ABSTRACT

Although most chitons (Mollusca: Polyplacophora) are shallow-water molluscs, diverse species also occur in deep-sea habitats. We investigated the feeding strategies of two species, Leptochiton boucheti and Nierstraszella lineata, recovered on sunken wood sampled in the western Pacific, close to the Vanuatu Islands. The two species display distinctly different associations with bacterial partners. Leptochiton boucheti harbours Mollicutes in regions of its gut epithelium and has no abundant bacterium associated with its gill. Nierstraszella lineata displays no dense gut-associated bacteria, but harbours bacterial filaments attached to its gill epithelium, related to the Deltaproteobacteria symbionts found in gills of the wood-eating limpet Pectinodonta sp. Stable carbon and nitrogen isotope signatures and an absence of cellulolytic activity give evidence against a direct wood-feeding diet; both species are secondary consumers within the wood food web. We suggest that the distinct associations with bacterial partners are linked to niche specialisations of the two species. Nierstraszella lineata is in a taxonomic family restricted to sunken wood and is possibly adapted to more anoxic conditions thanks to its gill-associated bacteria. Leptochiton boucheti is phylogenetically more proximate to an ancestral form not specialised on wood and may itself be more of a generalist; this observation is congruent with its association with Mollicutes, a bacterial clade comprising gut-associated bacteria occurring in several metazoan phyla.

ABSTRACT

The taxonomic position of three bacterial strains, Asr22-19T, NBRC 101035 and NBRC 101041, isolated from shellfish in Japan, was determined by using a polyphasic taxonomic approach. The strains were facultatively anaerobic, motile by gliding and Gram-staining-negative slender rods. Their major respiratory quinone was menaquinone-7 and their predominant cellular fatty acids were iso-C15:0, iso-C17:0 3-OH, iso-C15:0 3-OH, C16:0 3-OH, and C16:0. The G+C content of the genomic DNA was 42.0-42.7 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that the strains clustered with the genus Persicobacter in the family 'Flammeovirgaceae'. DNA-DNA relatedness values were higher than 68% among strains Asr22-19T, NBRC 101035 and NBRC 101041, and were lower than 28% between strain Asr22-19T and Persicobacter diffluens NBRC 15940T. The three novel strains could be differentiated from Persicobacter diffluens by several phenotypic characteristics. On the basis of these results, the novel species Persicobacter psychrovividus sp. nov. (type strain Asr22-19T=NBRC 101262T=CIP 109100T) is proposed and emended descriptions are given for the genus Persicobacter and for Persicobacter diffluens.

ABSTRACT

Strain FYK2218(T) was isolated from a specimen of the chiton Acanthopleura japonica, which had been collected from a beach on the Boso peninsula in Japan. Phylogenetic analyses based on 16S rRNA gene sequences revealed that the strain belonged to the phylum 'Acidobacteria'. The most closely related type strains to strain FYK2218(T) were Holophaga foetida TMBS4(T) (83.6 % 16S rRNA gene sequence similarity) and Geothrix fermentans H-5(T) (83.6 %) in subdivision 8 of the 'Acidobacteria'. Cells of FYK2218(T) were motile, rod-shaped, Gram-negative, mesophilic and strictly aerobic. The G+C content of the strain was 56.7 mol%. The strain had isoprenoid quinones MK-6 and MK-7 as major components. Major fatty acids of the strain were iso-C(15 : 0), iso-C(17 : 0), C(16 : 0) and C(20 : 5)omega3c (cis-5,8,11,14,17-eicosapentaenoic acid). From the taxonomic data obtained in this study, it is proposed that the new marine isolate be placed into a novel genus and species named Acanthopleuribacter pedis gen. nov., sp. nov. within the new family, order and class Acanthopleuribacteraceae fam. nov., Acanthopleuribacterales ord. nov. and Holophagae classis nov. The family Holophagaceae fam. nov. is also described. The type strain of Acanthopleuribacter pedis is FYK2218(T) (=NBRC 101209(T) =KCTC 12899(T)).
