ABSTRACT

The intensive use of anthelmintic drugs to control Fasciola hepatica infections in dairy cattle has resulted in the emergence of anthelmintic resistance. Cases of resistance to triclabendazole (TCBZ) have been reported worldwide. The main goal of this research was to evaluate the main five fasciolicides to control fasciolosis in dairy cattle in the Mantaro Valley, Peru. Two fecal egg count reduction tests were performed. In a first study, 24 naturally F. hepatica infected cattle were randomly grouped into three experimental groups (n = 8). Groups were treated with either TCBZ, nitroxynil (NTX) or closantel (CLOS). In a second experiment, 55 naturally infected cows were grouped into three experimental groups and treated with either TCBZ (n = 18), rafoxanide (RFX) + albendazole (ABZ) (n = 19) or clorsulon (CLN) + ivermectin (IVM) (n = 18). Therapeutic efficacy was determined following the WAAVP guidelines by measuring reduction in fluke egg output at days 15 and 30 post-treatment. Bootstrapping method was used to obtain the 95% confidence intervals. The efficacy of TCBZ was inadequate in both studies (≤80.8%). Closantel showed high efficacy (≥ 90%) at both days, while NTX showed 92.9% (83-100) and 82.1% (53.6-100), efficacy, at days 15 and 30, respectively. Efficacy for RFX were 92.1% (79.6-98.9) and 97.4% (94.1-99.4); and for CLN, 98.8% (97.6-100) and 80.1% (44.7-99.4), at days 15 and 30, respectively. The outcome of this study indicates reduced therapeutic efficacy of TCBZ against F. hepatica in an important dairy area of the Peruvian central highlands but also demonstrates the validity of four alternatives.

ABSTRACT

This work aimed to evaluate the thiol-disulphide homeostasis in serum of lambs naturally infected by gastrointestinal nematodes presenting different levels of parasite load indirectly indicated by faecal worm egg counts (EPG). Furthermore, the possible changes in the thiol-disulphide dynamic after different procedures to reduce the parasitic charge, such as the integrated crop-livestock system or anthelmintic treatment, were assessed. The results were compared with a panel of various oxidative stress and inflammatory biomarkers. The lambs were divided into three groups: animals highly infected (EPG higher than 5000) and packed cell volume (PCV) lower than 24% (G1); animals highly infected (EPG higher than 5000) and normal PCV (>24%) (G2); and animals presenting EPG lower than 5000 and normal PCV (>24%) (G3). The highly infected lambs (G1 and G2) showed lower total thiol (TT) and native thiol (SH) (p ≤ 0.01) than those from G3. After treatment, TT and SH increased significantly in all groups (p ≤ 0.01), and the disulphide (SS)/TT and SS/SH ratios decreased significantly (p < 0.01) in G1 and G2. These results show that the thiol-disulphide balance was impaired in lambs infected by gastrointestinal nematodes and that it could be potentially used as a biomarker to monitor this disease.

ABSTRACT

In this work, we present an evaluation of the fasciolicidal efficacy of a new injectable formulation of fosfatriclaben in comparison with the subcutaneous closantel and oral triclabendazole formulations currently used in veterinary practice as fasciolicides. The study was carried out in vivo on Fasciola hepatica at 2, 4, 6 and 8 weeks of age in experimentally infected sheep. To evaluate the formulation, the percent reduction of the parasite load was measured and the number of fluke eggs. Fosfatriclaben was used at 6 mg/kg/IM (dose equivalent to triclabendazole content), closantel at 5% at 10 mg/kg/SC, and triclabendazole at 10 mg/kg/PO; the control group received no treatment. Fosfatriclaben showed fasciolicidal efficacies of 95.5 %, 100 %, 100 % and 100 %, and triclabendazole showed similar efficacies of 97.4 %, 100 %, 100 % and 100 %, at the different treatment weeks (P > 0.05). Closantel showed limited efficacy against 2-, 4- and 6-week-old flukes but 100 % efficacy in adult flukes. All three evaluated formulations eliminated all 8-week-old F. hepatica trematode eggs. Although fosfatriclaben and triclabendazole showed similar fasciolicidal efficacy, the intramuscular administration of fosfatriclaben has several advantages over the oral administration of triclabendazole, such as ease of administration for veterinary use and a reduced risk of accidents for both the operator and the animals. In addition, the dose used in this injectable formulation is only 60 % of the oral dose, which reduces environmental contamination.

BACKGROUND AND AIM: Closantel is a widely used anti-parasitic drug that is known to cause ophthalmic problems that lead to blindness. The aim of this study was to investigate the possible electroretinographic changes in sheep that received closantel. MATERIALS AND METHODS: Twenty-four 30-day-old Suffolk sheep were split into control group (12 animals) and closantel group (12 animals). The latter group received 15 mg/kg of closantel subcutaneously immediately after the first electroretinography (ERG). The ISCEV protocol was used to perform the ERGs pre-dose (0), 7, and 30 days after treatment. Statistical analyses to compare ERG responses using t-test and analysis of variance were performed (p<0.05). Three months later, the animals were euthanized and the eyes and a part of optic nerve were collected for histopathology. Photography of the retina and optic nerve was taken, and measures of the retinal layers were made and analyzed by paired t-test. RESULTS: Closantel group showed a significant increase of the mean scotopic a-wave amplitude from 0 to 7 days after closantel administration, using a stimulus of 10,000 mcd.s/m2 and a decrease of the mean scotopic and photopic a-wave amplitude (from 7 to 30 days) using the same flash intensity, as well as a decrease in mean photopic b-wave amplitude (from 7 to 30 days) within the group. Control group showed a significant increase of the mean scotopic b-wave implicit time from pre to 30 days after treatment and an increase of the mean scotopic a-wave implicit time from pre to 7 days after treatment, with the stimulus of 10,000 mcd.s/m2. This group also showed a decrease in mean photopic b-wave implicit time (from pre to 30 days after treatment), using a stimulus of 10,000 mcd.s/m2 and a decrease in mean photopic a-wave implicit time from pre to 30 days after treatment, using a stimulus of 3000 mcd.s/m2. The no difference was found in images neither in the measurements of the retina layers. CONCLUSION: As observed by ERG responses and the histopathology, a dose of 15 mg/kg of closantel does not significantly affect retinal and optic nerve structures in sheep but the electroretinographic results, however, showed alterations on the phototransduction.

The results showed that all antiparasitic drugs had an in vitro inhibitory effect on nematophagous fungi, which could compromise their action as agents of biological control. ABSTRACT

This study aimed to evaluate the effect of veterinary pharmaceuticals (VPs) used to control endo- and ectoparasites in ruminants, on the survival and reproduction of the collembolan species Folsomia candida. Standard ecotoxicological tests were conducted in Tropical Artificial Soil and the treatments consisted of increasing dosages of four commercial products with different active ingredients: ivermectin, fipronil, fluazuron and closantel. Ecotoxicological effects were related to the class and mode of action of the different compounds. Fipronil and ivermectin were the most toxic compounds causing a significant reduction in the number of juveniles at the lowest doses tested (LOECreprod values of 0.3 and 0.2 mg kg-1 of dry soil, respectively) and similar low EC50 values (fipronil: 0.19 mg kg-1 dry soil, CL95% 0.16-0.22; ivermectin: 0.43 mg kg-1 dry soil, CL95% 0.09-0.77), although the effects observed in the former compound were possibly related to a low adult survival (LC50 of 0.62 mg kg-1 dry soil; CL95%: 0.25-1.06). For the latter compound no significant lethal effects were observed. Fluazuron caused an intermediate toxicity (EC50 of 3.07 mg kg-1 dry soil, CL95%: 2.26-3.87), and also here a decrease in adult survival could explain the effects observed at reproduction. Closantel, despite showing a significant reduction on the number of juveniles produced, no dose-response relationship nor effects higher than 50% were observed. Overall, all tested compounds, especially ivermectin, when present in soil even at sub-lethal concentrations, can impair the reproduction of collembolans and possibly other arthropods. However, the actual risk to arthropod communities should be further investigated performing tests under a more realistic exposure (e.g., by testing the dung itself as the contaminated matrix) and by deriving ecotoxicologically relevant exposure concentration in soil derived from the presence of cattle dung.

ABSTRACT

The influence of closantel on the rheological and physicochemical properties (particle size and by UV-Vis absorption spectroscopy) of PVA aqueous solutions is studied here. About 1% PVA aqueous solutions were prepared by varying the closantel content. The increase of closantel content led to a reduction in the particle size of final solutions. All the solutions were buffered at pH 7.4 and exhibited shear-thinning behavior. Furthermore, in oscillatory flow, a "solid-like" type behavior was observed for the sample containing 30 µg/mL closantel. Indicating a strong interaction between the dispersed and continuous phases and evidencing an interconnected network between the nanoparticle and PVA, this sample also showed the highest shear viscosity and higher shear thinning slope, indicating a more intrincate structure disrupted by shear. In conclusion, PVA interacts with closantel in aqueous solution and the critical concentration for closantel encapsulation by PVA was about 30 µg/mL; above this concentration, the average particle size decreased notoriously which was associated to closantel interacting with the surface of the PVA aggregates and thus avoiding to some extent direct polymer-polymer interaction.

ABSTRACT

An outbreak of Closantel intoxication in sheep in Uruguay is described. The outbreak occurred in a group of 1300 weaning lambs treated orally with a 10% solution of Closantel. One hundred forty eight lambs showed clinical signs of intoxication and 14 died. The clinical signs included mydriasis, nystagmus, and negative pupillary reflex, bilateral blindness, bump into objects, and lateral movement of the head. No macroscopic lesions were observed. The histological lesions of the retina were cytoplasmic vacuolization in ganglion cells and in cells of the inner and outer nuclear layers with different degrees of atrophy. Vacuolization and axonal degeneration were observed in the optic nerve, with multifocal areas of fibrosis and infiltration by lymphocytes and Gitter cells. To reproduce the intoxication, four sheep were given two, four and 10 times the therapeutic dose of Closantel (0.1g/kg of BW). Only the animals receiving 10 times the recommended dose showed clinical signs. The histological examination of the lesions in experimental sheep showed similar results to those described in the accidental outbreak, except for the absence of optic nerve fibrosis and inflammation, characterizing an acute phase. Axonal myelin sheaths loss, fibroblasts and collagen fibers were observed in the ultrastructural study of the optic nerve of accidental intoxicated animals. The optic nerve of experimentally intoxicated animals had vacuoles that separated the myelin sheaths of axons. To prevent outbreaks it is suggested to weigh the animals before Closantel administration to avoid errors in dose calculation.(AU)


Descreve-se um surto de intoxicação por Closantel em ovinos no Uruguai. O surto ocorreu em um lote de 1300 cordeiros que foram dosados com uma solução de Closantel 10%, por via oral. Do total, 148 apresentaram sinais clínicos de intoxicação e 14 morreram. Os sinais clínicos incluíam midríase, nistagmo, reflexo pupilar negativo, cegueira bilateral, pressão da cabeça contra objetos e desvio lateral da cabeça. Lesões macroscópicas não foram observadas. Histologicamente havia vacuolização citoplasmática das células ganglionares e nas células das camadas nuclear interna e externa. Na retina havia, também, diferentes graus de atrofia. Vacuolização e degeneração axonal foram observados no nervo óptico, com áreas multifocais de fibrose e infiltrado de linfócitos e células Gitter. Quatro ovinos receberam experimentalmente duas, quatro e 10 vezes a dose terapêutica de Closantel (0,1 g/kg de peso vivo). Apenas os animais que receberam 10 vezes a dose recomendada apresentaram sinais clínicos. O exame histológico nos ovinos experimentais mostrou resultados semelhantes aos do surto, com exceção da ausência de fibrose e infiltrado inflamatório do nervo óptico, caracterizando um quadro agudo. Foram observadas a perda da bainha de mielina dos axônios e a presença de fibroblastos e fibras colágenas no estudo ultra-estrutural do nervo óptico de animais intoxicados espontaneamente. No nervo óptico de animais intoxicados experimentalmente havia vacúolos que separavam as bainhas de mielina dos axônios. Para evitar surtos, sugere-se pesar os animais antes da administração de Closantel para evitar erros no cálculo da dose.(AU)

No nervo óptico de animais intoxicados experimentalmente havia vacúolos que separavam as bainhas de mielina dos axônios. Para evitar surtos, sugere-se pesar os animais antes da administração de Closantel para evitar erros no cálculo da dose.

ABSTRACT

This study reports poisoning in sheep wich have received two applications of closantel in therapeutic dosage (7.5mg/kg) within a preestablished 28-day period for effectiveness formulation. A high percentage of efficiency was observed after the first and second treatment, but around 72 hours after the second administration of closantel (D30), three sheep have showed mild apathy, anorexia, diarrhea, bilateral blindness, and dilated pupils without reaction. These three animals, were observed in the following 250 days, and the bilateral blindness remained. Numerous reports of animals poisoned by closantel emphasize that overdose and/or nutritional status of the animals are key factors to occur poisoning due closantel administration. However, in this experiment the signs of poisoning (mild apathy, anorexia, diarrhea and ophthalmic disturbance), observed in the three clinically healthy sheep, occurred when they received two applications of closantel (7.5mg/kg), in a interval of 28 days, highlighting the fact of obtaining a cumulative residual effect of closantel in the animal organism, caused by two consecutive applications, may also be a predisposing factor for sheep to demonstrate irreversible signs of intoxication.


O presente estudo notifica a intoxicação de ovinos que receberam duas aplicações de closantel, na dosagem terapêutica (7,5mg/kg), com intervalo de 28 dias pré-estabelecido pela eficácia da formulação. Referente aos resultados de eficácia foi possível observar elevado percentual após o primeiro e o segundo tratamento. Cerca de 72 horas após a administração do segundo tratamento (D+30), três animais que receberam closantel demonstraram leve apatia, anorexia, diarréia e cegueira bilateral com ausência de reflexo e midríase bilateral. Estes três ovinos foram observados por cerca de 250 dias após o segundo tratamento, e a cegueira bilateral não regrediu. Os inúmeros casos de animais intoxicados por closantel descritos na literatura enfatizam que a sobredosagem e/ou qualidade nutricional dos animais, são fatores determinantes para que ocorra intoxicação destes animais pelo closantel. Entretanto, no presente trabalho, os sinais de intoxicação (leve apatia, anorexia, diarréia e a alteração oftálmica) observada nos três ovinos clinicamente sadios, ocorreram quando estes receberam duas aplicações de closantel (7,5mg/kg) com o intervalo de 28 dias, evidenciando que o possível efeito residual cumulativo de closantel no organismo animal, desencadeado por duas aplicações consecutivas, também pode ser um fator predisponente para que ovinos demonstrem sinais irreversíveis de intoxicação pelo princípio ativo em questão.

ABSTRACT

An outbreak of oropharyngeal lesions and poisoning by closantel in sheep after improper handling and anthelmintic drug overdose is described here. Eleven sheep were affected out of 42 that were dewormed. Eight sheep died. Clinically, the animals showed abdominal recumbency, absent pupillary and palpebral reflexes, bilateral blindness, depression, apathy, emaciation, and corneal opacity. Three sheep exhibited swelling on the right side of the mandible. At necropsy, one sheep exhibited a purulent lesion located on the right, next to the root of the tongue. Microscopically, the main finding was status spongiosus in the central nervous system and optic nerve. The retina showed rod and cone degeneration, ganglionic cell layer vacuolation, karyorrhexis, and vacuolation of the outer plexiform layer. The liver of one sheep exhibited diffused coagulative necrosis of hepatocytes in the centrilobular region associated with high infestation of Haemonchus contortus.(AU)

ABSTRACT

Descrevem-se dois surtos de intoxicação por clo-santel, um em ovinos e outro em caprinos, no Estado de San-ta Catarina. No primeiro surto, de 12 ovinos que adoeceram 5 apresentaram cegueira, desses três morreram (Ovinos 1-3) e dois (Ovinos 4 e 5) foram eutanasiados, 6 meses após ficarem cegos. No segundo surto, de 26 caprinos que adoeceram, seis animais sobreviveram, porém ficaram cegos, e um deles foi eutanasiado. Clinicamente os animais apresentavam depressão, ataxia, incoordenação motora e reflexo pupilar diminuído a ausente. Em alguns animais esse quadro evoluiu para cegueira bilateral com ausência de reflexo de ameaça e midríase bilateral irresponsiva. Ao exame oftálmico verificou-se atrofia dos vasos da retina e hiperreflexia. Pelo exame histológico observou-se edema mielínico levando a status spongiosus no sistema nervoso central e neuropatia compressiva no nervo óptico, acompanhada de degeneração e/ou atrofia da retina. O objetivo deste trabalho é descrever os aspectos epidemiológicos, clínicos e patológicos da intoxicação por closantel em ovinos e caprinos.


Two outbreaks of closantel overdosage in sheep and goat flocks are described. In the first outbreak 12 sheep were affected, 5 of them showed blindness, three sheep died (Sheep 1-3) and two were euthanized 6 months after the onset of clinical manifestation (Sheep 4 and 5). In the second outbreak 26 goats were affected, from which six survived despite blindness and one was euthanized. Clinically the animals showed depression, ataxia, motor incoordination, decreased or absent pupil reflexes. In some animals this clinical picture developed to bilateral blindness, with no reaction to threat and bilateral irresponsive midriasis. In the ophthalmic examination retinal vessel atrophy and hyperreflexia were observed. The histological examination showed myelin edema leading to status spongiosus in the central nervous system and compressive neuropathy of the optic nerve, associated with retinal degeneration and/or atrophy. This report aims to describe the epidemiologic, clinic and pathologic aspects of closantel overdosage in sheep and goats.

As alterações crônicas mostravam disco óptico acinzentado, atrofia de vasos e da retina. Nos fundos tapetal e não-tapetal notavam-se áreas de despigmentação e lesões irregulares castanho-amareladas. As alterações histológicas consistiam em perda dos neurônios da camada ganglionar e das células da camada nuclear interna e externa da retina. As alterações agudas no nervo óptico e na substância branca do encéfalo foram de degeneração espongiforme. As alterações crônicas do nervo óptico caracterizavam-se por extensa necrose e infiltração de células Gitter.(AU)
