ABSTRACT

Although biochemical analytes have typically been measured using serum or whole blood samples, an increasing number of assays are validated for measurement of analytes from dried blood spots (DBS) on filter paper. DBS techniques are minimally invasive, require only a small sample volume, and simplify processing, storage, and shipment of samples. These qualities make DBS-based assays ideal for sampling of wildlife species in both captive and field settings. In this study, a liquid chromatography-tandem mass spectrometry assay was evaluated for measurement of 25-hydroxyvitamin D in sloths. Paired serum and DBS samples were collected from nine healthy captive Hoffmann's two-toed sloths (Choloepus hoffmanni). Statistical analysis using Passing-Bablok regression analysis, Bland-Altman plots, and the Wilcoxon signed-rank tests found good agreement between 25-hydroxyvitamin D3 measurements in serum and DBS samples. Constant and proportional bias were absent. Results from this study support the use of DBS samples for the evaluation of vitamin D status in Hoffmann's two-toed sloths and provide a foundation for further studies to validate this technique.

ABSTRACT

Two species of sloths in the family Megalonychidae, Hoffmann's (Choloepus hoffmanni) and Linnaeus's (Choloepus didactylus) two-toed sloths, are commonly held in zoological institutions. Despite frequent published reports of urinary tract disease in these species, reports of diagnostics are mostly limited to descriptions of hematology and serum chemistry. In this study, repeated urinalysis, urinary chemistry, serum chemistry, and radiographs were collected opportunistically from six Hoffmann's and five Linnaeus's sloths. Proteinuria, bacteriuria, low urine specific gravity, and crystalluria were common in the absence of other signs of urinary tract pathology.

ABSTRACT

Bradypus torquatus is a rare and endemic sloth species from the Atlantic Forest, Brazil. Due to a lack of medical information including hematologic reference parameters for the species, hematologic baseline values were determined using samples from 14 clinically healthy B. torquatus , under captive (n = 7) and free-living (n = 7) conditions in Bahia State, Brazil. Additionally, the morphology of the blood cells is presented, with a demonstration that the Barr body chromosome may assist with sex determination of the species. The Barr body chromosome was present in all seven females and absent in all males. Many erythrocytes were approximately the size of small lymphocytes, with red blood cells exhibiting anisocystosis, normochromia, and apparent macrocytosis, compared with domestic animals. This study provides the first published hematologic values and cell morphology for B. torquatus . However, further studies are suggested using an automated hematology analyzer with larger sample sizes so that reference intervals may be established and hematologic values better understood for sex, habitat type, and age cohorts.

ABSTRACT

Between 2005 and 2008, wild Bradypus variegatus and Choloepus hoffmanni inhabiting an agricultural landscape and captive animals from a rescue center in Northeastern Costa Rica were studied to assess exposure to pesticides. A total of 54 animals were sampled: 42 wild sloths captured at an agricultural landscape and 12 captive animals from a rescue center. Pesticides' active ingredients were determined in three sample matrices: hair, aqueous mixture (paws' wash) and cotton gauze (mouth clean) based on multi-residue gas chromatography methods. Recoveries tests ranged from 73 to 146% and relative standard deviations were less than 20% throughout all the recovery tests. Active ingredients detected in sloths samples were ametryn, chlorothalonil, chlorpyrifos, diazinon, difenoconazole, ethoprophos and thiabendazole. These active ingredients were used in intensive agricultural production for bananas, pineapples and other crops. Blood plasma cholinesterase activity (PChE) was determined by the Ellman method modified for micro plates. Enzyme activity determination was normalized to protein content in the samples according to Bradford method. Wild sloth PChE activity was similar for both species while sloths in captivity showed differences between species. Enzyme activity was significantly lower for two-toed sloths. This study showed that sloths were exposed to pesticides that caused acute and chronic effect in mammals and can also be a threat to other wildlife species. There is a need to better understand the potential effects of exposure to pesticides in sloths and other wild mammal populations, especially those threatened or endangered. More studies in this field must be carried out on the wildlife fauna inhabiting the agricultural landscape and its surroundings.

ABSTRACT

Dexmedetomidine, a highly selective alpha-2 adrenergic agonist and dextrorotary enantiomer of medetomidine, was combined with ketamine and used to immobilize 14 free-ranging Choloepus hoffmanni (Hoffmann's two-toed sloths) and 11 Bradypus variegatus (brown-throated three-toed sloths) in Upala, Costa Rica. Following intramuscular injection of ketamine (2.1 mg/kg) and dexmedetomidine (11 microg/kg), heart rate, respiratory rate, and indirect systolic blood pressure were measured every 5 min for a total of 25 min. An iStat (CG8+) was used to evaluate serum biochemical and hematologic values during anesthesia. After 30 min of anesthesia, atipamezole (0.13 mg/kg) was administered intramuscularly, which resulted in rapid and smooth recoveries. Mean heart rate and respiratory rate remained unchanged in both C. hoffmanni and B. variegatus over time. Progressive decreases in mean indirect systolic blood pressure were documented in both species. Results of this study suggest a combination of dexmedetomidne and ketamine is a safe and effective anesthetic protocol for use in free-ranging C. hoffmanni and B. variegatus. Similar to other alpha-2 adrenergic agonist-based immobilization protocols, close monitoring of cardiovascular and respiratory parameters are recommended. This study also provides serum biochemical and hematologic data in free-ranging C. hoffmanni and B. variegatus.

Standard hematology parameters were determined for 122 sloth bears (Melursus ursinus ursinus) at the Sur Sarovar Bird Sanctuary, Uttar Pradesh, India (27 degrees 0'N; 77 degrees 45'E), and the Bannerghatta Biological Park, Karnataka, India (12 degrees 48'N; 77 degrees 34'E) from March 2003 to July 2006. These two native sloth bear habitats have different climatic conditions and provided an opportunity to examine the effect of climate on the physiologic hematology values of these bears. We primarily analyzed the influence of age, sex, season, and body weight on the different hematology parameters. Several values were significantly different in sloth bear cubs (1 yr). The cubs had a lower erythrocyte count, hemoglobin concentration, packed cell volume (PCV), and mean cell hemoglobin (MCV) values when compared to adult and subadult bears. The cubs also had higher leukocyte counts, due to higher circulating neutrophils, as compared to adult and subadult bears. Within subadult and adult bears, we also identified a sexual dimorphic difference in leukocyte count in adult and subadult bears, wherein female bears had higher counts than males. This difference was the result of a significantly higher number of circulating neutrophils in female bears. Platelet counts were also higher in females as compared to males. On comparing different seasons, leukocyte counts were higher in winter as compared to the summer and monsoon seasons. When compared based on location, erythrocyte counts were higher in subadult and adult bears at Bannerghatta, which was at a higher altitude than Sur Sarovar. Within subadult and adult bears, we did not find any significant influence of age or body weight on the different hematologic parameters. In this study we have obtained mean hematologic values for sloth bears in their native habitat to serve as a reference for this species. This report will be useful to develop and evaluate health profiles of sloth bears under various ecological conditions.

ABSTRACT

The aim of the present study was to confirm whether feeding influences the resting breathing rate and to observe possible alterations in blood gas and pH levels produced by feeding in unanesthetized sloths (Bradypus variegatus). Five adult male sloths (4.1 +/- 0.6 kg) were placed daily in an experimental chair for a period of at least 4 h for sitting adaptation. Five measurements were made for each sloth. However, the sloths one, two and five were studied once and the sloths three and four were studied twice. Breathing rate was determined with an impedance meter and the output signal was digitized. Arterial blood samples were collected for blood gas analysis with a BGE electrolytes analyzer and adjusted for the animal's body temperature and hemoglobin content. The data are reported as mean +/- SD and were collected during the resting period (8:00-10:00 h) and during the feeding period (16:00-18:00 h). The mean breathing rate increased during mastication of ymbahuba leaves (rest: 5.0 +/- 1, feeding: 10 +/- 1 bpm). No significant alterations were observed in arterial pH (rest: 7.42 +/- 0.05, feeding: 7.45 +/- 0.03), PCO2 (rest: 35.2 +/- 5.3, feeding: 33.3 +/- 4.4 mmHg) or PO2 (rest: 77.5 +/- 8.2, feeding: 78.4 +/- 5.2 mmHg) levels. These results indicate that in unanesthetized sloths 1) feeding evokes an increase in breathing rate without a significant change in arterial pH, PCO2 or PO2 levels, and 2) the increase in breathing rate produced by feeding probably is due to the act of mastication.

ABSTRACT

Free-ranging southern two-toed sloths (Choloepus didactylus) were translocated during the flooding of a forest at a hydroelectric dam site in French Guiana. Over an 11 mo period blood samples were collected from 90 sloths (38 males, 52 females) in order to determine hematological and serum chemistry reference values. Mean values and range of values were calculated for 13 hematological and 21 serum chemistry parameters. Variations associated with sex, age and reproductive status were identified. Males had a significantly lower red blood cell count than females. Immature animals had more monocytes while adults had more neutrophils and higher mean corpuscular hemoglobin concentration. Aspartate aminotransferase and triglyceride values were higher in young than in adult sloths but uric acid was lower. Lactating females showed lower red blood cells count and iron levels than non-lactating females. These profiles will help to provide reliable baseline data for medical evaluation of sloths.

ABSTRACT

The hemoglobin of the Pale-Throated Three-Toed Sloth (Bradypus tridactylus, Xenarthra) was separated into two components (ratio 4:1) with identical amino-acid analyses for the alpha- and beta-chains. The primary structures of both chains from the major component are given. They could be isolated by chromatography on carboxymethyl cellulose CM-52. The sequences have been determined by automatic Edman degradation of the native chains and their tryptic peptides. The comparison with human hemoglobin showed 27 substitutions in the alpha-chains and 33 in the beta-chains. In the alpha-chains one amino-acid exchange involves an alpha 1/beta 1-contact. In the beta-chains two heme- and four alpha 1/beta 1-contacts are substituted. The hemoglobin of the Sloth is compared to that of the Nine-Banded Armadillo (Dasypus novemcinctus), another representative of the order Xenerthra.
