ABSTRACT

OBJECTIVE: Present an approach to the safe and efficient provision of anesthesia and birth control measures to a large group of primates. ANIMALS: 98 hamadryas baboons (Papio hamadryas) held in a German zoological institution. METHODS: A group of 12 veterinarians, 2 zookeepers, and 6 volunteers anesthetized all animals within 2 days. The baboons were orally premedicated with midazolam (0.1 to 0.5 mg/kg) and anesthetized with medetomidine (40 to 60 µg/kg, IM) and ketamine (2 to 4 mg/kg, IM); isoflurane at rates of 1.5% to 2% was used for maintaining anesthesia if necessary. All animals received a physical examination, prophylactic medication, and tuberculin testing. For population management, the animals received a contraceptive implant (adult females), orchiectomy (young males), or vasectomy (breeding males). Young males received intratesticular blocks with lidocaine. All animals received atipamezole (125 to 150 µg/kg) before recovery. RESULTS: Premedication resulted in anxiolysis, which facilitated separating and darting. Median time from darting to access to the animal was 10 minutes. Mean anesthetic times were 25 minutes for females and 55 minutes for males. The depth of anesthesia was appropriate for the procedures. No fatalities were recorded. One animal was injured by other baboons but recovered after treatment. CLINICAL RELEVANCE: Health management and birth control measures are necessary in baboon troops under human care. Anesthesia and/or contraception of individual animals often leads to intraspecific aggression. This case series describes how to provide anesthesia and contraception to an entire troop as an alternative approach that can be adopted to future similar interventions.

ABSTRACT

Reproductive management significantly impacts dairy farm productivity, necessitating accurate timely pregnancy detection in cattle. This paper presents a novel handheld and portable fluorescence imaging system designed for quantitative assessment of pregnancy-specific biomarkers, addressing the limitations of current detection methods. The objective was to develop a cost-effective, at-farm solution for detecting pregnancy-specific protein B (PSPB) in bovine plasma samples. The system integrates an imaging module and a custom software application, enabling image capture, data processing, and PSPB concentration determination. Calibration utilizing known PSPB concentrations achieved a 0.6 ng/mL limit of detection. Validation encompassed a comparison with a standard ELISA method using 100 bovine plasma samples; minimal bias and good agreement were observed within the linear range of the calibration curve for both methods. The system offers portability, user-friendliness, and potential for multiplex detection, promising real-time, at-farm reproductive management. This study demonstrates the successful development and validation of a portable fluorescence imaging system, offering an efficient and accurate approach to detecting pregnancy-specific biomarkers in cattle. Its implications extend to improving dairy farm productivity by enabling timely and reliable reproductive management practices.

ABSTRACT

The objective of this study was to compare the reproductive performance of lactating dairy cows submitted to first AI after combination of estrus detection and fixed timed AI (FTAI) and FTAI only. Cows were randomly assigned to receive AI at detected estrus between 50 and 70 d in milk (DIM), if not detected in estrus, were enrolled in either Ovsynch (ED-Ov, n = 485) or PRIDsynch (ED-PR, n = 505) protocols; or received FTAI at 80 DIM after Double-Ovsynch protocol (DO, n = 501). Cows were body condition scored (BCS) at calving and at 43 DIM; and evaluated for postpartum disorders within 7 d postpartum; clinical mastitis, lameness and bovine respiratory disease were recorded until first AI. Ovarian cyclicity was monitored at 43 and 50 DIM, and at 70 and 77 DIM. Pregnancy diagnoses (PD) were performed at 32 and 63 d after AI. Overall prevalence of postpartum anovulation was 7.8%. Pregnancy per AI (P/AI) did not differ between reproductive strategies at 32 d PD (ED-Ov = 43.2%; ED-PR = 41.7%; DO= 45.3%). Primiparous cows had greater P/AI than multiparous cows (53.7% vs 36.8%). Cows on farm 1 had lower P/AI compared with their counterparts on farm 2 (42.1% vs 45.4%). Cows with BCS > 2.5 at 43 DIM had greater P/AI compared with cows with BCS ≤ 2.5 (44.5% vs 34.7%). Similar P/AI for cow's receiving AI at detected estrus and FTAI, low prevalence of disease anovulation may have contributed to the similar performance of ED-Ov, ED-PR and DO.

ABSTRACT

The objective of this randomized controlled experiment was to evaluate the effect of reproductive management programs that prioritized artificial insemination (AI) at detected estrus (AIE) or timed AI (TAI) during the first lactation on the economic performance of dairy cows of different genomically enhanced predicted transmitting ability for fertility. Lactating primiparous Holstein cows from 6 commercial farms were stratified into high, medium, and low fertility groups based on a reproduction index value calculated from multiple genomically enhanced predicted transmitting abilities to predict the number of days to achieve pregnancy. Within herd and fertility group, cows were randomly assigned either to a program that prioritized AIE (P-AIE; n = 1,416) and used TAI for cows not AIE for all AI services or another that prioritized TAI and had an extended voluntary waiting period for first service and prioritized TAI for second and greater AI services (P-TAI; n = 1,338). Cash flow (CF) per cow accumulated for the experimental (first) and second calving interval (CIN) and cash flow per slot per 28 mo after calving in the experimental lactation were calculated. Market and rearing heifer cost values were used for estimating CF. For cows in the high fertility group, a positive effect of delayed pregnancy on milk income during the first lactation was observed (+$248 for P-TAI) but was insufficient to generate significant differences in CF between treatments mainly because of milk income compensation in the second lactation (+$125 for P-AIE) and minor reductions in reproductive cost and gains in calf value for the P-AIE treatment. In this regard, CF for 2 CIN was greater for the P-TAI treatment by $61 and $86 for market and rearing replacement heifer cost, respectively. Similarly, CF per slot was favorable to the P-TAI treatment but only by $13 and $47 for market and rearing replacement heifer cost, respectively. For cows in the low fertility group, CF was numerically in favor of the P-AIE treatment due to a pregnancy and herd exit dynamics that resulted in gains in milk income over feed cost during the first ($29) and second ($113) lactation. Differences in CF for the 2 CIN were $58 and $47 for market or rearing heifer value, respectively, and $77 and $19 for market and rearing heifer values, respectively for the slot analysis. Differences in CF between cows of different genetic merit for fertility were consistent across treatment and estimation method. Of note, cows in the low fertility group had greater CF than cows in the high fertility group in all comparisons, ranging from $198 per cow for 2 CIN to as much as $427 per slot. For the low fertility group, greater milk production contributed directly (milk income over feed cost) and indirectly (reduced culling) to increased CF. We concluded that genetic merit for fertility and CF are associated because cows of inferior genetic potential for fertility had greater CF than cows of superior genetic for fertility despite some increased costs and reduced revenues. Also, the magnitude of the CF differences observed for cows of different genetic merit for fertility managed with the P-AIE or P-TAI program may be valuable to commercial dairy farms but did not allow to conclusively support the choice of a type of reproductive management strategy for cows of different genetic merit for fertility.

ABSTRACT

This analysis introduces a stochastic herd simulation model and evaluates the estimated reproductive and economic performance of combinations of reproductive management programs for both heifers and lactating cows. The model simulates the growth, reproductive performance, production, and culling for individual animals and integrates individual animal outcomes to represent herd dynamics daily. The model has an extensible structure, allowing for future modification and expansion, and has been integrated into the Ruminant Farm Systems model, a holistic dairy farm simulation model. The herd simulation model was used to compare outcomes of 10 reproductive management scenarios based on common practices on US farms with combinations of estrous detection (ED) and artificial insemination (AI), synchronized estrous detection (synch-ED) and AI, timed AI (TAI, 5-d CIDR-Synch) programs for heifers; and ED, a combination of ED and TAI (ED-TAI, Presynch-Ovsynch), and TAI (Double-Ovsynch) with or without ED during the reinsemination period for lactating cows. The simulation was run for a 1,000-cow (milking and dry) herd for 7 yr, and we used the outcomes from the final year to evaluate results. The model accounted for incomes from milk, sold calves, and culled heifers and cows, as well as costs from breeding, AI, semen, pregnancy diagnosis, and calf, heifer, and cow feed. We found that the interaction between heifer and lactating dairy cow reproductive management programs influences herd economic performance primarily due to heifer rearing costs and replacement heifer supply. The greatest net return (NR) was achieved when combining heifer TAI and cow TAI without ED during the reinsemination period, whereas the lowest NR was obtained when combining heifer synch-ED with cow ED.

ABSTRACT

This paper reports findings regarding patterns of reproductive management in 325 sheep flocks and 119 goat herds throughout Greece. The objectives were to describe the patterns of reproductive management in sheep and goat farms, to highlight factors that were associated with these management patterns and the clinical application of the various reproductive management approaches in the farms and to establish potential associations with production outcomes. The median months of the start of the mating period for adult sheep and goats were May and June, respectively and for ewe lambs and replacement goats these were August and September, respectively. The median duration of the mating period was 2 months for adults and 1 month for replacements. The median ratio of female to male animals was 22:1 and 25:1 in sheep and goat farms, respectively. Control of reproduction was applied in 33.2% and 16.8% of sheep and goat farms, respectively, mostly by intravaginal progestagen sponges and less often by melatonin implants; multivariable analysis indicated five (availability of milking parlour, number of ewes in a farm, number of daily milkings, daily period spent by farmers at the farm, farming tradition in the family) and two (number of daily milkings, availability of milking parlour) predictors in sheep and goat farms, respectively. Pregnancy diagnosis ultrasonographically was performed in 36.6% and 16.8% of sheep and goat farms, respectively; multivariable analysis indicated as significant three (management system applied in farms, age of farmer, farming tradition in the family) and two (management system applied in farms, availability of milking parlour) predictors in sheep and goat farms, respectively. The control of reproduction and pregnancy diagnosis were associated with a higher number of vaccinations during pregnancy: 2.6 and 2.7, respectively, versus 2.2 and 2.1 in farms where they were not performed. The average per farm number of lambs/kids born per female animal was 1.33 ± 0.01 and 1.30 ± 0.02 for sheep and goat farms, respectively; five (control of reproduction, location of the farm, presence of working staff, collaboration with veterinary practice, breed of ewes) and two (control of reproduction, breed of female goats) predictors were identified in sheep and goat farms, respectively, for high numbers of lambs/kids. Lambs/kids were taken away from their dams at the age of 50 and 65 days, respectively. The average culling age (females, males) was 5.9 and 4.4 years and 6.9 and 4.9 years for sheep and goats, respectively. Farmers sourced animals to be used as replacements for those removed from the farm, mostly from the animals in their own farms, considering criteria related to milk production. Finally, the application of the control of reproduction was associated with increased milk production and the number of newborns, whilst an inverse correlation between the culling age of animals and milk production was seen.

ABSTRACT

Pregnancy success represents a major issue for the economic income of cattle breeders. Early detection of pregnant and non-pregnant animals, as well as the prediction of early pregnancy failure, can influence farm management decisions. Several diagnostic tools for pregnancy are currently available. Among these, pregnancy-associated glycoproteins (PAGs) have been shown to be useful for identifying the presence of vital embryos and for pregnancy follow-up monitoring. This review presents an overview of the PAGs' functions, their pregnancy trends, and their use as a tool to improve reproductive management in bovine and other dairy livestock, such as small ruminants and buffalos.

ABSTRACT

Capybaras are the primary hosts of Amblyomma sculptum tick, vectors of Rickettsia rickettsia bacteria, and the zoonotic agent of Brazilian Spotted Fever (BSF). In this context, contraceptive methods have been suggested for population control in order to reduce the number of free-ranging capybaras cohabiting with humans in urban and rural areas and acting as disease amplifiers. To maintain the group's expected behavior and social hierarchy, sterilization techniques that preserve the gonads are recommended. On 126 female capybaras in the Brazilian state of São Paulo, a new surgical technique named "Passos Nunes" uterine horn ligature was performed after adequate general anesthesia. It achieved effective surgical sterilization, with an incision length of about 3 cm in the periumbilical linea alba, cranial to the pubis. After entering the abdominal cavity, the urinary bladder is pulled laterally to access the uterine horns and the cervix. The uterine horn is folded up, forming a strap; the distal portion of the strap is ligated and its distal end sectioned. The exact process is performed on the opposite horn. After the surgical procedure, the musculature is sutured in a sultan pattern and the subcutaneous tissue with a horizontal mattress pattern. The skin is sutured in a separate simple format, using nylon 2.0 for all steps. The wide exposure of the uterine horns facilitates the confirmation of pregnancy, allowing the surgeon to choose between salpingo hysterectomy or ligature of the uterine horns. The present study presents a new technique of surgical sterilization that can be used in female free-ranging mammals in which maintenance of the gonads is recommended, and births of offspring should not occur.

ABSTRACT

The primary objective of this randomized controlled experiment was to evaluate the insemination dynamic and reproductive performance of cows managed with a targeted reproductive management (TRM) program designed to prioritize artificial insemination (AI) at detected estrus (AIE) and optimize timing of AI by grouping cows based on detection of estrus during the voluntary waiting period (VWP). Our secondary objective was to evaluate reproductive outcomes for cows with or without estrus during the VWP. Lactating Holstein cows fitted with an ear-attached sensor for detection of estrus were randomly assigned to a TRM treatment that prioritized AIE based on detection of estrus during the VWP (TP-AIE; n = 488), a non-TRM treatment that prioritized AIE (P-AIE; n = 489), or an all timed AI (TAI) treatment with extended VWP (ALL-TAI; n = 491). In TP-AIE, cows with or without automated estrus alerts (AEA) recorded during the VWP received AIE if detected in estrus for at least 31 ± 3 or 17 ± 3 d after a 49 d VWP, respectively. Cows not AIE with or without AEA during the VWP received TAI after Ovsynch with progesterone supplementation and 2 PGF2α treatments (P4-Ov) at 90 ± 3 or 74 ± 3 d in milk (DIM), respectively. In P-AIE, cows received AIE if detected in estrus for 24 ± 3 d after a 49 d VWP, and if not AIE received TAI at 83 ± 3 DIM after P4-Ov. In ALL-TAI, cows received TAI at 83 ± 3 DIM after a Double-Ovsynch protocol. Data were analyzed by logistic and Cox's proportional hazard regression. The proportion of cows AIE did not differ for TP-AIE (71.0%) and P-AIE (74.6%). Overall P/AI at 39 d after first service was greater for the ALL-TAI (47.6%) than for the P-AIE (40.2%) and TP-AIE (39.5%) treatments. The hazard of pregnancy up to 150 DIM was greater for cows in TP-AIE (hazard ratio = 1.2; 95% confidence interval: 1.1-1.4) and P-AIE (hazard ratio = 1.2; 95% confidence interval: 1.1-1.4) than for cows in the ALL-TAI treatment which resulted in median time to pregnancy of 89, 89, and 107 d. Conversely, the proportion of cows pregnant at 150 DIM did not differ (ALL-TAI 78.5%, P-AIE 76.3%, TP-AIE 76.0%). Except for a few outcomes for which no difference was observed, cows detected in estrus during the VWP had better performance than cows not detected in estrus. Cows with AEA during the VWP were more likely to receive AIE, had greater P/AI, and greater pregnancy rate up to 150 DIM regardless of first service management. We conclude that a TRM program designed to prioritize AIE by grouping cows based on detection of estrus during the VWP was an effective strategy to submit cows for first service resulting in similar or improved performance than a non-TRM program that prioritized AIE or an all-TAI program with extended VWP. Also, AEA recorded during the VWP might be used as a strategy for identifying subgroups of cows with different reproductive performance.

ABSTRACT

Repeat breeding is a substantial problem in heifer and cow breeding leading to greater infertility for female dairy herds. The aim of present study was to investigate the impact of corpus luteum (CL) presence and category and the first gonadotropin-releasing hormone (GnRH) administration concurrent with exogenous progesterone (P4) treatment on the largest follicle (LF) size and pregnancy rate (PR) in repeat-breeder crossbred dairy heifers submitted to the fixed-time artificial insemination (AI) protocol. Heifers (n= 243) were synchronised with (+GnRH) or without (-GnRH) first GnRH in the 7-day P4-GnRH-prostaglandin F2α-based programme. Each GnRH group was divided on presence of CL into two groups (+CL and -CL) in a 2 × 2 factorial arrangement. The PR was similar among -GnRH-CL (20.7%), -GnRH+CL (68.8%), +GnRH-CL (30.4%), and +GnRH+CL (68.3%) groups. However, presence of CL in heifers produced a 43.6% increase in PR compared to PR of heifers without CL (odds ratio = 6.550). Heifers bearing large-sized CL had greater large-sized LF on the day of fixed-time AI and PR. Plasma P4 concentration was positively related with CL diameter (r= 0.845; p < 0.001). The diameter of ovarian LF on the day of fixed-time AI was positively associated with P4 concentrations (r= 0.512; p < 0.001). We highlight that ovarian CL presence and category at the time of exogenous P4 treatment alters pre-ovulatory follicle size and PR but not initial GnRH treatment in repeat-breeder crossbred dairy heifers submitted to service with the 7-day fixed-time AI programme.

ABSTRACT

Here we revise circumstances of non-infectious causes in which the uterus may be associated with pregnancy loss during the late embryo/early fetal period (following a positive pregnancy diagnosis in lactating dairy cows). As the uterine size increases with parity and pregnant heifers with no detrimental effects of a previous parturition, a primigravid uterus is proposed as a reference for identifying risk factors that negatively influence pregnancy in lactating cows. Cows suffering placenta retention or with a large uterus at insemination were selected as topics for this revision. Retained placenta, that occurs around parturition, has a long-lasting influence on subsequent pregnancy loss. Although retained placenta is a particularly predisposing factor for uterine infection, farm conditions along with cow factors of non-infectious cause and their interactions have been identified as main factors favoring this disorder. A large uterus (cervix and uterine horns lying outside the pelvic cavity) with no detectable abnormalities has been associated with low fertility and with a greater incidence of pregnancy loss. A large reproductive tract may well derive from an inadequate uterine involution. Therefore, peripartum management and strategies to reduce the incidence of uterine disorders should reduce their associated financial losses in the herds.

ABSTRACT

In cattle, early detection of gestation is very important from an economic and management point of view in all types of farming. However, due to the poor efficiency of oestrous detection, it is essential to determine non-pregnant cows as early as possible, in order to minimize the inter-insemination interval, thus de facto, reducing herd open days. Direct and indirect gestation diagnostic methods have been developed with the aim of improving the reproductive performance of the herd. Today, the most accurate method for making an early diagnosis of gestation from 28 to 30 days post-insemination is B-mode ultrasound. In recent years, indirect methods have included techniques that allow non-pregnant cows to be identified with a minimum margin of error, the most widely utilized of which is the colour Doppler. This technique is rapidly becoming established for the diagnosis of non-pregnancy, which allows for the identification of non-pregnant animals earlier compared with the pregnancy diagnosis. Some limitations of this technique in dairy cow have been presented.

ABSTRACT

This study aimed to determine common pregnancy rates and effect sizes with meta-analysis of studies investigating the effect of different synchronization protocols applied to sheep during breeding and non-breeding seasons on pregnancy rates. Common pregnancy rates were estimated by coalescing pregnancy rates of studies performed independently, and heterogeneity between the studies was investigated. The meta-analysis included 24 studies that determined pregnancy rates in 78 different groups consisting of 1934 sheep with five different synchronization protocols in Turkey between 2001 and 2020. Among the different synchronization methods, the P4+PMSG group (90.37%) during the breeding season and P4+PGF2α (69.77%) and P4 (68.75%) groups during the non-breeding season showed the highest pregnancy rate. Also, the effect size of P4+PMSG application during the breeding season was calculated as 0.934 (95% confidence interval: 0.901-0.967), and the effect size of P4+PGF2α application during the non-breeding season was calculated as 0.709 (95% confidence interval: 0.406-1.013). To conclude, the combination of P4+PMSG during the breeding season and progestogen and other hormone applications during the non-breeding season are the most effective methods for estrus synchronization and for achieving the desired pregnancy rates.

ABSTRACT

Reproductive efficiency of farm animals has central consequences on productivity and profitability of livestock farming systems. Optimal reproductive management is based on applying different strategies, including biological, hormonal, nutritional strategies, as well as reproductive disease control. These strategies should not only guarantee sufficient reproductive outcomes but should also comply with practical and ethical aspects. For example, the efficiency of the biological- and hormonal-based reproductive strategies is mainly related to several biological factors and physiological status of animals, and of nutritional strategies, additional factors, such as digestion and absorption, can contribute. In addition, the management of reproductive-related diseases is challenged by the concerns regarding the intensive use of antibiotics and the development of antimicrobial resistant strains. The emergence of nanotechnology applications in livestock farming systems may present innovative and new solutions for overcoming reproductive management challenges. Many drugs (hormones and antibiotics), biological molecules, and nutrients can acquire novel physicochemical properties using nanotechnology; the main ones are improved bioavailability, higher cellular uptake, controlled sustained release, and lower toxicity compared with ordinary forms. In this review, we illustrate advances in the most common reproductive management strategies by applying nanotechnology, considering the current challenges of each strategy.

ABSTRACT

Factors related to the thermal physiology and activity of Zebu animals close to calving are still unknown. The aims of this study were 1) to describe the pattern of reticulo-rumen temperature and activity variation in nulliparous Gyr heifers close to calving, and 2) to evaluate the predictive potential of these traits for calving in Gyr heifers. Forty pregnant Gyr heifers that had calved between August and December 2017 at the Getúlio Vargas Experimental Station, Empresa de Pesquisa Agropecuária de Minas Gerais (Epamig), Brazil, were used. The animals received a rumen bolus to monitor reticulo-rumen temperature (Trr) and activity (Act) at intervals of 10 min. Mixed linear models were used. A decrease in Trr and an increase in Act were observed on the days preceding calving. Differences in Trr and Act were more evident during the final 21 and 11 h previous to calving compared to 14 days before calving, measured at the same time of day. There was a decrease of about 0.20 °C in Trr at the time of calving when compared to baseline (14 days before calving measured at the same time of day). Environmental variables, such as temperature and air humidity, as well as biological factors such as circadian rhythm, may influence the interpretation of the data. In conclusion, variations exist in the Trr and Act of Gyr heifers in the hours before calving, which is preceded by a decrease in Trr and an increase in Act. Particularities in the thermal physiology of Zebu cattle must be considered when prediction devices previously validated only for European breeds are used.

ABSTRACT

Progesterone (P4) concentration during follicular growth has a major impact on fertility response in timed artificial insemination (TAI) protocols. Luteal presence at the beginning of a TAI protocol and ovarian response after the first gonadotropin-releasing hormone (GnRH) injection (G1) affect P4 concentration and subsequently pregnancy per artificial insemination (P/AI). A systematic review of the literature and meta-analytical assessment was performed with the objective of evaluating the magnitude of the effect of luteal presence and ovarian response at the beginning of a TAI protocol on P/AI in lactating dairy cows. We considered only studies using synchronisation protocols consisting of GnRH and prostaglandin F 2α. The time interval between G1 and prostaglandin F 2α (PGF 2α) had to range from 5 to 7 d. The time interval between the PGF 2α injection and G2 had to range from 48 to 72 h. We used 28 controlled experiments from 27 published manuscripts including 16,489 cows with the objective of evaluating the effect size of having a functional corpus luteum (CL) at G1 on P/AI. Information regarding ovulatory response after G1 was available for 5676 cows. In a subset of cows (n = 4291), information was available for luteal presence and ovulatory response at the initiation of the TAI protocol. A functional CL at G1 increased (p < 0.001) the relative risk of conceiving (RR (relative risk) = 1.32; 95% CI = 1.21-1.45) in lactating dairy cows. Ovulation after G1 increased (p < 0.001) the relative risk of conceiving (RR = 1.29; 95% CI = 1.20-1.38) in lactating dairy cows. The effect of ovulatory response on P/AI after G1 was affected by luteal presence at G1. In summary, there was a clear benefit on P/AI for cows starting a TAI protocol with a functional CL (+10.5 percentage units) and cows ovulating at the beginning of a TAI protocol (+11.0 percentage units).

ABSTRACT

Reproductive management practices that use hormones, sociosexual biostimulation, nutritional management, or abiotic factors are used to induce the resumption of reproduction in anestrous does. However, their overall efficacy remains uncertain; therefore, the identification of evidence-based management recommendations to manipulate anestrous in goats is important. Electronic databases were searched to retrieve reports on studies using interventions based on hormonal, sociosexual, nutritional, and abiotic factors. Only experimental studies in which a group of anestrous does was treated and compared against an untreated group were included. Estrus, ovulation, and pregnancy were primary outcomes, whereas the onset of estrus after treatment, the ovulation rate, and the number of anovulatory days were secondary outcomes. Odds ratio (OR) and mean differences were used to synthesize pooled data, and random effects models were used to calculate them. Seventy studies involving 3974 goats met the inclusion criteria. Unclear risk of bias for random sequence generation and allocation concealment predominated across studies. Pooled data for hormonal, sociosexual, and abiotic interventions showed a significant, though variable, increase in estrus (OR range 7.15-144.80), ovulation (OR range 6.08-56.95), and pregnancy (OR range 3.94-30.8). Hormonal treatments significantly reduced the onset of estrus, whereas abiotic interventions failed to reduce the number of anovulatory days. Secondary outcomes were not assessed in trials using sociosexual approaches. Finally, except for pregnancy, no significant efficacy was observed for studies using nutritional management. In conclusion, reproductive management practices using sociosexual approaches showed the highest efficacy for restoring reproductive activity in anestrous does.

ABSTRACT

The in-line milk analysis system (IMAS) is an automated biosensor technology that samples and quantifies milk progesterone concentrations (P4c) at frequent intervals starting early postpartum until pregnancy. The objective was to validate the use of pregnancy notifications (PregN) generated by an IMAS based on P4c profiles after artificial insemination (AI) to determine pregnancy and nonpregnancy status in dairy cows. Records of 1,821 AI events from 715 Holstein cows that had milk P4c (ng/mL) measured every 2.2 ± 1.9 d (mean ± standard deviation) between 24.5 ± 8.2 and 173.4 ± 49.3 d in milk through a real-time IMAS (Herd Navigator, DeLaval International, Tumba, Sweden) were evaluated. Based on variations in adjusted milk P4c (< vs. ≥ the 5.0 ng/mL threshold), the system determined the sampling frequency, onset and cessation of luteal phases, and pregnancy. If a luteal phase initiated (P4c increased to ≥5.0 ng/mL) after AI and remained uninterrupted, a PregN was generated starting at (mean ± standard deviation) 31.0 ± 4.3 d until 53.4 ± 7.9 d after AI, when sampling stopped, unless a decline in P4c (to <5.0 ng/mL) occurred indicating nonpregnancy and imminent estrus. The assessment of IMAS PregN at 4 weekly intervals was tested, and a confirmed calving occurrence between 262 and 296 d after AI, with no other subsequent AI recorded, was the gold standard for pregnancy. In total, 14.1 (256/1,821), 41.0 (746/1,821), and 50.7% (924/1,821) of AI events were followed by a decline in P4c before 19, 23, and 30 d after AI, respectively. Frequency of the last 3 sampling events preceding P4c decline was greater if P4c decline occurred between 18 and 25 d after AI (1.4 ± 0.5 samples per day) compared with before 17 or beyond 26 d after AI (1.0 ± 0.5 samples per day). At 30 ± 3 (27 to 33) d after AI, PregN occurred in 46.8% (853/1,821) of AI events, of which 15.2% (130/853) had a decline in P4c between 30 and 55 d after AI and 17.1% (146/853) was later confirmed nonpregnant based on the gold standard. A total of 40.7% (742/1,821) of AI events was confirmed pregnant by the gold standard, which was no different than the proportion of PregN at 51 ± 3 (48 to 54) d (40.9%; 744/1,821). At any time point between 27 and 54 d after AI, sensitivity and negative predictive values for PregN were greater than 95.0 and 96.0%, respectively, whereas specificity values were less than 90.0% for PregN before 40 d but greater than 94.0% for PregN beyond 41 d after AI. In conclusion, IMAS is able to diagnose pregnancy based on P4c profiles with high precision and determine early nonpregnancy based on the spontaneous cessation of the luteal phase. However, for accuracy greater than 95.0%, pregnancy declaration based on IMAS notifications alone should occur no earlier than 41 d after AI.

ABSTRACT

The production of captive fish is only possible through artificial reproduction, but manipulation is a known stressor stimulus. Thus, the objective of the present study was to evaluate the effects of different eugenol concentrations (0, 30, 40, 50 and 60 mg/L) during reproductive management of Rhamdia quelen. Seventy-five mature male R. quelen were randomly distributed among the five treatments, and blood samples were collected at the time of semen collection to measure plasma cortisol. The following parameters were evaluated in the fresh semen samples: motility, motility duration, concentration and fertilization rate. The following parameters were evaluated in the frozen semen samples: motility, motility duration, morphology, membrane integrity, DNA integrity and mitochondrial functionality. The animals anesthetized with eugenol at concentrations of 40 and 50 mg/L had lower levels of plasma cortisol (88.4 and 83.3 ng/mL, respectively) than the control (147.1 ng/mL). For fresh semen, the control treatment presented the highest rate and time of motility but differed (P < 0.05) only from the animals treated with 60 mg/L eugenol. For the cryopreserved semen the highest rates and motility time were observed in the control treatment and in the animals anesthetized with 40 mg/L eugenol, differing (P < 0.05) from anesthetized animals with 50 and 60 mg/L. Mitochondrial functionality was higher in fish anesthetized with 30 mg/L eugenol differing only for animals anesthetized with 60 mg/L. There was no difference between treatments for sperm concentration and fertilization rate of fresh semen. There were no differences (P > 0.05) between treatments in the parameters of membrane integrity, DNA integrity and% of normal spermatozoa after thawing of the cryopreserved semen samples. The use of 30, 40 and 50 mg/L eugenol maintained the seminal quality of the fresh semen, and the quality of the thawed semen was maintained with 30 and 40 mg/L eugenol. These results show that stress reduction can be reconciled with reproductive management without compromising reproductive performance.

ABSTRACT

This study compared estradiol cypionate (ECP) or GnRH as ovulation inducers at the end of a timed AI (TAI) protocol in Angus heifers. On day 0, heifers (n = 415), between 22 and 24 months of age, were treated with an intravaginal 1 g progesterone (P4) insert and 2 mg of estradiol benzoate. On day 8, heifers had P4 removed, received 500 µg cloprostenol, and were randomized into two groups: ECP [n = 214; 0.5 mg of ECP on day 8] or GnRH [n = 201; 25 µg of GnRH analog licerelin acetate on day 10]. All heifers received TAI on day 10; 48-50 h after P4 insert withdrawal. Estrus was determined by removal of tail paint. Ovaries of heifers were evaluated by ultrasound on day 0 to determine CL presence (with CL = 213, without CL = 202) and on day 10 to measure preovulatory follicle size. Heifers were divided into three categories based on preovulatory follicle diameter: <8.5 mm (smaller than deviation), 8.5-10.9 mm, or ≥11 mm. Pregnancy diagnosis was performed 32 days after TAI. Heifers treated with ECP had greater expression of estrus than GnRH-treated heifers (93.9% vs 67.7%; P < 0.0001), regardless of CL presence at beginning of protocol. Heifers with CL at beginning of protocol had larger preovulatory follicle diameter (10.4 mm vs 9.6 mm; P = 0.0058) and greater pregnancy per AI (P/AI; 61.0% vs 50.5%; P = 0.032) than heifers without CL at day 0. In heifers with CL at day 0, GnRH treatment increased P/AI compared to ECP treatment (68.0% vs 54.9%; P = 0.0498). Expression of estrus was greater in ECP-treated than GnRH-treated heifers that had small (<8.5 mm; 77.1% vs 5.6%; P < 0.001) or medium-sized (8.5-10.9 mm; 98.4% vs. 61.7%) follicles, but not in heifers with large follicles (≥11 mm; 97.9% vs 98.3%). The P/AI was very low in both treatments for heifers with follicles <8.5 mm (ECP-14.3% vs GnRH-16.7%). In heifers with medium-sized follicles (8.5-10.9 mm), ECP treatment tended to increase P/AI compared with GnRH-treated heifers (62.9 vs 46.7%; P = 0.074). In contrast, P/AI was greater for GnRH-treated than ECP-treated heifers with large preovulatory follicles (≥11 mm; 79.7% vs 60.4%; P = 0.032). Thus, the optimal inducer of ovulation in a TAI protocol for beef heifers appears to depend on the presence of a CL at beginning of protocol (GnRH > ECP if CL present) and size of the preovulatory follicle, with ECP increasing expression of estrus and tending to increase fertility in heifers with medium-sized follicles but GnRH increasing fertility in heifers having large preovulatory follicles.
