ABSTRACT
Despite the record speed of developing vaccines and therapeutics against the SARS-CoV-2 virus, it is not a given that such success can be secured in future pandemics. In addition, COVID-19 vaccination and application of therapeutics remain low in developing countries. Rapid and low cost mass production of antiviral IgY antibodies could be an attractive alternative or complementary option for vaccine and therapeutic development. In this article, we rapidly produced SARS-CoV-2 antigens, immunized hens and purified IgY antibodies in 2 months after the SARS-CoV-2 gene sequence became public. We further demonstrated that the IgY antibodies competitively block RBD binding to ACE2, neutralize authentic SARS-CoV-2 virus and effectively protect hamsters from SARS-CoV-2 challenge by preventing weight loss and lung pathology, representing the first comprehensive study with IgY antibodies. The process of mass production can be easily implemented in most developing countries and hence could become a new vital option in our toolbox for combating viral pandemics. This study could stimulate further studies, optimization and potential applications of IgY antibodies as therapeutics and prophylactics for human and animals.

This retrospective cohort study aimed to identify risk factors for the failure of transfer of passive immunity (FPI) in preweaned dairy calves, explore its associations with morbidity, mortality, genetics, and determine a standardized cut-off point for FPI. Analyzing data from 6011 calves, factors such as sire predicted transmitted ability for milk (PTA), birth season, retained placenta (RP), total serum protein concentration (TSP), morbidity (neonatal calf diarrhea - NCD, bovine respiratory disease - BRD, tick-borne disease - TBD), mortality, and average daily weight gain (ADG) were considered. The calves were categorized into predominantly Gyr (PG) and predominantly Holstein (PH) genetic composition groups. Multivariate mixed logistic regression revealed optimal TSP cut-off points for predicting morbidity (7.6 g/dL) and mortality (6.9 g/dL). PH calves exhibited 1.35 times higher odds of FPI and 1.48 times greater odds of disease. Calves from multiparous cows and those born to dams with RP had increased FPI odds. Disease prevalence was 53%, with 41% NCD, 18% BRD, and 10% TBD. Season, parity, PTA, and birth weight were associated with disease odds, though FPI was not a reliable predictor. The mortality percentage was 6%, with PH calves and those with a positive PTA having higher odds. The ADG was 0.64 kg, and FPI-affected calves gained less weight. The study compared and identified various risk factors that potentially impact calf immunity. However, the use of a standardized cut-off point for FPI assessment was not effective in predicting morbidity and mortality at this specific farm.

A colostragem é essencial para a saúde dos bezerros neonatos, uma vez que não há a transferência de imunidade através da placenta, ou seja, o contato inicial do organismo com anticorpos se dá através da primeira mamada, onde há transferência da imunidade passiva. Sendo assim, o objetivo do trabalho é revisar os benefícios da acidificação ou silagem do colostro para otimizar a conservação da dieta líquida em fazendas; analisar o impacto no desempenho de bezerros em comparação com o colostro tradicional refrigerado em aleitamentos convencionais. O volume ideal preconizado é, no mínimo, 10% do peso vivo do animal nas primeiras duas horas e mais 5% do peso vivo nas seis a oito horas seguintes a primeira ingestão, para que se obtenha um bom desenvolvimento durante o crescimento, caso contrário, a falta da administração do colostro nas primeiras horas de vida predispõe enfermidades, tais como pneumonia e diarreia, prejudicando assim a saúde e consequentemente o desempenho em relação a outros animais que receberam uma colostragem adequada. Problemas com a qualidade do colostro surgem devido ao armazenamento inadequado, especialmente em propriedades sem refrigeração. A falta de sistemas de congelamento resulta em administração de leite em temperatura ambiente por períodos prolongados, prejudicando assim a imunidade e nutrição dos bezerros durante a colostragem. Todavia, há alternativas para o problema tal como o fornecimento de silagem de colostro. Em alguns casos, o processo de acidificação demanda a adição de ácidos no leite, a fim de evitar o crescimento de microrganismos patogênicos. O principal aspecto positivo do leite acidificado é a manutenção em temperatura ambiente, ou seja, não há a necessidade de passar por processos de refrigeração.
Colostrum is essential for the health of newborn calves, since there is no transfer of immunity through the placenta, that is, the body's initial contact with antibodies occurs through the first feeding, where there is a transfer of passive immunity. Therefore, the objective of the work is to review the benefits of acidifying or colostrum silage to optimize the conservation of liquid diets on farms; analyze the impact on calf performance compared to traditional refrigerated colostrum in conventional sucklers. The recommended ideal volume is at least 10% of the animal's live weight in the first two hours and a further 5% of its live weight in the six to eight hours following the first ingestion, so that good development is achieved during growth, otherwise, the lack of colostrum administration in the first hours of life predisposes diseases, such as pneumonia and diarrhea, thus harming health and consequently performance in relation to other animals that received adequate colostrum. Problems with colostrum quality arise due to inadequate storage, especially in unrefrigerated properties. The lack of freezing systems results in milk being administered at room temperature for prolonged periods, thus damaging the calves immunity and nutrition during colostrum. However, there are alternatives to the problem such as the supply of colostrum silage. In some cases, the acidification process requires the addition of acids to the milk in order to prevent the growth of pathogenic microorganisms. The main positive aspect of acidified milk is that it remains at room temperature, that is, there is no need to undergo refrigeration processes.
El calostro es esencial para la salud de los terneros recién nacidos, ya que no existe transferencia de inmunidad a través de la placenta, es decir, el contacto inicial del cuerpo con los anticuerpos ocurre a través de la primera alimentación, donde existe una transferencia de inmunidad pasiva. Por lo tanto, el objetivo del trabajo es revisar los beneficios de acidificar o ensilar el calostro para optimizar la conservación de dietas líquidas en granjas; analizar el impacto en el rendimiento de los terneros en comparación con el calostro refrigerado tradicional en lechones convencionales. El volumen ideal recomendado es al menos el 10% del peso vivo del animal en las dos primeras horas y otro 5% de su peso vivo en las seis a ocho horas siguientes a la primera ingesta, para que se consiga un buen desarrollo durante el crecimiento, en caso contrario. la falta de administración de calostro en las primeras horas de vida predispone a enfermedades, como neumonía y diarrea, perjudicando la salud y consecuentemente el rendimiento en relación a otros animales que recibieron el calostro adecuado. Los problemas con la calidad del calostro surgen debido a un almacenamiento inadecuado, especialmente en propiedades no refrigeradas. La falta de sistemas de congelación provoca que la leche se administre a temperatura ambiente durante períodos prolongados, dañando así la inmunidad y la nutrición de los terneros durante el calostro. Sin embargo, existen alternativas al problema como el suministro de ensilaje de calostro. En algunos casos, el proceso de acidificación requiere la adición de ácidos a la leche para evitar el crecimiento de microorganismos patógenos. El principal aspecto positivo de la leche acidificada es que se mantiene a temperatura ambiente, es decir, no es necesario someterse a procesos de refrigeración.

Resumen OBJETIVO: Identificar la coexistencia de anticuerpos contra la proteína "Spike" del SARS-CoV-2 en sangre del cordón umbilical de recién nacidos de madres vacunadas contra la COVID-19 durante el embarazo. MATERIALES Y MÉTODOS: Estudio transversal, llevado a cabo en un centro de atención privada de la Ciudad de México, entre junio del 2021 y febrero del 2022, al que se incluyeron mujeres que completaron el esquema de vacunación contra SARS-CoV-2 durante el embarazo y en quienes en el trascurso de la finalización del embarazo parto o cesárea se haya obtenido una muestra de sangre de cordón umbilical para identificar la proteína Spike. RESULTADOS: Se registraron 219 pacientes y se analizaron 5 tipos de vacuna de dos plataformas diferentes; Pfizer®, Moderna®, AstraZeneca®, J&J/Janssen® y Sputnik®. La concentración de anticuerpos fue mayor en pacientes vacunadas con Moderna® y Sputnik®. Mediante el análisis de regresión de Cox se evaluó un modelo predictivo, sin mostrar diferencia estadísticamente significativa en la cantidad de anticuerpos generados luego de la primera y segunda dosis de la vacuna. CONCLUSIONES: La vacunación prenatal contra la COVID-19 induce una fuerte respuesta humoral materna, que se transfiere de manera efectiva al feto, incluso con una sola dosis.
Abstract OBJECTIVE: Identification of the presence of SARS-CoV-2 Spike protein antibodies in cord blood of newborns following maternal COVID-19 vaccination during pregnancy. MATERIALS AND METHODS: Cross-sectional study in vaccinated women during pregnancy who had a cord blood sample taken during the surgical procedure Vaginal birth/Cesarean section to measure spike protein antibodies. RESULTS: We registered 219 patients in the study: five types of vaccines, from two different platforms, were tested: Pfizer®, Moderna®, AstraZeneca®, J&J/Janssen® and Sputnik®. The levels of antibodies were greater in those patients vaccinated with Moderna® and Sputnik®. Through a Cox regression we made a predictive model where we observed that there are no differences in the number of antibodies generated after the first and second vaccine dose. CONCLUSIONS: Prenatal vaccination against COVID-19 induces a strong maternal humoral response that is effectively transferred to the fetus, even with a single dose.
ABSTRACT
This study evaluated the vaccine-induced serological response after administering four commercial formulations containing Bovine Viral Diarrhea Virus (BVDV) type-1, BVDV-2, and Bovine Herpesvirus type 1 (BoHV-1) to young heifers with circulating maternal antibody titers. The study also determined the anamnestic response to vaccinations after the complete metabolization of maternal antibodies when the calves reached six months. Eighty-seven Holstein heifers were selected and randomly distributed into four experimental groups based on the vaccine administered. The four vaccine-based experimental groups were as follows: vaccine A (n = 22), vaccine B (n = 22), vaccine C (n = 24), and vaccine D (n = 19), given on the 60th, 90th, and 180th day of life, respectively. Virus-neutralization (VN) tests were performed at baseline (day 60) and 30 days after administering the second (day 120) and third doses (day 210). We analyzed the effect of vaccine group (P ≤ 0.001), day of vaccination (P ≤ 0.001), and group versus vaccine day interaction (P ≤ 0.001) for antibody titers produced against BVDV-1, BVDV-2, and BoHV-1 using the PROC MIXED method (Statistical Analysis System -SAS 9.4). Antibody titers against BVDV-1, BVDV-2, and BoHV-1 were similar at baseline and on day 60 of life. The mean antibody titers were constant and persisted against BVDV-1 in heifers immunized with vaccines A and C. Heifers immunized with vaccine A alone had a similar effect against BVDV-2. Regarding BoHV-1, the antibody titers decreased between days 60 and 210 in groups B, C, and D. The antibody titer for heifers in group A also decreased between days 60 and day 120, and an intense increase in titers was observed on day 210. After being immunized with formulations B, C, and D, the frequency of animals with titers above protective levels for BVDV-1, BVDV-2, and BoHV-1 was very low or null. Indicators of anamnestic response were observed in heifers vaccinated with formulation A only. Therefore, it can be concluded that passive immunity negatively interferes with antibody production after vaccination.
O objetivo desse estudo foi avaliar a resposta sorológica induzida após vacinação com quatro formulações comerciais contendo o BVDV-1, BVDV-2 e BoHV-1 em novilhas jovens com títulos de anticorpos maternos circulantes, assim como analisar uma possível resposta anamnéstica em vacinações, após a completa metabolização dos anticorpos maternais aos seis meses de idade. Foram selecionadas 87 novilhas Holandesas, distribuídas aleatoriamente em quatro grupos experimentais de acordo com as vacinas: vacina A (n = 22), vacina B (n = 22), vacina C (n = 24) e vacina D (n = 19), aplicadas aos dias 60, 90 e 180 de vida. Testes de vírus-neutralização (VN) foram realizados no momento basal (D60) e 30 dias após a aplicação da 2ª dose (D120) e 3ª dose (D210). Observou-se efeito do grupo (P ≤ 0,001), dia da vacinação (P ≤ 0,001), e interação grupo versus dia (P ≤ 0,001) para os títulos de anticorpos produzidos contra o BVDV-1, BVDV-2 e BoHV-1, por meio do comando PROC MIXED (Statistical Analysis System, versão 9,4). Os títulos de anticorpos contra o BVDV-1, BVDV-2 e BoHV-1 eram semelhantes no momento basal aos 60 dias de vida. Os títulos médios de anticorpos foram constantes e persistentes contra o BVDV-1 nas novilhas imunizadas com as vacinas A e C, porém apenas a vacina A teve este mesmo perfil contra o BVDV-2. Em relação ao BoHV-1, os grupos B, C e D apresentaram queda nos títulos de anticorpos do D60 ao D210, enquanto as novilhas do grupo A apresentaram queda do D60 ao D120, com aumento intenso dos títulos no D210. A frequência de animais com títulos acima dos protetores contra o BVDV-1, BVDV-2 e BoHV-1 foram muito baixos ou nulos após as vacinações nos animais imunizados com as formulações B, C e D. Sugere-se que a imunidade passiva interferiu negativamente na indução de anticorpos pelas vacinas, observando-se indicadores de resposta anamnéstica apenas para as novilhas vacinadas com a formulação A.

Birth and the first few weeks of age are critical periods of developing the immune system of puppies and kittens and adapting to an environment containing a variety of infectious agents. The survival rate during these periods depends mainly on the newborn's immune capacity to prevent and combat infections. Although most components of innate and adaptive immunity are present at birth, responses are slow and immature compared to adults. Due to immunological immaturity and the endotheliochorial placental structure, circulating concentrations of immunoglobulins in dogs and cats at birth are quite low. Thus, newborns need a prompt and immediate immune response, which is essentially provided by defense cells and maternal antibodies via colostrum. Failure to ingest colostrum is correlated with high mortality rates in the neonatal period. Concurrently, factors related to pregnant, such as pregnancy physiological immunosuppression and nutritional and health states, can directly influence newborn immunity and health. Therefore, understanding the maternal and neonatal immunological aspects, importance of colostrum, risk factors for failure to transfer passive immunity and colostrum substitute strategies are essential to ensure the survival of the litter.

Objective: To understand the dynamics of Zika virus (ZIKV)-specific antibody immunity in children born to mothers in a flavivirus-endemic region during and after the emergence of ZIKV in the Americas. Methods: We performed serologic testing for ZIKV cross-reactive and type-specific IgG in two longitudinal cohorts, which enrolled pregnant women and their children (PW1 and PW2) after the beginning of the ZIKV epidemic in Nicaragua. Quarterly samples from children over their first two years of life and maternal blood samples at birth and at the end of the two-year follow-up period were studied. Results: Most mothers in this dengue-endemic area were flavivirus-immune at enrollment. ZIKV-specific IgG (anti-ZIKV EDIII IgG) was detected in 82 of 102 (80.4%) mothers in cohort PW1 and 89 of 134 (66.4%) mothers in cohort PW2, consistent with extensive transmission observed in Nicaragua during 2016. ZIKV-reactive IgG decayed to undetectable levels by 6-9 months in infants, whereas these antibodies were maintained in mothers at the year two time point. Interestingly, a greater contribution to ZIKV immunity by IgG3 was observed in babies born soon after ZIKV transmission. Finally, 43 of 343 (13%) children exhibited persistent or increasing ZIKV-reactive IgG at ≥9 months, with 10 of 30 (33%) tested demonstrating serologic evidence of incident dengue infection. Conclusions: These data inform our understanding of protective and pathogenic immunity to potential flavivirus infections in early life in areas where multiple flaviviruses co-circulate, particularly considering the immune interactions between ZIKV and dengue and the future possibility of ZIKV vaccination in women of childbearing potential. This study also shows the benefits of cord blood sampling for serologic surveillance of infectious diseases in resource-limited settings.

The intensification of ewe production, focusing on increasing prolificity, results in a higher incidence of metabolic disorders, especially in the transition period, which can lead to production losses with consequences for the health of the animals. The objective of this study was to evaluate the metabolic profile in transition period Lacaune ewes according to the parity and litter size born and its influence on the transference of passive immunity. Nineteen Lacaune ewes, with initial mean body weight of 76.36 ± 10.37 kg (SD), kept in a semi-extensive system and receiving the same diet, were used. Ewes were classified according to parity (primiparous and multiparous) and the litter size born at parturition (1, 2, and 3). On days - 14, - 7, and - 4 of the expected date of parturition and on days 1, 4, 7, and 14 days postpartum, blood sampling, the evaluation of the body condition score (BCS), and the weighing of animals were performed. Blood samples were taken from the lambs 48 h after birth. Colostrum was collected up to 6 h postpartum to determine the brix percentage. Serum concentrations of total plasma proteins (TPP), albumin, aspartate aminotransferase (AST), gamma-glutamyl transferase (GGT), beta-hydroxybutyrate (BHB), non-esterified fatty acids (NEFA), paraoxonase (PON1), calcium (Ca), and magnesium (Mg) were evaluated. In lambs, the blood parameters evaluated were TPP, albumin, GGT, and PON1. Lamb plasma was used to estimate colostrum efficiency, brix percentage immunity, and total plasma protein (PPT). Productive data, pre- and postpartum metabolic variables for ewes, lambs, and those related to colostrum brix were analyzed. The BCS had a reduction (p < 0.0001) in the postpartum period, which is evidenced on days 7 and 14 (2.88 and 2.73, respectively). Serum concentration of BHB presented the highest concentrations in the prepartum period for ewes that gave birth to triplets, from day - 14, and for ewes that gave birth to twins on day - 4 (p = 0.0245). Serum concentration of NEFA demonstrated the highest value on day 14 in primiparous (1.07; p = 0.0402). Calcium had the lowest concentrations on day 1 (8.85 mg/dl, p < 0.0001) and magnesium on day 3 (1.79 mg/dl, p < 0.0001) postpartum. The multiparous with two lambs had higher brix percentages (30.5%), and the lowest value was observed in primiparous ewes that gave birth to a single lamb (27.06%; p = 0.0395). Triplet lambs had the lowest weight (3.73 kg, p = 0.0007), and the best brix percentage in plasma was observed in twins (10.29%, p = 0.0174). Regardless of the parity, the ewes that presented the greatest metabolic challenge were those that gave birth to triplets, and these lambs presented the worst immunity and the lowest live weight. The quality of colostrum was influenced by the parity, and multiparous ewes that gave birth to triplets had the lowest brix percentage in the colostrum.

The limited ability of newborn piglets to produce cytokines may influence lymphocyte development and response to antigen exposure. As a result, colostrum intake is crucial because it contains nutrients that contribute to immune system development in piglets. Our goal was to investigate the effect of sow parity on the transfer of maternal cytokines to nursing piglets. Sixty piglets from nine sows were divided into six groups: piglets from gilts or sows kept with their dams and allowed to suckle normally; piglets from gilts or sows having their dams exchanged and then allowed to suckle normally; piglets from gilts or sows isolated from their dams and bottle-fed a commercial milk replacer formula for pigs. All piglets remained in the diet groups for 24 hours after birth. Concentrations of cytokines in colostrum and serum of gilt/ sows and serum of piglets were then evaluated. The 13 evaluated cytokines had higher concentrations in colostrum and serum of sows than in gilts. Concentrations of GM-CSF, IFNγ, IL-1α, IL-1ß, IL-2, IL-4, IL-6, IL-10, IL-12, IL-18, and TNFα were higher in piglets suckling sows. Piglets that received commercial formula showed higher concentrations of the cytokines IL1-RA and IL-8 than piglets fed colostrum. This outcome can influence piglets' development into adulthood. In short, our findings demonstrated that maternal parity influenced colostrum cytokine composition and its maternal transfer patterns.(AU)
A capacidade limitada dos leitões recém-nascidos de produzir citocinas pode influenciar o desenvolvimento de linfócitos e a resposta à exposição ao antígeno. Portanto, a ingestão de colostro é importante porque contém nutrientes, que contribuem para o desenvolvimento do sistema imunológico do leitão. O objetivo do estudo foi investigar o efeito da paridade da porca na transferência de citocina materna para leitões lactentes. Sessenta leitões de nove porcas foram divididos em seis grupos: leitões de marrãs/porcas mantidas com suas próprias mães e amamentadas normalmente; leitões de marrãs/porcas que foram trocados de mães e amamentados normalmente; leitões de marrãs/porcas que foram isolados das mães e alimentados com mamadeira com substituto do leite para suínos. Os leitões permaneceram nos grupos por 24 horas após o nascimento. Foram avaliadas as concentrações de citocinas no colostro e plasma das marrãs/porcas e no plasma dos leitões. O colostro e o plasma das porcas apresentaram maiores concentrações das 13 citocinas analisadas do que as marrãs. No mesmo sentido, as concentrações de GM-CSF, IFNγ, IL-1α, IL-1ß, IL-2, IL-4, IL-6, IL-10, IL-12, IL-18 e TNFα foram significantemente maiores nos leitões que mamaram o colostro de porcas. Os leitões que receberam fórmula comercial apresentaram, em especial, concentrações das citocinas IL1-RA e IL-8 superiores aos leitões amamentados com colostro. Isso pode influenciar o desenvolvimento até a fase adulta. Portanto, nossos dados demonstraram que a paridade materna influenciou a composição das citocinas do colostro, bem como as características das citocinas na transferência materna.(AU)

Vaccination has been used to prevent the losses associated with Bovine alphaherpesvirus 1 (BoHV-1) infection but passively acquired antibodies may compromise vaccine efficacy. Intranasal immunization (IN) of calves with modified live viral BoHV-1 vaccines has proven to overcome the acquired passive antibodies and confer adequate protection. Herein, we evaluated the safety and immunogenicity of a glycoprotein E-deleted Brazilian BoHV-1 strain (BoHV-1gEΔ) for IN immunization of calves. Ten 1-to-2 months-old calves with virus-neutralizing titers (VN) ranging from 2-64 were immunized IN with viable BoHV-1gEΔ (107.1 TCID50) and four remained as unvaccinated controls (VN titers 8-32). After IN immunization, calves presented a transient (2-6 days) mild nasal secretion and shed the vaccine virus in nasal secretions in low titers (<102.6TCID50/mL) for 4-8 days. Interestingly, the vaccinated calves did not show an increase in VN titers after vaccination. Rather, they presented a gradual reduction in serum VN antibodies in the following weeks - similarly to unvaccinated controls. Upon IN challenge with a virulent heterologous BoHV-1 strain at day 55 post-immunization (107.63TCID50), vaccinated calves shed significantly less virus from day 6 post-challenge onwards (p < 0.07) and for a shorter period of time than the controls (p < 0.0024). Importantly, both the duration and intensity of clinical signs were reduced in vaccinated animals. In addition, vaccinated calves showed an abrupt raise in VN titers post-challenge, indicating adequate immunological priming by vaccination. In summary, immunization of calves harboring passive antibodies with BoHV-1gEΔ by the IN route was able to prime the immunity to afford partial virological and clinical protection upon challenge.
A vacinação tem sido usada para prevenir perdas associadas à infecção pelo alfaherpesvírus bovino 1 (BoHV-1), embora anticorpos adquiridos passivamente possam comprometer a eficácia das vacinas. A imunização intranasal (IN) de bezerros com vacinas de BoHV-1 vivas modificadas pode contornar o obstáculo relacionado à presença de anticorpos adquiridos passivamente, conferindo proteção aos animais vacinados. Nesse contexto, avaliou-se a segurança e imunogenicidade de uma cepa brasileira de BoHV-1 com deleção no gene da glicoproteína E (BoHV-1gEΔ) na imunização IN de bezerros. Dez bezerros, de um a dois meses de idade e com títulos neutralizantes (VN) variando de 2-64, foram inoculados IN com BoHV-1gEΔ (107,1TCID50), e quatro permaneceram como controles não vacinados (títulos de VN 8-32). ABSTRACT: Vaccination has been used to prevent the losses associated with Bovine alphaherpesvirus 1 (BoHV-1) infection but passively acquired antibodies may compromise vaccine efficacy. Intranasal immunization (IN) of calves with modified live viral BoHV-1 vaccines has proven to overcome the acquired passive antibodies and confer adequate protection. INTRODUCTION: South America is one of the regions most affected by the COVID-19 pandemic. Specific and affordable treatments are needed to treat SARS-CoV-2 infection. Evidence regarding the use of convalescent plasma in COVID-19 patients is still limited. We compared the safety and efficacy of COVID-19-convalescent plasma administration as a complement to standard treatment in the early management of patients with moderate SARS-CoV-2 infection. METHODS: We carried out a random double blinded, placebo-controlled trial that compared standard treatment plus convalescent plasma (CP) or plus non-convalescent plasma in the management of COVID-19 patients. The main outcome was survival and secondary endpoints included: length of hospitalisation (LOH), days from treatment to discharge, time to clinical improvement or death within a 28-day period, and adverse reactions to treatment. RESULTS: Administration of CP with antibodies against SARS-CoV-2 did not affect patient survival, RR = 1.003, 95% CI (0.3938, 2.555). These results led to terminate the RCT prematurely. However, early treatment of COVID-19 patients with CP tended to decrease the LOH while the delay in CP treatment was associated with longer hospitalisation. In addition, delay in CP treatment negatively affected the recovery of the respiratory rate. CONCLUSION: Use of CP for the treatment of COVID-19 patients is safe and its early use can decrease the LOH and improve respiratory function. Early administration of antibody-rich CP could contribute to decrease the negative impact of COVID-19 pandemic in patients with impaired immune response.

Background: Limited data are available regarding the differences between immunological, biochemical, and cellular contents of human colostrum following maternal infection during pregnancy with coronavirus 2 disease (COVID-19). Objective: To investigate whether maternal COVID-19 infection may affect immunological, biochemical, and cellular contents of human colostrum. Methods: Using a case-control study design, we collected colostrum from 14 lactating women with a previous diagnosis of COVID-19 during pregnancy and 12 without a clear diagnosis during September 2020 to May 2021. Colostrum samples were analysed for some enzymes and non-enzymatic oxidative stress markers (SOD, CAT, GPx, MDA, GSH, GSSG, H2O2, MPO) and for IL-1ß, IL-6, tumour necrosis factor (TNF)-α, protein induced by interferon gamma (IP)-10, IL-8, IFN-λ1, IL12p70, IFN-α2, IFN-λ2/3, granulocyte macrophage colony stimulating factor (GM-CSF), IFN-ß, IL-10 and IFN-γ, along with IgA and IgG for the SARS-CoV-2 S protein. We perform immunophenotyping to assess the frequency of different cell types in the colostrum. Results: Colostrum from the COVID-19 symptomatic group in pregnancy contained reduced levels of H2O2, IFN-α2, and GM-CSF. This group had higher levels of GSH, and both NK cell subtypes CD3-CD56brightCD16-CD27+IFN-γ+ and CD3-CD56dimCD16+CD27- were also increased. Conclusion: The present results reinforce the protective role of colostrum even in the case of mild SARS-Cov-2 infection, in addition to demonstrating how adaptive the composition of colostrum is after infections. It also supports the recommendation to encourage lactating women to continue breastfeeding after COVID-19 illness.

The aims of this study were: 1) determine total specific IgG and subisotypes in serum and colostrum of pregnant mares vaccinated against Theileria equi and Streptococcus equi subspecies equi; 2) determine total specific IgG and subisotypes in foals born from these mares. In mares, the highest total serum IgG value for T. equi was at 335 days of gestation declining 30 days postpartum, while for S. equi was at 328 days of gestation remaining high up to 30 days postpartum. Transfer of passive immunity against both antigens was observed with specific IgG values in colostrum and foals' serum proportional to mares' values. The most detected specific IgG subisotypes were IgG3/5 and IgG4/7 for both antigens. Foals born from mares immunized with T. equi kept maternal IgG values until 2 months of age, while those born from mares immunized with S. equi kept maternal IgG values until 3-4 months of age. These results suggest that foals should be vaccinated after this period.
Os objetivos deste estudo foram: 1) determinar IgG total específica e seus subisotipos no soro e colostro de éguas vacinadas contra Theileria equi e Streptococcus equi subspécie equi; 2) determinar IgG total específica e seus subisotipos em potros provenientes dessas éguas. Em éguas vacinadas contra T. equi, o maior nível sérico de IgG total específico ocorreu aos 335 dias de gestação, decaindo 30 dias pós parto, enquanto nas vacinadas contra S. equi, ocorreu aos 328 dias de gestação e manteve-se pelo mesmo período. A avaliação de IgG total específica no colostro de éguas vacinadas demonstrou níveis de IgG proporcionais aos títulos do soro materno. Os isotipos de IgG mais detectados foram IgG3/5 e IgG4/7 para ambos antígenos avaliados. Potros provenientes de éguas vacinadas com antígeno de T. equi mantiveram os níveis de IgG específica até dois meses de idade, enquanto potros provenientes de éguas vacinadas com antígeno de S. equi mantiveram por três a quatro meses, sugerindo que esse seja o período ideal para início do esquema vacinal em potros.

Avian coccidiosis is the first to most economically important parasite disease affecting poultry industries worldwide. Current prevention measures are largely based upon prophylactic chemotherapy supplemented by the application of live attenuated or wild-type parasite vaccines. However, the rising appearance of drug resistance, consumer's concern for antibiotics use in poultry production and higher manufacturing cost of live vaccines has driven to adopt new technologies aimed at increasing animal health and production efficiency. Supplementing chickens with egg yolk Eimeria sp.-specific immunoglobulins can be a viable alternative to avoid severe outbreaks of the disease. Twelve-week-old SPF White Leghorn chickens were experimentally infected with a large dose of E. tenella. During the prepatent period, the birds were supplemented by oral gavage with 60 or 120 mg/bird of hyperimmune egg yolk Eimeria species-specific immunoglobulins Y (Supracox®, SC) on a daily basis. The animals were euthanized 7 days post-infection (PI) and their passive immune protection was evaluated. Birds treated with 120 mg/bird of SC showed more viability, increased body weight gain (BWG), a normal hematocrit level (HCT), reduced oocyst output per gram of feces (OPG) or cecal tissue (OPGC), and fewer cecal lesions compared to the untreated infected (UI) control group. Birds supplemented with 60 mg/bird of SC did not show any significant difference on BWG, HCT, OPG, OPGC, and cecal lesion score when compared with the UI group. An ELISA test of the SC showed a weak cross-reactivity of IgY toward two asexual zoite stages of E. tenella. Western blot analysis of the sporozoite with SC showed few antigens barely recognized, while more stained bands were detected in the merozoite (≈82, ≈60, ≈54, ≈40, ≈38, ≈27.5, and ≈13 kDa). Oral immunotherapy using egg yolk polyclonal IgYs against Eimeria sp. represents an effective and natural resource against severe E. tenella infection favoring the gradual withdrawal of the anticoccidial drugs and antibiotics.

Neonatal calf diarrhea (NCD) and mortality cause significant losses to the dairy industry. The preweaning dairy calf mortality risk in Uruguay is high (15.2%); however, causes for these losses are largely unknown. This study aimed to assess whether various pathogens were associated with NCD and death in Uruguayan dairy calves and whether these infections, diarrhea, or deaths were associated with the failure of transfer of passive immunity (FTPI). Contemporary diarrheic (n = 264,) and non-diarrheic (n = 271) 1- to 30-day-old calves from 27 farms were sampled. Feces were analyzed by antigen-capture ELISA for Cryptosporidium spp., rotavirus, bovine coronavirus, and Escherichia coli F5+, RT-PCR for bovine astrovirus (BoAstV), and bacterial cultures for Salmonella enterica. Blood/serum was analyzed by RT-PCR or antigen-capture ELISA for bovine viral diarrhea virus (BVDV). Serum of ≤ 8-day-old calves (n = 95) was assessed by refractometry to determine the concention of serum total proteins (STP) as an indicator of FTPI. Whether the sampled calves died before weaning was recorded. At least one pathogen was detected in 65.4% of the calves, and this percentage was significantly higher in diarrheic (83.7%) versus non-diarrheic (47.6%) calves. Unlike the other pathogens, Cryptosporidium spp. and rotavirus were associated with NCD. Diarrheic calves, calves infected with any of the pathogens, and calves infected with rotavirus had significantly lower concentrations of STP. Diarrheic calves had higher chances of dying before weaning than non-diarrheic calves. Diarrheic calves infected with S. enterica were at increased risk of mortality. Controlling NCD, salmonellosis, cryptosporidiosis, and rotavirus infections, and improving colostrum management practices would help to reduce calf morbi-mortality in dairy farms in Uruguay.

Resumen En el momento actual no existen opciones terapéuticas específicas o vacuna para la enfermedad por coronavirus 2019 (COVID-19), causada por el coronavirus 2 del síndrome respiratorio agudo grave (SARS-CoV-2). La opción con plasma convaleciente resulta atractiva, dados sus precedentes en pandemias previas, su rápida disponibilidad y su bien conocido perfil de bioseguridad. Se propone la eficacia del uso de inmunidad pasiva por medio de la transfusión de plasma convaleciente mediante distintos mecanismos de acción (neutralización, citotoxicidad mediada por anticuerpos, activación del complemento, inmunomodulación). El uso de plasma convaleciente está aprobado en pacientes con datos de gravedad o criterios de complicabilidad. Los resultados disponibles de experiencias internacionales y locales demuestran un adecuado perfil de bioseguridad (< 1% de eventos adversos graves reportados). La evidencia de la eficacia de la intervención es limitada a no más de seis series de casos, destacando la necesidad de una administración temprana, un seguimiento estrecho de variables de desenlace y la comunicación de series más robustas y con una mayor fortaleza metodológica. En nuestro país diversos grupos se encuentran realizando esfuerzos colaborativos y/o metacéntricos, lo que nos ayudará a dilucidar la relevancia de esta intervención en un panorama aún complejo de una epidemia que se encuentra en su cénit sin que se cuente con herramientas específicas para su tratamiento.
Abstract At the present time, there are no specific therapeutic options or vaccine for coronavirus disease 2019 (COVID-19), produced by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The convalescent plasma option is attractive given its precedents in the context of previous pandemics, its rapid availability in areas where the epidemic is active, and its well-known biosecurity profile. There is emerging evidence that proposes the effectiveness of the use of passive immunity through convalescent plasma transfusion, which would exert its effect through different mechanisms of action (neutralization, antibody-mediated cytotoxicity, complement activation, immunomodulation). Globally, the use of convalescent plasma is approved in patients with severe disease criteria or high risk of complication. The available results of international and local experiences demonstrate an adequate biosecurity profile of the use of convalescent plasma with a rate of serious adverse events related to the intervention of less than 1%. So far, the evidence for the efficacy of the intervention is limited to no more than 6 case series, highlighting the need for early administration, close monitoring of outcome variables, and communication of more robust series with greater methodological strength. In our country, several groups are making collaborative and/or metacentric efforts, which will help us to clarify the relevance of this intervention in a still complex panorama of an epidemic that is its highest point without specific tools for its treatment.

Background: Passive immunity acquired by colostrum ingestion is essential to prevent neonatal infections. Failure ofpassive transfer (FPT) of maternal immunity occurs in foals that fail to absorb enough immunoglobulins within 24 h afterbirth. Foals with FPT are at increased risk of infections and death. Serum samples from neonatal foals might be examinedfor FPT using the zinc sulphate turbidity (ZST) test. The aim of this study was to investigate the accuracy of the ZST test,performed at two different times after first suckling (12 and 18 h), to detect FPT in newborn foals. The effect of temperatureon the turbidity intensity resulting from the ZST reaction was also investigated.Materials, Methods & Results: Blood samples were collected from 112 newborn foals at 12 h after the first colostrumintake. In 36 foals, additional serum samples were collected at 18 h after first colostrum intake. The serum samples weretested with the ZST test and, later, in the laboratory setting, the ZST test was repeated. The IgG levels were measured bysingle radial immunodiffusion (SRID), which was used as the reference method. The standard solution used for the interpretation of results had a turbidity corresponding to approximately 800 mg/dL of immunoglobulins (IgG). The mean IgG concentration measured at 12 and 18 h after the first colostrum intake was analyzed using the t-test for paired samples.Values of absorbance of ZST test under different temperatures were analyzed using a one-way analysis of variance, andmeans were compared using the Tukey test. The relationship between the temperature of the solution and absorbance wasdetermined using the Pearsons correlation coefficient. Based on SRID results, 12 foals (10.7%) had serum IgG concentration < 400 mg and 26 foals (23.2%) had IgG levels between 400 and 800 mg/dL. Serum blood samples from 50 Murrah buffalo calves were examined in this study. The animals were allocated into three groups according to the number of parturitions of their mothers: G1 (n= 15) calves from primiparous buffaloes, G2 (n= 19) calves from buffaloes with two to four parturitions, and G3 (n= 16) calves from buffaloes with five or more parturitions. Blood samples were taken at birth, before colostrum ingestion, at 24h, 48h, and 72h after birth, and at 7, 14, 21, and 30 days after birth for determination of levels of gammaglutamyl transferase (GGT), alkaline phosphatase (ALP), aspartate aminotransferase (AST), creatine kinase, total protein, albumin, globulins (including immunoglobulin G), iron, total calcium, ionized calcium, phosphorus, sodium, and potassium. The age of the calves was found to influence all of the biochemical parameters, with the exception of ionized calcium and potassium in the calves in groups G1 and G3. The calving order was found to influence AST, GGT, total protein, albumin, and globulins, including IgG. The high serum ALP activity in the first two days after birth indicates that measurement of the levels of this enzyme may be used as an indirect method of assessing passive immunity transfer.(AU)
Amostras de sangue de 50 bezerros de búfalo Murrah foram examinados nesse estudo. Os animais foram distribuídos em três grupos de acordo com a paridade de suas genitoras: G1 (n=15) bezerros de búfalas primíparas, G2 (n=19) bezerros de búfalas com 2 a 4 gestações, e G3 (n=16) bezerros de búfalas com cinco ou mais gestações. Amostras de sangue foram colhidas ao nascimento, antes da ingestão de colostro e 24h, 48h, e 72h após o nascimento e 7, 14, 21 e 30 dias após nascimento para determinar níveis de gammaglutamil transferase (GGT), fosfatase alcalina (ALP), aspartato aminotrasferase (AST), creatina quinase, proteínas totais, albumina, globulina (inclusive imunoglobulina G), ferro, cálcio total, cálcio ionizado, fósforo, sódio e potássio. A idade dos bezerros influenciou todos os parâmetros bioquímicos, exceto cálcio ionizado e potássio nos bezerros dos grupos G1 e G3. A ordem de nascimento influenciou AST, GGT, proteínas totais, albumina e globulinas, inclusive IgG. Intensa atividade ALP no soro nos primeiros dois dias após nascimento indica que medidas dos níveis dessa enzima podem ser utilizados como método indireto de avaliar transferência passiva de imunidade.(AU)
