinical studies is needed to prove applicability in humans.

Introduction: Infections in hematological cancer patients are common and usually life-threatening; avoiding them could decrease morbidity, mortality, and cost. Genes associated with antineoplastics' pharmacokinetics or with the immune/inflammatory response could explain variability in infection occurrence. Objective: To build a pharmacogenetic-based algorithm to predict the incidence of infections in patients undergoing cytotoxic chemotherapy. Methods: Prospective cohort study in adult patients receiving cytotoxic chemotherapy to treat leukemia, lymphoma, or myeloma in two hospitals in Santiago, Chile. We constructed the predictive model using logistic regression. We assessed thirteen genetic polymorphisms (including nine pharmacokinetic-related genes and four inflammatory response-related genes) and sociodemographic/clinical variables to be incorporated into the model. The model's calibration and discrimination were used to compare models; they were assessed by the Hosmer-Lemeshow goodness-of-fit test and area under the ROC curve, respectively, in association with Pseudo-R2. Results: We analyzed 203 chemotherapy cycles in 50 patients (47.8 ± 16.1 years; 56% women), including 13 (26%) with acute lymphoblastic and 12 (24%) with myeloblastic leukemia. Pharmacokinetics-related polymorphisms incorporated into the model were CYP3A4 rs2242480C>T and OAT4 rs11231809T>A. Immune/inflammatory response-related polymorphisms were TLR2 rs4696480T>A and IL-6 rs1800796C>G. Clinical/demographic variables incorporated into the model were chemotherapy type and cycle, diagnosis, days in neutropenia, age, and sex. The Pseudo-R2 was 0.56, the p-value of the Hosmer-Lemeshow test was 0.98, showing good goodness-of-fit, and the area under the ROC curve was 0.93, showing good diagnostic accuracy. Conclusions: Genetics can help to predict infections in patients undergoing chemotherapy. This algorithm should be validated and could be used to save lives, decrease economic costs, and optimize limited health resources.

Oat (Avena sativa) is one of the most cultivated and consumed cereals worldwide. Recognized among cereals for its high protein content (12%-24%), it makes it an excellent source of bioactive peptides, which could be modified during processes such as heating and gastrointestinal digestion (GID). This work aims to evaluate the impact of heat treatment on the proteolysis of oat proteins and on the evolution of antioxidant peptide released during in vitro static GID, in terms of comparative analysis between cooked oat protein concentrate (COPC) and non-heated oat protein concentrate (OPC) samples. The protein extraction method and cooking procedure used showed no detrimental effects on protein quality. After GID, the proportion of free amino acids/dipeptides (<0.2 â€‹kDa) reached >40% for both samples (OPC and COPC), thus producing peptides with low molecular weight and enhanced bioactivity. Furthermore, during GID, the amino acid profile showed an increase in essential, positively-charged, hydrophobic and aromatic amino acids. At the end of GID, the reducing power of OPC and COPC increased >0.3 and 8-fold, respectively, in comparison to the non-digested samples; while ABTS•+ and DPPH• showed a >20-fold increase. Fe2+ chelating capacity of OPC and COPC was enhanced >4 times; similarly, Cu2+ chelation showed a >19-fold enhancement for OPC and >10 for COPC. ß-carotene bleaching activity was improved 0.8 times in OPC and >9 times in COPC; the oxygen radical antioxidant capacity assay increased 2 times in OPC and >4.7 times in COPC, respectively. This study suggests that OPC after cooking and GID positively influenced the nutritional and bioactive properties of oat peptides. Thus, COPC could be used as a functional food ingredient with health-promoting effects, as hydrothermal treatment is frequently used for this type of cereals.
