ABSTRACT

The objective of this study was to evaluate the chemical composition of two chickpea varieties, 'Costa 2004' and 'El Patrón', and to characterize their proteins to determine their technological potential for the food industry. For this purpose, chickpea samples of both varieties from the 2019 harvest region of Guanajuato, Mexico, were obtained and chemically characterized to determine the protein fractions using electrophoretic and amino acid profiling. The chickpea variety 'Costa 2004' contained 3% less protein and 7% less dietary fiber content than the variety 'El Patrón'; whereas, the carbohydrate content of 'Costa 2004' was 4% greater. Additionally, the chickpeas demonstrated an antioxidant capacity ranging from 319 to 387 µMET/g and total phenol levels exceeding 500 mg/g. Among the protein fractions, globulins represented the highest proportion in both varieties of chickpea, at approximately 8.73 g/100 g ('Costa 2004') and 10.42 g/100 g ('El Patrón'), followed by albumin, at approximately 1.24 g/100 g and 1.47 g/100 g, respectively. The chickpea proteins ranged in molecular weight between 100 and 25 kDa, with particularly strong signals in the albumin and globulin bands. Regarding the amino acid profile, histidine was predominant in both varieties. In conclusion, both varieties of chickpea have high nutritional value and broad potential for technological use in the food industry.

BACKGROUND: Electrophoresis can be used to aid in the diagnosis of different diseases in avian species. Capillary zone electrophoresis (CZE) is an automated method that is proposed to be superior to the dye methods used in agarose gel electrophoresis (AGE). However, reference intervals (RIs) for CZE in avian species and comparison studies between electrophoretic methods are lacking. OBJECTIVES: The goals of the current study were to compare AGE and CZE methods and determine reference intervals for CZE using plasma from bald eagles (Haliaeetus leucocephalus). METHODS: Heparinized plasma samples from 44 bald eagles (mean age 18.7 years) under managed care were examined by AGE and CZE. Method comparison analyses were completed, as well as the generation of preliminary RIs using the CZE method and ASVCP guidelines. RESULTS: Passing-Bablok regression and Bland-Altman plots demonstrate that these methods are not equivalent. All fractions were significantly correlated between the methods except for alpha 1 globulin. Inter-assay and intra-assay CVs for CZE were lower or comparable to AGE and ranged from 2.4% to 15.4%, and 0.8% to 8.3%, respectively. CZE resolved more fractions than AGE with two fractions observed in the beta and gamma region vs one for AGE in each region. CONCLUSIONS: CZE provided improved resolution and reproducibility for the quantitation of protein fractions in the bald eagle. Although most fraction results correlated with AGE, these methods were judged as not equivalent, necessitating method-specific Rls. Reference intervals generated from a limited number of mostly aged individuals under managed care should be considered preliminary; additional studies will aid in the production of more robust intervals.

ABSTRACT

The objective was to study the productive and biological traits of young cattle of the Kalmyk breed and its crosses. The experiment was conducted on 4 groups of steers, 10 heads each, at OAO Shurupovskoe, Frolovsky District, Volgograd Region. The experimental steers were kept in rooms separately in groups on a deep bed of straw and had free access to walking yards with mounds. The hemoglobin content and the numbers of erythrocytes and leukocytes were determined on a Medonic CA 530 hematological analyzer. The total protein in blood serum and protein fractions were found on an automatic biochemical analyzer Stat Faks 1904. The digital experimental data were processed by the variation statistics method in terms of the significance of the compared parameters using the Student's test accepted in biology and animal science. The study evidenced that the contents of morphological elements and proteins in blood reflected the metabolism and potential for crossbred steers to gain muscle mass.


O objetivo era estudar os traços produtivos e biológicos do gado jovem da raça Kalmyk e seus cruzamentos. A experiência foi conduzida em 4 grupos de bois, 10 cabeças cada, na OAO Shurupovskoe, Distrito de Frolovsky, Região de Volgograd. Os bois experimentais foram mantidos em quartos separados em grupos em uma cama de palha profunda e tiveram livre acesso a pátios de caminhada com montes. O conteúdo de hemoglobina e o número de eritrócitos e leucócitos foram determinados em um analisador hematológico Medonic CA 530. A proteína total em soro sanguíneo e frações proteicas foram encontradas em um analisador bioquímico automático Stat Faks 1904. Os dados experimentais digitais foram processados pelo método de estatística de variação em termos da significância dos parâmetros comparados usando o teste de Student aceito em biologia e ciência animal. O estudo evidenciou que o conteúdo de elementos morfológicos e proteínas no sangue refletia o metabolismo e o potencial de bois cruzados para ganhar massa muscular.

ABSTRACT

This study evaluated the solubility profiles of quinoa grain proteins and applied a complete process for the isolation of its main protein fractions, namely: albumins, globulins, prolamins and glutelins, which corresponded to 26.96%, 41.3%, 1.7% and 23.16% of the total protein content, respectively. When these fractions were digested with pepsin followed by pancreatin, the degrees of hydrolysis achieved varied between 26.62% (for unheated globulin fraction) and 38.97% (for unheated glutelin), with casein reached 33.73% hydrolysis. After heating, the globulin hydrolysis degree increased to 34.7%, not significantly differing from casein. These results reflect its good susceptibility to hydrolysis by digestive enzymes, and this observation is reinforced with assays with pepsin, trypsin and chymotrypsin tested separately. Globulins, the largest protein fraction, showed promising results in additional assays regarding the amino acid profile, with limitation only for lysine in relation to the FAO standard, and the potential for releasing bioactive peptides after digestion. Although pepsin-digested globulin inhibited only 5% of ACE activity under the conditions tested, after 24h with the addition of pancreatin, the inhibition was 100%. Antioxidant activity (DPPH assay) also indicated very similar results, when hydrolysis with pepsin was inefficient in releasing antioxidant peptides, while hydrolysis by pancreatin led to 35 times greater results.

Resumo Fundamento A fetuína-A é um fator anti-inflamatório e anticalcificação envolvido no curso da doença arterial coronariana (DAC). Em alinhamento com essas funções, investigou-se a fetuína-A como marcador de risco cardiovascular em vários estudos. Porém, a associação entre a fetuína-A e o prognóstico dos pacientes com DAC ainda é controversa. Objetivos O presente estudo foi conduzido para identificar a associação entre o nível de fetuína-A sérica e doença cardiovascular (DCV) de longo prazo e a mortalidade global por infarto do agudo do miocárdio por supradesnivelamento do segmento ST (STEMI). Métodos Foram cadastrados no estudo cento e oitenta pacientes consecutivos com STEMI. A população do estudo foi dividida em subgrupos (mais baixo, ≤288 µg/ml; e mais alto, >288 µg/ml) de acordo com a mediana do nível de fetuína-A. Dados de acompanhamento clínico foram obtidos por contato telefônico anual com pacientes ou familiares. As causas das mortes também foram confirmadas pelo banco de dados de saúde nacional. P-valores bilaterais <0,05 foram considerados estatisticamente significativos. Resultados Durante um acompanhamento médio de 10 anos, foram registradas 71 mortes, das quais 62 foram devidas a DCV. Identificou-se um índice de mortalidade global e por DCV significativamente mais alto no grupo com nível de fetuína-A mais baixo que no grupo com nível de fetuína-A mais alto (44% versus 24%, p= 0,005; 48% versus 31%, p= 0,022, respectivamente). Nas análises de risco proporcionais por regressão de Cox, detectou-se que a fetuína-A era um preditor independente de mortalidade global e por DCV. Conclusões A baixa concentração de fetuína-A está associada ao prognóstico de longo prazo ruim pós-STEMI, independentemente de fatores de risco cardiovascular tradicionais. Nossos achados fortaleceram estudos prévios demonstrando consistentemente o papel determinante dos mediadores anti-inflamatórios em síndromes coronárias agudas.


Abstract Background Fetuin-A is an anti-inflammatory and anti-calcification factor involved in the course of coronary artery disease (CAD). In line with these functions, fetuin-A has been investigated as a cardiovascular risk marker in many studies. However, the association between fetuin-A and the prognosis of CAD patients is still controversial. Objectives The present study was conducted to identify the association between serum fetuin-A level and long-term cardiovascular disease (CVD) and all-cause mortality of ST-elevation acute myocardial infarction (STEMI). Methods One hundred eigthy consecutive patients with STEMI were enrolled in the study. The study population was divided into subgroups (lower, ≤288 µg/ml; and higher, >288 µg/ml) according to the median fetuin-A level. Clinical follow-up data was obtained by annual contact with the patients or family members by telephone. The causes of death were also confirmed by the national health database. Two-sided p-values<0.05 were considered statistically significant. Results During a median follow-up of 10 years, 71 deaths were recorded , 62 of whom died from CVD. Both CVD and all-cause mortality were found to be significantly higher in the lower fetuin-A group than the higher fetuin-A group (44% vs 24%, p= 0.005; 48% vs 31%, p= 0.022, respectively). In Cox regression proportional hazard analyses, fetuin-A was found to be an independent predictor of CVD and all-cause mortality. Conclusions Low fetuin-A concentration is associated with a poor long-term prognosis after STEMI, regardless of the traditional cardiovascular risk factors. Our findings have strengthened previous studies that consistently demonstrate the determining role of anti-inflammatory mediators in acute coronary syndromes.

ABSTRACT

Carotenoids are pigmented compounds acquired through diet that have important functions as antioxidants and immune modulators. We studied the association between immunity and circulating carotenoids in Andean condors (Vultur gryphus). We evaluated the relationship between α-, ß-, and γ-globulin blood concentrations and different circulating carotenoids in two groups of Andean condors that differ in their mean health status, rehabilitating (suffering different pathologies), and wild individuals (trapped when displaying their physiological behavior). In rehabilitating individuals, α-, ß-, and γ-globulin concentrations were higher than in wild individuals. This shows that rehabilitating individuals were developing an immune response associated with the pathologies that they were suffering at the time of sampling. In addition, circulating carotenoids were lower in rehabilitating than in wild individuals. We found negative correlations between α-, ß-, and γ-globulins and different circulating carotenoids in rehabilitating individuals, but not in wild condors. Xanthophylls were strongly related to α-, ß-, and γ-globulin blood concentrations in rehabilitating, but not in wild condors. Our results suggest that there is a potential relationship between circulating carotenoids and immunity in the Andean condor. Given that this species may display a carotenoid-based pigmentation, our results could suggest that a trade-off between the immune system and external coloration could operate in this species, which may have implications in their access to food resources and mate selection and, thus, in their conservation.

ABSTRACT

RESUMEN La pandemia de COVID-19 ha traído una nueva afección grave e inusual denominada Síndrome Inflamatorio Multisistémico en niños, de la cual aún hay mucho por conocer. Presentamos una serie de 8 casos atendidos en el Instituto Nacional de Salud del Niño, Lima Perú. La edad media fue 5,1 años. La presentación clínica incluyó fiebre, problemas gastrointestinales agudos, afectación ocular y mucocutánea. Cuatro cumplieron criterios para Enfermedad de Kawasaki clásica. Todos tuvieron serología positiva para SARS-CoV-2, hemograma patológico, marcadores inflamatorios elevados y pruebas de coagulación alteradas. Cinco casos presentaron hipertransaminasemia y tres retención nitrogenada. Cuatro casos cumplieron criterios para Síndrome de Activación Macrófagica. Todos recibieron inmunoglobulina intravenosa, corticoides y ácido acetil salicílico. Ninguno desarrolló aneurismas coronarios. Solo uno presentó miocarditis, shock y requirió ingreso a Unidad de Cuidados Intensivos. La mayoría evolucionaron favorablemente. En todo niño con fiebre, síntomas gastrointestinales y dermatológicos; asociado a exposición al SARS-CoV-2, debe investigarse compromiso multisistémico.


ABSTRACT During the COVID-19 pandemic, a new, severe and unusual condition called Multisystem Inflammatory Syndrome in children emerged, from which there is still much to learn. We report 8 children admitted to Instituto Nacional de Salud del Niño, in Lima, Perú. Their mean age was 5,1 years. Their clinical presentation included fever, acute gastrointestinal symptoms, ocular and mucocutaneous involvement. Four patients met criteria for classic Kawasaki Disease. All the patients had positive serology for SARS-CoV-2, abnormal complete blood counts and coagulation tests, and elevated inflammatory markers. Five had elevated liver enzymes and three had kidney involvement. Four patients met criteria for Macrophage Activation Syndrome. All of them received intravenous immune globulin, corticosteroids and aspirin. No coronary aneurysms were identified. Only one developed miocarditis, shock and was admitted to the Pediatric Intensive Care Unit. Most patients recovered successfully. Every child with fever, gastrointestinal and dermatological symptoms, associated with prior exposure to SARS-CoV-2, should be investigated for multi-systemic compromise.

Introducción: El lupus eritematoso sistémico es el modelo clásico de enfermedad autoinmune. En el desarrollo de la enfermedad intervienen varios tipos de inmunoglobulinas, con predominio de la IgG, IgM e IgA. Objetivo: Describir la utilidad del cociente albúmina/globulina como un indicador de actividad en el lupus eritematoso sistémico. Desarrollo: Se estima que el 50 por ciento de los pacientes con lupus eritematoso sistémico muestran una hipoalbuminemia con una hipergammaglobulinemia. La hipoalbuminemia en mayor medida está relacionada con la presencia de nefritis lúpica. La mitad de los pacientes con nefritis lúpica presentan proteinuria en el orden del síndrome nefrótico. Esta proteinuria iguala o invierte parcialmente el valor del cociente albúmina/globulina. El cociente albúmina/globulina invertido por sí solo es insuficiente para afirmar la presencia de actividad en el lupus eritematoso sistémico. Se deben excluir otras entidades clínicas causantes de hipergammaglobulinemia policlonal. Los criterios de actividad del lupus eritematoso sistémico incrementan la sensibilidad del cociente albúmina/globulina invertido. Conclusiones: La interpretación del cociente albúmina/globulina debe ir aparejada a la estimación de actividad por los criterios clínicos de mayor uso (SLICC, SLEDAI, BILAG). No en todos los pacientes con lupus eritematoso sistémico puede interpretarse como criterio de actividad, por lo que es necesario excluir otras entidades clínicas(AU)


Introduction: Systemic lupus erythematosus is the model of autoimmune disease. Several types of immunoglobulins are involved in the development of the disease, mainly IgG, IgM and IgA. Objective: To describe the potential use of the albumin/globulin ratio as an indicator of activity in systemic lupus erythematosus. Development: fifty percent of patients with systemic lupus erythematosus exhibit hypoalbuminemia with hypergammaglobulinemia. Hypoalbuminemia is mainly related to the presence of lupus nephritis. The half of patients with lupus nephritis develops proteinuria with values of nephrotic syndrome. The proteinuria equals or partially reverses the albumin/globulin ratio. The inverted albumin/globulin ratio is insufficient to establish the presence of lupus activity. Other clinical entities producing polyclonal hypergammaglobulinaemia should be excluded. The systemic lupus erythematosus activity criteria increase the sensitivity of the inverted albumin/globulin ratio. Conclusions: The interpretation of the albumin/globulin ratio requires the activity estimation by different clinical criteria (SLICC, SLEDAI, BILAG). The inverted albumin/globulin ratio cannot be interpreted as a stand-alone indicator of disease activity in every systemic lupus erythematosus patients(AU)

The aim of this study to measure the fractions of the total serum proteins of the Campeiro horse and identify the influences of biological variants. Blood samples were taken in 138 horses of the breed Campeiro for measuring the concentration of total serum protein by the biuret method. Serum concentrations of protein fractions were measured by electrophoresis using agarose gel. Groups were formed according to age, sex and reproductive condition. The average values of serum fractions: albumin (2.85±0.36g/dl), alpha 1 (0.28±0.11g/dl), alpha 2 (0.26±0.08g/dL) beta 1 (0.57±0.15g/dl), beta 2 (0.89±0.28g/dL), gamaglobulinas (1.86±0.34g/dL), albumin/globulin ratio (0.75±0.18) and 2.5% percentile and 97.5% had slight differences in relation to the reference interval proposed for the species. They observed higher values of alpha 1 and 2 globulins in the group from that had six to eight years old and gammaglobulins in group above 13 years old. Serum protein concentrations were similar in horses and mares and between non-pregnant and pregnant. Sex and pregnancy status did not affect serum proteinogram. Alpha and gammaglobulins have higher values as the age increases. Serum proteinogram of Campeiro horses shows variations that have to be considered in the interpretation of laboratory tests.(AU)


Este trabalho tem por objetivo mensurar as frações das proteínas totais séricas de equinos Campeiros e identificar as influências de variantes biológicas. Foram colhidas amostras de sangue de 138 equinos, machos e fêmeas da raça Campeiro. A determinação da concentração de proteínas totais séricas foi realizada pelo método de biureto. As concentrações séricas das frações proteicas foram determinadas por eletroforese, utilizando-se gel de agarose. Formaram-se grupos em relação à idade, ao sexo e à condição reprodutiva. Os valores médios das frações séricas albumina (2,85±0,36g/dL), alfa 1 (0,28±0,11g/dL), alfa 2 (0,26±0,08g/dL), beta 1 (0,57±0,15g/dL), beta 2 (0,89±0,28g/dL), gamaglobulinas (1,86±0,34g/dL), relação albumina/globulina (0,75±0,18) e os percentis 2,5% e 97,5% apresentaram diferenças pontuais em relação aos intervalos propostos para a espécie. Observaram-se maiores valores de alfa 1, alfa 2 globulinas, no grupo de seis a oito anos, e de gamaglobulinas, no grupo acima de 13 anos de idade. O proteinograma sérico foi similar entre machos e fêmeas e entre fêmeas vazias e gestantes. Sexo e estado gestacional não afetaram o proteinograma sérico. Alfa e gamaglobulinas têm incrementos em função de idades crescentes. O proteinograma sérico de equinos Campeiros tem variações que devem ser consideradas em exames laboratoriais.(AU)

BACKGROUND: Amaranth is a plant naturally resistant to various types of stresses that produces seeds of excellent nutritional quality, so amaranth is a promising system for food production. Amaranth wild relatives have survived climate changes and grow under harsh conditions, however no studies about morphological and molecular characteristics of their seeds are known. Therefore, we carried out a detailed morphological and molecular characterization of wild species A. powellii and A. hybridus, and compared them with the cultivated amaranth species A. hypochondriacus (waxy and non-waxy seeds) and A. cruentus. RESULTS: Seed proteins were fractionated according to their polarity properties and were analysed in one-dimensional gel electrophoresis (1-DE) followed by nano-liquid chromatography coupled to tandem mass spectrometry (nLC-MS/MS). A total of 34 differentially accumulated protein bands were detected and 105 proteins were successfully identified. Late embryogenesis abundant proteins were detected as species-specific. Oleosins and oil bodies associated proteins were observed preferentially in A. cruentus. Different isoforms of the granule-bound starch synthase I, and several paralogs of 7S and 11S globulins were also identified. The in silico structural analysis from different isoforms of 11S globulins was carried out, including new types of 11S globulin not reported so far. CONCLUSIONS: The results provide novel information about 11S globulins and proteins related in seed protection, which could play important roles in the nutritional value and adaptive tolerance to stress in amaranth species.

ABSTRACT Purpose: To investigate the prognostic role of preoperative albumin/globulin ratio (AGR) in predicting disease-free survival (DFS) and overall survival (OS) in localized and locally advanced clear cell renal cell carcinoma (RCC) patients. Patients and Methods: 162 patients who met the criteria specified were included in the study. The DFS and OS ratios were determined using the Kaplan-Meier method. Univariate and multivariate Cox regression analyses were performed to determine the prognostic factors affecting DFS and OS. Results: Median follow-up period was 27.5 (6-89) months. There was a statistically significant relationship between low AGR and high pathological tumor (pT) stage, presence of collecting system invasion, presence of tumor necrosis, and a high platelet count (p = 0.012, p = 0.01, p = 0.001, and p = 0.004, respectively). According to the Kaplan-Meier survival analysis, both OS and DFS were found to be significantly lower in the low AGR group (p = 0.006 and p = 0.012). In the multivariate Cox regression analysis, collecting system invasion and tumor necrosis were found to be independent prognostic factors in predicting OS and pT stage was found to be an independent prognostic factor in predicting DFS (HR: 4.08, p = 0.043; HR: 8.64, p = 0.003 and HR: 7.78, p = 0.041, respectively). Conclusion: In our study, low AGR was found to be associated with increased mortality and disease recurrence in localized and locally advanced RCC.

ABSTRACT

Isolated 7S and 11S globulins obtained from defeated soy flour were complexated with folic acid (FA) in order to generate nano-carriers for this important vitamin in human nutrition. Fluorescence spectroscopy and dynamic light scattering were applied to follow the nano-complexes formation and for their characterization. Fluorescence experimental data were modeled by the Stern-Volmer and a modified double logarithm approach. The results obtained confirmed static quenching. The number of binding sites on the protein molecule was ~1. The values obtained for the binding constants suggest a high affinity between proteins and FA. Particle size distribution allowed to study the protein aggregation phenomenon induced by FA bound to the native proteins. Z-average manifested a clear trend to protein aggregation. 11S-FA nano-complexes resulted in more polydispersity. ζ-potential of FA nano-complexes did not show a remarkable change after FA complexation. The biological activity of nano-complexes loaded with FA was explored in terms of their capacity to enhance the biomass formation of Lactobacillus casei BL23. The results concerning to nano-complexes inclusion in culture media showed higher bacterial growth. Such a result was attributed to the entry of the acid by the specific receptors concomitantly by the peptide receptors. These findings have technological impact for the use of globulins-FA based nano-complexes in nutraceutical, pharmaceutical and food industries.

This research aimed to evaluate the influence of different levels of salinity on growth performance, haematological parameters and the cells of Nile tilapia chloride. 144 fish were used during a 97 day trial period, submitted to four experimental treatments of 0%o, 10%o, 20%o and 25%o salinity. Sampling were collected for monitoring of performance parameters, blood samples to determine the erythrocyte count, packed cell volume, hemoglobin concentration, the erythrocyte indices, the count of thrombocytes and leukocytes and the electrophoretic profile of serum proteins. Gill samples were also collected for counting the number of cells chloride. Significant differences were found in hemoglobin concentration, the total plasma proteins and the number of erythrocytes (p < 0.05). There was no difference in the number of white blood cells and thrombocytes. Serum proteins showed differences (p < 0.05) from pre-albunima fractions, alfa1-globulin, alfa2-globulin, beta-globulin, and gama-globulin. There was a direct relationship between the number of chloride cells and increase in salinity levels (p < 0.05). The CBC constituents, the electrophoretic profile of proteins as well as quantitative analysis of gill chloride cells can serve as indicares mechanism of adaptation and tolerance to salinity in Nile tilapia.


Esta pesquisa teve por objetivo avaliar a influência de diferentes níveis de salinidade sobre o desempenho zootécnico, parâmetros hematológicos e células de cloreto de tilápia do Nilo. 144 peixes, durante 97 dias, foram submetidos a quatro tratamentos experimentais de 0%o, 10%o, 20%o e 25%o de salinidade. Realizaram-se biometrias para acompanhamento dos índices zootécnicos, coletas de amostras sanguíneas para determinação do número de eritrócitos, volume globular, concentração de hemoglobina, índices hematimétricos, contagem de trombócitos e de leucócitos total e diferencial, além do perfil eletroforético das proteínas séricas. As amostras branquiais foram para contagem do número de células de cloreto. Diferenças significativas foram encontradas na concentração de hemoglobina, nas proteínas plasmáticas totais e no número de eritrócitos (p < 0,05). Não foi observada diferença no número de leucócitos e de trombócitos. As proteínas séricas apresentaram diferenças (p < 0,05) nas frações pré-albunima, alfa1-globulina, alfa2-globulina, beta-globulina e gama-globulina. Verificou-se relação direta entre o número de células de cloreto e o aumento nos níveis de salinidade (p < 0,05). Os constituintes do hemograma, o perfil eletroforético de proteínas, bem como a análise quantitativa de células de cloreto branquiais, podem servir como indicares do mecanismo de adaptação e de tolerância à salinidade em tilápia do Nilo.

Background: Some reports indicate that calves produced by in vitro fertilization (IVF) may have excessive birth weights, increasing the rates of dystocia. Prolonged labor can cause the birth of lethargic calves that slowly ingest colostrum; this may compromise the intestinal absorption of immunoglobulins. Although the number of beef calves produced by IVF is high in Brazil, there have been few physiological studies examining the neonatal period in such cases. The aim of this study was to evaluate the passive transfer of immunity, and characterize the variation in serum protein concentrations, in Nelore calves up to 35 days of age, comparing naturally conceived calves with those conceived by IVF. Materials, Methods & Results: Forty healthy newborn Nelore calves were randomly selected and allocated into two groups (n = 20) according to their method of conception (natural versus IVF), and each group was composed of ten males and ten females. All calves were born and raised on the same farm, and kept with their mothers on extensive management condition. Blood samples were collected between 24 and 36 h after birth, and at 7, 14, 21, 28, and 35 days of age. Total protein (TP) concentration in serum was measured by the biuret method, and gamma-glutamyl transferase (GGT) serum activity was measured by the kinetic method. ABSTRACT

The role of seed proteins, especially soyabean 7S globulins, in controlling dyslipidaemia is widely acknowledged. Amino acid sequence homology among the proteins of this family could reflect similar biological functions in other species. The aim of the present study was to unveil a hypolipidaemic effect of the 7S globulins from cowpeas (7S-C) and adzuki beans (7S-A), administered orally to rats fed a hypercholesterolaemic (HC; high cholesterol and TAG) diet for 28 d. A total of forty-five rats were divided into five groups (nine rats per group): (1) standard (STD) diet; (2) HC diet; (3) HC diet + 7S-C (300 mg/kg per d); (4) HC diet + 7S-A (300 mg/kg per d); and (5) HC diet + simvastatin (SVT; 50 mg/kg per d), as a control. Significant decreases in food intake and final body weight of rats receiving HC + 7S-C and HC + 7S-A diets compared with groups fed the HC and STD diets were observed. Significant decreases in serum total and non-HDL-cholesterol of 7S-C, 7S-A and SVT groups were also observed. HDL-cholesterol levels increased in the 7S-C, 7S-A and SVT groups, while hepatic cholesterol and TAG concentrations were significantly lower than in the HC diet group for the 7S-C-supplemented group only. Faecal excretions of fat and cholesterol in HC diet groups were considerably higher in animals consuming the 7S globulins. The results show that cowpea and adzuki bean 7S globulins promote cholesterol-decreasing effects in hypercholesterolaemic rats even at low dosages, as already observed for other legume seed storage proteins of this family. This main effect is discussed in relation to the possible mechanisms of action.

ABSTRACT

Las altas temperaturas ambientales propician susceptibilidad al estrés calórico en los pollos de engorde, lo que genera cambios metabólicos. Se buscó determinar los cambios en la bioquímica sanguínea y la concentración plasmática de corticosterona del pollo de engorde sometido a estrés calórico crónico y a las condiciones de temperatura ambiental del piedemonte amazónico colombiano, sobre las concentraciones plasmáticas de corticosterona, glucosa, proteínas totales, albumina, globulina, sodio, cloro, potasio, magnesio, fósforo y calcio. Se utilizaron pollos machos de 2 estirpes de 21 días de edad, distribuidos en un diseño experimental irrestrictamente al azar, en un esquema bifactorial, con 4 tratamientos. Se hicieron 5 repeticiones por tratamiento y 25 animales por unidad experimental. Las aves fueron alimentadas con una dieta básica de maíz y torta de soya con 3,100 Kcal de EM y 19,5% de proteína hasta los 42 días de edad. El factor estirpe no tuvo efecto en las variables evaluadas (p ≥ 0,05). No obstante, existió diferencia estadística significativa (p ≤ 0,05) en todas las variables evaluadas al ser comparadas las concentraciones de los metabolitos de las aves en condiciones de estrés calórico crónico con las de las aves expuestas a las condiciones de temperatura del piedemonte amazónico colombiano. Se concluyó que la bioquímica sanguínea sufrió modificaciones significativas bajo las 2 temperaturas experimentales, con mayor detrimento fisiológico de las aves con estrés calórico crónico. La concentración de corticosterona se constituyó en el indicador más sensible y constante de la condición fisiológica de estrés crónico por calor.


High ambient temperatures cause susceptibility to heat stress in broiler chickens, generating metabolic changes. This paper seeks to determine the changes in blood biochemistry and plasma corticosterone concentration, as well as in glucose, total protein, albumin, globulin, sodium, chlorine, potassium, magnesium, phosphorus, and calcium in broiler chickens under chronic heat stress and at ambient temperature conditions at the Colombian Amazonian piedmont. 21-days-old male chickens of two lines were studied, distributed in an unrestricted random design, in a two-factor scheme, with four treatments. Five repetitions per treatment were performed, and 25 animals per experimental unit examined. Broilers were fed a basic diet of corn and soybean meal with 3,100 kcal ME and 19.5% protein until they reached 42 days of age. The line factor had no effect on the evaluated variables (p ≥ 0.05). However, there was statistically significant difference (p ≤ 0.05) in all variables when concentrations of metabolites in broilers under chronic heat stress were compared to those of chickens exposed to ambient temperatures at the Colombian Amazon piedmont. In conclusion, blood biochemistry suffered significant changes under both experimental temperatures, with more physiological detriment in broilers under chronic heat stress. Concentration of corticosterone became the most sensitive and consistent indicator of the physiological condition of chronic heat stress.

Buscou-se determinar as mudanças na bioquímica sanguínea e a concentração plasmática de corticosterona do frango de engorde sometido a estresse calórico crónico e às condições de temperatura ambiental do sopé amazônico colombiano, sobre as concentrações plasmáticas de corticosterona, glucose, proteínas totais, albumina, globulina, sódio, cloro, potássio, magnésio, fósforo e cálcio. Utilizaram-se frangos machos de 2 estirpes de 21 dias de idade, distribuídos em um desenho experimental irrestritamente aleatório, em um esquema bivariado, com 4 tratamentos. Realizaram-se 5 repetições por tratamento e 25 animais por unidade experimental. As aves foram alimentadas com uma dieta básica de milho e torta de soja com 3,100 Kcal de EM e 19,5% de proteína até os 42 dias de idade. O fator estirpe não teve efeito nas variáveis avaliadas (p ≥ 0,05). Não obstante, existiu diferença estatística significativa (p ≤ 0,05) em todas as variáveis avaliadas ao ser comparadas as concentrações dos metabolitos das aves em condições de estresse calórico crônico com as das aves expostas às condições de temperatura do sopé amazônico colombiano. Concluiu-se que a bioquímica sanguínea sofreu modificações significativas sob as 2 temperaturas experimentais, com maior detrimento fisiológico das aves com estresse calórico crônico. A concentração de corticosterona se constituiu no indicador mais sensível e constante da condição fisiológica de estresse crônico por calor.
