ABSTRACT

The intestinal epithelium dynamically controls cell cycle, yet no experimental platform exists for directly analyzing cell cycle phases in non-immortalized human intestinal epithelial cells (IECs). Here, we present two reporters and a complete platform for analyzing cell cycle phases in live primary human IECs. We interrogate the transcriptional identity of IECs grown on soft collagen, develop two fluorescent cell cycle reporter IEC lines, design and 3D print a collagen press to make chamber slides for optimal imaging while supporting primary human IEC growth, live image cell cycle dynamics, then assemble a computational pipeline building upon free-to-use programs for semi-automated analysis of cell cycle phases. The PIP-FUCCI construct allows for assigning cell cycle phase from a single image of living cells, and our PIP-H2A construct allows for semi-automated direct quantification of cell cycle phase lengths using our publicly available computational pipeline. Treating PIP-FUCCI IECs with oligomycin demonstrates that inhibiting mitochondrial respiration lengthens G1 phase, and PIP-H2A cells allow us to measure that oligomycin differentially lengthens S and G2/M phases across heterogeneous IECs. These platforms provide opportunities for future studies on pharmaceutical effects on the intestinal epithelium, cell cycle regulation, and more.

ABSTRACT

The objective of this paper was to determine the impact of EEG-biofeedback training on the motivation and efficiency of powerlifters during the bench press exercise in relation to the external load and the level of training. The study included 18 trained powerlifters who were divided into the intermediate (IG) and the advanced (AG) groups. EEG-biofeedback training was conducted every three days, lasting 27 minutes each time (5 × 3-minute intervals with recovery periods - lying on a bench - between them 4 × 3 minutes), and ended with a final EEG measurement in the second cycle of research. The repeated measures ANOVA showed intra-group differences due to external loading for the FAI (Frontal Alpha Asymmetry) obtained in the EEG both before and after biofeedback training. In AG group analysis revealed significant differences between 65%1RM and 35%1RM. In the IG group between 35%1RM and 50, 65 and 80%1RM. One of the major variables influencing the efficiency of strength training, including bench press workouts, is the level of training. The more successfully an athlete uses motivation when exercising, the better their training, which translates into greater results and a lower chance of injury.

ABSTRACT

This study aims to improve press equipment for safflower oil production by using a mechanism that optimizes pressure distribution within screw turns. A detailed analysis of the main components of the produced safflower oil was performed, encompassing both quantitative and qualitative assessments. Through the exploration of dependencies governing the safflower oil pressing process on the screw press, the optimal parameters were determined. As a result of the research, the optimal diaphragm gap between the gape cylinder and the pressing screw was determined, with the optimal oil yield percentage achieved at ω = 6.2 rad/s and δ = 5 mm. The study also compared the performance of the existing Dream Modern ODM-01 screw press and its upgraded version by analyzing the extracted oil. The results reveal changes in the quantitative and qualitative composition of the main oil components following the operation of the existing and the modernized screw presses. For instance, the amount of unsaturated fatty acids, such as oleic acid (7.7 ± 0.566%), linoleic acid (85.3 ± 1.185%), and linolenic acid (1.2 ± 0.223%), increased. There was an increase in the presence of inorganic substances in safflower oil: iron (0.023 ± 0.031 mg/kg), phosphorus (0.086 ± 0.059 mg/kg), silicium (0.136 ± 0.075 mg/kg), and others. The findings of this study hold significant commercial value and offer promising prospects for global market implementation.

ABSTRACT

BACKGROUND: To examine data quality and reproducibility using ISTHMUS, which has been implemented as the standardized MR spectroscopy sequence for the multi-site Healthy Brain and Child Development (HBCD) study. METHODS: ISTHMUS is the consecutive acquisition of short-TE PRESS (32 transients) and long-TE HERCULES (224 transients) data with dual-TE water reference scans. Voxels were positioned in the centrum semiovale, dorsal anterior cingulate cortex, posterior cingulate cortex and bilateral thalamus regions. After acquisition, ISTHMUS data were separated into the PRESS and HERCULES portions for analysis and modeled separately using Osprey. In vivo experiments were performed in 10 healthy volunteers (6 female; 29.5±6.6 years). Each volunteer underwent two scans on the same day. Differences in metabolite measurements were examined. T2 correction based on the dual-TE water integrals were compared with: 1) T2 correction based on the default white matter and gray matter T2 reference values in Osprey and 2) shorter WM and GM T2 values from recent literature. RESULTS: No significant difference in linewidth was observed between PRESS and HERCULES. Bilateral thalamus spectra had produced significantly higher (p<0.001) linewidth compared to the other three regions. Linewidth measurements were similar between scans, with scan-to-scan differences under 1 Hz for most subjects. Paired t-tests indicated a significant difference only in PRESS NAAG between the two thalamus scans (p=0.002). T2 correction based on shorter T2 values showed better agreement to the dual-TE water integral ratio. CONCLUSIONS: ISTHMUS facilitated data acquisition and post-processing and reduced operator workload to eliminate potential human error.

ABSTRACT

Enhancing the sensitivity of capacitive pressure sensors through microstructure design may compromise the reliability of the device and rely on intricate manufacturing processes. It is an effective way to solve this issue by balancing the intrinsic properties (elastic modulus and dielectric constant) of the dielectric layer materials. Here, we introduce a liquid metal (LM) hybrid elastomer prepared by a chain-extension-free polyurethane (PU) and LM. The synergistic strategies of extender-free and LM doping effectively reduce the elastic modulus (7.6 ± 0.2-2.1 ± 0.3 MPa) and enhance the dielectric constant (5.12-8.17 @1 kHz) of LM hybrid elastomers. Interestingly, the LM hybrid elastomer combines reprocessability, recyclability, and photothermal conversion. The obtained flexible pressure sensor can be used for detecting hand and throat muscle movements, and high-precision speech recognition of seven words has been using a convolutional neural network (CNN) in deep learning. This work provides an idea for designing and manufacturing wearable, recyclable, and intelligent control pressure sensors.

ABSTRACT

BACKGROUND: Biomedtrix BFX® cementless total hip replacement (THR) requires the use of femoral broaches to prepare a press-fit envelope within the femur for subsequent stem insertion. Current broaches contain teeth that crush and remove cancellous bone; however, they are not particularly well-suited for broaching sclerotic (corticalized) cancellous bone. In this study, three tooth designs [Control, TG1 (additional V-grooves), TG2 (diamond tooth pattern)] were evaluated with a quasi-static testing protocol and polyurethane test blocks simulating normal and sclerotic bone. To mimic clinical broaching, a series of five sequential broach insertions were used to determine cumulative broaching energy (J) and peak loads during broach insertion. To determine the effect of broach tooth design on THR stem insertion, a BFX® stem was inserted into prepared test blocks and insertion and subsidence energy and peak loads were determined. RESULTS: Broach tooth design led to significant differences in broaching energy and peak broaching loads in test blocks of both densities. In low density test blocks, TG1 required the lowest cumulative broaching energy (10.76 ±0.29 J), followed by Control (12.18 ±1.20 J) and TG2 (16.66 ±0.78 J) broaches. In high density test blocks, TG1 required the lowest cumulative broaching energy (32.60 ±2.54 J) as compared to Control (33.25 ±2.16 J) and TG2 (59.97 ±3.07 J).  During stem insertion and subsidence testing, stem insertion energy for high density test blocks prepared with Control broaches was 14.53 ± 0.81 J, which was significantly lower than blocks prepared with TG1 (22.53 ± 1.04 J) or TG2 (19.38 ± 3.00 J) broaches. For stem subsidence testing in high density blocks, TG1 prepared blocks required the highest amount of energy to undergo subsidence (14.49 ± 0.49 J), which was significantly greater than test blocks prepared with Control (11.09 ±0.09 J) or TG2 (12.57 ± 0.81 J) broaches. CONCLUSIONS: The additional V-grooves in TG1 broaches demonstrated improved broaching performance while also generating press-fit envelopes that were more resistant to stem insertion and subsidence. TG1 broaches may prove useful in the clinical setting; however additional studies that more closely simulate clinical broach impaction are necessary prior to making widespread changes to THR broaches.

ABSTRACT

PURPOSE: The J-difference edited γ-aminobutyric acid (GABA) signal is contaminated by other co-edited signals-the largest of which originates from co-edited macromolecules (MMs)-and is consequently often reported as "GABA+." MM signals are broader and less well-characterized than the metabolites, and are commonly approximated using a Gaussian model parameterization. Experimentally measured MM signals are a consensus-recommended alternative to parameterized modeling; however, they are relatively under-studied in the context of edited MRS. METHODS: To address this limitation in the literature, we have acquired GABA-edited MEGA-PRESS data with pre-inversion to null metabolite signals in 13 healthy controls. An experimental MM basis function was derived from the mean across subjects. We further derived a new parameterization of the MM signals from the experimental data, using multiple Gaussians to accurately represent their observed asymmetry. The previous single-Gaussian parameterization, mean experimental MM spectrum and new multi-Gaussian parameterization were compared in a three-way analysis of a public MEGA-PRESS dataset of 61 healthy participants. RESULTS: Both the experimental MMs and the multi-Gaussian parameterization exhibited reduced fit residuals compared to the single-Gaussian approach (p = 0.034 and p = 0.031, respectively), suggesting they better represent the underlying data than the single-Gaussian parameterization. Furthermore, both experimentally derived models estimated larger MM fractional contribution to the GABA+ signal for the experimental MMs (58%) and multi-Gaussian parameterization (58%), compared to the single-Gaussian approach (50%). CONCLUSIONS: Our results indicate that single-Gaussian parameterization of edited MM signals is insufficient and that both experimentally derived GABA+ spectra and their parameterized replicas improve the modeling of GABA+ spectra.

ABSTRACT

This report provides an updated analysis for patients with osteoporosis following total hip arthroplasty (THA). The comorbidities of alcohol abuse, chronic kidney disease, cerebrovascular disease, obesity, and rheumatoid arthritis continue to be significant risk factors for periprosthetic femur fracture (PPFFx) and aseptic loosening in the population with osteoporosis. Patients with dual-energy x-ray absorptiometric (DEXA) scans were at risk for PPFFx regardless of femoral fixation method, and patients with DEXA scans with cementless fixation were at risk of aseptic loosening after THA. The patient population with severe osteoporosis may have higher risks for aseptic loosening and PPFFx than previously recognized.

ABSTRACT

This article presents a framework to reduce energy consumption in a floor shop press based on Industrializable Industrial Internet of Things (I3oT). The I3oT proposes the development of IIoT tools using the information available in the system, without adding any additional sensors. Based on this philosophy, we proposed to develop the C360 criterion in our previous works, which allowed to extract all the information available in the stamping presses for the development of I3oT applications. In this article, we propose the development of a framework to optimize the parameters accessible from the C360 criterion for energy saving in the stamping process. Regarding the three parameters that can be modified and that affect energy consumption, that is, counterbalance pressure, tonnage and press speed, we will work with the first two in this paper. At the end of the article, the results obtained from the presses installed at Ford factory in Almussafes (Valencia) are shown based on their adjustment.

ABSTRACT

Athletic development centers on optimizing performance, including technical skills and fundamental motor abilities such as strength and speed. Parameters such as maximum contraction force and rate of force development, influence athletic success, although performance gains become harder to achieve as athletic abilities increase. Non-invasive transcranial direct current stimulation of the cerebellum (CB-tDCS) has been used successfully to increase force production in novices, although the potential effects in athletes remain unexplored. The present study examined the effects of CB-tDCS on maximum isometric voluntary contraction force (MVCiso) and isometric rate of force development (RFDiso) during a bench press task in well-trained athletes. 21 healthy, male, strength-trained athletes participated in a randomized, sham-controlled, double-blinded crossover design. Each participant completed the isometric bench press (iBP) task on two separate days, with at least 5 days between sessions, while receiving either CB-tDCS or sham stimulation. Electromyography (EMG) recordings of three muscles involved in iBP were acquired bilaterally to uncover differences in neuromuscular activation and agonist-antagonist co-contraction between conditions. Contrary to our hypothesis, no significant differences in MVCiso and RFDiso were observed between CB-tDCS and sham conditions. Furthermore, no tDCS-induced differences in neuromuscular activation or agonist-antagonist co-contraction were revealed. Here, we argue that the effects of CB-tDCS on force production appear to depend on the individual's training status. Future research should study individual differences in tDCS responses between athletes and novices, as well as the potential of high-definition tDCS for precise brain region targeting to potentially enhance motor performance in athletic populations.

Rice bran oil is one of oryzanol source oils. Oryzanol is an antioxidant compound that is related to the absorption of cholesterol, and is used in hyperlipidemia treatment and menopause problems. RBO extraction, purification and its γ-oryzanol content have been carefully reviewed. The quality and concentration of γ-oryzanol depend on the extraction process and purification. In selecting the extraction method to obtain the highest oryzanol content, in addition to comparing the concentration of oryzanol obtained and it can also be done by comparing the extraction kinetics parameters. Modeling according to physical or empirical kinetics can contribute in increasing the result of extraction. This study aims to determine the highest oryzanol content in rice bran oil, comparing several extraction methods and studies of rice bran oil extraction kinetic is necessary for scale up purposes. In this study is conducted Rice Bran Oil Extraction with n-Hexane solvent using several different methods, such as maceration, ultrasonication, and pneumatic press extractions. Independent variable that is used is the extraction time and yield as dependent variable. The study shows that the best extraction method to get the highest yield is 10.34 % by ultrasonicator and oryzanol content is 5.09 mg/g by a pneumatic press machine. According to kinetic parameter k2 is 0.001546, Cs is 0.0589, and h is 0.4707, R2 = 0.9715 obtained from extraction using ultrasonicator.

ABSTRACT

Pheochromocytomas are rare neuroendocrine tumors that can present as hypertensive crises or serious cardiac and cerebrovascular complications that endanger the patient's life. Two unusual cases of adrenergic crises induced by pheochromocytoma with cardiovascular manifestations are presented, one with multiple complications/multiorgan failure, fatal outcome and definitive diagnosis in the post mortem autopsy, and another with a satisfactory evolution after diagnosis and appropriate treatment.

Mean propulsive velocity (MPV) has been associated with neuromuscular fatigue; however, its suitability for strength training in Paralympic powerlifting (PP) remains uncertain. The objective of this work was to evaluate the MPV in two training methods (traditional-TRAD and eccentric-ECC). Eleven PP athletes were evaluated pre, during the intervention and post intervention at a load of 80% of the 1RM for TRAD and 110%-80% of 1 RM for ECC. The results demonstrated that there was no significant neuromuscular fatigue for the TRAD (~5% performance loss), as well as no significant decline in MPV during the intervention. For the ECC, there is a significant reduction in MPV before and after training (~12% loss of performance). A difference between TRAD and ECC after the intervention was also identified (0.87 m/s±0.22, 95% CI 0.72-1.02 vs. 0.72±0.20, 95% CI 0.59-0.86 p=0.042, F(3.30)=10.190, η2p=0.505 - very high effect). During the intervention for ECC, no significant decline in MPV was observed. The results of this study suggest that the mechanical indices of MPV do not seem to be effective indicators of neuromuscular fatigue in the sample studied or in the context of this specific training regime, being more indicated as a control of training volume.

ABSTRACT

In the present study, press mud (PM), a major waste by-product from sugar industries, was subjected to hydrothermal pretreatment (HTP) to create resource recovery opportunities. The HTP process was performed with the PM samples in a laboratory scale high pressure batch reactor (capacity = 0.7 L) at 160 °C and 200 °C temperatures (solids content = 5 % and 30 %). The pretreatment resulted in separation of solid and liquid phases which are termed as solid hydrochar (HC) and process water (PW), respectively. High heating value (HHV) of HC was âˆ¼14-18 MJ kg-1, slightly higher than that of PM (14 MJ kg-1). The thermogravimetric analysis showed about 1.5-1.7 times higher heat release from HC burning compared to that observed from combustion of PM. Apart from this, the HC and PM showed no phytotoxicity during germination of mung bean (Vigna radiata). Moreover, the biochemical methane potential test on the PW showed a generation of 167-245 mL biogas per gram of chemical oxygen demand added. Hence, the HTP offers several resource recovery opportunities from PM which may also reduce the risks of environmental degradation.

ABSTRACT

Despite the development of implant-supported prostheses, there are still patients for whom conservative treatments such as resin-bonded fixed dental prostheses (RBFDPs) are more appropriate. This study's objective was to analyze the available research on full-ceramic RBFDPs. In this study, Web of Science, MEDLINE/PubMed, Scopus, Embase, Cochrane Library, and Google Scholar databases were searched for articles published in English between 2010 and 2020. A total of 14 studies were reviewed based on the eligibility criteria. The results showed that using a cantilever design with one abutment had an advantage over two abutments. Additionally, it was proposed that preparations designed with retentive aids, such as a proximal box, groove, and pinhole, could improve RBFDP survival rates. IPS e.max ZirCAD, In-Ceram alumina, and zirconia CAD/CAM were the most commonly used framework materials. Most studies used air abrasion, salinization, or hydrofluoric acid for surface treatment. Adhesive resin cements were the most frequently used type of cement. The survival rate of In-Ceram ceramics (85.3%-94.8%) was lower than that of In-Ceram zirconia and IPS e.max ZirCAD. Debonding, followed by framework fracture, was the leading cause of failure. Following 3-10 years follow-up, the survival percentage of all-ceramic RBFDPs ranged from 76% to 100%. Although RBFDPs have demonstrated satisfactory success as a conservative treatment, long-term follow-ups and higher sample sizes in clinical research are required to gain more reliable outcomes on the clinical success rate of various RBFDP designs.

ABSTRACT

To promote the development of the 'Bamboo as a Substitute for Steel' proposal, rotary cut bamboo veneers were applied to prepare a kind of high-strength laminated bamboo composite, which was achieved through the hot press molding method in this study. Orthogonal experiments of L9 (33) were performed, with hot-pressing temperature, pressure, and time considered as three influencing factors. Physical properties like density and moisture content, and mechanical properties like modulus of rupture (MOR), modulus of elasticity (MOE), shear strength, and compressive strength were tested for the samples. It can be obtained from the results of range analysis and ANOVA that higher density and lower moisture content were correlated with higher mechanical strength. Within the selected range of tested factors, a hot-pressing temperature and time of 150 °C and 10 min can contribute to higher density and lower moisture content, and the combination of 150 °C and 50 MPa can produce greater mechanical strength. In the thickness direction, the laminated bamboo composites displayed a notable compressed structure.

ABSTRACT

Mineral extraction of coal in the carboniferous region of southern Santa Catarina (Brazil) plays a significant role in the regional economy. However, this activity has severe environmental impacts, with approximately 65% of the extracted mineral being discarded as a rejected material (deposited in landfills). The identification of the technological potential of these materials, based on the geological aspects of the extraction site and the beneficiation operations applied to obtain coal, provides the opportunity to add value to different residual fractions that can be reused. Thus, waste valorization, the main objective of this work, has recently become a strategy for the application of these minerals in the production of clay ceramics using a systematic approach named CPQvA, which means "classification, potentiality, quantity/viability, and applicability". The use of these materials as secondary mineral sources can avoid the deposition of these materials in industrial landfills and help to reduce the pressure on landfills, which receive an average of four million tons of material annually. In this study, the residual fraction, part of the tailing from coal beneficiation, known as coal fines, was evaluated for formulation valorization in clay ceramics. This residual fraction was classified as non-hazardous (class II-A, non-inert). X-ray fluorescence spectrometry, X-ray diffraction, and thermogravimetric analysis were performed to characterize the moisture content, particle-size distribution, and coal content to be used in the development of different formulations using the residual fraction of coal tailings (ranging from 0 to 40%) that are of technological interest to the sector. Processing parameters, such as firing at different temperatures (750, 800, 850, and 900 °C), were also correlated with these formulations. The results were compared with those of a reference ceramic formulation from the local productive arrangement of Morro da Fumaça (Arranjo Produtivo Local Cerâmica Vermelha de Morro da Fumaça). The various relationships between the materials were characterized in terms of their thermal shrinkage, water absorption, and mechanical resistance. Leaching and solubilization environmental tests revealed that both the industrial standard formulation and formulations with the application of the residual fraction were classified as non-hazardous materials. Thus, the method of using a mining residual fraction in the formulation of clay ceramics proved to be beneficial for the circular economy in the regional mineral sector through productive and environmental gains; the primary mineral resource and energy consumptions and the impacts related to waste generation were reduced. The results of this study can be applied to similar situations in other parts of the world.

ABSTRACT

Cold-pressed moringa, milk thistle, and jujube seed oils were investigated in terms of their characteristic profiles, thermal properties, and oxidative stability. The findings proved that the extracted oils were characterized by high nutritional values, which encourages their use in various fields. Results showed significant differences between the obtained oils. Overall, jujube seed oil exhibited the best quality parameters, with acidity equal to 0.762 versus 1% for the moringa and milk thistle seed oils. Milk thistle seed oil showed absorbance in the UV-C (100-290 nm), UV-B (290-320 nm), and UV-A (320-400 nm) ranges, while the moringa and jujube seed oils showed absorbance only in the UV-B and UV-A ranges. Concerning bioactive compounds, jujube seed oil presented the highest content of polyphenols, which promoted a good scavenging capacity (90% at 10 µg/mL) compared to the moringa and milk thistle seed oils. Assessing the thermal properties of the obtained oils showed the presence of four groups of triglycerides in the moringa and milk thistle seed oils, and two groups of triglycerides in the jujube seed oil. The thermograms were constant at temperatures above 10 °C for milk thistle seed oil, 15 °C for jujube seed oil, and 30 °C for moringa seed oil, which corresponded to complete liquefaction of the oils. The extinction coefficients K232 and K270, monitored during storage for 60 days at 60 °C, proved that jujube seed oil had the highest polyphenols content and was the most stable against thermal oxidation.

ABSTRACT

Objective: Although pediatric otolaryngology providers are reported to garner lower patient satisfaction than adults, this difference is not well characterized. This study investigates whether patient satisfaction differences exist in providers who treat both pediatric and adult patients. Study Design: Retrospective review. Setting: Tertiary medical center. Methods: In this cross-sectional study, Press Ganey surveys (PGS) completed by patients or parents on their first-time visit with 5 general otolaryngology providers from July 2014 to March 2022 were analyzed. Surveys were categorized by child (<18 years old) or adult and consisted of 14 items including 6 service domains of access, visit, nursing, provider, personal issues, and assessment. Analysis was performed with Walsh's t test and analysis of variance. Multivariable logistic regression, controlling for wait times and provider, evaluated the likelihood of highest satisfaction scores (HI-SCORES) based on age. Results: A total of 2549 patients (135 pediatric, 2414 adults) completed the PGS on their initial visit. There was no significant difference in the mean overall satisfaction scores between pediatric and adult patients. Further analysis of service domains among pediatric patients found the mean score in the access domain to be higher for the 6- to 11-year-old age group (0-5 years old: 85.5 ± 20.5 [mean ± SD], 6-11 years old: 94.7 ± 11.5, 12-17 years old: 87.3 ± 15.4, P = .03). Pediatric patients did not have a significantly higher likelihood (odds ratio = 1.1, 95% confidence interval: 0.8-1.6, P > .05) of reporting HI-SCORES compared to adults after covariate adjustment. Conclusion: There was no significant difference in patient satisfaction scores for providers who treat pediatric and adult patients utilizing the same facility and scheduling team.
