ABSTRACT

Extracellular vesicles are recognized as signaling mediators between cells both in physiological and pathological communication. In this work, we explored the potential effect of citicoline to modify relevant proteins or miRNAs for cardioprotection in the smallest population of such microvesicles; i.e., in exosomes from patients diagnosed with ST-segment elevation myocardial infarction (STEMI) undergoing coronary angioplasty. The plasma-exosome-enriched fraction from these patients was characterized. Their cellular origin was assessed by flow cytometry and Western blot, whereas miRNA expression was evaluated by real-time polymerase chain reaction (qRT-PCR). The content of caveolin-1, caveolin-3, and hnRNPA2B1, which play a relevant role in selective transport of miRNAs into microvesicles, along with the effect on cell viability of the exosomes obtained from citicoline-treated and untreated groups were also analyzed. Our results showed that hypoxic stress increases exosome release into the circulation. Moreover, we found that CD146+ increased in exosomes from citicoline-treated patients, while CD142+ decreased in these patients compared to the placebo group. No changes were detected in the protein levels of caveolin-1, caveolin-3, and hnRNPA2B1. Citicoline administration modified the expression of miR233-3p, miR92, and miR21-5p in exosomes. Cell viability decreased in the presence of exosomes from infarcted patients, while incubation of H9c2 cells with exosomes from patients reperfused with citicoline did not affect cell viability. In conclusion, citicoline administration modifies the expression of specific miRNAs related to cardioprotection in exosomes.

ABSTRACT

We report the first optical biosensor for the novel and important cardiac biomarker, galectin-3 (Gal3), using the anti-Gal3 antibody as a biorecognition element and surface plasmon resonance (SPR) for transducing the bioaffinity event. The immunosensing platform was built at a thiolated Au surface modified by self-assembling four bilayers of poly(diallyldimethylammonium chloride) and graphene oxide (GO), followed by the covalent attachment of 3-aminephenylboronic acid (3ABA). The importance of GO, both as the anchoring point of the antibody and as a field enhancer for improving the biosensor sensitivity, was critically discussed. The advantages of using 3ABA to orientate the anti-Gal3 antibody through the selective link to the Fc region were also demonstrated. The new platform represents an interesting alternative for the label-free biosensing of Gal3 in the whole range of clinically relevant concentrations (linear range between 10.0 and 50.0 ng mL-1, detection limit of 2.0 ng mL-1) with successful application for Gal3 biosensing in enriched human serum samples.

ABSTRACT

Background: Atresia coli is an intestinal anomaly. Generally, absence of defecation in calves less than 10 days old, diminishing appetite, progressing abdominal distention and time-developing depression are reported in atresia coli. Although a calf consumes its milk normally in first days, loss of appetite develops over time. Occasional minor pain indications are observed. The objective of this study was to evaluate of hematologic, blood gases, cardiac biomarkers and serum biochemical parameters before operation in calves with atresia coli, and to investigate relationship with prognosis of these parameters.Materials, Methods & Results: Thirty calves with atresia coli and 6 healthy calves for control were used as material. Poor suction reflex, anorexia, dehydration, abdomen-kicking, frequent leg movement, abdominal distension and depression were observed in calves brought to the clinic. Some calves presented swashing sound depending on content of intestines when abdomen was shaken. Intestine atresia was diagnosed by history and clinical examination. After clinical examination, operation was performed. All calves were observed during 10 days after operation. Fifteen of 30 calves died during 10 days after operation. The blood lactate, WBC, LDH and CK-MB levels in non-surviving calves with atresia coli were significantly higher compared to control calves. The venous blood pH level was significantly lower in non-surviving calves compared to surviving calves. In addition, when compared to the control group, the blood pCO2 level was significantly higher in non-surviving and surviving calves with atresia coli while the pO2 and O2 saturation levels were significantly lower.Discussion: In calves with intestinal atresia, hematologic parameters are normal in first 48 h of disease. The blood lactate, WBC, LDH and CK-MB levels in non-surviving calves with atresia coli were significantly higher compared to control calves. The venous blood pH level was significantly lower in non-surviving calves compared to surviving calves. In addition, when compared to the control group, the blood pCO2 level was significantly higher in non-surviving and surviving calves with atresia coli while the pO2 and O2 saturation levels were significantly lower.Discussion: In calves with intestinal atresia, hematologic parameters are normal in first 48 h of disease. Some calves presented swashing sound depending on content of intestines when abdomen was shaken. Intestine atresia was diagnosed by history and clinical examination. ABSTRACT

OBJECTIVES: The red blood cell distribution width has been associated with an increased risk of cardiovascular events. In the present study, we assessed the relationship between red cell distribution width values and cardiac troponin I levels in patients admitted with non-ST-elevation acute coronary syndrome. METHODS: We analyzed blood parameters in 251 adult patients who were consecutively admitted to the intensive coronary care unit with non-ST-elevation acute coronary syndrome over a 1-year period. For all patients, a baseline blood sample was collected for routine hematological testing. Cardiac troponin I was measured at baseline and after 6 h. The patients were diagnosed with non-ST-elevation myocardial infarction or unstable angina based on the elevation of cardiac troponin I levels. RESULTS: The red cell distribution width was higher in the group with non-ST-elevation myocardial infarction compared with the patient group with unstable angina (14.6±1.0 vs 13.06±1.7, respectively; p = 0.006). Coronary thrombus was detected more frequently in the group of patients with non-ST-elevation myocardial infarction than in the patients with unstable angina (72% vs 51%, respectively; p = 0.007). Using receiver operating characteristic curve analysis for the prediction of non-ST-elevation myocardial infarction based on the red cell distribution width, the area under the curve was 0.649 (95% confidence interval: 0.546-0.753; p = 0.006), suggesting a modest model for the prediction of non-ST-elevation myocardial infarction using the red cell distribution width. At a cut-off value of 14%, the sensitivity and specificity of the red cell distribution width were 73% and 59%, respectively. Additionally, the red cell distribution width was positively correlated with cardiac troponin I (r = 0.19; p = 0.006). Hemoparasitoses são causas frequentes de anemia e miocardite em cães. Em virtude da anemia poder provocar distúrbios hemodinâmicos relacionados à hipóxia e da miocardite secundária à hemoparasitose causar arritmias cardíacas, esperam-se alterações eletrocardiográficas e aumento dos níveis de biomarcadores cardíacos como troponina I cardíaca em cães com hemoparasitose. Por conseguinte, este estudo teve como objetivo investigar as características eletrocardiográficas de cães anêmicos por hemoparasitose e a relação da anemia com alterações eletrocardiográficas e aumento da troponina I cardíaca. Para tanto, foram avaliados vinte e oito cães com hemoparasitose, sorologicamente positivos para Ehrlichia canis/ Babesia canis, com diferentes níveis de anemia. Cães com anemia muito grave (hematócrito abaixo de 13%) apresentaram aumentos significativos da frequência cardíaca, comparados aos cães com anemia grave, moderada ou leve. Houve predomínio de arritmia sinusal respiratória em cães com anemia leve, moderada e grave e alta ocorrência de ritmo sinusal em cães com anemia muito grave, demonstrando diminuição do tônus parassimpático neste grupo. Because anemia can trigger hypoxia or hemodynamic disturbances and myocarditis secondary to blood parasitosis can develop arrhythmias, electrocardiogram changes and increased levels of cardiac biomarkers, such as cardiac troponin I, are expected in dogs with blood parasitosis. Thus, this study aimed to investigate the electrocardiographic characteristics and the relationship to troponin I levels in anemic dogs with blood parasitosis. Twenty-eight dogs with blood parasitosis (seropositive for Ehrlichia canis/ Babesia canis) and varying levels of anemia were evaluated. Dogs with very severe anemia (hematocrit below 13%) showed significant increases in heart rate when compared to dogs with severe, moderate or mild anemia. There was a predominance of respiratory sinus arrhythmia in dogs with mild, moderate and severe anemia and a high prevalence of sinus rhythm in dogs with very severe anemia, which showed a decrease in parasympathetic activity in this group. In all of the groups, electrocardiographic changes were detected showing compatibility with left and right atrial and left ventricular enlargement.
