ABSTRACT

Objective To observe the counteraction of Ganoderma lucidum polysaccharide(GLP) on the inhibition of mice splenocyte proliferation and IL-1? and IL-2 mRNA expression induced by prostaglandin E2(PGE2).Methods Mixed lymphocyte culture reaction(MLR) method was used for the experiment.The lymphocyte proliferation was determined by MTT method,and the levels of IL-1? and IL-2 mRNA expression were evaluated by semi-quantitative reverse transcriptase polymerase chain reaction(RT-PCR).Results After co-culture with PGE2 for 48 hours,the splenocyte proliferation was inhibited to some extent,and the difference was significant when the concentration of PGE2 was over 10?mol/L(P

ABSTRACT

【Objective】To investigate the therapeutic effect of Shen Tao Ruangan Pill(STRP) for postoperative recurred liver cancer by a randomized and controlled trial.【Methods】Forty patients with postoperative recurred liver cancer were equally randomized into groups A and B.The two groups received routine symptomatic treatment,and group A was additionally treated with STRP(mainly composed of Herba Artemisiae Scopariae,Herba Hedyotis Diffusae,Herba Scutellariae Barbatae,Rhizoma Curcumae,Semen Persicae,Radix Angelicae Sinensis,Radix Salviae Miltiorrhizae,Radix Ginseng,Poria,Carapax et Plastrum Testudinis) and group B with Pingxiao Capsule.After 3-month treatment,the therapeutic effect was evaluated in the two groups.【Results】(1)After treatment,the symptoms were relieved in both of groups A and B,and group A had a better effect in relieving abdominal distention,anorexia and lassitude than group B(P0.05).(3) The non-progressive survival period was(172?11.64) days in group A and(143?10.95) days in group B;the survival rate within 3,6 and 10 months was 82.31%,70.42% and 50.33% in group A respectively,and 72.70%,65.70% and 46.33% in group B respectively;the median survival time was(203?20.87) days in group A and(167?19.58) days in group B,the difference being significant(P

ABSTRACT

ive] To observe the therapeutic effects of Xiaoji Decoction (XD) in treating intermed iate and late stages of lung cancer. [ Methods] One hundred and twelve patients with lung cancer in stage ? -? were randomly allocated to Group A, Group B and Group C. Group A ( n = 49) was treated with XD alone, Group B ( n = 33 ) was treated with various chemotherapeutic regimens according to the histo-logical types of lung cancer: CAP regimen for lung squamous carcinoma, EP regimen for lung adenocarci-noma and CE regimen for small cell carcinoma of lung, In Group C, 30 patients were treated with chemo therapy combined with XD. Symptomatic relief, life quality, tumor size, distant metastasis, sub-types of T lymphocytes, survival period and side effects in the three groups were evaluated after one course of treatment. [Results] The effects of XD on tumor size were evaluated by the effective rate and stabilized rate. ABSTRACT

Objective To compare the pharmacological actions of Liandai Tablet(LT) and its main active components on apoptosis of gastric cancer cells and DNA damage. Methods Effects of Liandai Tablet(LT) and its main active components,berberine and indirubin,on growth and apoptosis of gastric cancer cell strain MGC_803 were explored and their effects on DNA damage were also studied. Results LT serum in high and low dosages and berberine could inhibit the growth of MGC_803 as compared with the control group,and typical morphological features of apoptosis were found in the MGC_803 by methyl green pyronin stain assay.But indirubin at various concentrations showed no obvious inhibitory effects. Agarose gel electrophoresis assay revealed that the MGC_803 cell DNA was split into large fragments when treated with berberine. Conclusion LT serum exerts a similar inhibitory effect on the growth and apoptosis of gastric cancer cells as compared with berberine.The effects of LT at various serum concentrations on MGC_803 DNA was less than that of berberine,and indirubin at the given concentration had no this effect.

ABSTRACT

【Objective】To observe the effect of Herba Hedyotis Diffusae(HHD)on the histological features of living murine ascites hepatoma H22 cells as well as CD+4 and CD+8 expression in T lymphocytes.【Methods】Special pathogen free(SPF)Kunming mice were randomized into 4 groups:model group,HHD(25 g/kg)group,Radix Codonopsis(RC,25 g/kg)group,heat-shock group.The murine ascites hepatoma H22 model was established by abdominal transplantation of H22 cells.The histological features of H22 cells from living murine ascites were examined through living mice ascites smear and giemsa staining.The CD+4 and CD+8 expression in ascites T lymphocytes was detected by immunohistochemical method.【Results】H22 cell deformation such as cell membrane incomplete and vesicant,some nucleus being migration and pycnosis was obvious in HHD and heat-shock groups.The expression of both CD+4 and CD+8 expression in T lymphocytes of HHD groups was increased,but only CD+4 expression in RC group and only CD+8 expression in heat-shock group was increased.【Conclusion】HHD exerts some inhibitory-killing effect on living murine H22 cells,and its mechanism may be related to the increase of immune function and to the cytotoxic effect.

ABSTRACT

【Objective】To investigate the influence of serum containing Hechan Tablets(HT)on the proliferation and apoptosis of human lung adenocarcinoma cell(LAC)line.【Methods】Serum containing HT was prepared.Human LAC line was cultured by the routine method firstly,and then was planted respectively in the culture medium including serum containing HT at the concentration of 30%(group A),in the culture medium including blank serum at the same concentration(group B)and in RPMI-1640 culture medium(group C).The growth of LAC was monitored under inverted phase contrast microscope,histological features of apoptotic LAC nucleus examined under fluorescence microscope and the apoptosis of LAC observed by flow cytometry with the methods of DNA content analysis and terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL).【Results】After treatment with serum containing HT,the apoptotic changes of LAC were as follows: apoptotic rate was 5.6% in group A,lower than 0.9% in group B and 0.1% in group C;DNA content analysis showed that the number of LAC which was in phase S and phase G2-M was less in group A than that in groups B and C 72 hours after the culturing,so did the cell proliferation index;the results of TUNEL examination showed that the incidence of cell apoptosis was 12.3% in group A,5.7% in group B and 1.2% in group C 72 hours after the culturing.【Conclusion】Serum containing HT has an inhibitory effect on the proliferation of LAC,and its mechanism may be related to inhibiting DNA synthesis and division of LAC,restraining LAC at the phase G0 and G1 and promoting LAC apoptosis.

ABSTRACT

[Objective] To investigate the effect of Lycium barbarum polysaccharide (LBP) on T-lymphocyte subsets levels, dendritic cells (DCs) counting and antigen CD80 expression in peripheral blood and tumor stroma of H22-bearing mice. [Methods] H22-bearing mice models were established. LBP in the dosages of 1.25 and 0.625 g?kg-1?d-1 was orally administered to the mice models for two weeks. The changes of T-lymphocyte subsets levels, DCs counting and antigen CDgo expression in peripheral blood and tumor stroma were detected by flow cytometry (FCM) . [ Results ] LBP increased the numbers of CD4+ and CD8+ T-cells, promoted the proliferation of CD4+ T-cells, and elevated the ratio of CD4+ /CD8+ in peripheral blood. ABSTRACT

Objective: To evaluate the antitumor effect of polysaccharide from spirulina platensis(PSP), and its synergic and attenuated effects on chemotherapeutic drugs in tumor-bearing mice. Methods: Mouse models of transplanting tumor were established. Antitumor effects of PSP and PSP combined with chemotherapeutic drugs were observed and the effect of PSP on leukopenia induced by chemotherapeutic drugs was also investigated. Results: PSP significantly inhibited S180 and H22, the inhibitory rate being 50 %-58 %and 43 %-51 %respectively. PSP evidently increased the antitumor action of cytoxan, PDD and 5-Fu, and elevated the number of white blood cells in leukopenia. Conclusion: PSP can obviously inhibit the tumor growth, also has synergic and attenuated effects on chemotherapeutic drugs.

ABSTRACT

Tumor-inhibiting experiment in vivo and tumor_cell_killing experiment in vitro were carried out in mice for Cortex Moutan Radicis, Caulis Lonicerae and Herba Lycopi.The results showed that the tumor_inhibiting rate of Cortex Moutan Radicis was over 50%,IC 50 38.403 mg/L,and its 95% confidence limit in 31.19 ~ 47.29 mg/L. The tumor_inhibiting rate of Caulis Lonicerae was more than 30%,IC 50 7.306 mg/L and its 95% confidence limit in 4.56 - 11.71 mg/L.The tumor_inhibiting rate of Herba Lycobi was less than 30%,IC 50 18.11 mg/L and its 95% confidence limit in 11.23 ~ 29.20 mg/L.It is suggested that Cortex Moutan Radicis,Caulis Lonicerae have certain antineoplastic effect.

ABSTRACT

[Objective] To compare the effects of heat-clearing and detoxifying herbs with that of tonifying herbs on mice hepatoma cells line H22, and to explore their immunologic mechanisms. [Methods] Herba Rabdosiae Rubescentis (HRR) and Herba Scutellariae Barbatae (HSB) were selected as the heat-clearing and detoxifying herbs, and Radix Astragali (RA) as the representative of tonifying herbs. Fifty KM mice were inoculated with hepatoma cells line H22 and then were randomized into five groups: HRR group (30 g?kg-1?d-1 by gavage for 12 days), HSB group (15 g?kg-1?d-1 by gavage for 12 days), RA group (15 g?kg-1?d-1 by gavage for 12 days) , heat-shock group (treated 20 min at 43℃, once every 4 days and three times in all) and model group. Twelve days later, the mice were executed and the ascites smears were prepared. The cell feature in ascites was observed under light microscope and the expression of CD4+ and CD8+ on the cellular membrane of the T lymphocyte in ascites was detected with immunohistochemical method. [Results] A large amount of damaged and apoptotic H22 cells were found in HRR, HSB and heat-shock groups, and the changes of cell feature in RA group were not obvious. The expressions of CD4+ and CD8+ on the membrane of T cells in ascites was increased in HRR and HRB groups, but the expression of CD4+ was increased in RA group, and CD8+ expression was increased in heat-shock group.[Conclusion] Heat-clearing and detoxifying herbs can induce the necrosis and apoptosis of mice hepatoma cells line H22 and the mechanism may be related to the increase of cellular immunity, thus enhancing the body immunity to kill the tumor cells. RA can not kill the tumor cells directly and its antitumor action may be related to the increase of CD4+ expression.

Objective To investigate the objectivity and comprehensiveness of Response Evaluation Criteria of Traditional Chinese Medicine for Solid Tumor(REC-TCM-ST) in the therapeutic effect evaluation of Chinese medicine for late non-small-cell lung cancer(NSCLC).Methods A retrospective study was carried out in 104 NSCLC patients in the stages of Ⅲ~Ⅳ.Of 104 patients,53 received traditional Chinese medicine(TCM group),and 51 received western medicine(WM group).The therapeutic effect in the two groups was evaluated with both REC-TCM-ST and Response Evaluation Criteria in Solid Tumor(RECIST).Results The REC-TCM-ST evaluation results showed that the effective rate on relieving tumor mass in TCM group was lower than that in WM group(P0.05);the symptom of weakness was much relieved,and effect on Karnofsky score was good in TCM group as compared with that in WM group(P0.05).As for the survival patients,TCM group had a median survival period of(8.0?1.2) months,and survival rates for half a year,one year and two years were 60.38%,24.53% and 3.77% respectively;WM group had a median survival period of(5.9?0.4) months,and survival rates for half a year,one year and two years were 41.18%,15.69% and 1.96% respectively.The RECIST evaluation results showed that the total effective rate and the total stable rate were 9.62% and 72.12% respectively,while were 34.62% and 84.62% respectively when evaluated with REC-TCM-ST,the difference being significant(P
