ABSTRACT

Gastric cancer (GC) is one of the most common tumors worldwide and is the leading cause of tumor-related mortality. Traditional biomarkers and screening methods cannot meet the clinical demands. There is an urgent need for highly sensitive diagnostic markers as well as accurate quantification methods for early gastric cancer (EGC) screening. Here a dual-target cooperatively responsive fluorescent nanomachine by the innovative application of two targets─responsive strand migration system with a single-amplification-cycle element was developed for the simultaneous detection of GC biomarkers miR-5585-5p and PLS3 mRNA, which were selected by next-generation sequencing and RT-qPCR. It was also an RNA extraction-free, PCR-free, and nonenzymatic biosensor to achieve tumor cell imaging and serum diagnosis. Requiring only a 20 µL serum sample and 20 min incubation time, the nanomachine achieved an ultrasensitive detection limit of fM level with a broad linear range from fM to nM. More importantly, a higher AUC value (0.884) compared to the clinically used biomarker CA 72-4 was obtained by the nanomachine to distinguish GC patients successfully. Notably, for the key concerns of diagnosis of EGC patients, the nanomachine also achieved a satisfactory AUC value of 0.859. Taken together, this work has screened and obtained multiple biomarkers and developed a fluorescent nanomachine for combination diagnosis of GC, providing an ingenious design of a functionalized DNA nanomachine and a feasible strategy for the transformation of serum biomarkers into clinical diagnosis.

ABSTRACT

A new species of soft scale (Hemiptera: Coccomorpha; Cocccidae) from South Africa, Coccus giliomeei Lagowska Hodgson sp. n., collected on Gymnosporia buxifolia (L.) Szyszyl, is described and illustrated. Also, Coccus rhodesiensis (Hall) is recorded for the first time from South Africa and is redescribed and illustrated based on the adult females of the type series and fresh South African specimens. An updated key to the species of Coccus and similar species known from Africa is included. Based on this latter study, (i) Marsipococcus proteae (Brain) and M. durbanensis (Brain) are considered not to be congeneric with Marsipococcus marsupialis (Green), the type species of Marsipococcus Cockerell Bueker, and are placed in a new genus Proteacoccus Lagowska Hodgson, gen. n. with Lecanium proteae Brain as the type species; (ii) it is considered that Coccus asiaticus Lindinger is clearly not a junior synonym of Parasaissetia nigra (Nietner) and is accepted as a full species, rev. stat., and (iii) Neoplatylecanium adersi (Newstead) is considered to be non-conspecific with N. cinnamomi Takahashi, the type species of Neoplatylecanium Takahashi, and is transferred to Maacoccus Tao Wong, as Maacoccus adersi (Newstead), comb. n.

Objective To evaluate the differentiating diagnostic value of single use and combined use of contrast-enhanced ultrasound(CEUS)and acoustic radiation force impulse imaging(ARFI) in TI-RADS class 3 and 4 thyroid nodules.Methods A total of 68 inpatients with thyroid nodules treated by operation in the First Affiliated Hospital of Henan University of Science and Technology (according with TI-RADS class 3 and 4 thyroid nodules suggested by Ji-Young Park in 99 cases) were collected and their imaging data were retrospectively analyzed.All cases simultaneously conducted ARFI and CEUS examinations before operation.The pathogenic results served as the golden standard.The differentiating diagnostic values of single use and combined use of CEUS and ARFI in TI-RADS class 3 and 4 thyroid nodules were comparatively analyzed by using the statistical method.Results Both ARFI and CEUS had high differential diagnosis value in TI-RADS class 3 and 4 thyroid nodules,however,the single use of ARFI and CEUS no statistically significant difference in diagnosis value (P>0.05),the combination use of ARFI and CEUS significantly improved the sensitivity and accuracy of diagnosis.Conclusion The combination of CEUS and ARFI can be applied to differentiate TI-RADS class 3 and 4 thyroid nodules.

ABSTRACT

Objective To explore the value of combined use of laboratory indicators for diagnosis of early renal functional damage. Methods Eighty-six patients with various kidney diseases were enrolled in the Second Affiliated Hospital of Guangzhou Medical University. On admission,the serum Cystatin C(Cys C),creatinine(Cr), Urea,etc were determined. The value of using combined laboratory indicators in the diagnosis of renal functional damage was obtained through the analysis of the receiver operating characteristic curve(ROC curve);multiple variable indicators were grouped to establish multiple logistic regression models to be compared and evaluated. Results In the early and late renal injury groups(group B of 32 cases and group C of 12 cases),the serum levels of Cys C,Cr, and Urea were significantly higher than those in the normal renal function control group(group A of 42 cases),the elevation in level in group C being the most significant〔Cys C(mg/L):3.47±0.75 vs. 1.59±1.29,Cr(μmol/L):669±466 vs. 214±173,Urea(mmol/L):21.22±13.10 vs. 11.04±8.24,PCys C+Urea>Cys C>Cr+Urea(0.920=0.920>0.911>0.908>0.809). In this sequence,the AUC made by Cys C+Cr+Urea and Cys C+Cr were equal,both 0.920, whose sensitivity was 75.0%,specificity 100.0%,positive predictive value 100.0%,negative predictive value 80.0%and diagnostic accuracy rate 87.5%. So,Cys C + Cr combination could be used to substitute Cys C + Cr + Urea, and the former clinical diagnostic effect was the best,much higher than that by using AUC whose curve was made by Cys C alone. Conclusion The value of using only one laboratory indicator for diagnosis of patients with early renal functional damage is not high,while applying Cys C+Cr combination can improve the diagnostic effect greatly,and its sensitivity and specificity are higher.
