ABSTRACT

Studies on fish embryo models are widely developed in research. They are used in several research fields including drug discovery or environmental toxicology. In this article, we propose an entirely automated assay to detect cardiac arrest in Medaka (Oryzias latipes) based on image analysis. We propose a multi-scale pipeline based on mathematical morphology. Starting from video sequences of entire wells in 24-well plates, we focus on the embryo, detect its heart, and ascertain whether or not the heart is beating based on intensity variation analysis. Our image analysis pipeline only uses commonly available operators. It has a low computational cost, allowing analysis at the same rate as acquisition. From an initial dataset of 3192 videos, 660 were discarded as unusable (20.7%), 655 of them correctly so (99.25%) and only 5 incorrectly so (0.75%). The 2532 remaining videos were used for our test. On these, 45 errors were made, leading to a success rate of 98.23%.

ABSTRACT

OBJECTIVE: To describe an exceptional case of conjoined twins in a triplet pregnancy after intracytoplasmic sperm injection (ICSI) and blastocyst transfer. DESIGN: Case report. SETTING: University teaching hospital reproductive endocrinology department and infertility practice. PATIENT(S): A 34-year-old woman underwent ICSI and received two blastocysts transferred. INTERVENTION(S): Transvaginal ultrasonography performed sequentially during early pregnancy. MAIN OUTCOME MEASURE(S): Ultrasound images of the fetus in gestational sac. RESULT(S): Two gestational sacs in the uterus were revealed at the 5th week. At the 8th week of gestation, a single fetus was seen in one sac, whereas thoracopagus conjoined twins was diagnosed in the other sac. At the 10th week, the conjoined twins had a spontaneous cardiac arrest confirmed by color Doppler. The subsequent course was uneventful, and a healthy child was born at 39th week. CONCLUSION(S): To date, a small number of cases of conjoined twins in IVF/ICSI pregnancies have been reported, in which most cases were treated with manipulations causing possible trauma in the zona pellucida. Our case is unique in that the transferred embryos were blastocysts that might have had additional damage on the zona pellucida from the longer culture.

ABSTRACT

Ten-day-old embryos were exposed to 28, 18 and 8 degrees C environments and their electrocardiograms (ECG) monitored. Embryos in 28 and 18 degrees C environments maintained a constant heart rate averaging 97 and 25 beats/min, respectively, followed by arrhythmias and cardiac arrest at 101 and 59 hr. Embryos in an 8 degrees C environment went into cardiac arrest after 2-4 hr, but recovered 20 hr later upon rewarming to 38 degrees C. Six to 20-day-old embryos exposed to 8 degrees C were examined for tolerance time after cardiac arrest. The younger the embryo the longer its tolerance to prolonged cardiac arrest.
