ABSTRACT

Serum vitamin D level and vitamin D receptor (VDR) gene FokI polymorphisms were detected in 200 single full-term pregnant women who gave birth in our hospital during 2016 to 2018. The newborns with birth weight<2500 g were diagnosed as fetal growth restriction (FGR), there were 100 cases in FGR group and 100 cases in control group. The average level of vitamin D in pregnant women in FGR group was significantly lower than that of the control group [(30.1±10.9) vs. (36.1±15.8) nmol/L, P<0.05]. In the FGR group, the birth weight of infants in mother carrying ff genotype was significantly lower than that in mother carrying Ff and FF genotypes [(2073±90) g vs. (2242±122) g and (2349±96) g, P<0.05]. Pregnant women carrying Ff and FF genotypes had lower risk of FGR than those of carrying ff genotype (ORFf=0.31, 95% CI: 0.17-0.76; ORFF=0.28, 95%CI: 0.11-0.46). The pregnant women with serum 25(OH)D level>30 nmol/L carrying F allele (FF+Ff) were set as reference, the risk of FGR in pregnant women with serum 25 (OH)D level ≤ 30 nmol/L carrying ff genotype was increased (OR=6.14, 95%CI: 2.13-13.23). The polymorphism of VDR gene FokI may be associated with the occurrence of FGR. In the case of vitamin D deficiency, the influence of ff genotype on FGR is more tangible.

ABSTRACT

Objective:To construct an expression vector containing RNA interfering(RNAi) sequence targeting human vitamin D_3 receptor(VDR) protein gene, and to investigate its inhibitory effect on the expression and function of VDR in a human osteosarcoma cell line. Methods: Two small interfering RNA sequences targeting VDR were designed by online screening and were cloned into the expression vector(pSilencer-2.1-U6).Then the products,pSilencer-2.1-U6-VDR1 and pSilencer-2.1-U6-VDR2,were separately transfected into a human osteosarcoma cell line(HOS-8603) by liposome.Western blot was employed to determine their effect on VDR protein expression and 1,25(OH)_2D_3-induced phosphorylation of PKC.Cell counting and MTT were used to evaluate their influence on the inhibitory effects of 1,25(OH)_2D_3 on HOS-8603 cells proliferation. Results: Compared to HOS-8603 cells transfected with control vectors, those transfected with pSilencer2.1-U6-VDR1 or pSilencer2.1-U6-VDR2 had significant decreases in the expression of VDR protein, in the inhibitory effects of 1,25(OH)_2D_3 on the cell proliferation, and in quick phosphorylation of PKC induced by 1,25(OH)_2D_3.Conclusion: The vectors constructed in the present study(pSilencer2.1-U6-VDR1 and pSilencer2.1-U6-VDR2) can significantly suppress the VDR expression in HOS-8603 cells, and block some biological activities of 1,25(OH)_2D_3 mediated by VDR, which paves a way for further study on the biological functions of VDR and vitamin D_3.

ABSTRACT

Objective To investigate the association of estrogen receptor-? (ER-?) and vitamin D receptor (VDR) gene polymorphisms with peak bone mass in Shanghai women. Methods The ER-? PvuⅡ and XbaⅠ genotypes and VDR ApaⅠ genotypes were determined by PCR-RFLP in 515 unrelated healthy women aged 19-40 years of Han nationality in Shanghai. Bone mineral density (BMD) was measured by dual-energy X-ray absorptiometry. Results Frequencies of ER-? PvuⅡ genotype PP, Pp and pp were 13.2%, 49.3% and 37.5% respectively. Frequencies of ER-? XbaⅠ genotype XX, Xx and xx were 4.7%, 40.4% and 54.9% respectively. Frequencies of VDR ApaⅠ genotype AA, Aa and aa were 5.8%, 41.9% and 52.3% respectively. Hardy-Weinberg equilibrium was evident for both ER-? and VDR gene polymorphisms. No association was found between ER-? PvuⅡ and XbaⅠ genotypes and BMD of various sites in women. Only a significant association was found between VDR ApaⅠ genotype and BMD at L 1-4(P
