ABSTRACT

Rhabdias pocoto n. sp. is herein described from the lungs of the swamp frog Pseudopaludicola pocoto Magalhães, Loebmann, Nogueira, Kokubum, Baptista, Haddad & Garda, 2014, from the Caatinga biome in the state of Ceará, in north-eastern Brazil. The new species is characterized by a body that dilates posteriorly, six small lips (protuberances) and two rounded lateral expansions of cuticular inflation on the anterior end, each containing an amorphous gland-like structure inside and a short and conical tail. Additionally, molecular analysis and comparison of the partial mitochondrial cytochrome c oxidase I sequence of R. pocoto n. sp. revealed genetic divergence between the new species and the sequences of Rhabdias spp. previously deposited in GenBank. Phylogenetic analysis grouped the new taxon into the R. pseudosphaerocephala species complex + R. glaurungi clade. The new discovery represents the 19th species of Rhabdias spp. described in the Neotropical region, the ninth in Brazil and the first species of Rhabdias found parasitizing South American frogs of the genus Pseudopaludicola, as well as the first Caatinga biome species of Rhabdias.

ABSTRACT

Terrestrial gastropod molluscs are widely distributed and are well known as pests of many types of plants that are notoriously difficult to control. Many species of nematodes are able to parasitize land snails and slugs, but few of them are lethal to their host. Species and/or populations of mollusc-parasitic nematodes (MPNs) that kill their hosts are promising for biological control purposes. The recent discovery of new nematode species of the genus Phasmarhabditis in Europe and the associations between Alloionema spp. and slugs are expanding the possibilities of using MPNs as control agents. However, very little is known about the distribution and ecology of these species. Using molecular techniques based on qPCR methods for quick identification and quantification of various species of MPN isolated directly from the soil or from infected hosts can assist in providing information on their presence and persistence, as well as the composition of natural assemblages. Here, we developed new primers and probes for five species of the genus Phasmarhabditis and one species of the genus Alloionema. We employed these novel molecular techniques and implemented a published molecular set to detect MPN presence in soil samples coming from natural and agricultural areas in Switzerland. We also developed a method that allows the detection and quantification of Phasmarhabditis hermaphrodita directly from the tissues of their slug host in a laboratory experiment. The new molecular approaches were optimized to a satisfactory limit of detection of the species, with only few cross-amplifications with closely related species in late cycles (>32). Using these tools, we detected MPNs in 7.5% of sampled sites, corresponding to forest areas (P. hermaphrodita and Alloionema appendiculatum) and wheat-oriented agricultural areas (Phasmarhabditis bohemica). Moreover, we confirmed that the method can be used to detect the presence of P. hermaphrodita inside slug hosts, with more detections in the susceptible slug Deroceras larvae compared to the resistant Arion vulgaris. These primers/probe sets provide a novel and quick tool to identify MPNs from soil samples and infected slugs without having to culture and retrieve all nematode life stages, as well as a new tool to unravel the ecology of nematode-slug complexes.

ABSTRACT

O presente estudo teve como objetivo descrever a ocorrência de otite parasitária causada por Rhabditis blumi em bovinos leiteiros de raça Gir de uma fazenda da região Norte do Brasil. Foram coletadas amostras de 42 bovinos por swab e lavado dos condutos auditivos externos (CAE). Ao exame clínico, 9,5% (4/42) dos bovinos apresentavam sintomatologia nervosa, como leve a moderada rotação da cabeça, apatia, flacidez dos lábios e ptose palpebral unilateral, alopecia das regiões da cabeça e cupim, causados pelo desconforto e prurido da região auricular, alteração na mastigação e acúmulo de alimento na cavidade oral. Adicionalmente, 71,4% (30/42) dos abovinos mostraram a presença do parasita no cerúmen dos condutos auditivos. À análise microscópica do material do saco conjuntival foi observado presença do parasita em 90% (9/10) dos bovinos avaliados. Os 30 bovinos positivos para Rhabditis spp. foram distribuídos aleatoriamente em três grupos de 10 animais: (G1) Bovinos controle, (G2) Bovinos tratados com ivermectina 1% pour on e (G3) Bovinos submetidos a lavado dos condutos auditivos externos (CAE). Cada tratamento foi repetido três vezes com intervalo de sete dias. No G1 os 10 bovinos mantiveram-se infectados durante todo o estudo. No G2 20% dos bovinos foram negativos após os dois primeiros tratamentos, porém, mostraram-se positivos na terceira avaliação. No G3 todos os bovinos mantiveram-se positivos, sendo observada apenas diminuição da carga parasitária. A identificação por análise molecular por meio de fragmentos amplificados da expansão D2/D3 do 28S rDNA confirmou a presença apenas da espécie Rhabditis blumi nos animais. Baseado nas observações clínicas, morfológicas e moleculares pode-se relatar o primeiro caso de R. blumi em bovinos da raça Gir no Estado do Pará, através da compra de animais oriundos de áreas onde a otite parasitária tem sido diagnosticada, principalmente de Minas Gerais, para formar animais mestiços (Gir x Holandês). Desta forma ressalta-se a importância do exame clínico prévio dos animais a serem transferidos para outras propriedades ou regiões. Este relato também parece ser o primeiro sobre a presença de R. blumi no saco conjuntival de bovinos. O tratamento com ivermectina no G2 não surtiu melhora clínica dos bovinos.(AU)


This study aimed to describe the occurrence of parasitic otitis caused by Rhabditis blumi in dairy cattle of the Gir race from a farm in northern Brazil. Forty-two samples were collected from cattle by swab washed from the external auditory canal (EAC). On clinical examination, in 71.4% (30/42) of the cattle the parasite was found in the cerumen of the ear canal, along with alopecia of head and hump caused by discomfort and itching of the auricular region. At microscopic analysis of material from the conjunctival sac the parasite was found in 90% (9/10) of the evaluated cattle. In addition, 9.5% (4/42) of the cattle showed nervous symptoms, such as mild to moderate rotation of the head, apathy, flaccid lips and unilateral ptosis, change in chewing and food accumulation in the oral cavity. Thirty cattle positive for Rhabditis spp. were randomly divided into three groups of 10 animals each: (G1) Cattle Control, (G2) Cattle treated with ivermectin 1% pour-on, and (G3) Cattle undergoing wash of the external auditory canal (EAC). Each treatment was repeated three times with intervals of seven days. In G1, 10 cattle remained infected throughout the study. In G2, 20% of the cattle were negative after the first two treatments, however were positive at the third evaluation. In G3, all cattle remained positive, but with decrease in parasite load. Identification by molecular analysis of amplified fragments through the expansion D2/D3 28S rDNA confirmed the presence of only Rhabditis blumi. Based on clinical, morphologic and molecular examination, it appears to be the first report of the occurrence of R. blumi infection in Gir cattle in the State of Pará, due to the purchase of cattle from areas where parasitic otitis has been diagnosed, as from Minas Gerais, to produce crossbred animals (Gir x Holstein). This emphasizes the importance of prior clinical examination by the veterinary service in order to transfer only healthy animals to other properties or regions. This appears also to be the first report on R. blumi infection of the conjunctival sac in cattle. Treatment with ivermectin in G2 did not produce clinical improvement.(AU)

In this study, we assessed the effect of the saprobic fungus, Fusarium oxysporum (Ascomycota: Hypocreales) on the fitness of the entomopathogenic nematode Heterorhabditis sonorensis (Caborca strain). Sand column assays were considered to evaluate the effect of fungal mycelia on infective juvenile (IJ) movement and host access. Additionally, we investigated the effect of fungal spores on the nematodes' ability to search for a host, its virulence, penetration efficiency and reproduction. Three application timings were considered to assess interactions between the fungus and the nematodes. In vitro assays were also considered to determine the effect of fungal extracts on the nematode's symbiotic bacteria. Our observations indicate that presence and age of fungal mycelia significantly affect IJ movement in the sand columns and their ability to establish in the host. These results were also reflected in a reduced insect mortality. In particular, treatments with the 15 days old mycelia showed a significant reduction in insect mortality and penetration efficiency. Presence of fungal spores also impacted nematode virulence and reproduction. In particular, two of the application timings tested (simultaneous [EPN and fungal spores applied at the same time] and alternate I [EPN applied first, fungus applied 24h later]) resulted in antagonistic interactions. Moreover, IJ progeny was reduced to half in the simultaneous application. In vitro assays revealed that fungal extracts at the highest concentration tested (10mg/ml) inhibited the growth of the symbiotic bacteria. Overall, these results suggest that saprobic fungi may play an important role in regulating. EPN populations in the soil, and that they may be one of the factors that impact nematode survival in the soil and their access to insect hosts.

ABSTRACT

O consumo de vegetais in natura é recomendado como alimentação saudável em razão de seuconsiderável percentual de vitaminas, sais minerais e fibras alimentares. Frequentemente as verduras são adubadas com dejetos ou irrigadas com águas contaminadas com matéria fecal. Em razão da ocorrência de enteroparasitos em consumidores de hortaliças, avaliou-se, por meio deste trabalho, a presença de parasitos em alfaces hidropônicas e convencionais comercializadas na cidade de Anápolis-GO. Utilizado o método de sedimentação espontânea, foram encontradas larvas de nematoides do gênero Rhabditis. Neste gênero, há espécies de vida livre que, eventualmente, podem parasitar o homem e animais domésticos. Diante dessas evidências, recomenda-se a higienização adequada destes alimentos tanto pelos consumidores domésticos como pelos manipuladores de hortaliças a serem servidas em restaurantes e lanchonetes. Trata-se do primeiro relato de Rhabditis em hortaliças no Brasil.

ABSTRACT

The purpose of this study was to evaluate the efficacy of orally administered albendazole sulphoxide and pour-on ivermectin for the treatment of bovine parasitic otitis caused by rhabditiform nematodes. Eighteen Gyr cows presenting clinical otitis were divided in three groups with six animals each. The first one did not receive any treatment (control group). The second one was treated with 0.5 percent pour-on ivermectin, 500µg/kg of body weight, and the third group was treated with oral 6 percent albendazole sulphoxide, at 6.0mg/kg. Both ear canals of each animal were reexamined on days 7 and 21 post treatment. The animals in the control group remained infected throughout the days of observation. Ivermectin treatment did not show effectiveness on days 7 or 21 post treatment. The albendazole sulphoxide treatment had an efficacy of 16.7 and 25 percent on days 7 and 21, respectively. Further studies are required to assess an effective treatment for this parasitic disease, especially via alternative administration routes, because of its significant impact on Bos taurus indicus cattle breeding in Tropical and Subtropical Regions.


O objetivo deste estudo foi avaliar a eficácia do sulfóxido de albendazol administrado oralmente e da ivermectina "pour-on" no tratamento da otite parasitária bovina causada por nematóides rhabditiformes. Dezoito vacas Gir apresentando otite clínica foram divididas em três grupos de seis animais cada. O primeiro não recebeu tratamento (grupo controle). O segundo foi tratado com ivermectina "pour-on" a 0,05 por cento na dose de 500µg/kg de peso vivo. O terceiro grupo foi tratado com sulfóxido de albendazol oral a 6 por cento na dose 6,0mg/kg. Os condutos auditivos de todos os animais foram reexaminados nos dias 7 e 21 pós-tratamento. Os animais do grupo controle permaneceram infectados nos dias de observação. O tratamento com ivermectina não demonstrou eficácia alguma para os dias 7 e 21 pós-tratamento. O tratamento com sulfóxido de albendazol obteve 16,7 e 25 por cento de eficácia nos dias 7 e 21, respectivamente. Mais estudos são necessários para determinação de tratamentos eficazes para tal doença parasitária, especialmente através de vias alternativas de administração, por causa de seu significante impacto na criação de Bos taurus indicus nas Regiões Tropical e Subtropical.

BACKGROUND: Heterorhabditis bacteriophora is applied throughout the world for the biological control of insects and is an animal model to study interspecies interactions, e.g. mutualism, parasitism and vector-borne disease. H. bacteriophora nematodes are mutually associated with the insect pathogen, Photorhabdus luminescens. The developmentally arrested infective juvenile (IJ) stage nematode (vector) specifically transmits Photorhabdus luminescens bacteria (pathogen) in its gut mucosa to the haemocoel of insects (host). The nematode vector and pathogen alone are not known to cause insect disease. RNA interference is an excellent reverse genetic tool to study gene function in C. elegans, and it would be useful in H. bacteriophora to exploit the H. bacteriophora genome project, currently in progress. RESULTS: Soaking L1 stage H. bacteriophora with seven dsRNAs of genes whose C. elegans orthologs had severe RNAi phenotypes resulted in highly penetrant and obvious developmental and reproductive abnormalities. The efficacy of postembryonic double strand RNA interference (RNAi) was evident by abnormal gonad morphology and sterility of adult H. bacteriophora and C. elegans presumable due to defects in germ cell proliferation and gonad development. The penetrance of RNAi phenotypes in H. bacteriophora was high for five genes (87-100%; Hba-cct-2, Hba-daf-21, Hba-icd-1; Hba-nol-5, and Hba-W01G7.3) and moderate for two genes (usually 30-50%; Hba-rack-1 and Hba-arf-1). RNAi of three additional C. elegans orthologs for which RNAi phenotypes were not previously detected in C. elegans, also did not result in any apparent phenotypes in H. bacteriophora. Specific and severe reduction in transcript levels in RNAi treated L1s was determined by quantitative real-time RT-PCR. These results suggest that postembryonic RNAi by soaking is potent and specific. CONCLUSION: Although RNAi is conserved in animals and plants, RNAi using long dsRNA is not. These results demonstrate that RNAi can be used effectively in H. bacteriophora and can be applied for analyses of nematode genes involved in symbiosis and parasitism. It is likely that RNAi will be an important tool for functional genomics utilizing the high quality draft H. bacteriophora genome sequence.

ABSTRACT

Rhabdias kongmongthaensis sp. n. is described based on specimens found in the lungs of the tree frog Polypedates leucomystax (Gravenhorst) (Amphibia: Rhacophoridae) from Kanchanaburi Province, western Thailand. The new species is similar to two North-American species, Rhabdias ranae and R. americanus, by presence of two lateral pseudolabia, each with two inner submedian protuberances. R. kongmongthaensis differs from both species by relative length and shape of the tail, and by its distribution and host specificity. Presence of lateral pseudolabia distinguishes the new species from the geographically closest Rhabdias species as well as from those parasitizing other rhacophorid frogs.
