ABSTRACT

Frogs evolved terrestrial development multiple times, necessitating mechanisms to avoid ammonia toxicity at early stages. Urea synthesis from ammonia is a key adaptation that reduces water dependence after metamorphosis. We tested for early expression and plasticity of enzymatic mechanisms of ammonia detoxification in three terrestrial-breeding frogs: foam-nest-dwelling larvae of Leptodactylus fragilis (Lf) and arboreal embryos of Hyalinobatrachium fleischmanni (Hf) and Agalychnis callidryas (Ac). Activity of two ornithine-urea cycle (OUC) enzymes, arginase and CPSase, and levels of their products urea and CP in tissues were high in Lf regardless of nest hydration, but reduced in experimental low- vs. high-ammonia environments. High OUC activity in wet and dry nests, comparable to that under experimental high ammonia, suggests terrestrial Lf larvae maintain high capacity for urea excretion regardless of their immediate risk of ammonia toxicity. This may aid survival through unpredictably long waiting periods before rain enables their transition to water. Moderate levels of urea and CP were present in Hf and Ac tissues and enzymatic activities were lower than in Lf. In both species, embryos in drying clutches can hatch and enter the water early, behaviorally avoiding ammonia toxicity. Moreover, glutamine synthetase was active in early stages of all three species, condensing ammonia and glutamate to glutamine as another mechanism of detoxification. Enzyme activity appeared highest in Lf, although substrate and product levels were higher in Ac and Lf. Our results reveal that multiple biochemical mechanisms of ammonia detoxification occur in early life stages of anuran lineages that evolved terrestrial development.

ABSTRACT

This study evaluated the multivariate relationship among the crude protein (CP) intake and digestibility, the neutral detergent fiber (NDF) intake and digestibility, and the nitrogen excretion in hair sheep fed Mombasa grass silage mixed with açai palm seeds using canonical correlation analysis (CCA). For this purpose, a dataset was constructed using the information derived from a metabolic experiment with Santa Ines sheep (BW ± SE: 33.9 ± 1.37 kg) allocated into a 5 × 5 Latin square. The following two multivariate relationships were explored using CCA: (1) the N in both feces (Nfeces) and urine (Nurine) (canonical variate U) versus the intake of both CP (CPI) and NDF (NDFI) (canonical variate V). (2) the Nfeces and Nurine (canonical variate W) versus the digestibility of both CP (CPD) and NDF (NDFD) (canonical variate Z). Canonical loadings for relationship No. 1 revealed that Nfeces (0.971) and Nurine (- 0.249) had a positive and negative correlation, respectively, with U. Both CPI (0.960) and NDFI (0.997) had a positive and similar correlation with V. Regarding relationship No. 2, both Nfeces (0.860) and Nurine (0.500) had a positive correlation with W. Conversely, both CPD (- 0.910) and NDFD (- 0.889) had a negative and similar association with Z. In conclusion, our data showed that when a multivariate approach is used, an increase in both CPI and NDFI produces an increase in Nfeces, as well as a decrease in Nurine. Additionally, a decrease in both CPD and NDFD produces an increase in both Nfeces and Nurine.

ABSTRACT

O objetivo deste estudo foi avaliar o balanço de nitrogênio e a concentração de ureia no plasma em cabritos alimentados com dietas contendo farinha de pupunha em substituição ao milho (0, 10, 40, 60 e 85% MS). Trinta cabritos Boer × SRD, com 90 dias de idade e peso corporal inicial de 16,7 ± 3,5kg, foram distribuídos em um delineamento inteiramente ao acaso, com seis repetições. As dietas foram fornecidas diariamente ad libitum para permitir 10% das sobras. Três coletas foram realizadas a cada 28 dias. As dietas foram constituídas por milho, farelo de soja, farinha de pupunha, suplemento mineral e feno de Tifton-85, com a relação volumoso: concentrado de 30:70. O consumo de nitrogênio (N) diminuiu linearmente à medida que a farinha de pupunha substituiu o milho no concentrado. Com relação ao N digerido, os animais alimentados com níveis de farinha de pupunha apresentaram valores inferiores aos alimentados com o milho como única fonte de energia, com médias de 14,9 e 17,1g dia-1, respectivamente. A excreção urinária de N diminuiu à medida que a farinha de pupunha substituiu o milho no concentrado. O nível de 39% de farinha de pupunha reduziu a excreção urinária de ureia. Houve retenção máxima de N em 7,83g dia-1 para o nível de substituição de milho de 28,9% em farinha de pupunha.(AU)

The growth of jundiá (Rhamdia quelen Quoy and Gaimard) juveniles (31.54 ± 4.92 g) fed five diets comprising increasing levels of digestible protein (DP: 24, 29, 34, 39, and 44 %) and digestible energy (DE: 12.13, 13.39, 13.81, 14.64, and 15.06 MJ kg−1) with DP:DE ratios: 20, 22, 25, 27, and 29 g MJ−1, respectively, was evaluated. Each diet was fed to three groups of 25 fish for 75 days. Regression analysis showed that the different DP:DE ratios affected both growth and economic performance as well as excretion of total ammonia nitrogen. Fish fed diets with DP:DE ratios between 22 and 27 g MJ−1 achieved a higher daily weight gain, superior apparent net protein utilization, feed conversion rates, and lower feed costs. For the economically viable and environmentally friendly farming of jundiá juveniles in the initial grow–out stage (30 to 80 g average weight), the best DP:DE ratio was 25 g MJ−1 DP:DE (34.05 % DP and 13.81 MJ kg−1 DE).(AU)

ABSTRACT

The excretion of urinary nitrogen (N), one of the most important environmental contaminants from livestock systems, is highly correlated with milk urea N content. The objective of this research was to evaluate the use of different types of corn supplementation on milk urea N in grazing dairy cows. Twelve Holstein × Jersey lactating dairy cows were divided into six uniform groups according to milk production, lactation stage and live weight. Treatments were compared according to a 3 × 3 replicated Latin square experimental design, with three periods of seventeen days (twelve days to adaptation and five to measurements). The experimental treatments were exclusively grazing (G); grazing + supplementation with 4.2 kg DM of corn silage (CS) and grazing + supplementation with 3.2 kg DM of ground corn (GC). The pasture used was annual ryegrass (Lolium multiflorum L.) and white oats (Avena sativa L.). The milk protein production increased 65 g/day in the GC treatment group compared to the G and CS groups. The supplemented dairy cows showed lower milk urea N (-2.8 mg/dL) than unsupplemented cows, but the N utilization efficiency (g N output in milk/ g N intake) did not change between treatments (average = 0.26). Additionally, there was a relationship between milk and plasma urea nitrogen concentrations (R2 = 0.64). In conclusion, for dairy cows grazing annual temperate pastures, corn ground supplementation increased milk protein production and reduced the excretion of milk urea N, whereas corn silage reduced the excretion of milk urea N without affecting milk protein production.(AU)


A excreção de nitrogênio (N) urinário é um dos mais importantes contaminantes ambientais nos sistemas de produção animal, e possui alta correlação com as concentrações de N ureico no leite. Objetivou-se avaliar a suplementação com grão ou silagem de milho na excreção de N ureico e produção de proteína no leite de vacas em pastos anuais de inverno. Doze vacas em lactação cruza Holandês × Jersey foram divididas em seis lotes uniformes de acordo com a produção de leite, estádio de lactação e peso vivo. Os tratamentos foram comparados de acordo com um delineamento experimental em duplo Quadrado Latino 3 × 3, com três períodos de dezessete dias (doze dias de adaptação e cinco de avaliações). Os tratamentos experimentais foram exclusivamente pastejo (G), suplementação com 4.2 kg de MS de silagem de milho (CS) e 3.2 kg de MS de milho moído (GC). O pasto utilizado foi Azevém anual (Lolium multiflorum L.) e Aveia branca (Avena sativa L.). A produção de proteína no leite aumentou 65 g/dia nos animais do grupo GC quando comparados aos dos grupos G e CS. As vacas suplementadas reduziram os teores de N ureico do leite em comparação às não suplementadas (-2,8 mg/dL), mas a eficiência de uso do N (g N produzido no leite/g N ingerido) foi semelhante entre os tratamentos (média = 0,26). Além disso, houve uma relação positiva entre as concentrações de N ureico no leite e no plasma (R2 = 0.64). Balancing diets for protein affects both protein and energy use efficiency in cattle. This study aimed to evaluate the effects of dietary protein level on intake, digestibility and energy expenditure in Holstein heifers. Four Holstein heifers with a mean BW of 266.5 ± 10.7 kg were distributed in a 4 x 4 balanced Latin square design. Each period was comprised of 14 d for adaptation and 6 d for samplings. The heifers were fed diets with 9, 12, 15 and 18% of crude protein (CP). Total feces and urine were collected. Rumen fluid was collected at 2, 4, 6 and 8 h after feeding. The energy expenditure was evaluated at 6 a.m., 12 a.m., 6 p.m. and 12 p.m. by a closed-circuit mask technique. The intake of dry matter (DM), organic matter (OM) and neutral detergent fiber (NDF) were similar between treatments (P > 0.05). Although the NDF digestibility was not changed (P > 0.05), the digestibilities of DM, OM, CP, non fibrous carbohydrates, and total digestible nutrients increased linearly (P < 0.05) with increasing CP levels in the diet. Urinary urea nitrogen excretion and nitrogen retained increased linearly (P < 0.01) with increasing CP levels. Dietary CP levels did not affect (P =0.53) energy expenditure in heifers. Ruminal ammonia-nitrogen concentration peaked at 2-4 h after feeding. Increasing dietary CP levels from 9 to 18% does not affect feed intake and energy expenditure in heifers. O balanceamento das dietas proteicas afeta tanto a eficiência no uso de proteínas quanto a eficiência no uso de energia em bovinos. Este estudo teve como objetivo avaliar os efeitos dos níveis de proteína na dieta sobre consumo, digestibilidade e gasto de energia em novilhas Holandesas. Quatro novilhas Holandesas com um peso corporal (PC) médio de 266,5 ± 10,7 kg foram distribuídas em delineamento quadrado latino 4 x 4. Cada período era composto de 14 d para adaptação e 6 d para amostragens. As novilhas foram alimentadas com 9,12,15 e 18% de proteína bruta (PB). Fezes e urina totais foram coletadas. O líquido ruminal foi coletado às 2, 4, 6 e 8 h após a alimentação. O gasto de energia foi avaliado às 6h, 12h, 18h e 12h por uma técnica de máscara de circuito fechado. O consumo de matéria seca(MS), matéria orgânica (MO) e fibra em detergente neutro (FDN) foram similares entre os tratamentos(P > 0,05). Embora a digestibilidade do FDN não tenha sido alterada (P > 0,05), a digestibilidade de MS, MO, PB, carboidratos não-fibrosos e nutrientes digestíveis totais aumentou linearmente (P < 0,05)com o aumento dos níveis de PB na dieta. A excreção de uréia urinária e o nitrogênio retido aumentaram linearmente (P < 0,01). Os níveis de PB na dieta não afetaram (P = 0,53) o gasto de energia em novilhas. A concentração de nitrogênio amoniacal atingiu o pico em 2-4 h após a alimentação. O aumento dos níveis dietéticos de PB de 9 para 18% não afeta o consumo de ração e o gasto de energia em novilhas. Entretanto, melhora a digestibilidade da MO e a retenção de nitrogênio.(AU)

The objective of this work was to evaluate whether changes in time of herbage allocation and herbage mass (HM) (low (L) or medium (M)) modify the diurnal pattern of urinary nitrogen (N) concentration and ruminal ammonia (NH3) of lactating dairy cows. Four Holstein-Friesian cows fitted with rumen cannula were randomly allocated to one of four treatments: 1) low herbage mass in the morning (L-AM) (Access to new herbage allocation after morning milking with a herbage mass (HM) of 2000 kg DM/ha); 2) low herbage mass in the afternoon (L-PM) (Access to new herbage allocation after afternoon milking with a HM of 2000 kg DM/ha); 3) medium herbage mass in the morning (M-AM) (Access to new herbage allocation after morning milking with a HM of 3000 kg DM/ha); and 4) medium herbage mass in the afternoon (M-PM) (Access to new herbage allocation after afternoon milking with a HM of 3000 kg DM/ha). A four by four Latin Square design with four treatments, four cows, and four experimental periods was used to evaluate treatment effects. Rumen NH3 concentration was greater for L-AM compared to L-PM and M-PM at 13:00 and 16:00 h. Urine urea and N concentrations were lower for M-AM compared to L-AM. Urine N concentration was greater for L-AM than other treatments at 10:00 hours and greater for M-PM compared to M-AM at 16:00 hours. Results suggest that maintaining the cows in the holding pen at the milking parlor for two hours after morning grass silage supplementation for L-AM and for two hours after afternoon grass silage supplementation for M-PM, could allow collection of urine from cows at the holding pen and storage of urine in the slurry pit during the time of peak N concentration, returning cows to the pasture at a time of day when urinary N concentration is decreased.

ABSTRACT

The aim of this study was to evaluate the effects of the order of grass silage (GS) and maize silage (MS) supplementation on milk yield, grazing behavior and nitrogen (N) partitioning of lactating dairy cows during autumn. Thirty-six Holstein-Friesian dairy cows were randomly assigned to one of three treatments, and cows remained on these treatments for a 62 days period: (1) MIX; cows supplemented with 3 kg of dry matter (DM) of silage containing 1.5 kg DM of MS and 1.5 kg DM of GS in both the morning and afternoon; (2) GS-MS; cows supplemented with 3 kg DM of GS in the morning and 3 kg DM of MS in the afternoon; (3) MS-GS; cows supplemented with 3 kg DM of MS in the morning and 3 kg DM of GS in the afternoon. All cows received a pasture allowance of 17 kg DM/cow/d and 3 kg DM of concentrate. Grazing time and pasture intake were unaffected by treatment; however, milk production was greater for MS-GS, while milk protein was greater for GS-MS. Urinary N excretion was greater for MS-GS than MIX. In conclusion, MS-GS resulted in high milk yield but also high urinary N excretion, while MIX resulted in low urinary N excretion but also decreased milk yield.

ABSTRACT

Experiment I: T1-1 =  basal diet with 25% crude protein (CP) + limiting amino acids (LA); T1-2  = 20% CP + LA; T1-3  = 20% CP + LA + L-glycine; T1-4  = 20% CP + LA + L-glutamate; T1-5  = 20% CP + LA + L-glycine + L-glutamate. Experiment II: T2-1  = basal diet with 22% CP + LA; T2-2  = 20% CP + LA; T2-3  = 17.6% CP + LA + L-glycine; T2-4  = 17.6% CP + LA + L-glutamate; T2-5  = 17.6% CP + LA + L-glycine + L-glutamate. The reduction of dietary protein based on the concept of ideal protein decreases nitrogen excretion in quails when L-glycine is added to the diets. Quails fed diets supplemented with L-glutamate as the non-specific nitrogen source equivalent to the nitrogen level of the control diet had increased nitrogen excretion. However, quails had reduced nitrogen excretion in both experiments when L-glycine was added to diets with L-glutamate. Carcass fat was increased by reducing dietary protein, but fat deposition was reduced by adding L-glutamate and L-glycine, or both. The dietary addition of L-glutamate and L-glycine in quails based on the ideal protein concept is not necessary (Exp. I). Although the total nitrogen, electrolytic balance and glycine level were adjusted in diets, quails had decreased performance. Therefore, other hypotheses besides protein reduction need to be studied (Exp. II). Protein reduction with supplementation of only limiting essential amino acids does not affect quail performance. Dietary addition of L-glycine reduces nitrogen excretion.

ABSTRACT

The objective was to quantify the flow of intestinal nutrients and nitrogen excretion and retention in sheep receiving isoproteic diets. Eight Texel x Lacaune wethers (average body weight = 25±2.5 kg) were fitted with duodenal cannula and housed in metabolic cages. Wethers were assigned to the treatments in a crossover design with two periods of 20 days each, and all feces and urine produced by the wethers were collected. The treatments consisted of two isoproteic (160 g kg−1 of crude protein on dry matter basis) diets composed of red clover (RC) or lucerne (LU; Medicago sativa) silages plus corn silage and concentrate feed. The digestible organic matter and metabolizable energy intake did not differ between treatments. The intestinal non-ammonia N (NAN) flow was 5.9 g day−1 (37%) higher in RC wethers than in those of the LU treatment. This result was a consequence of both an increase in the efficiency of microbial protein synthesis (12.7% higher) and a decrease in ruminal degradable protein (RDP) content (20% lower) of the diet. However, the increase in the intestinal NAN flow was accompanied by a reduction in intestinal digestibility of N, resulting in similar daily N retention between treatments. The reduction of RDP content was probably the main reason for reductions in N urinary excretion in RC wethers compared with those in the LU treatment, showing that RC silage may be a tool for mitigating the impact of N excretion in ovine production systems, without changes in N retention.(AU)

Condensed tannins can reduce protein degradation in the rumen by increasing the duodenal flow of metabolizable proteins and consequently improving animal performance. The objective of this study was to evaluate the effect of the tannin extract of Acacia mearnsii on lactating ewes receiving a total mixed ration (TMR) containing two concentrations of crude protein. The experimental treatments consisted of a TMR containing 16.4% or 22.3% of crude protein in dry matter either with or without the tannin extract of Acacia mearnsii at 20 g kg-1 dry matter. The extract concentration that guaranteed the daily intake of approximately 15 g of the extract per animal was determined. Eight lactating ewes of the Texel × Lacaune breed in the second third of lactation were distributed in a 4 × 4 Latin square experimental design, with four periods of 19 days, including 14 days of adaptation and 5 days of sample collection. Urinary nitrogen excretion was increased in the animals fed a diet containing a relatively higher protein concentration compared to those receiving a diet with a lower protein concentration. However, milk production and the concentration of milk solids did not vary significantly among treatments. The tannin extract of Acacia mearnsii decreased urinary nitrogen excretion, and consequently reduced the environmental impact of the production system, but did not significantly improve animal performance.(AU)


Os taninos condensados podem reduzir a degradação proteica no rúmen, aumentando o fluxo duodenal de proteína metabolizável com melhorias no desempenho animal. Objetivou-se avaliar o efeito do extrato tanífero de Acacia mearnsii em ovelhas lactantes recebendo ração totalmente misturada (RTM) com dois níveis de proteína bruta. Os tratamentos experimentais constituíram-se de RTM com 16,4 e 22,3% de PB na MS, com ou sem a adição de extrato tanífero de Acacia mearnsii na proporção de 20 g kg-1 na matéria seca. A quantidade de extrato foi calculada para permitir a ingestão de aproximadamente 15 g dia-1 por animal. Foram utilizadas oito ovelhas cruza da raça Texel × Lacaune no terço médio de lactação distribuídas em delineamento experimental em Quadrado latino 4 × 4, com quatro períodos de 19 dias, sendo 14 de adaptação e cinco de coleta. A excreção urinária de N aumentou nos animais recebendo a dieta com maior nível de proteína em comparação aos que receberam a dieta com menor nível de proteína. Entretanto, a produção de leite e de sólidos não variou entre os tratamentos. A suplementação com extrato tanífero de Acacia mearnsii reduziu a excreção urinária de N, o que reduz o impacto ambiental do sistema, mas não foi suficiente para melhorar o desempenho animal.(AU)

The estimated protein retention rate showed a positive quadratic response to dietary CP with maximum retention at the 510.20g kg-1 protein level, and increasing levels of protein resulted in a greater total ammonia excretion rate in water. The mean apparent protein digestibility coefficient values increased with increasing levels of dietary protein up to 520g kg-1, where they stabilized. As determined by regression, the level of dietary CP that promotes the best digestibility was 495.62g kg-1.(AU)

ABSTRACT

Three hundred and twenty-four DeKalb White laying hens aged 42 weeks were distributed in a completely randomised design with nine treatments and six replicates of six birds in each treatment. The experiment lasted 112 days. Diets were: T1 = 16.02% crude protein - CP [Met + Lys + Thr + Trp + Val]; T2 = 14.02% CP [Met + Lys + Thr + Trp + Ile + Val]; T3 = 14.02% CP [no amino acid supplementation]; T4 = 14.02% CP [Met + Lys + Thr + Trp]; T5 = 14.02% CP [Met + Lys + Thr]; T6 = 14.02% CP [Met]; T7 = 14.02% CP [Lys]; T8 = 14.02% CP [Thr]; T9 = 14.02% CP [Trp]. Regarding the quality of the eggs, the percentage of yolk and albumen, shell thickness and Haugh unit were not affected by the different diets. The percentage of shell, specific gravity and albumen height showed significant differences. We found that supplementation of only one amino acid in the diet (T7, T8 or T9), with the exception of methionine (T6), worsened performance relative to the control. Supplementation of three amino acids (methionine, lysine and threonine; T5) or four amino acids (methionine, lysine, threonine and tryptophan; T4) worsened egg production and conversion per mass and per dozen eggs; however, feed intake and egg weight and mass were similar to the control treatment. Trezentos e vinte e quatro galinhas poedeiras DeKalb White com idades de 42 semanas foram distribuídas em um delineamento experimental inteiramente casualizado, com nove tratamentos e seis repetições de seis aves em cada tratamento. O experimento teve duração de 112 dias. As dietas foram: T1 = 16,02% - PB [Met + Lys + Thr + Trp + Val]; T2 = 14,02% PB [Met + Lys + Thr + Trp + Ile + Val]; T3 = 14,02% de PB [sem suplementação de aminoácidos]; T4 = 14,02% CP [Met + Lys + Thr + Trp]; T5 = 14,02% CP [Met + Lys + Thr]; T6 = 14,02% CP [Met]; T7 = 14,02% CP [Lys]; T8 = 14,02% CP [Thr]; T9 = 14,02% CP [Trp]. Em relação à qualidade dos ovos, a porcentagem de gema e albúmen, espessura de casca e a unidade Haugh não foram afetadas pelas dietas diferentes. A percentagem de casca, gravidade específica e altura do albúmen mostraram diferenças significativas. Verificou-se que a suplementação de apenas um aminoácido na dieta (T7; T8 ou T9), com a exceção de metionina (T6), piorou o desempenho das aves em relação a aves que consumiram a dieta controle. A suplementação de três aminoácidos (metionina, lisina e treonina; T5) ou quatro aminoácidos (metionina, lisina, treonina e triptofano; T4) agravou a produção de ovos e conversão por massa e por dúzia de ovos; no entanto, o consumo de ração e peso dos ovos e massa de ovos, o desempenho foi semelhante ao tratamento controle T1.
