ABSTRACT

Chagas disease and leishmaniasis are neglected diseases of high priority as a public health problem. Pharmacotherapy is based on the administration of a few drugs, which exhibit hazardous adverse effects and toxicity to the patients. Thus, the search for new antitrypanosomatid drugs is imperative to overcome the limitations of the treatments. In this work, 46 2-nitroimidazole 3,5-disubstituted isoxazole compounds were synthesized in good yields by [3 + 2] cycloaddition reaction between terminal acetylene (propargyl-2-nitroimidazole) and chloro-oximes. The compounds were non-toxic to LLC-MK2 cells. Compounds 30, 35, and 44 showed in vitro antichagasic activity, 15-fold, 12-fold, and 10-fold, respectively, more active than benznidazole (BZN). Compounds 30, 35, 44, 45, 53, and 61 acted as substrates for the TcNTR enzyme, indicating that this might be one of the mechanisms of action involved in their antiparasitic activity. Piperazine series and 4-monosubstituted compounds were potent against T. cruzi parasites. Besides the in vitro activity observed in compound 45, the in vivo assay showed that the compound only reduced the parasitemia levels by the seventh-day post-infection (77%, p > 0.001) compared to the control group. However, 45 significantly reduced the parasite load in cardiac tissue (p < 0.01) 11 days post-infection. Compounds 49, 52, and 54 showed antileishmanial activity against intracellular amastigotes of Leishmania (L.) amazonensis at the same range as amphotericin B. These findings highlight the antitrypanosomatid properties of 2-nitroimidazole 3,5-disubstituted isoxazole compounds and the possibility in using them as antitrypanosomatid agents in further studies.

ABSTRACT

Amitriptyline was first introduced as a medication to treat depression. Over time, this substance has been used to treat other conditions, such as gastrointestinal disorders, fibromyalgia, neuropathic pain, and analgesia, among others. However, there are no published studies that provide a broad view of the possible motivations that have led to changes in the use of amitriptyline. In this study, we have identified the landscape of use for amitriptyline based on knowledge mapping of the 100 most-cited articles about this drug. We searched Web of Science Core Collection without time and language restrictions. We obtained 14,446 results, but we only used the 100 most-cited articles that had amitriptyline as the object of study. We collected the following information from each article: authors, country of the corresponding authors, year of publication, citation count, citation density (number of citations per year), and keywords. In addition, we seek to map in the chosen articles study design and research findings. We found that since 1980, the use of amitriptyline has expanded beyond depression, moving to off-label use to treat a variety of diseases and conditions, including post-herpetic neuralgia, neuropathic pain, primary fibrosis, fibromyalgia, and migraine, can be considered a drug with more clinical applicability than its original clinical indication.

ABSTRACT

There is a trend toward the use of natural substances present in plants and vegetables. In general, foods rich in antioxidants are complex matrices; therefore, understanding its absorption effects is extremely relevant to know its bioactive potential. Thus, this systematic review focused on clinical trials involving plants (or compounds) registered on the National List of Medicinal Plants of Interest to the Unified Health System (RENISUS) with antioxidant properties. Following the reporting guidelines of Preferred Reporting Items for Systematic Reviews and Meta-Analyzes studies of interest indexed in the PubMed and ClinicalTrials.gov databases were analyzed. Of the 59 clinical trials found, Allium sativum and Curcuma longa are the plant species with the highest percentage of clinical research. Prevention/attenuation of oxidative stress was one of the main antioxidant mechanisms indicated in the studies. The most tested compounds of the RENISUS plants in clinical trials were curcumin and soy isoflavone. In this review, we selected studies in advanced stages that highlight plants' value in optimizing antioxidant status; however, even with high-quality studies, it is not prudent to overstate the clinical efficacy of these plants.

ABSTRACT

ABSTRACT To explore the clinical effect of music therapy in the treatment of middle-aged and elderly patients with depression. From June 2019 to June 2020, 100 patients with depression in a community were selected as the object of this study, and were divided into a control group and an observation group according to the order of visits, with 50 patients in each group. There was no significant difference between the two groups in terms of gender, age, education level and drug treatment (medication dose, type) (P>0.05). The patients in the control group were treated with conventional antidepressant drugs. The patients in the observation group added music therapy to the conventional therapy. The music therapist needs to fully understand the basic situation of the patient and formulate a suitable music treatment plan according to the actual state of the patient. Compare the HANA and PSP scores between the two groups of depressive patients after treatment to judge the treatment effect on the patients. The Hana and PSP scores in the observation group after treatment were significantly higher than those in the control group (P<0.05). To treat depression, in addition to selecting appropriate drugs, alleviating the emotional state of patients is more important. Music therapy as an auxiliary treatment of traditional therapy, in the treatment of depression in the elderly, has been effectively verified, we should actively promote its application.

ABSTRACT

Nonalcoholic fatty liver disease (NAFLD) is the most common liver disease in the world. Despite the impact of this pathology in the population, nowadays there is no specific treatment for this disease, focusing its treatment on risks factors. However, it is imperative the existence of a specific treatment, due to this, the aim of this study was to determine the therapeutic effect of treatment with metformin, 4-hydroxychalcone or co-treatment on male Wistar rats with NAFLD. Wistar rats were divided into two groups with free access to either tap water or 50% sucrose (NAFLD) during 25 weeks. After 20 weeks of induction each were divided into four groups that received daily p.o. administration of: i) saline solution (1 ml); ii) metformin (200 mg/kg/day); iii) 4-hydroxychalcone (80 mg/kg/day) and i.v.) co-treatment (metformin plus 4-hydroxychalcone at the doses mentioned above), for 5 weeks. In healthy rats: metformin and co-treatment modified food and total caloric intake and induced diarrhea; but none of the treatments changed the other parameters evaluated. Meanwhile in rats with NAFLD: i) metformin inhibited hepatic total cholesterol and TGF-ß, increased diarrhea frequency, and slightly decreased liver steatosis, and fibrosis; ii) 4-hydroxychalcone decreased IL-6, TNF-α and TGF-ß, increased IL-10, and markedly decreased liver steatosis and fibrosis; and iii) co-treatment markedly decreased food intake, total caloric intake, and body weight, increased diarrhea; increased IL-10, showing and intermediate effect on decrease TNF-α, TGF-ß, liver steatosis and fibrosis. Our results showed that 4-hydroxychalcone treatment was the most effective among the treatments tested against NAFLD.

ABSTRACT

(AU)

The devices of intense pulsed light, similar to lasers, has the basic principle to absorption of photons by endogenous or exogenous chromophores in the skin. The pulsed light is converted into heat energy, which coagulates the desired target (photo-selective thermolysis). The intense pulsed light technology involves a system of parallel xenon lamps and capacitors contained in an articulated arm that is applied directly to the skin surface. Its emission spectrum ranges from 500 to 1,300 nm, thus presenting a great versatility of treatments. For the construction of this review, the methodology adopted was a literature search in scientific journals healthcare indexed in Pubmed (between the years 1989 to 2016) that address the physiological and therapeutic effects of intense pulsed light therapy. Among the therapeutic effects of the intense pulsed light therapy, we highlight hair removal, pigmented lesions, vascular lesions, acnes treatment, and finally skin rejuvenation. One of the major advantages of the intense pulsed light is in relation to its versatility and economy. Moreover, the devices of intense pulsed light are more advantageous due to their ability to treat large areas of skin. However, more comparative controlled trials with extended follow-up period are needed to determine significant differences between the intense pulsed light and other therapies. (AU)

ABSTRACT
