ABSTRACT
BACKGROUND: Benign neoplasms exhibit most of the cellular phenomena considered hallmarks of cancer, except the capacity to metastasize. Thus, the elucidation of the mechanisms associated with the progression of benign neoplasms may complement and clarify the mechanisms involved in carcinogenesis. Benign odontogenic tumours often result in facial deformities and morbidities, and have complex pathogenesis, mainly due to the diversity of interactions between the odontogenic epithelium and the ectomesenchyme. Primary cell culture of such tumours is not only difficult to be established and maintained, but also tumour cells lose characteristic cellular morphology. Considering gene expression, growth, migration, proliferation and cellular morphology are controlled by cell-cell interactions and cell-extracellular matrix interactions, cell culture in 3D substrates has gained space as a way to overcome some of the limitations of traditional monolayer cell culture systems. METHODS: In this study, fragments obtained from mesenchymal odontogenic tumours were cultured in type I collagen scaffolds. Invasion tests were performed in these models, as well as phenotypic characterization of the cultured tumours. RESULTS: The results obtained for the odontogenic myxoma and the cemento-ossifying fibroma demonstrate a good reproduction of the growth pattern of these tumours under ex vivo conditions. Microscopic evaluation showed maintenance of cell viability in the explants for more than 30 days, without the presence of necrosis. CONCLUSION: This is the first study involving long-term 3D primary cultures of benign odontogenic tumours, which is expected to support complex approaches to cell and molecular biology, and to serve as an experimental model for testing molecular therapies.

As lesões fibro-ósseas constituem um grupo de patologias caracterizadas pela substituição de tecido ósseo normal por tecido fibroso benigno contendo quantidades variadas de material mineralizado. A displasia fibrosa, o fibroma cemento-ossificante e a displasia cemento-óssea são as mais frequentes. Na literatura, a classificação e a terminologia das lesões fibro-ósseas apresentam-se ainda confusas e controversas, pois essas patologias exibem, frequentemente, características clínicas e morfológicas muito semelhantes. O fibroma cemento-ossificante é uma neoplasia benigna que faz parte do grupo das lesões fibro-ósseas. Caracteriza-se por crescimento bem delimitado e eventualmente encapsulado, apresentando quantidades variáveis de tecido mineralizado semelhante ao osso e ao cemento. São lesões incomuns, que tendem a ocorrer durante a terceira e a quarta décadas de vida, com uma predileção pelo gênero feminino, sendo a mandíbula o sítio de maior ocorrência. Este trabalho tem por objetivo descrever e discutir um caso clínico-cirúrgico sintomático de fibroma cemento-ossificante.
Fibro-osseous lesions are a group of diseases characterized by replacement of normal bone tissue for benign fibrous tissue containing different amounts of mineralized material. The most frequent lesions are fibrous dysplasia, cemento-ossifying fibroma and cemento-osseous dysplasia. In the literature, classification and terminology of these entities remain confusing and controversial, since these lesions show some clinical and morphologic similarities. Cemento-ossifying fibroma is a benign neoplasia that belongs to the group of fibro-osseous lesions. It is characterized by well delimited growth and occasionally is encapsulated, presenting changeable amounts of mineralized tissue similar to the bone and cementum. They are uncommon lesions that tend to occur during the 3rd and 4th decades of life, mainly in females and preferentially in the mandible. The aim of this work is to describe and discuss a clinical surgical case of symptomatic cemento-ossifying fibroma.

Peripheral cement-ossifying fibroma is a relatively common gingival growth of a reactive rather than neoplastic nature, whose pathogenesis is uncertain. It predominantly affects adolescents and young adults, with peak prevalence between 10 and 19 years. We report here the clinical case of a 5-year-old girl with disease duration of 3 years, who was followed up for 4 years, showing a gingival health and normal radiopacity of bone.
