ABSTRACT

This study aimed to investigate seven outbreaks of A. marginale infection in two regions of Brazil, affecting taurine, zebu, and crossbred cattle. We assessed the possible causes, treatment measures, and genetic diversity of A. marginale. These outbreaks occurred in two states (Goiás: outbreaks 1-7; Mato Grosso do Sul: outbreak 3), breeds (Holstein, Nellore, and crossbreed), age groups (beef cattle: 18-25 days old and 7-8 months; dairy cattle: 18-25 days old, 13-14 months, and cow after the first birth) and rearing systems (feedlot, pasture, pen in a wood shaving bedding system and compost bedded-pack barns). Metaphylactic or prophylactic treatments varied according to outbreak (imidocarb dipropionate: outbreaks 1-4 and 6; enrofloxacin: outbreaks 5 and 7; diminazene diaceturate: outbreak 5). In outbreaks 6 and 7, the packed cell volume was monitored. In all outbreaks, the practice of needle/syringe sharing was discontinued. For outbreaks 1-3, clinical signs and mortality (range, 4.8-13.3%) occurred 36-45 days after entry into the feedlot. In outbreak 4, A. marginale was diagnosed in 66.2% of the calves (bacteremia, 0-4.5%), with a mortality of 8.6%. Among nursing calves aged 60 days during outbreak 5, 53.8% were infected with A. marginale, with average bacteremia of 2.7% (range, 0-21.3%), and a mortality of 13.8%. In dairy heifers aged 14 months, raised in paddocks lacking vegetation cover and infested with R. microplus, then transitioned to a rotational grazing system also infested with R. microplus, the A. marginale bacteremia ranged from 3.2 to 6.7%, with a mortality of 20%. Before monitoring during outbreak 7, the mortality was 17.9%, but no further deaths were observed after monitoring initiation. In conclusion, possible causes triggering the outbreaks included primary tick infestation, needle/syringe sharing, and stress factors which may have affected the immunological statues of animals in the feedlots. Control measures performed in all outbreaks were effective. The partial msp4 gene sequences of A. marginale generated herein belonged to two haplotypes, but further research would be needed to investigate if this finding has any clinical significance.

ABSTRACT

This retrospective cohort study aimed to identify risk factors for the failure of transfer of passive immunity (FPI) in preweaned dairy calves, explore its associations with morbidity, mortality, genetics, and determine a standardized cut-off point for FPI. Analyzing data from 6011 calves, factors such as sire predicted transmitted ability for milk (PTA), birth season, retained placenta (RP), total serum protein concentration (TSP), morbidity (neonatal calf diarrhea - NCD, bovine respiratory disease - BRD, tick-borne disease - TBD), mortality, and average daily weight gain (ADG) were considered. The calves were categorized into predominantly Gyr (PG) and predominantly Holstein (PH) genetic composition groups. Multivariate mixed logistic regression revealed optimal TSP cut-off points for predicting morbidity (7.6 g/dL) and mortality (6.9 g/dL). PH calves exhibited 1.35 times higher odds of FPI and 1.48 times greater odds of disease. Calves from multiparous cows and those born to dams with RP had increased FPI odds. Disease prevalence was 53%, with 41% NCD, 18% BRD, and 10% TBD. Season, parity, PTA, and birth weight were associated with disease odds, though FPI was not a reliable predictor. The mortality percentage was 6%, with PH calves and those with a positive PTA having higher odds. The ADG was 0.64 kg, and FPI-affected calves gained less weight. The study compared and identified various risk factors that potentially impact calf immunity. However, the use of a standardized cut-off point for FPI assessment was not effective in predicting morbidity and mortality at this specific farm.

ABSTRACT

The aim of this study was to evaluate a host-tick distancing control system using Rhipicephalus microplus in cattle. Its impact on the profiles of the pathogens Anaplasma marginale and Babesia bigemina was also assessed using enzyme-linked immunosorbent and real-time polymerase chain reaction. Experiments were conducted in Mato Grosso do Sul, Brazil. A total of 37 Senepol animals with natural tick infestation were used in this study. The ticks on the animals were counted, and pasture rotation was performed at 28-day intervals. This approach was repeated until the end of the experiment to maintain a low number of ticks, resulting in a final average of 6.3 ticks/host. Cattle maintained an immune response to the cattle tick fever (CTF) agents. DNA copies of A. marginale (X̄ 1.1) and B. bigemina (X̄ 0.05) did not vary significantly with the decrease in tick count throughout the study period. Based on these results, we conclude that a distancing period of 84 days between ticks and cattle in conditions of the Cerrado biome can control ticks in animals and maintain enzootic stability. Thus, it is possible to create productive breeds with lower tick control costs.

ABSTRACT

Domestic cats are susceptible to infection with at least 11 species of Babesia. In Hong Kong, where dogs are commonly infected with B. gibsoni, a single infection in a cat by a novel species, B. hongkongensis, was reported previously. The aim of this study was to investigate the frequency of Babesia spp. detection in cats in Hong Kong. Residual blood-derived DNA from healthy free-roaming community cats (n = 239), and privately-owned cats with and without anaemia undergoing diagnostic investigations (n = 125) was tested for Babesia spp. DNA using a pan-Babesia PCR targeting mitochondrial Cytochrome B, and a B. hongkongensis specific PCR targeting 18S rRNA. Positive samples were confirmed by sequencing and comparative sequence analysis against the GenBank nucleotide database. Babesia hongkongensis was detected in 4/239 (1.7 %) community cats, and 0/125 (0.0 %) privately-owned cats. Babesia gibsoni was detected in 0/239 community cats and 1/125 (0.8 %) privately-owned cats. Cats infected with B. hongkongensis were clinically healthy at the time of sampling. The B. gibsoni-infected cat was anaemic and thrombocytopenic. Cats in Hong Kong can be infected with B. hongkongensis and B. gibsoni, albeit at low frequency. The tick vector for B. hongkongensis is yet to be identified.

ABSTRACT

Colorado tick fever virus is an understudied tick-borne virus of medical importance that is primarily transmitted in the western United States and southwestern Canada. The virus is the type species of the genus Coltivirus (Spinareoviridae) and consists of 12 segments that remain largely uncharacterized. Patterns of viral distribution are driven by the presence of the primary vector, the Rocky Mountain wood tick, Dermacentor andersoni. Infection prevalence in D. andersoni can range from 3% to 58% across the geographic distribution of the tick. Infection in humans can be severe and often presents with fever relapses but is rarely fatal. Here, we review the literature from primary characterizations in the early 20th century to current virus/vector research being conducted and identify vacancies in current research.

ABSTRACT

Tick-borne haemoparasite Babesia gibsoni has been detected rarely in cats, in surveys of apparently healthy animals. In stored blood from a 6-year-old male-neutered domestic shorthair cat in Hong Kong, B. gibsoni DNA was detected retrospectively using PCR for Babesia spp. 18S rRNA and mitochondrial cytochrome B genes, followed by sequencing and basic local alignment search tool (BLAST) analysis. The cat presented with severe haemolytic anaemia and thrombocytopenia. The cat responded to supportive care and glucocorticoids and was clinically normal despite persistent subclinical thrombocytopenia until six months after presentation, when it succumbed to a fatal haemorrhagic episode. Necropsy revealed severe intestinal and pulmonary haemorrhage and hypocellular bone marrow with megakaryocytosis but no other causes of immune-mediated thrombocytopenia (IMTP) or immune-mediated haemolytic anaemia (IMHA). Blood stored on days 158 and 180 tested PCR negative for Babesia spp. This report demonstrates that geographic range of B. gibsoni detection in cats includes Hong Kong. The exclusion of other causes suggests that B. gibsoni might have potentially played a role in triggering immune-mediated disease in this case.

In 2020, Montana, USA, reported a large increase in Colorado tick fever (CTF) cases. To investigate potential causes of the increase, we conducted a case-control study of Montana residents who tested positive or negative for CTF during 2020, assessed healthcare providers' CTF awareness and testing practices, and reviewed CTF testing methods. Case-patients reported more time recreating outdoors on weekends, and all reported finding a tick on themselves before illness. No consistent changes were identified in provider practices. Previously, only CTF serologic testing was used in Montana. In 2020, because of SARS-CoV-2 testing needs, the state laboratory sent specimens for CTF testing to the Centers for Disease Control and Prevention, where more sensitive molecular methods are used. This change in testing probably increased the number of CTF cases detected. Molecular testing is optimal for CTF diagnosis during acute illness. Tick bite prevention measures should continue to be advised for persons doing outdoor activities.

ABSTRACT

This study examined 400 tick-infested cattle from the following four governorates in Egypt: Faiyum, Beni Suef, Giza, and Minya. These cattle were examined for blood parasites between January 2021 and April 2022. The infected cattle were classified into four groups based on tick infestations and clinical signs. Blood was drawn for assessing oxidative stress markers as well as for parasitological examination and molecular analysis of the 18S rRNA gene of Babesia bigemina (B. bigemina). We performed a comparison of the levels of tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6) between B. bigemina-infected blood samples and non-infected blood samples used as negative controls. Babesia spp. infection increases hemolysis, which in turn increases oxidative stress marker levels and cell-mediated immune response.

In the past few decades, reported human cases of Colorado tick fever in the western United States have decreased dramatically. The goal of this study was to conduct surveillance for Colorado tick fever virus (CTFV) in Dermacentor ticks in recreational sites in Colorado, Wyoming, and California to determine whether the virus is still present in Dermacentor ticks from these states. Surveillance focused on regions where surveys had been conducted in the 1950s, 1960s, and 1970s. Adult Rocky Mountain wood ticks (Dermacentor andersoni), Pacific Coast ticks (Dermacentor occidentalis), and winter ticks (Dermacentor albipictus) were tested by PCR. A subset of PCR-positive D. andersoni ticks (n = 7) were cultured in Vero cells. CTFV-positive Rocky Mountain wood ticks were found in all states: Colorado (58% prevalence), Wyoming (21%), and California (4%). Although no winter ticks tested positive, Pacific Coast ticks tested positive in one county (Siskiyou County, 15% prevalence) and were positive only in a location that also maintained Rocky Mountain wood ticks and golden mantled ground squirrels, a known CTFV host. In summary, CTFV is prevalent in D. andersoni and D. occidentalis in regions where they are sympatric in California and in D. andersoni in Colorado and Wyoming. Although the number of human CTFV cases has declined dramatically, this decrease in reported disease does not appear to be due to the disappearance or even the decline in prevalence of this virus in ticks in historically endemic regions of the country.

ABSTRACT

Bovine parasitic sadness, comprised of the diseases babesiosis and anaplasmosis, has a large impact on cattle farming in several countries, as it compromises animal productivity. Using systematic review (SR)-meta-analysis (MA) methodology, our objective was to summarize and to investigate study characteristics associated with prevalence of Babesia spp. and Anaplasma marginale infection in Latin American cattle herds. Five electronic databases were used. The inclusion criteria were studies that assessed the prevalence of Babesia spp. and A. marginale in cattle in Brazil, Argentina, Uruguay, Paraguay, Colombia, and Mexico. The reported prevalences were converted to logistic scale and summarized using random effects MA. The heterogeneity was assessed, separately, for papers from Brazil and from the remaining countries. A total of 28 papers were included in this MA with a high heterogeneity (I2 > 95%). The prevalences of A. marginale, Babesia spp., and their co-infections, in Latin America, were 48.9% (95% CI: 30.3-67.8%), 39.8% (95% CI: 24.6-57.2%), and 26.1% (95% CI: 9.1-55.8%), respectively. The prevalence of A. marginale was higher due to the high prevalence of this agent in Mexico (67.1%). In Brazil, the prevalence was 36.6% for A. marginale, 62.6% for Babesia spp., and 8.2% for their co-infections. The North region of Brazil was reported with the highest prevalence for A. marginale (71.9%), while for Babesia spp., the prevalence was considerably divergent between regions, with the highest values also observed in the North region (97.4%), and the lowest in the South region (9.5%). For studies of Babesia spp. in cattle in Latin America, the heterogeneity was mainly explained by the diagnostic method (98.0%), and country (54.8%). When looking at Brazil alone, a similar pattern was observed, but with lower values (i.e., diagnostic method, 31.5%; region, 25.3%; and climate, 12,4%). The evaluated regions presented different levels of prevalence infections, with most of them being classified as high degree of enzootic instability, which can predispose to the occurrence of outbreaks.

ABSTRACT

The type species of the genus Coltivirus, Colorado tick fever virus (CTFV), was discovered in 1943 and is the most common tick-borne viral infection in the Western US. Despite its long history, very little is known about the molecular diversity of viruses classified within the species Colorado tick fever coltivirus. Previous studies have suggested genetic variants and potential serotypes of CTFV, but limited genetic sequence information is available for CTFV strains. To address this knowledge gap, we report herein the full-length genomes of five strains of CTFV, including Salmon River virus and California hare coltivirus (CTFV-Ca). The sequence from the full-length genome of Salmon River virus identified a high genetic identity to the CTFV prototype strain with >90% amino acid identity in all the segments except segment four, suggesting Salmon River virus is a strain of the species Colorado tick fever coltivirus. Additionally, analysis suggests that segment four has been associated with reassortment in at least one strain. The CTFV-Ca full-length genomic sequence was highly variable from the prototype CTFV in all the segments. The genome of CTFV-Ca was most similar to the Eyach virus, including similar segments six and seven. These data suggest that CTFV-Ca is not a strain of CTFV but a unique species. Additional sequence information of CTFV strains will improve the molecular surveillance tools and provide additional taxonomic resolution to this understudied virus.

ABSTRACT

Colorado tick fever virus (CTFV) belongs to the genus Coltivirus of the Reoviridae family, and it is the causative agent of Colorado tick fever. Symptoms of the infection are characterized by sudden biphasic fever, headache, and petechial rash, while severe forms of the disease can include meningoencephalitis, hemorrhagic fever, and death in children. However, the mechanisms underlying CTFV induced pathology and severe complications remain unknown. As CTFV is spread by tick bites and disseminates systemically via hematogenous routes, we performed in vitro analysis examining the interactions between endothelial cells (ECs) and CTFV. Our findings indicate that dermal microvascular ECs, HMEC-1, are susceptible and permissive to CTFV infection. To investigate the role of CTFV infection on endothelial barrier function, we assessed transendothelial electrical resistance (TEER) by xCELLigence and observed a dose-dependent decrease in cell index, indicating increased vascular permeability starting at approximately hour 18 (MOI=1) and hour 26 (MOI=0.1). Since CTFV induced cytopathic effect and increased vascular permeability in HMEC-1 cells, we hypothesized that CTFV causes apoptotic cell death. Our results showed that HMEC-1 cells infected with CTFV at 48 h caused a significant increase in Annexin V staining with reduced viability compared to uninfected cells suggesting CTFV induces apoptotic cell death in human ECs. Electron microscopy also was consistent with apoptotic features, including chromatin condensation and cell blebbing. Furthermore, CTFV induced caspase-3/7 activation at 24 and 48 h post-infection (hpi). The inhibition of caspase activity using Z-VAD-FMK reduced CTFV induced cell death and significantly reduced viral titer. These results indicated that CTFV can infect ECs, exerting direct adverse effects, leading to vascular permeability and cell death. Overall, our data suggest that caspase-mediated apoptosis is a critical mechanism by which CTFV induces disease in the host and enhances viral replication. Future studies will examine the viral and cellular determinants involved in CTFV induced apoptosis in human ECs.

Abstract Anaplasma marginale is a vector-borne pathogen that causes a disease known as anaplasmosis. No sequenced genomes of Brazilian strains are yet available. The aim of this work was to compare whole genomes of Brazilian strains of A. marginale (Palmeira and Jaboticabal) with genomes of strains from other regions (USA and Australia strains). Genome sequencing of Brazilian strains was performed by means of next-generation sequencing. Reads were mapped using the genome of the Florida strain of A. marginale as a reference sequence. Single nucleotide polymorphisms (SNPs) and insertions/deletions (INDELs) were identified. The data showed that two Brazilian strains grouped together in one particular clade, which grouped in a larger American group together with North American strains. Moreover, some important differences in surface proteins between the two Brazilian isolates can be discerned. These results shed light on the evolutionary history of A. marginale and provide the first genome information on South American isolates. Assessing the genome sequences of strains from different regions is essential for increasing knowledge of the pan-genome of this bacteria.


Resumo Anaplasma marginale é um patógeno transmitido por vetores que causam uma doença conhecida como anaplasmose. Até a presente data, não há genomas sequenciados de cepas brasileiras. O objetivo deste estudo foi comparar o genoma completo das cepas brasileiras de A. marginale (Palmeira e Jaboticabal) com os genomas de cepas de outras regiões (cepas dos EUA e Austrália). As sequências dos genomas das cepas brasileiras foram obtidas mediante sequenciamento de nova geração. As "reads" foram mapeadas usando-se como referência o genoma de A. marginale da cepa Florida. Foram identificados polimorfismos de nucleotídeo único (SNPs) e analisadas inserções/deleções (INDELs). As duas linhagens brasileiras se agruparam em um clado particular que, por sua vez, agrupou-se em um grupo maior junto com as linhagens norte-americanas. Além disso, foram identificadas diferenças significativas nas proteínas de superfície entre os dois isolados brasileiros. Esses resultados lançam luz sobre a história evolutiva de A. marginale e fornecem as primeiras informações de genomas de isolados sul-americanos. Avaliar as sequências de genomas de cepas de diferentes regiões é essencial para aumentar o conhecimento do pan-genoma dessa bactéria.

ABSTRACT

While investigating patients with gastrointestinal (GI) and/or hepatic symptoms, tickborne diseases are only rarely considered to be the cause. However, the Czech Republic is an endemic region for several of tickborne diseases and, therefore, they should be a part of differential diagnosis of GI symptoms of unknown origin. This article describes GI and hepatic symptoms of several tickborne diseases - Lyme disease, ehrlichiosis, Rocky mountain spotted fever, tularemia, Colorado tick fever, tickborne relapsing fever, Q fever and babesiosis. GI and hepatic symptoms are quite common in Lyme disease patients. The prognosis is generally favourable with antibiotics treatment, however, serious courses have been described. Lyme disease should be a part of differential diagnosis of liver tests elevation and GI symptoms in patients from endemic regions regardless erythema migrans presence. Ehrlichiosis should be a part of differential diagnosis of acute febrile illness with GI symptoms especially in the presence of leukopenia/thrombocytopenia and/or liver tests elevation. Tularemia should be considered as a rare etiology of cholestatic hepatopathy and a history of a tick bite. In general, the importance of careful patient interviewing, including the history of a tick bite, can be highlighted also as a part of investigation of patients with seemingly unrelated GI and/or hepatic symptoms.

ABSTRACT

The causes of death of cattle kept in pre-export feedlots (PEFs) and in feedlot for finishing for slaughter are described. Two studies were conducted: a retrospective study of mortality cases in feedlots from 2000 to 2017 registered at the "Laboratório Regional de Diagnóstico" of the "Faculdade de Veterinária" of the "Universidade Federal de Pelotas"; and a prospective study from January 2018 to August 2019, following up 22 feedlots for finishing and six PEFs for the export of live cattle. From January 2000 to August 2019 samples of 150 cases of diseases that affected feedlot cattle were received from 22 feedlots for finishing and 115 of the six PEFs followed. Mortality considering all diagnosed diseases was significantly higher in feedlots for finishing (p<0.05), than in PEFs for the export of live cattle, of 1% and 0.12%, respectively. Diseases of the digestive system were the most important causes, of death in feedlots regardless of its purpose. Acidosis presented the highest mortality rates both in feedlot for finishing (3.33%) as in PEFs for export (0.95%). In all cases the disease occurred due to failure in the adaptation of animals to the ingestion of concentrated foods. Bovine tick fever and pneumonia presented mortality rates of 0.13% and 0.09%, respectively in PEFs. In the feedlot for finishing seneciosis was the second cause of death due to cattle coming from areas with high infestation by the plant. In the present study, it was possible to identify the main diseases that occur in cattle feedlots for finishing or for the export of live animals in the southern region of Rio Grande do Sul. These diseases are known in other systems of cattle breeding and can be prevented or controlled through management, chemoprophylaxis or vaccination, minimizing losses due to mortality.(AU)


Descrevem-se as causas de morte de bovinos mantidos confinados em estabelecimentos pré-embarque (EPEs) para exportação de animais vivos e em estabelecimentos de terminação para abate. Foram realizados dois estudos: um retrospectivo dos casos de mortalidade em confinamentos de 2000 a 2017 registrados no Laboratório Regional de Diagnóstico da Faculdade de Veterinária da Universidade Federal de Pelotas; e um estudo prospectivo de janeiro de 2018 a agosto de 2019, acompanhando-se 22 confinamentos de terminação e seis EPEs para exportação de bovinos vivos. No total, de janeiro de 2000 a agosto de 2019 foram recebidos no LRD-UFPel amostras de 150 casos de enfermidades que afetaram bovinos confinados, 35 provenientes de 22 confinamentos de terminação e 115 dos seis EPEs acompanhados. A mortalidade considerando-se todas as enfermidades diagnosticadas foi significativamente maior nos confinamentos para terminação (p<0,05), do que nos EPEs para exportação de bovinos vivos, de 1% e 0,12%, respectivamente. As doenças do sistema digestivo foram as causas de morte mais importante nos confinamentos, independente da finalidade. Acidose apresentou as maiores taxas de mortalidade tanto nos confinamentos para terminação (3,33%) como nos EPEs para exportação (0,95%). Em todos os casos a doença ocorreu devido a falha na adaptação dos animais à ingestão de alimentos concentrados. Tristeza parasitária bovina e pneumonias apresentaram taxas de mortalidade de 0,13% e 0,09, respectivamente em EPEs de exportação. Nos confinamentos para terminação a seneciose foi a segunda causa de morte devido aos bovinos serem provenientes de áreas com alta infestação pela planta. Este estudo permitiu identificar as principais enfermidades diagnosticadas em confinamentos para terminação de bovinos ou para exportação de bovinos vivos na região Sul do Rio Grande do Sul. Observou-se que são doenças que ocorrem em outros sistemas de criação de bovinos e que podem ser prevenidas ou controladas por meio de manejo, quimioprofilaxia ou vacinação, minimizando prejuízos por mortalidade de animais.(AU)

ABSTRACT

OBJECTIVES: We evaluated trends in non-Lyme disease tick-borne disease (NLTBI) testing at a national reference laboratory. METHODS: Testing data performed at Quest Diagnostics during 2010 to 2016 were analyzed nationally and at the state level. RESULTS: Testing and positivity for most NLTBIs increased dramatically from 2010 through 2016 based on testing from a large reference laboratory. The number of positive cases, though not as stringent as criteria for public health reporting, generally exceeds that reported by the Centers for Disease Control and Prevention. The frequency of NLTBI in the US is seasonal but testing activity and positive test results are observed throughout all months of the year. Positive results for NLTBI testing mostly originated from a limited number of states, indicating the geographic concentration and distribution of NLTBIs reported in this study. CONCLUSIONS: This report provides an important complementary source of data to best understand trends in and spread of NLTBI.

ABSTRACT

Abstract Cattle tick fever (CTF) causes significant economic losses in the livestock sector. The pathogenic action of the hemoparasites is associated with anemia, weight loss, abortion and reduced productivity, which result with animal death. Programs to prevent CTF involve several procedures, including immunization, chemoprophylaxis and use of ectoparasiticides, together with the vector control in the environment. The objective of this study was to report an acute outbreak of CTF in a group of 157 Hereford cattle from a farm without presence of the vector, that were moved to a farm in the same state with a high tick infestation (Rhipicephalus microplus). On the day before the transportation, the animals received a chemoprophylaxis with imidocarb dipropionate (3 mg/kg, SC), which was repeated 21 days after the first application. After 42 days, some animals showed signs compatible with CTF, which was confirmed through clinical examination, necropsy, histopathological and hemoparasitological analyses. The morbidity rate was 37.6% and the mortality rate was 24.8%. Calves that were recently weaned were the group most affected with the tick fever, morbidity (100% and mortality (73%). Chemoprophylaxis in association with use of ectoparasiticides was not sufficient to control the outbreak of the disease.


Resumo A "tristeza parasitária bovina" (TPB) gera importantes perdas econômicas na bovinocultura mundial. A ação patogênica dos hemoparasitas resulta em anemia, perda de peso, abortos e diminuição da produtividade, culminando, muitas vezes, em óbito dos animais. Um programa de prevenção para TPB necessita de medidas integradas, como a imunização, quimioprofilaxia e utilização de ectoparasiticidas, em conjunto com ações que visem ao controle ambiental dos vetores. Este artigo tem em vista o relato de um surto de TPB em uma fazenda de produção de bovinos de corte e com alta infestação do carrapato (Rhipicephalus microplus). A fazenda adquiriu 157 animais puros de origem, da raça Hereford, proveniente de uma fazenda sem presença do vetor. No dia anterior ao transporte, os animais receberam quimioprofilaxia com dipropionato de imidocarb (3mg/Kg/SC), repetindo-se 21 dias após a primeira aplicação. Aos 42 dias, alguns bezerros manifestaram sinais clínicos compatíveis com TPB, sendo confirmado pelo exame clínico, necropsia, análises histopatológicas e hemoparasitológicas. A morbidade foi de 37,6% (59/157), e a letalidade de 24,8% (39/157). A categoria de bezerros recém desmamados foi a mais acometida, com morbidade de 100% (52/52) e letalidade de 73% (38/52). A quimioprofilaxia associada à utilização de ectoparasiticidas foram insuficientes para evitar a ocorrência do surto da enfermidade.

ABSTRACT

Pacific Coast tick fever is a febrile illness associated with the bite of Dermacentor occidentalis and results from an infection due to the intracellular pathogen Rickettsia 364D (also known by the proposed name "Rickettsia philipii"). Current molecular methods for the detection of this pathogen rely on the amplification of a conserved spotted fever group rickettsial gene (ompA) followed by DNA sequencing of the amplicon to identify the species. This work describes the development of a Rickettsia 364D-specific TaqMan assay to simplify and accelerate the detection and identification processes. The assay demonstrated a sensitivity of 1 genomic copy per 4-µl sample and is highly specific for Rickettsia 364D. The utility of this assay for ecological and diagnostic samples was evaluated using banked specimens collected in a single-blind manner and yielded a clinical sensitivity and specificity of 100%. In conclusion, we describe the development and evaluation of a novel TaqMan real-time PCR assay for the detection and identification of Rickettsia 364D suitable for ecological and diagnostic applications.

Rhipicephalus microplus causes high economic losses for Uruguay; the country is divided into a R. microplus-free area and an endemic area. The aim of this research was to evaluate the R. microplus and tick fever situation in Uruguay and the feasibility of implementing R. microplus control or elimination programs to reduce the number of infested farms. A representative survey was performed to determine the number of infested farms, the number of farms with outbreaks of tick fever and the control and preventive measures being used. Control or elimination programs using generational treatment were implemented on 30 farms. Previously, a situation diagnosis was performed by evaluating the background of each farm, identifying acaricide resistance, serologically evaluating the enzootic stability of tick fever, and analyzing the risk of R. microplus introduction to farms. According to the survey, R. microplus was present on 9544 (36%) farms, and 3436 (36%) of these farms experienced outbreaks of tick fever. Only 323 (9.4%) farmers used the tick fever vaccine. Of the 30 farms for which control or elimination programs were carried out, 18 reduced the number of treatments applied per year and six achieved the elimination of the cattle tick. It is concluded that by administering elimination or control programs, it is possible to reduce the number of infested farms and the number of treatments to maintain control. New surveys should be performed to measure the degree of technology adoption by farmers and to analyze the advances in tick control or elimination programs on farms.

ABSTRACT

The global impact of bovine babesiosis caused by the tick-borne apicomplexan parasites Babesia bovis, Babesia bigemina and Babesia divergens is vastly underappreciated. These parasites invade and multiply asexually in bovine red blood cells (RBCs), undergo sexual reproduction in their tick vectors (Rhipicephalus spp. for B. bovis and B. bigemina, and Ixodes ricinus for B. divergens) and have a trans-ovarial mode of transmission. Babesia parasites can cause acute and persistent infections to adult naïve cattle that can occur without evident clinical signs, but infections caused by B. bovis are associated with more severe disease and increased mortality, and are considered to be the most virulent agent of bovine babesiosis. In addition, babesiosis caused by B. divergens has an important zoonotic potential. The disease caused by B. bovis and B. bigemina can be controlled, at least in part, using therapeutic agents or vaccines comprising live-attenuated parasites, but these methods are limited in terms of their safety, ease of deployability and long-term efficacy, and improved control measures are urgently needed. In addition, expansion of tick habitats due to climate change and other rapidly changing environmental factors complicate efficient control of these parasites. While the ability to cause persistent infections facilitates transmission and persistence of the parasite in endemic regions, it also highlights their capacity to evade the host immune responses. Currently, the mechanisms of immune responses used by infected bovines to survive acute and chronic infections remain poorly understood, warranting further research. Similarly, molecular details on the processes leading to sexual reproduction and the development of tick-stage parasites are lacking, and such tick-specific molecules can be targets for control using alternative transmission blocking vaccines. In this review, we identify and examine key phases in the life-cycle of Babesia parasites, including dependence on a tick vector for transmission, sexual reproduction of the parasite in the midgut of the tick, parasite-dependent invasion and egression of bovine RBCs, the role of the spleen in the clearance of infected RBCs (IRBCs), and age-related disease resistance in cattle, as opportunities for developing improved control measures. The availability of integrated novel research approaches including "omics" (such as genomics, transcriptomics, and proteomics), gene modification, cytoadhesion assays, RBC invasion assays and methods for in vitro induction of sexual-stage parasites will accelerate our understanding of parasite vulnerabilities. Further, producing new knowledge on these vulnerabilities, as well as taking full advantage of existing knowledge, by filling important research gaps should result in the development of next-generation vaccines to control acute disease and parasite transmission. Creative and effective use of current and future technical and computational resources are needed, in the face of the numerous challenges imposed by these highly evolved parasites, for improving the control of this disease. Overall, bovine babesiosis is recognised as a global disease that imposes a serious burden on livestock production and human livelihood, but it largely remains a poorly controlled disease in many areas of the world. Recently, important progress has been made in our understanding of the basic biology and host-parasite interactions of Babesia parasites, yet a good deal of basic and translational research is still needed to achieve effective control of this important disease and to improve animal and human health.
