ABSTRACT

<b>Background and Objective:</b> Expert biological pest control is thought to be safer and to have fewer side effects. The use of plants carrying antifertility compounds is one method that can be developed to biologically reduce rat pest populations and thwart reproduction. This study aims to determine whether the toxicity of patiwala leaf extract (<i>Lantana camara</i> Linn.) can interfere with preimplantation stage rat (<i>Mus musculus</i> L.) pregnancy. <b>Materials and Methods:</b> This study was an experimental study arranged in a completely randomized design (CRD) consisting of 4 groups with 6 replications namely control (no treatment), patiwala leaf extract concentrations of 2 g/kg b.wt. (P1), 4 g/kg b.wt. (P2) and 6 g/kg b.wt. (P3). Mice were treated orally by gavage using a 1 mL disposable syringe on the 1st day of pregnancy (preimplantation) for three consecutive days. Mice were dissected at 16 days of gestation, observations were made of the percentage of implantation (%IM), the percentage of gestational loss (%KGE) and the percentage of postimplantation mortality (%KPI). Data were analyzed by Analysis of Variance (ANOVA) using KaleidaGraph 95% confidence level and Tukey's HSD test. <b>Results:</b> The toxicity of patiwala leaf extract significantly interfered with the preimplantation stage of pregnancy in rats. The P3 treatment (6 g/kg b.wt.) gave a higher toxicity effect, namely, the percentage of implantation success was only 25.67%, the percentage increase in gestational loss was 74.26% and the percentage of post-implantation death was 66.67%. <b>Conclusion:</b> The patiwala leaf extract at a concentration of 6 g/kg b.wt., interfered with the pregnancy of preimplantation stage rats because it could reduce the number of live fetuses.

ABSTRACT

This study describes an outbreak of acute necrotic hepatopathy associated with spontaneous poisoning by Lantana camara L. in dairy cattle. A herd of 15 cows and heifers was introduced into a native pasture with limited food supply, and, sixteen days later, eight animals had spontaneous nasal hemorrhage, fever, lethargy, jaundice, and dry, dark stools with mucus and blood. The clinical course varied from two to five days. In the pasture where the cattle were kept, abundant adult specimens of L. camara L. with evident signs of consumption were observed. In total, seven cattle died and necropsy was performed in three of them. All animals had moderate jaundice, hemorrhage in the subcutaneous tissue and on the surface of different organs. The liver was slightly enlarged, with orange discoloration and enhanced lobular pattern. Histologically, multifocal areas of coagulative necrosis of hepatocytes in the centrilobular area, occasionally extending to the midzonal area, were observed, as well as marked hepatocellular degeneration and prominent cholestasis. The current study suggests that L. camara L. poisoning should be considered a differential diagnosis of acute and necrotic hepatotoxicity in cattle, despite the absence of photosensitization.(AU)


Esse estudo descreve um surto de hepatopatia necrótica aguda associada a intoxicação espontânea por Lantana camara L. em bovinos leiteiros. Um lote de 15 vacas e novilhas foram introduzidas em um piquete com campo nativo, com escassa oferta de alimento. Após dezesseis dias, oito animais manifestaram epistaxe, febre, apatia, icterícia, fezes ressecadas e escuras com muco e sangue. A evolução do quadro clínico variou de dois a cinco dias. No piquete em que os bovinos estavam alojados havia grande quantidade de L. camara L. com sinais evidentes de consumo. No total, sete bovinos morreram, e destes, o exame post mortem foi realizado em três. Os bovinos exibiam moderada icterícia, hemorragias no tecido subcutâneo e na superfície de diferentes órgãos. O fígado estava discretamente aumentado, com coloração alaranjada e padrão lobular evidente. As lesões histológicas consistiam em acentuada necrose de coagulação de hepatócitos em região centrolobular, por vezes se estendendo a região mediozonal, além de acentuada degeneração dos hepatócitos e evidente Colestase. O presente trabalho alerta para que intoxicação por L. camara L. seja levada em consideração nos diagnósticos diferenciais de hepatotoxicidade necrótica aguda em bovinos, mesmo sem indícios de fotossensibilização.(AU)

Background and objectives: Natural products such as essential oils with antioxidant potential can reduce the level of oxidative stress and prevent the oxidation of biomolecules. In the present study, we investigated the antioxidant potential of Lantana montevidensis leaf essential oil (EOLM) in chemical and biological models using Drosophila melanogaster. Materials and methods: in addition, the chemical components of the oil were identified and quantified by gas chromatography coupled to mass spectrometry (GC-MS), and the percentage compositions were obtained from electronic integration measurements using flame ionization detection (FID). Results: our results demonstrated that EOLM is rich in terpenes with Germacrene-D (31.27%) and ß-caryophyllene (28.15%) as the major components. EOLM (0.12-0.48 g/mL) was ineffective in scavenging DPPH radical, and chelating Fe(II), but showed reducing activity at 0.24 g/mL and 0.48 g/mL. In in vivo studies, exposure of D. melanogaster to EOLM (0.12-0.48 g/mL) for 5 h resulted in 10% mortality; no change in oxidative stress parameters such as total thiol, non-protein thiol, and malondialdehyde contents, in comparison to control (p > 0.05). Conclusions: taken together, our results indicate EOLM may not be toxic at the concentrations tested, and thus may not be suitable for the development of new botanical insecticides, such as fumigants or spray-type control agents against Drosophila melanogaster.

BACKGROUND: In the field conditions, animals regularly consume small quantities of lantana leaves either while grazing or due to mixing with regular fodder. The hypothesis of this study was that consumption of lantana toxins over a long period of time leads to progression of sub-clinical disease. Toxicopathological effects of sub-chronic (90 days) administration of lantadenes of L. camara were investigated in guinea pigs. For this, a total of 40 animals were divided into 5 groups whereby groups I, II, III and IV were orally administered lantadenes, daily at the dose of 24, 18, 12, and 6 mg/kg bw, respectively while group V was control. The animals were evaluated by weekly body weight changes, haematology, serum liver and kidney markers, tissue oxidative markers and histopathology. RESULTS: The results of significant decrease in weekly body weights, haematology, liver and kidney marker enzymes (alanine aminotransaminase, aspartate aminotransaminase, acid phosphatase and creatinine), oxidation stress markers (lipid peroxidation, reduced glutathione, superoxide dismutase and catalase) in liver and kidneys, histopathology, and confirmation of fibrous collagenous tissue proliferation by Masson's Trichome stain showed that lantadenes led to a dose-dependent toxicity in decreasing order with the highest dose (24 mg/kg bw) producing maximum lesions and the lowest dose (6 mg/kg bw) producing minimum alterations. CONCLUSIONS: The study revealed that lantadenes which are considered to be classical hepatotoxicants in acute toxicity produced pronounced nephrotoxicity during sub-chronic exposure. Further studies are needed to quantify the levels of lantadenes in blood or serum of animals exposed to lantana in field conditions which would help to assess the extent of damage to the vital organs.

BACKGROUND: Diarrheal disease remains a public health problem in developing countries, including Ethiopia. In order to alleviate this disease, Ethiopian traditional healers use a wide range of medicinal plants from which Lantana camara is one of them. The stem of this plant is traditionally used for the treatment of diarrhoea. In addition, this plant is scientifically evaluated to have an antispasmodic effect on in vitro study. The aim of this study was to evaluate the antidiarrheal activity of the aqueous stem extract of L. camara Linn in mice. METHODS: The antidiarrheal activity of the extract was investigated using castor oil induced diarrhoea, enteropooling and small intestine transit models. The test groups received various doses (100, 200, and 400 mg/kg) of the extract, whereas positive controls received Loperamide (3 mg/kg) and negative controls received distilled water (10 ml/kg). RESULTS: In castor oil induced diarrhoea model, the extract, at all test doses, significantly (p < 0.001) prolonged diarrhoea onset, decreased the frequency of defecation, and weight of faeces. Similarly, the extract produced a significant (p < 0.001) decline in the weight and volume of intestinal contents at all tested doses. In addition, a significant (P < 0.001) reduction in the gastrointestinal motility in charcoal meal test was also observed in all doses of the extract. Phytochemical screening of the extract revealed the presence of flavonoids, alkaloids, tannins, and phytosterols that may play a key role in its antidiarrheal activity. CONCLUSION: The obtained results of the present study confirm antidiarrheal activity of the stem of L. camara, thus provide the scientific basis for the traditional uses of this plant as a treatment for diarrhoea.

Introdução: O uso de plantas como inseticidas podem substituir muitos produtos sintéticos por apresentarem baixa toxicidade para os animais e biodegradação no ambiente. A espécie Lantana montevidensis (Spreng.) Briq. (Verbenaceae), chumbinho, é um vegetal subarbustivo rico de compostos secundários para a sua defesa. O modelo experimental com Drosophila melanogaster tem sido usada para estudos, por apresentar vantagens, pois tem fácil manuseio e rápida taxa de reprodução. Objetivo: Avaliar a composição química e efeito inseticida do óleo essencial (OE) de L. montevidensis contra D. melanogaster. Metodos: O OE das folhas secas provenientes do Crato-CE, Brasil, foi extraído por hidrodestilação e foi analisado por CG/EMS para identificação dos constituintes. No ensaio foi avaliada a mortalidade e geotaxia negativa das moscas, em que elas foram expostas a diferentes concentrações do OE (3-30,5 µg/mL do óleo essencial/ar). Um total de vinte moscas adultas foram submetidas à diapausa por meio de resfriamento e em seguidas foram colocadas em frascos de 300 mL, cuja parte inferior havia papel filtro impregnado com 1 mL de sacarose a 20 por cento em água destilada. Na parte superior (tampa) foi afixado um papel filtro para a aplicação do produto a ser testado, nesse caso o OELM. As leituras foram realizadas a cada 3, 6, 12 e 24 h. Resultados: Os resultados mostraram que o OE apresentou o ß-Cariofileno (34,96 por cento), o Germacreno D (25,49 por cento) e o Biciclogermacreno (9, 78 por cento) como componentes majoritários, no ensaio inseticida houve uma CL50 na concentração de 15,14 µg/mL em 3 h de exposição, e houve um efeito significativo nos danos do aparelho locomotor das moscas.mConclusão: Este estudo fornece evidências que esse OE apresenta-se como um potencial bioinseticida(AU)

The species Lantana montevidensis (Spreng.) Briq. (Verbenaceae), chumbinho, is a subshrub rich in secondary metabolites it uses for its own defence. The experimental model for Drosophila melanogaster has been used in several types of studies. Some of its advantages are its easy management and its fast rate of reproduction. Objective: Evaluate the chemical composition and insecticidal effect of the essential oil (EO) of L. montevidensis against D. melanogaster. Method: EO of dry leaves from Crato-CE, Brazil, was extracted by hydrodistillation and analyzed by GC/EMS to identify its constituents. As part of the test, evaluation was conducted or the mortality and negative geotaxis of the flies, to attain which they were exposed to different concentrations of the EO (3-30.5 µg/mL essential oil/air). A total 20 adult flies were subjected to cold diapause and placed in 300 mL bottles with filter paper in their lower section impregnated with 1 mL of 20 percent saccharose in distilled water. Filter paper was also attached to the upper section (lid) to apply the test product OELM. Readings were taken every 3, 6, 12 and 24 h. Results: Results showed that the principal components of the EO are ?-caryophyllene (34.96 percent), germacrene D (25.49 percent) and bicyclogermacrene (9.78 percent). In the insecticide test there was a CL50 for the concentration of 15.14 µg/ml in 3 h exposure, and significant damage to the locomotor system of the flies. Conclusion: The study provides evidence that the EO analyzed is a potential biopesticide(AU)

ABSTRACT

In evidently the first study of its kind, vermicompost derived solely from a weed known to possess plant and animal toxicity was used to assess its impact on the germination and early growth of several plant species. No pre-composting or supplementation of animal manure was done to generate the vermicompost in order to ensure that the impact is clearly attributable to the weed. Whereas the weed used in this study, Lantana (Lantana camara), is known to possess strong negative allelopathy, besides plant/animal toxicity in other forms, its vermicompost was seen to be a good organic fertilizer as it increased germination success and encouraged growth of all the three botanical species explored by the authors - green gram (Vigna radiata), ladies finger (Abelmoschus esculentus) and cucumber (Cucumis sativus). In terms of several physical, chemical and biochemical attributes that were studied, the vermicompost appeared plant-friendly, giving best results in general when employed at concentrations of 1.5% in soil (w/w). Fourier transform infrared spectrometry revealed that the phenols and the sesquiterpene lactones that are responsible for the allelopathic impact of Lantana were largely destroyed in the course of vermicomposting. There is also an indication that lignin content of Lantana was reduced during its vermicomposting. The findings open up the possibility that the billions of tons of phytomass that is generated annually by Lantana and other invasives can be gainfully utilized in generating organic fertilizer via vermicomposting.

ABSTRACT

This study investigated biological activity of chloroform extract of dry Lantana camara 'Mozelle' leaves against the eastern subterranean termite, Reticulitermes flavipes (Kollar), an important structural pest. Repellent activity was assessed using a paper-disc choice test and a sand arena choice test. Antifeedant and toxic properties were assessed using a no-choice paper test and a topical application method. In the choice tests, significantly fewer termites made contact with treated paper-discs at test concentrations > or = 0.016 mg/cm2 (equivalent to 0.0023 wt:wt) or tunneled into treated sand at test concentrations > or = 0.125 mg/g, compared with control. In the no-choice tests, termite feeding activity was significantly reduced and termite mortality was greatly increased in treatments than control. Exposure to filter paper treated at 0.212 and 0.106 mg/cm2 (equivalent to 0.03 and 0.015 wt:wt) resulted in > 90% mortality and 78% reduction in feeding, and approximately 52% mortality and 40% reduction in feeding, respectively. Top-dorsal application led to > 60% mortality at 4 microg/termite. This study showed that the chloroform leaf extract of L. camara had excellent repellent and moderate toxic and antifeedant activities.

ABSTRACT

Lantana (Lantana camara Linn.) is a noxious weed to which certain medicinal properties have been attributed, but its ingestion has been reported to be highly toxic to animals and humans, especially in the liver. The main hepatotoxin in lantana leaves is believed to be the pentacyclic triterpenoid lantadene A (LA), but the precise mechanism by which it induces hepatotoxicity has not yet been established. This work addressed the action of LA and its reduced derivative (RLA) on mitochondrial bioenergetics. At the concentration range tested (5-25 microM), RLA stimulated state-4 respiration, inhibited state-3 respiration, circumvented oligomycin-inhibited state-3 respiration, dissipated membrane potential and depleted ATP in a concentration-dependent manner. However, LA did not stimulate state-4 respiration, nor did it affect the other mitochondrial parameters to the extent of its reduced derivative. The lantadenes didn't inhibit the CCCP-uncoupled respiration but increased the ATPase activity of intact coupled mitochondria. The ATPase activity of intact uncoupled or disrupted mitochondria was not affected by the compounds. We propose, therefore, that RLA acts as a mitochondrial uncoupler of oxidative phosphorylation, a property that arises from the biotransformation (reduction) of LA, and LA acts in other mitochondrial membrane components rather than the ATP synthase affecting the mitochondrial bioenergetics. Such effects may account for the well-documented hepatoxicity of lantana.

ABSTRACT

Lantana (Lantana camara Linn) is a noxious weed that grows in many tropical and subtropical parts of the world. Ingestion of lantana foliage by grazing animals causes cholestasis and hepatotoxicity. Both ruminants and nonruminant animals such as guinea pigs, rabbits, and female rats are susceptible to the hepatotoxic action of lantana toxins. The hepatotoxins are pentacyclic triterpenoids called lantadenes. Molecular structure of lantadenes has been determined. Green unripe fruits of the plant are toxic to humans. Lantana spp. exert allelopathic action on the neighboring vegetation. The allelochemicals have been identified as phenolics, with umbelliferone, methylcoumarin, and salicylic acid being the most phytotoxic. In addition to phenolics, a recent report indicates lantadene A and B as more potent allelochemicals. Management of lantana toxicosis in animals is achieved by drenching with activated charcoal and supportive therapy. Recent reports on the bilirubin clearance effect of Chinese herbal tea Yin Zhi Huang (decoction of the plant Yin Chin, Artemisia capillaries, and three other herbs) or its active ingredient 6,7-dimethylesculetin, in jaundice are very exciting and warrant investigations on its, possible, ameliorative effects in lantana intoxicated animals. Research is being conducted on new drug discovery based on natural products in different parts of the lantana plant.

ABSTRACT

The anti-inflammatory activity of Lantana trifolia (Verbenaceae) was determined by carrageenan, serotonin and histamine-induced rat paw edema and the analgesic activity of this plant was studied by acetic acid-induced writhings and tail flick tests in mice. Lantana trifolia extracts (at 30 mg/kg) inhibited carrageenan and histamine-induced rat paw edema. Although the extracts did not produce any effect on acetic acid-induced writhings, they all develop a significant increase on tail flick antinociceptive index (doses varying between 1 and 30 mg/kg), indicating a spinal antinociceptive effect. These results provide support for the use of Lantana trifolia in relieving inflammatory pain.

ABSTRACT

Neste estudo sobre a toxidez das lantanas para bovinos e ovinos, verificou-se situação semelhante à que ocorre na Austrália, que nem todas as espécies de Lantana e nem todos os taxa de Lantana camara que ocorrem no Brasil, são tóxicos. Verificou-se, que foram tóxicas as amostras de lantanas procedentes de Boa Vista (RO), Castanhal (PA), Cáceres (MT), Serra Talhada (PE), Cabo Frio e Quatis (RJ) e Canoinhas (SC). Outras amostras procedentes de Cruzeiro do Sul (Acre), Castanhal (PA), Chapada dos Guimarães e Lambari d Oeste (MT), Jaguaribe (CE), Vitória da Conquista e Wanderley (BA), Vitória (ES), Vassouras, Parati e Itaguaí (RJ) não revelaram toxidez nas doses administradas (40 g/kg). A dose letal das amostras submetidas à experimentação no Brasil foi bastante constante (40 g/kg), com duas exceções: as folhas frescas da lantana procedente de Canoinha (SC) foram muito mais tóxicas (10 g/kg) e as folhas frescas e as dessecadas da lantana de Serra Talhada (PE) foram muito menos tóxicas, pois somente causaram intoxicação não-letal com a dose de 40g/kg/dia administradas durante 30 dias. Históricos sobre a observação de fotossensibilização em bovinos, ligados à ingestão de lantanas foram obtidos em Serra Talhada (PE), Cáceres (MT), Cabo Frio (RJ), Quatis (RJ) e Canoinhas (SC). Verificou-se que não é possível estabelecer uma correlação entre a cor das inflorescências das lantanas e sua toxidez, confirmando a constatação feita na Austrália, que o potencial de intoxicar não está necessariamente relacionada com a cor das flores.
