ABSTRACT

BACKGROUND: In light of the exponential rise in global population, there is a critical requirement to reduce food waste on a global scale. According to studies, agricultural wastes such as oil-seed cakes offer great nutritional value. Acid precipitation (A) and alkaline extraction methods (traditional methods) were used to extract protein from oil-seed cakes; however, both procedures are linked to decreased protein quality and quantity, which prompted the development of a novel strategy known as the biological/microbial/probiotic (B) method. Therefore, the present study aimed to highlight the optimal way of protein extraction from oil-seed cakes and the effect of extraction methods on protein efficacy against obesity. The outcomes were also compared with milk proteins. RESULTS: In vitro study provided evidence that proteins from both sources (plant and milk) suppressed adipogenesis and stimulated adipolysis in 3T3L-1 cells. For the in vivo study, mice were fed with different protein extracts: soya protein preparation (SPP), ground protein preparation (GPP), whey protein (WP) and casein protein (CP) containing 40% of their calories as fat. Body weight decreased significantly in all the rats except CP-fed rats. Body mass index, atherogenic index, plasma triglyceride and very-low-density lipoprotein cholesterol level decreased significantly in all the groups in comparison to the model group (high-fat-diet group), but the decrease was more pronounced in plant proteins than milk proteins. In hepatocytes, the expression of fasting-induced adipose factor, carnitine palmitoyltransferase I and peroxisome proliferator-activated receptor α genes was increased significantly in SPP-fed groups. Adiponectin gene expression was upregulated significantly in visceral fat tissue in groups fed SPP-B, GPP-A and CP, whereas leptin gene was downregulated significantly in all groups except SPP-A. CONCLUSION: This study demonstrates that SPP-B showed the most effective anti-obesity property, followed by WP. Additionally, we found that the biological precipitation approach produced better outcomes for plant proteins isolated from oil-seed cakes than the acid precipitation method. © 2023 Society of Chemical Industry.

ABSTRACT

BACKGROUND: Prodigiosin is a naturally occurring red pigment by Serratia marcescens and having enormous medicinal properties. Recently, there is a need to develop a high-throughput and economically feasible bioprocess for the production of prodigiosin. In order to find a cost-effective alternative to individual fatty acids as substrate in industries, we tried to study the effect of different fatty acid containing oil seed cakes of peanut, sesame, and mustard as sources of substrate. The present study screened waste and unconventional bioresources for the production of prodigiosin using S. marcescens ATCC 13880. Sources with high oil content were screened for maximum production of prodigiosin. Also, various parameters like temperature, pH, and nutrient precursors were screened and optimized for the production of prodigiosin. RESULTS: Scaled-up of optimized media consisting of 4% peanut oil seed cake powder, 2% sucrose, pH 7.5, temperature 28 °C, and 72 h incubation time resulted in highest production of 15.5 g/L wet biomass and 0.9 g/L of dried prodigiosin. Further, UV scan of the pigment showed maximum absorbance at 538 nm which is physiological property of the pigment. Extraction and purification of the pigment at the commercial level using the chromatographic techniques and mass spectral analysis confirmed the presence of prodigiosin. CONCLUSION: Using oil-extraction leftover wastes might help in the commercial and cost-effective production of prodigiosin.

ABSTRACT

Soil application of organics has been explored as an alternative means of organic management of plant-parasitic nematodes. Efficiency of different oil-seed cakes of neem (Azadirachta indica), castor (Ricinus communis), groundnut (Arachis hypogaea), linseed (Linum usitatissimum), sunflower (Helianthus annuus) and soybean (Glycine max) were evaluated in field conditions with association of Pseudomonas fluorescens in relation to growth parameters of chickpea and population of plant-parasitic nematodes. Their efficacious nature was highly effective in reducing the population of these dominant soil nematodes. Significant improvement was observed in plant-growth parameters such as plant weight, percent pollen fertility, pod numbers, root-nodulation and chlorophyll content of chickpea, seemed to be due to reduction in disease incidence and might be due to growth promoting substances secreted by P. fluorescens. The multiplication rate of nematodes was less in the presence of P. fluorescens as compared to its absence. Most effective combination of P. fluorescens was observed with neem cake.
