ABSTRACT

Chelonoidis carbonarius is host to a great diversity of endoparasites. This research aimed to identify and taxonomically classify the parasitic forms of endoparasites found in the feces of C. carbonarius using coproparasitological methods. In total, 38 fecal samples were collected from animals captured in the municipalities of Cuiabá (n = 23) and Jataí (n = 15) in the Brazilian Ceraado. Fecal samples were submitted to at least one of the following techniques: Willis-Mollay flotation technique, zinc sulfate centrifugal flotation technique, and spontaneous sedimentation of Hoffman, and Pons, and Janer. Overall, 86.85% (33/38) of animals were positive in at least one of the diagnostic techniques. Of the 38 samples used, 50% of the animals were captive and 50% of the animals were free-living. Cysts, oocysts, and parasitic eggs were identified, as follows: Chapiniella spp. (65.78%; n = 24), Sauricola spp. (31.57%; n = 12), Ascaroidea (15.78%; n = 6), Oxyuridae (2.63%; n = 1), Trematoda (2.63%; n = 1), Acanthocephala (2.63%; n = 1), Balantidium spp. (39.47%; n = 15), Eimeria spp. (21.05%; n = 8), and Nyctotherus spp. (2.63%; n = 1). The study showed that both captive and free-living C. carbonarius are often parasitized by different endoparasite species, with nematode eggs being the most common.

Abstract The sexing of species is an important tool for population management. In tortoises and turtles, sexing is usually done according to secondary characteristics, such as plastron morphology or phallus eversion techniques. In this paper, we present the phallus eversion method and compare its efficiency to other existing techniques. This new method aims at shortening the management time and reducing the physical aftereffects on the animals, fostering the clinical and reproductive management of Geoffroys side-necked turtle (Phrynops geoffroanus) and related species. A sample of fifty individuals was taken in the central region of Piauí state, Northeastern Brazil. The proposed method consisted of moving the head from its resting position. The methods achieving a success rate higher than 80% were compared to each other as for the stimulation time required for phallus eversion. The method as here proposed achieved 100% efficiency in the sex determination of specimens with a linear carapace length between 10 and 34 cm. Also, when compared to the second most efficient method, it substantially reduced the time needed for managing the specimens (91%), avoiding possible sequelae in the individuals handled.


Resumo A sexagem das espécies é uma ferramenta importante para o manejo populacional. Em tartarugas, cágados e jabutis, a sexagem geralmente é feita de acordo com características secundárias, como morfologia do plastrão ou técnicas de eversão do falo. Neste artigo, apresentamos o método de eversão do falo e comparamos sua eficiência com outras técnicas existentes. Este novo método visa encurtar o tempo de manejo e reduzir os efeitos colaterais físicos nos animais, promovendo o manejo clínico e reprodutivo do cágado-de-barbicha (Phrynops geoffroanus) e espécies relacionadas. Uma amostra de cinquenta indivíduos foi coletada na região central do estado do Piauí, Nordeste do Brasil. O método proposto consiste em remover a cabeça de sua posição de repouso. Os métodos que obtiveram índice de sucesso superior a 80% foram comparados entre si quanto ao tempo de estimulação necessário para a eversão do falo. O método aqui proposto alcançou 100% de eficiência na determinação do sexo de espécimes com comprimento de carapaça linear entre 10 e 34 cm. Além disso, quando comparado ao segundo método mais eficiente, reduziu substancialmente o tempo necessário para o manejo dos espécimes (91%), evitando possíveis sequelas nos indivíduos manuseados.

ABSTRACT

The efficacy of the combination of milbemycin oxime and afoxolaner was evaluated in desert tortoises infested with fly larvae. Oral administration of the combination of milbemycin oxime and afoxolaner eliminated the infestation without generating any evident adverse effects on the tortoises.


L'efficacité de la combinaison de milbémycine oxime et d'afoxolaner a été évaluée chez des tortues du désert infestées par des larves de mouches. L'administration orale de la combinaison de milbémycine oxime et d'afoxolaner traite l'infestation sans générer d'effets indésirables évidents sur les tortues.


A eficácia da associação de milbemicina oxima e afoxolaner foi avaliada em tartarugas do deserto infestadas por larvas de moscas. A administração oral da combinação de afoxolaner e milbemicina oxima eliminou a infestação sem gerar nenhum efeito adverso evidente às tartarugas.


Se evaluó la eficacia de la combinación de milbemicina oxima y afoxolaner en tortugas del desierto infestadas con larvas de mosca. La administración oral de la combinación de milbemicina oxima y afoxolaner eliminó la infestación sin generar efectos adversos evidentes en las tortugas.

Reference intervals (RIs) are an increasingly valuable tool for monitoring captive and free-living wildlife populations. Galapagos tortoises are one of the most emblematic species on Earth with 9 of the 12 extant species considered endangered due to human activities. As part of a long-term health assessment within the Galapagos Tortoise Movement Ecology Programme, we sampled a total of 210 free-living Santa Cruz Galapagos tortoises (Chelonoidis porteri). We collected blood from the brachial vein and performed packed cell volume (PCV), total solids (TS), morphological evaluation, white blood cell (WBC) count estimates and differentials and a VetScan biochemistry panel for each individual. We calculated 95% RIs and 90% confidence intervals (CIs) using the Reference Value Advisor tool and following international standard guidelines. Tortoises were categorized by estimated age and sex, with RI and CI reported here for 164 adult tortoises and 46 sub-adult tortoises. We found significant differences between sexes, with adult females presenting a lower PCV and higher values for both calcium and potassium. Among age groups, adult tortoises presented higher PCV, TS and albumin and lower WBC counts, aspartate aminotransferase and creatine kinase than sub-adult tortoises. We also found that tortoises presented higher numbers of lymphocytes during the dry season, but higher basophils, eosinophils, phosphorus, potassium and TS during the humid season. The heterophil:lymphocyte ratio did not differ between groups. To the authors' knowledge, this is the first report of formal plasma biochemistry and haematology RI for free-living Galapagos tortoises. With the present study we provide an important diagnostic tool for captive-breeding programs in the Galapagos and zoological institutions globally that care for giant tortoises. The ultimate goal of this study is to contribute to the understanding of giant tortoise baseline health parameters and to inform local management decisions that help to conserve these emblematic species.

ABSTRACT

The general bacterial microbiota of the soft tick Ornithodoros turicata found on Bolson tortoises (Gopherus flavomarginatus) were analyzed using next generation sequencing. The main aims of the study were to establish the relative abundance of bacterial taxa in the tick, and to document the presence of potentially pathogenic species for this tortoise, other animals, and humans. The study was carried-out in the Mapimi Biosphere Reserve in the northern-arid part of Mexico. Bolson tortoises (n = 45) were inspected for the presence of soft ticks, from which 11 tortoises (24.4%) had ticks in low loads (1-3 ticks per individual). Tick pools (five adult ticks each) were analyzed through 16S rRNA V3-V4 region amplification in a MiSeq Illumina, using EzBioCloud as a taxonomical reference. The operational taxonomic units (OTUs) revealed 28 phyla, 84 classes, 165 orders, 342 families, 1013 genera, and 1326 species. The high number of taxa registered for O. turicata may be the result of the variety of hosts that this tick parasitizes as they live inside G. flavomarginatus burrows. While the most abundant phyla were Proteobacteria, Actinobacteria, and Firmicutes, the most abundant species were two endosymbionts of ticks (Midichloria-like and Coxiella-like). Two bacteria documented as pathogenic to Gopherus spp. were registered (Mycoplasma spp. and Pasteurella testudinis). The bovine and ovine tick-borne pathogens A. marginale and A. ovis, respectively, were recorded, as well as the zoonotic bacteria A. phagocytophilum,Coxiella burnetii, and Neoehrlichia sp. Tortoises parasitized with O. turicata did not show evident signs of disease, which could indicate a possible ecological role as a reservoir that has yet to be demonstrated. In fact, the defense mechanisms of this tortoise against the microorganisms transmitted by ticks during their feeding process are still unknown. Future studies on soft ticks should expand our knowledge about what components of the microbiota are notable across multiple host-microbe dynamics. Likewise, studies are required to better understand the host competence of this tortoise, considered the largest terrestrial reptile in North America distributed throughout the Chihuahuan Desert since the late Pleistocene.

Galápagos giant tortoises are an essential component of their ecosystem and evaluation of parasites in their populations is essential for the management of conservation processes. Coccidiosis is the most common intestinal infection in free-living and captive reptiles. The aim of this study was to characterize molecularly the presence of Eimeria sp. in captive reared giant tortoises from Santa Cruz, Santiago, Española, and Pinzon Islands hatched and housed at the tortoise rearing center on Santa Cruz Island, Galápagos, by sequencing of the 18S rRNA gene. Galápagos. All samples were previously analyzed by coproparasitoscopic flotation technique and PCR for molecular identification. The results obtained by microscopy examination showed oocysts in all samples. PCR and sequencing indicated the presence Eimeria sp., showing a similarity percentage of 98% with Eimeria environmental. In conclusion, we identified a group of coccidia of the genus Eimeria sp. (MK909931) in Galápagos tortoises.

ABSTRACT

Antimicrobial resistance (AMR), encoded by plasmid-mediated AMR genes (ARGs), is an increasing global public health threat. Wildlife play a fundamental role as sentinels, reservoirs and potential vectors of ARGs. For the first time in Galapagos, we have identified and quantified the presence of ARGs in free-living giant tortoises (Chelonoidis porteri). We performed ARG analyses by quantitative PCR of faeces collected from the cloaca of 30 tortoises widely distributed across Santa Cruz Island. Validated samples (n = 28) were analysed by a panel of up to 21 different ARGs and all 28 tortoise samples were positive to one or more genes encoding resistance. Thirteen of 21 tested ARGs were present in at least one sample, and 10 tortoises (35.7%) had a multi-resistant pattern. We recommend additional research so we may more fully understand resistance patterns across taxa and geographical locations throughout the Galapagos archipelago, and the implications of ARGs for the health of wildlife, domestic animals, and humans. In this study, we found 100% of sampled giant tortoises had ARGs present in their faeces, suggesting a large-scale distribution of these genes within the archipelago.

ABSTRACT

Conservation discourses change rapidly both at global and local scales. To be able to capture these shifts and the relationships between humans and nature, we focused on a local and iconic conservation case: the Galapagos giant tortoises (Chelonoidis spp.). We used the Q methodology to contextualize conservation for science and decision making and to explore the multidimensionality of the conservation concept in Galapagos. The results indicate four prevailing discourses: (1) Multi-actor governance; (2) giant tortoise and ecosystems conservation; (3) community governance; and (4) market and tourism centred. These findings allow us to identify foreseeable points of disagreement, as well as areas of consensus, and to discuss the implication of the findings to address socio-ecological conservation and sustainability challenges. This can help the different involved stakeholders (managers, scientists and local communities) to the design and apply contextualized conservation actions and policies to contribute to a better sustainable management of the archipelago.

ABSTRACT

O cágado, Trachemys scripta elegans, conhecido popularmente como "tigre d´água americano" é uma espécie nativa das regiões aladas da Flórida e do Mississipi, nos Estados Unidos. Foram utilizados 20 (vinte) cágados (Trachemys scripta elegans), machos e fêmeas adultos, os quais foram eutanasiados, segundo a Resolução 714 de 20 de junho de 2002 do Conselho Federal de Medicina Veterinária (CFMV). Após a eutanásia foram identificados as aortas direita e esquerda para a injeção de Neoprene látex 450, corado com pigmento específico. Para a obtenção do molde de vinil foi injetado através das aortas direita e esquerda o acetato de vinila, seguido por corrosão em ácido sulfúrico. Observou-se que o pâncreas está intimamente relacionado com o fígado, ventrículo gástrico, duodeno e vesícula biliar. Sua face cranial esta na região distal do piloro, enquanto sua porção caudal ao longo da região cranial do duodeno. Anatomicamente o pâncreas é um órgão de estrutura alongada apresentando uma superfície levemente lobada. Quanto à vascularização arterial o pâncreas é irrigado em seu plano transversal por duas arteriais oriundas da artéria celíaca, e em cada antímero por duas artérias pancreaticoduodenais na região cranial, próximo ao piloro, pela artéria pancreaticoduodenal cranial e na porção caudal, ao longo do duodeno, irrigado pela artéria pancreaticoduodenal caudal.


The turtle Trachemys scripta elegans popularly known as "American tiger water" are native regions winged Florida and Mississippi, in the United States. We used twenty (20) turtles (Trachemys scripta elegans), adult males and females, which were euthanized under Resolution 714 of June 20, 2002 the Federal Council of Veterinary Medicine (CFMV). After euthanasia were identified aortas right and left to the injection of Neoprene latex 450, stained with specific pigment. To obtain the vinyl mold aorta was injected through the right and left vinyl acetate, followed by corrosion in sulfuric acid. It was observed that the pancreas is closely related to the liver, gizzard, gall bladder, and duodenum. His face cranial this distal region of the pylorus, while its caudal along the cranial region of the duodenum. Anatomically, the pancreas is an elongated body structure featuring a lightly lobed. As to the pancreas arterial vasculature is flushed in its transverse plane of two arteries arising from the celiac artery, and each antimere two pancreaticoduodenal artery in the cranial region, close to the pylorus by the pancreaticoduodenal artery flow in the caudal portion along the duodenum.
