ABSTRACT

This study evaluated the potential of Aspergillus niger as an inoculant for growth promotion of vegetable seedlings. Seven vegetable species were evaluated in independent experiments carried out in 22 + 1 factorial schemes, with two doses of conidia (102 and 106 per plant) applied in two inoculation methods (seed treatment and in-furrow granular application), plus an uninoculated control. Experiments were carried out in a greenhouse. Growth parameters evaluated were shoot length, stem diameter, root volume, total root length, shoot and root fresh mass, shoot and root dry mass, and total dry mass. Regardless of the dose and inoculation method, seedlings inoculated with A. niger showed higher growth than uninoculated ones for all crops. The highest relative increase promoted by the fungus was observed for aboveground parts, increasing the production of shoot fresh mass of lettuce (61%), kale (40%), scarlet eggplant (101%), watermelon (38%), melon (16%), pepper (92%), and tomato (42%). Aspergillus niger inoculation also increased seedling root growth of lettuce, pepper, scarlet eggplant, watermelon, and tomato. This research shows that A. niger boosts the growth of all analyzed vegetables, appearing as a promising bio-input for vegetable seedling production.

ABSTRACT

Seeds with high vigor have greater capacity for hydrolysis and mobilization of stored reserves, which results in the formation of vigorous seedlings, and this behavior is observed under abiotic stress conditions. This study proposes to investigate the relationship of the enzyme alpha-amylase in lots of common-bean seeds with contrasting vigor, when subjected to the absence and presence of salt stress, aiming to identify the relationship of this enzyme with the vigor of the seed lot under these conditions. Seven common-bean cultivars were used. Physiological quality was determined by germination, vigor index and seedling length. The mobilization of reserves was evaluated under absence and presence of salt stress simulated with a NaCl solution with a concentration of 50 mmol L-1. The analyzed variables regarding reserve mobilization were reserve reduction, reserve reduction rate, seedling dry weight, reserve mobilization rate, starch, starch reduction rate and alpha-amylase activity. Results showed that the stress condition negatively affected all the evaluated variables; however, the cultivars classified as having greater vigor showed better physiological performance under stress. Salt stress in common-bean seeds affects seedling performance and reduces alpha-amylase activity during germination, and high-vigor seed lots exhibited higher enzyme activity in the no-stress condition.(AU)


As sementes com alto vigor possuem maior capacidade de hidrólise e mobilização das reservas armazenadas resultando na formação de plântulas vigorosas, sendo esse comportamento observado em condições de estresse abiótico. O objetivo deste trabalho foi avaliar a relação da enzima alfa-amilase em lotes de sementes de feijão com contraste no vigor quando submetidos a condições de ausência e presença de estresse salino, buscando identificar a relação desta enzima com o vigor do lote de sementes nessas condições. Sete cultivares de feijão foram utilizadas. A qualidade fisiológica foi determinada pela germinação, índice de vigor e comprimento de plântulas. A mobilização de reservas foi avaliada em condições com ausência e presença de estresse salino simulado com solução de NaCl com concentração de 50 mmol L-1. As variáveis analisadas referentes a mobilização de reservas foram a redução das reservas, taxa de redução das reservas, massa seca de plântulas, taxa de mobilização de reservas, amido, taxa de redução de amido e atividade da alfa-amilase. Os resultados demonstraram que a condição de estresse afetou negativamente todas as variáveis avaliadas, contudo, as cultivares classificadas como de maior vigor apresentaram melhor desempenho fisiológico sob o estresse. O estresse salino em sementes de feijão afeta o desempenho de plântulas e reduz a atividade da alfa-amilase durante a germinação e, os lotes de sementes com alto vigor apresentaram maior atividade da enzima em condições de ausência de estresse.(AU)

The refinement of vigor tests and the possibility of utilizing computer resources for the effective evaluation of the seed physiological potential have attracted considerable interest from research and seed technologists. The aim of this study was to evaluate the physiological potential of maize seeds using the newly-created Automated Analysis of Seed Vigor System (Vigor-S) compared with other recommended seed vigor tests; two maize hybrids were used, each represented by seven seed lots. Germination and vigor (cold test, saturated salt accelerated aging, and field seedling emergence) evaluations were conducted. For the evaluation of seed vigor with the use of seedling image analysis resources, two systems were compared: the Seed Vigor Imaging System (SVIS®), developed by Ohio State University, USA and the Vigor-S, resulting from collaboration between USP/ESALQ and EMBRAPA (Embrapa Instrumentation). Using these two systems, three day old seedlings were scanned and the images were analyzed. Similar results for the vigor index, uniformity of development, and seedling length were obtained. The computerized image analysis of seedlings using Vigor-S has advantages with respect to accuracy, speed, and the possibility of automatic application to a worksheet. It is a consistent alternative for the evaluation of maize seed vigor, and produces information compatible with that obtained by the accelerated aging test and SVIS®.

ABSTRACT: Citrus seeds normally have low storability. Identifying an optimal fruit harvest time for production of high vigor seeds is important for nurseries; however, identifying this stage for Swingle citrumelo fruits has been based only on visual color examination, and research related to this parameter has been inconsistent. The main objective of this study was to evaluate a red-green-blue (RGB) color measurement system for successful identification of Swingle citrumelo fruits possessing seeds with maximum physiological potential and storability. Fruits were harvested at three ripening stages identified as green (G), greenish-yellow (GY), and yellow (Y) pericarp, photographed, and the images processed using ImageJ software. Data were expressed as the average pixel value of R, G and B color components and a mean RGB pixel value (R+G+B/3). After harvest, seeds were evaluated for water content, germination, seedling emergence, length, dry mass and vigor as measured by the Seed Vigor Imaging System - SVIS (uniformity and vigor indexes) after 0 and 5 months storage (5 °C and 65 % relative humidity). Percentage of ruptured coat seeds was also evaluated after extraction. The R color component provided the best identification of each G, GY and Y fruits. Seeds extracted from GY fruits had higher storage potential compared with seeds from G and Y fruits. Thus, precise fruit ripening classification can be generated using the RGB color system to identify the best time for harvest to obtain seeds with greater physiological and storage potential.

ABSTRACT

A obtenção de cariopses de cana-de-açúcar por meio de cruzamento de parentais é de primordial importância para os programas de melhoramento de cultivares. Objetivou-se, neste trabalho comparar o ambiente de germinação de cariopses da casa de vegetação do Programa Cana IAC com um ambiente de condições controladas, a fim de avaliar sua eficiência, no período de 30/09 a 09/10/2004. Foram desenvolvidos dois experimentos, em delineamento experimental de blocos casualizados com quatro repetições, em Jaú, SP. Um deles foi montado sob condições de laboratório em câmara de germinação e outro em casa de vegetação em condições não controladas, sendo cada tratamento constituído por 100 cariopses por repetição, provenientes de seis cruzamentos. Foram realizadas avaliações do índice de velocidade de germinação (IVG), da porcentagem ( por centoG) e dos dias para emergência (DE), em intervalos de 24 horas, após a instalação dos testes. A massa de 100 sementes de cada cruzamento também foi avaliada com o propósito de verificar sua influência nos demais atributos da germinação. Os resultados foram submetidos à análise de variância e a comparação de médias foi feita pelo teste de Tukey, a 0,05 de significância. A massa de 100 sementes foi diferente entre os cruzamentos, e interferiu, de maneira geral, nos atributos IVG e por cento G. Concluiu-se que a casa de vegetação utilizada pelo Programa Cana IAC proporciona porcentagens de germinação semelhantes às da câmara de germinação, porém condições controladas favorecem o IVG e os DE.


The production of true seeds of sugarcane through parental crossing is primordial for the breeding programs of cultivars. With the objective of comparing the efficiency of the environment greenhouse used for seed germination in the IAC Sugarcane Program with an environment with controlled conditions (germination chamber), two experiments were carried out in a completely randomized block with four replicates of 100 seeds each, in Jaú (SP), from 09/30 to 10/09/2004. In both places the treatments were composed of seeds from six crossings. The evaluations accomplished were speed of germination index (SGI), germination percentage ( percent G) and days for emergence (DE) in 24 h intervals after set up treatments. The mass of 100 seeds of each crossing was also evaluated with the purpose of measuring its influence on other germination attributes. The data were submitted to the analysis of variance and averages were compared by the Tukey test at 0.005 significance. The weight of 100 seeds differed among the crossings, and, in general, it interfered with the attributes SGI and percent G. It was concluded that the greenhouse used by the IAC Sugarcane Program provides germination percentages similar to the germination chamber; however, controlled conditions improve SGI and DE.
