ABSTRACT

AIM To investigate the effects of Tea Po lyp henols on induction of apoptosis in human liver cancer cell etc. Telomerase acti vity of BEL-7404 cells was dramatically declined during the cell incuba tion with Tea Polyphenols. METHODS MTT colorimetric assay and trypan b lue exclusion method were used to examine the growth inhibition ofTea Polyphenols on BEL-7404 cells. Telomerase activity in cell was determined by PCR-ELISA method. Cell morphology and DNA gel electrophoresis were used to observe the apoptosis. RESULTS The solution of Tea Polyphenols showed dose-dependent and time-dependent inh ibitory effects on the proliferation on BEL-7404 cell line. After treatment wit h 0 2 and 0 1 g?L -1 tea polyphenols for 48 hours, cells displayed DNA ladder bands and typical morphological change of apoptosis included: ce ll shrinkage, membrane blebbing, chromatin condensation, nuclear fragmentation etc. Telomerase activit y of BEL-7404 cells was dramatically declined during the cell incubation with Tea Polyphenols. CONCLUSION Tea Polyphenols has growth inhibiti ng effect and may induce apoptosis in BEL-7404 in a certain dose range. The ant icancer mechanism of Tea Polyphenols might be related to the inhibition of telom erase activity.

ABSTRACT

Objective To investigate the effects of nu cleus transcription factor (NF)-?B, survivin, Bcl-2 and Caspase-3 on apoptos is of gastric cancer cells induced by tumor necrosis factor related apoptosis in ducing ligand(TRAIL). Methods Gastric cancer cells were cultured in RPMI-1640 and the proportions of apoptosis were analyzed by flow cytometry. The expressions of N F-?B, survivin, Bcl-2 and Caspase-3 in gastric cancer cells were evaluated b y Western blot. Results After gastric cancer cells were exposed to 50 ng/ml TRAIL f or 24 hours, the proportions of apoptosis were 24.05% in line MKN28, 7.83% in MK N45, 8.05% in AGS and 3.17% in SGC-7901 respectively; after 300 ng/ml TRAIL for 24 ho urs, the proportions of apoptosis were 36.05% in MKN28, 20.27% in MKN45, 16.50% in AGS and 11.80% in SGC-7901 respectively. The expressions of NF-?B and surv ivin under Western blot were lower in MKN28 than that in MKN45, AGS and SGC-79 0 1. In contrast, the expression of Caspase-3 was higher in MKN28 than that in MK N45, AGS a nd SGC-7901. The expression of Bcl-2 had no difference among the 4 lines of ga stric cancer cells. Conclusions The anti-tumor sensitivity of TRAIL to gastric cancer c ell may be associated with both increased expressions of NF-?B and survivin an d decreased expression of Caspase-3.

ABSTRACT

ObjectiveTo study the effect of apoptosis in the p athogenesis of great saphenous varicose(GSV) resulting from primary deep ven ous insufficiency (PDVI). Methods Apoptosis and Bcl-2 expres sion in the segment of first valve sinus of the GSV of PDVI of lower limbs w ere detected by transmission electron microscopy(TEM), agarose gel electrophore sis, TUNEL and immunohistochemistry. ResultsThere were 38 case s of PDVLs in experment group and 5 normal GSV in control group. In experiment group, the apoptosis cells(AC) (6.30? 2.70 )and apoptosis rate (AR)(0.42?2 .12) in the first valve sinus of GSV were significantly higher than those in control group(1.60?0.81,0.21?1.10,respectively)(all P
