ABSTRACT

This report describes a case of notomelia in a female piglet. The extra limb was attached at the interscapular region and was significantly undergrown. Its appendage was bifurcated with a unilateral keratinized tip. This is probably the first report of notomelia in a pig. The observations were compared with the bovine and ovine cases of notomelia reported so far.

ABSTRACT

Obstructive urolithiasis is a disease characterized by the presence of uroliths in the urinary tract, with consequent obstruction of excretion pathways. This paper described the epidemiological and clinical-pathological findings of 22 outbreaks of urolithiasis in growing-finishing pigs in Southern Brazil. All affected pigs were male and clinical presentation consisted of lethargy, dysuria, rectal prolapse, abdominal distention, peripheral cyanosis and reluctance to move. Clinical progression course ranged from 12 hours to one week, and the lethality rate was 100%. Gross changes were characterized by urinary bladder rupture associated with marked amount of yellowish liquid with ammoniacal odor (urine) in the abdominal cavity (uroperitoneum), as well as mild fibrin deposition on the surface of abdominal organs and hydronephrosis. Urinary uroliths ranging from 0.3 to 1cm in diameter were often observed obstructing the lumen of the penile urethra and sometimes those were free in the abdominal cavity. Histopathological findings included diffuse and marked urinary bladder edema and hemorrhage associated with inflammatory infiltrate of lymphocytes, plasma cells, and macrophages. Diffuse and marked necrosis of the mucosal epithelium was observed in the penile urethra. Intense fibrin deposition and inflammatory infiltrate of neutrophils were noted in the peritoneum, as well as in the serosa of the organs in the abdominal cavity. Uroliths were submitted to the method of qualitative determination of the mineral components, and were compatible with calcium carbonate and magnesium ammonium phosphate. Growing pigs ration analysis revealed low levels of calcium in relation to phosphorus, resulting in a Ca:P ratio of approximately 0.35:1. Histological findings and mineral analysis suggest that outbreaks of urolithiasis were related to a nutritional imbalance in the proportions of dietary calcium and phosphorus. The main cause of mortality was related to dehydration and uroperitoneum.(AU)


Urolitíase obstrutiva é uma enfermidade caracterizada pela presença de urólitos no trato urinário, com consequente obstrução das vias de excreção. Este artigo descreve os achados epidemiológicos e clínico-patológicos de 22 surtos de urolitíase em suínos de crescimento e terminação no Sul do Brasil. Os suínos afetados eram machos e clinicamente apresentavam letargia, disúria, prolapso retal, abaulamento do abdômen, extremidades cianóticas e relutância em movimentar-se. A duração dos sinais clínicos variou de 12 horas a uma semana, e a letalidade foi de 100%. As alterações macroscópicas caracterizaram-se por ruptura da bexiga com acentuada quantidade de líquido de coloração amarelada e odor amoniacal (urina) livre na cavidade abdominal (uroperitônio), além de discreta deposição de fibrina sobre os órgãos e hidronefrose. Frequentemente obstruindo o lúmen da uretra peniana e por vezes livre na cavidade abdominal, era possível observar urólitos urinários que variavam de 0,3 a 1cm de diâmetro. Os achados histopatológicos incluíram edema e hemorragia difusos e acentuados na bexiga, associado a infiltrado inflamatório predominante de linfócitos, plasmócitos e macrófagos. Na uretra peniana havia necrose difusa e acentuada do epitélio da mucosa. No peritônio e nas serosas dos órgãos da cavidade abdominal havia intensa deposição de fibrina e infiltrado neutrofílico. Os urólitos foram submetidos ao método de determinação qualitativa dos componentes minerais, os quais foram compatíveis com carbonato de cálcio e fosfato de amônio magnesiano. A análise da ração de crescimento revelou baixos níveis de cálcio, em relação ao fósforo, perfazendo uma relação Ca:P de aproximadamente 0,35:1. Os achados histológicos e as dosagens minerais sugerem que os surtos de urolitíase foram relacionados a um desequilíbrio nutricional nas proporções de cálcio e fósforo dietético. A principal causa da morte dos suínos foi relacionada à desidratação e ao uroperitônio.(AU)

ABSTRACT

O enriquecimento ambiental é uma ferramenta importante dentro dos sistemas de produção, a fim de promover o bem-estar e favorecer a saúde dos animais. Portanto, o objetivo deste trabalho foi avaliar o efeito do enriquecimento ambiental sobre o estresse de suínos na fase de creche. Foram utilizados 32 leitões, alojados em granja experimental, distribuídos em quatro grupos (n= 8): corda, corrente, garrafa PET e controle negativo. Amostras de sangue foram coletadas no início e no final do experimento para contagem de leucócitos circulantes e determinação de antioxidantes não enzimáticos, óxido nítrico, malondialdeído, e de saliva para avaliação do cortisol. Foi aplicado etograma e fez-a ganho médio de peso diário e a conversão alimentar. Os parâmetros avaliados no primeiro dia do experimento não variaram entre os grupos (P>0,05). No último dia do experimento, os valores de neutrófilos e da relação neutrófilo/linfócito foram mais elevados nos leitões do grupo corrente, assim como os valores de cortisol salivar (P<0,05). O ácido úrico apresentou-se mais elevado nos leitões do grupo corrente e o malondialdeído (MDA) nos do grupo garrafa (P< 0,05). Os enriquecimentos ambientais estimularam comportamentos positivos nos leitões, tendo a corda se destacado como o mais atrativo. Por outro lado, a corrente apresentou efeito negativo sobre a fisiologia dos animais, gerando estresse, assim como a garrafa, que induziu a peroxidação lipídica e um menor ganho de peso nos leitões.(AU)


Environmental enrichment is an important tool within production systems to promote welfare and animal health. The aim of this study was to evaluate the effect of enrichment objects on stress of piglets in nursery phase. 32 piglets housed in experimental farm were distributed in 4 groups (n= 8): rope, chain, pet bottle and negative control. Blood samples were collected at the beginning and end of the experiment to count circulating leukocytes and determine non-enzymatic antioxidants, nitric oxide, malondialdehyde, and saliva to evaluate cortisol. At the same time, an etogram and evaluation of mean daily gain and feed conversion were applied. The evaluated parameters on first day of the experiment did not vary between groups (P> 0.05). On the last day, neutrophil and neutrophil /lymphocyte ratios were higher in chain group piglets, as were salivary cortisol values (P< 0.05). Uric acid was higher in chain group and MDA in bottle group (P< 0.05). Environmental enrichment stimulated positive behaviors in piglets, where the rope stood out as the most attractive. On the other hand, chain had a negative effect on animal physiology, generating stress, as well as the bottle that induced lipid peroxidation and a lower weight gain in piglets.(AU)

ABSTRACT

Com o objetivo de avaliar a situação epidemiológica da cisticercose suína e bovina na zona rural de cinco municípios, distribuídos em três distintas regiões do estado de Minas Gerais, foi determinada a prevalência das cisticercoses bovina e suína nas propriedades rurais e os principais fatores de risco que poderiam favorecer a transmissão e manutenção da doença nas referidas propriedades. A pesquisa abrangeu 388 propriedades, localizadas em 91 diferentes comunidades rurais, sendo coletadas 1.792 amostras de sangue bovino e 554 de suíno. Além da avaliação das referidas amostras laboratoriais (ELISA indireto e imunoblot), também foram analisados os dados obtidos da aplicação de um questionário para cada propriedade, no qual constava as informações sobre as condições socioeconômicas, higiênico-sanitárias e as relacionadas ao sistema de criação animal. A prevalência da cisticercose variou de 0,3 a 5,9% nos bovinos e de 0 a 2,8% nos suínos criados artesanalmente nesses municípios pesquisados. Os principais fatores de risco de transmissão identificados foram a fonte de água consumida, a origem da carne, a natureza do município e da região, para a cisticercose bovina, e o destino do esgoto e o sistema de criação, para a cisticercose suína.(AU)


In order to evaluate the epidemiological situation of porcine and bovine cysticercosis in the rural area of five municipalities, distributed in three different regions of the state of Minas Gerais, Brazil, the prevalence of bovine and porcine cysticercosis was determined in the rural properties as well as the main risk factors that may favor the transmission and maintenance of the disease in these properties. The research covered 388 farms, located in 91 different rural communities, with 1,792 samples of bovine blood and 554 of swine collected. In addition to the evaluation of these laboratory samples (indirect ELISA and Imunoblot), the data obtained from the application of a questionnaire for each property were also analyzed, with information on socioeconomic, hygienic-sanitary and animal-related conditions. The prevalence of cysticercosis ranged from 0.3 to 5.9% in cattle and from 0 to 2.8% in the pigs raised in these municipalities. Among the main risk factors for transmission of bovine cysticercosis were the source of water consumed, the origin of the meat, the nature of the municipality and the region. Regarding the pigs, risk factors were the sewage disposal and animal management system.(AU)

ABSTRACT

The primary objective of the current study was to compare the pharmacokinetic (PK) of florfenicol (FFL) in pulmonary epithelial lining fluid and the plasma in swine. The second objectives were to evaluate the effect of anesthesia with ketamine and propofol on the PK of FFL in plasma. Bronchoaveolar lavage was utilized for quantification of PELF volume and the urea dilution method was used to determine the concentration of FFL in PELF. FFL was administered intramuscularly (IM) to swine in a single dose of 20mg/kg body weight. The main PK parameters of FFL in plasma and PELF were as follows: the area under the concentration-time curve, maximal drug concentration, elimination half-life and mean residence time were 69.45±4.36 vs 85.03±9.26µg·hr/ml, 4.65±0.34 vs 5.94±0.86µg/ml, 9.87±1.70 vs 10.69±1.60hr and 12.75±0.35 vs 14.46±1.26hr, respectively. There was no statistically significant difference between the PK profiles of FFL for the anesthetized and unanesthetized pigs. This study suggest that (i) FFL penetrated rapidly into the pulmonary and the drug concentration decay faster in plasma than in the pulmonary, (ii) the PK profile of FFL in swine was not interfered after administration of anesthetic agent.(AU)


O objetivo primário desse estudo foi comparar a farmacocinética de florfenicol (FFL) em fluido epitelial pulmonar à farmacocinética (PK) de FFL em plasma suíno. O segundo objetivo foi avaliar o efeito de anestesia com ketamina e propofol no PK de FFL em plasma. Lavagem broncoalveolar foi utilizada para quantificar volume de fluido epitelial pulmonar (PELF) e método de diluição de uréia para determinar FFL em PELF. Injeção de FFL foi administrada intramuscular a suínos em dose única de 20mg/kg de peso corporal. Os principais parâmetros de PK em FFL em plasma e PELF foram os seguintes: a área sob a curva de concentração-tempo, concentração máxima da droga, eliminação de meia-vida e média de tempo de permanência foram 69,45±4,36 vs 85,03±9,26µg·hr/ml, 4,65±0,34 vs 5,94±0,86µg/ml, 9,87±1,70 vs 10,69±1,60hr e 12,75±0,35 vs 14,46±1,26hr, respectivamente. Não houve diferença estatisticamente significante entre os perfis de PK de FFL para os porcos anestesiados e não anestesiados. Esse estudo sugere que (i) FFL penetrou rapidamente no pulmão e concentração da droga sofre queda mais veloz em plasma que líquido pulmonar, (ii) o perfil de PK de FFL em suínos não modificou após administração de agente anestésico.(AU)

ABSTRACT

Knowledge of the incidence of spontaneous congenital abnormalities is critical for the accurate interpretation of findings in teratogenicity studies in any species. In this paper, results of the examination of 1739 neonatal Göttingen Minipigs are presented. Over the 2-year period under consideration, the incidence of external and visceral malformations was less than 0.2 and 0.1%, respectively. The most common external malformations were syndactyly, limb hyperflexion, domed head and scoliosis. The most common internal malformations were undescended testes, ventricular septal defect, diaphragmatic hernia and atrial septal defects. Pentadactyly and variation in the aortic arch's bifurcation (absent truncus bicaroticus) were the most common variations. These data will help support the use of the Göttingen Minipig as a non-rodent species in embryofetal development studies where concerns persist about the availability of background data.

ABSTRACT

Important factors contributing to the well-known high mortality of piglets produced by SCNT are gross malformations of vital organs. The aim of the present retrospective study was to describe malformations found in cloned piglets, transgenic or not, dying or culled before weaning on Day 28. Large White (LW) embryos were transferred to 78 LW recipients, while 72 recipients received Göttingen embryos (67 transgenic and five not transgenic) and 56 received Yucatan embryos (43 transgenic and 13 not transgenic). Overall pregnancy rate was 76%, and there were more abortions in recipients with minipig embryos than in those with LW embryos (26% and 24% vs. 6%). Piglets (n = 815) were born from 128 sows with 6.5 ± 0.4 full-born piglets per litter. The overall rate of stillborn piglets was 21% of all born with the number of stillborn piglets ranging from one to nine in a litter. The mortality of the surviving piglets during the first month was 48%. Thus, altogether 58% of the full-born piglets died before weaning. In 87 of the 128 litters (68%), one to 12 of the piglets showed major or minor malformations. Malformations were found in 232 piglets (29.5% of all born). A single malformation was registered in 152 piglets, but several piglets showed two (n = 58) or more (n = 23) malformations (7.4% and 2.8% of all born, respectively). A significantly higher malformation rate was found in transgenic Göttingen and Yucatan piglets (32% and 46% of all born, respectively) than in nontransgenic LW (17%). There was a gender difference in the transgenic minipigs because male piglets had a higher rate of malformations (49.1%) than females (29.7%). The most common defects in the cloned piglets were in the digestive (12.2%), circulatory (9.4%), reproductive (11.3%), and musculoskeletal (9.1%) systems. Malformations of the musculoskeletal system were most frequent in Göttingen (16.3% vs. approximately 5.5% in the two other breeds), whereas abnormal cardiopulmonary systems were most frequent in Yucatan piglets (26.9% vs. 2.1% in LW and 5.3% in Göttingen). In conclusion, these results show that pig cloning results in a considerable loss of piglets and that many of these can be related to various malformations that all are also seen in noncloned piglets. Because approximately half of the cloned piglets still survive, even with eventual unknown minor malformations, use of pigs as models for human diseases is still realistic. However, continued efforts are needed to further reduce the level of malformations.

ABSTRACT

Congenital skeletal abnormalities may be genetic, teratogenic, or nutritional in origin; distinguishing among these different causes is essential in the management of the disease but may be challenging. In some cases, teratogenic or nutritional causes of skeletal abnormalities may appear very similar to genetic causes. For example, chondrodysplasia associated with intrauterine zinc or manganese deficiency and mild forms of hereditary chondrodysplasia have very similar clinical features and histologic lesions. Therefore, historical data are essential in any attempt to distinguish genetic and acquired causes of skeletal lesions; as many animals as possible should be examined; and samples should be collected for future analysis, such as genetic testing. Acquired causes of defects often show substantial variation in presentation and may improve with time, while genetic causes frequently have a consistent presentation. If a disease is determined to be of genetic origin, a number of approaches may be used to detect mutations, each with advantages and disadvantages. These approaches include sequencing candidate genes, single-nucleotide polymorphism array with genomewide association studies, and exome or whole genome sequencing. Despite advances in technology and increased cost-effectiveness of these techniques, a good clinical history and description of the pathology and a reliable diagnosis are still key components of any investigation.

ABSTRACT

Two outbreaks of poisoning by selenium in swine in southern Brazil are described. Piglets were affected in the post-weaning period, between 27 and 22 days, with mortality rates ranging from 16% to 15.3% (Outbreak 1 and 2 respectively). The pigs had focal symmetrical poliomielomalacia and hoof lesions, which were initially characterized by a reddish line at the coronary band that evolved in surviving pigs to release of the hooves. São descritos dois surtos de intoxicação por selênio em suínos na região Sul do Brasil. Foram acometidos leitões em fase de creche, entre 27 e 22 dias, com mortalidade variando de 16% a 15,3% (Surto 1 e 2 respectivamente). Os suínos apresentaram poliomielomalacia simétrica focal e lesões de casco, que inicialmente eram caracterizadas por uma linha avermelhada na borda coronária que evoluía nos suínos sobreviventes para desprendimento do casco. ABSTRACT

In the modern pig industry, the increasing number of piglets born per litter augments the importance of the number of functional teats in the sow. The aim of this study was to evaluate the function and importance of inverted teats during nursing and to analyze structural and functional differences between the mammary glands of inverted teats versus normal teats. Nine farrowing gilts (8 purebred Swedish Yorkshire gilts and 1 cross between Swedish Yorkshire and Norwegian Landrace) and 94 piglets (59 piglets suckling normal teats, 32 piglets suckling protruded teats [i.e., previously inverted], 2 piglets suckling inverted teats, and 3 piglets suckling considerably smaller teats) were included in the study. Teat fidelity (keeping the same teat between the nursings) was registered, excluding the first 48 h postpartum. Piglet weight was recorded daily during the first week of life and thereafter once a week until weaning at 4 wk of age. Weight and growth rate were analyzed using repeated observation mixed-model analysis of variance. The 2 piglets that suckled the inverted teats were not able to emerge the teats and they were euthanized 4 and 8 d after birth, respectively, due to loss of BW. The average weight at weaning (28 d of age) was 8.1 kg (range 3.2-13.8 kg). In the normal teats (n = 53), the weight of the corresponding mammary gland tissue at necropsy was positively correlated to the piglet average daily weight gain during wk 2 (r = 0.33, P < 0.05), 3 (r = 0.55, P < 0.001), and 4 (r = 0.47, P < 0.001). In the protruded teats (n = 32), the weight of the corresponding mammary gland tissue was positively correlated to the piglet average daily weight gain during wk 2 (r = 0.63, P < 0.001) and 3 (r = 0.43, P < 0.05). Among the piglets nursing normal teats, 82% kept fidelity to its teat and the corresponding percent for the protruded teats was 26%. In 7 of the 9 sows, the weaning weight of the piglets suckling protruded teats was numerically lower compared to the piglets suckling normal teats, although the difference was not statistically significant. Piglets nursing small teats had lower weaning weight (4.8 kg) and the corresponding mammary tissue also had lower weight (335 g). This study indicates the importance of an adequate number of well-developed and well-functioning teats in healthy sows for the growth and development of the piglets but also emphasizes the complexity of the interaction between the nursing sow and her piglets.

A Salmonella pode estar presente em carcaças suínas ao final da tecnologia de abate, sendo que não existem procedimentos de inspeção especificamente direcionados para o controle desta bactéria, consideradas um risco potencial para a saúde pública. O objetivo do trabalho foi verificar a presença de Salmonella em carcaças suínas e caracterizar os pontos críticos de controle (PCCs) da tecnologia de abate como subsídio ao sistema APPCC. Foram realizados swabs de carcaças após a escaldagem/depilação, antes da evisceração, após evisceração e serragem da carcaça e após 24 horas de refrigeração, gerando 120 amostras (30 carcaças de cinco lotes). Isolou-se Salmonella em 53,3% dos PCs analisados, sendo 40% antes da eviscerarão e 30% após a escaldagem/depilação e evisceração/serragem. Nas carcaças resfriadas por 24 horas não foi isolada Salmonella. Os sorovares identificados foram Typhimurium (96,7 %) e Panama (3,22%). O isolamento de Salmonella reforça a necessidade de programas de monitoria da bactéria, desde as granjas até os abatedouros, enquanto o não isolamento nas carcaças resfriadas indica a importância da lavagem final e adequação da cadeia do frio, o que minimiza a contaminação pela intensificação das medidas de higiene e controle de pontos críticos para subsidiar o sistema APPCC...


Salmonella can be present in swine carcasses at the end of slaughtering and there is no specifically guided inspection procedure for the control of this bacterium, considered a potential risk for public health. The objective of this paper is to verify the presence of Salmonella in carcasses and to characterize the critical points of control (CPCs) of the slaughtering technology as a subsidy to the HACCP system. The carcasses were swabbed at different points during the slaughtering process, ant after 24 hours of refrigeration, generating 120 samples (30 carcasses from five different batches). Salmonella was isolated in 53.3% of the CPs analyzed, being 40% before evisceration and 30% after scalding/depilation and evisceration/ sawing. No Salmonella was isolated in the carcasses refrigerated for 24 hours. Typhimurium (96.7%) and Panama (3.22%) were the serovars identified. Salmonella isolation indicates the need of programs to monitor the bacterium from the farms to the slaughterhouse, while the non-isolation in the refrigerated carcasses shows the relevance of the cold chain, which minimizes the contamination by the intensification of hygiene measures and the control of critical points to subsidize the HACCP system...


La Salmonella puede estar presente en carcasas porcinas al final de la matanza, siendo que no existen procedimientos de inspección específicamente direccionados para el control de esta bacteria, consideradas un riesgo potencial a la salud pública. El objetivo del trabajo ha sido verificar la presencia de Salmonella en carcasas porcinas y caracterizar los puntos críticos de control (PCCs) en la tecnología de matanza como subsidio al sistema APPCC. Se han realizado swabs de carcasas tras escaldadura/depilación, antes de evisceración, después de la evisceración y serrín de la carcasa y después de 24 horas de refrigeración, generando 120 muestras (30 carcasas de cinco lotes) Se ha aislado Salmonella en 53,3% de los PCs analizados, siendo 40% antes de evisceración y 30% después de la escaldadura/depilación y evisceración/serrín. En las carcasas enfriadas por 24 horas no se ha aislado Salmonella. Los serovares identificados fueron Typhimurium (96,7%) y Panama (3,22%). El aislamiento de Salmonella refuerza la necesidad de programas de monitoria de la bacteria, desde las granjas hasta los mataderos, el no aislamiento en las carcasas enfriadas indica la importancia del lavado final y adecuación de la cadena del frío, lo que minimiza la contaminación por la intensificación de las medidas de higiene y control de puntos críticos para subsidiar el sistema APPCC...

ABSTRACT

The inverted teat defect is the most common disorder of the mammary complex in pigs. It is characterized by the failure of teats to protrude from the udder surface, preventing normal milk flow and thus limiting the rearing capacity and increasing the risk of mastitis. The inverted teat defect is a liability trait with a complex mode of inheritance. We previously identified QTL for inverted teats. As a complementary approach that integrates map-based efforts to identify candidate genes for the inverted teat defect with function-driven expression analysis, application-specific microarrays were constructed that cover 1525 transcripts mapping in QTL regions on pig chromosomes 2, 3, 4, 6 and 11. About 950 transcripts were expressed in epithelial and mesenchymal teat tissue. The expression of three categories of teats was compared: normal teats of both non-affected and affected animals and inverted teats of affected animals. In epithelium and mesenchyme, 62 and 24 genes respectively were significantly differentially expressed (DE). The majority of biofunctions to which a significant number of DE genes were assigned are related to the following: (1) cell maintenance, proliferation, differentiation and replacement; (2) organismal, organ and tissue development; or (3) genetic information and nucleic acid processing. Moreover, the DE genes belong almost exclusively to canonical pathways related to signaling rather than metabolic pathways. This is in line with findings obtained by genome-wide catalogue microarrays. This study adds another piece to the puzzle of the etiology of inverted teats by indicating that causal genetic variation leading to the disorder is likely among the genes encoding for members of the signaling cascades of growth factors.

ABSTRACT

This study reports on changes in the number of somatostatin-like immunoreactive (SOM-LI) endocrine cells in the porcine descending colon, caused by chemically driven inflammation, axotomy and proliferative enteropathy (PE). The distribution pattern of SOM-LI endocrine cells has been studied using the routine single-labelling immunofluorescence technique. Semi-quantitative evaluation of the number of the SOM-immunostained endocrine cells within the mucosal layer of the porcine descending colon has been based on counting of all endocrine cells immunoreactive to SOM per unit area (0,1 mm²). Under physiological conditions the number of SOM-LI endocrine cells has been shown to constitute 3,30±0,22. All applied pathological processes resulted in changes in the SOM-like immunoreactivity, which varied in particular processes studied. The number of SOM-LI endocrine cells increased to 6,28±0,31 and 4,43±0,35 during chemically driven inflammation and proliferative enteropathy, respectively, and decreased to 1,17%±0,16 after axotomy. The obtained results suggest that SOM-LI endocrine cells may participate in various pathological states within porcine descending colon and their functions probably depend on the type of pathological factor.(AU)

ABSTRACT

To delineate the anomaly and frequency of their occurrence in a pig model, we reported the topography of the vagus laryngeal branches and compared the differences with humans. Thirty sides of cervical vagus nerve in 15 fresh cadavers (Sus scrofa) were microdissected. We measured the branch diameters and lengths of the laryngeal branches using a Vernier caliper with a resolution of 0.01 mm. Two patterns of the vagus laryngeal branches were shown: 56.7% with the cranial laryngeal nerve (CLN) and 43.3% without the CLN. The diameters and the length of the CLN were not affected by the side of the neck (P > 0.05), but the diameters of the recurrent laryngeal nerve (RLN) and the nodose ganglion were significantly different between left and right sides (P < 0.05). The left RLN was thinner than the right side in diameter (P < 0.05). Four of the 30 sides had anastomoses between the vagus and the cervical sympathetic chain. There were some differences between the pig anatomy and human anatomy, but the patterns were largely similar. The similarities support the utility of this model, which is closer in size to humans than the standard rodent models.

ABSTRACT

Gene expression profiling of compromised umbilical cords (CUCs) derived from somatic cell nuclear transfer (scNT) clones was performed to determine why scNT-derived clones often exhibit malformed umbilical arteries. Umbilical cord samples were obtained from 65 scNT piglets, and of these, nine displayed a CUC. Microscopic analyses of the scNT clones with CUCs (scNT-CUCs) revealed complete occlusive thrombi that were not detected in the arteries of scNT clones with normal umbilical cords (scNT-Ns). Moreover, whereas the allantoic ducts of the scNT-Ns contained columnar epithelium, the scNT-CUCs lacked this epithelial layer. Compared to scNT-Ns, the scNT-CUCs exhibited severe histological damage, including tissue swelling and vein and arterial damage with complete occlusive thrombi. To investigate functional abnormality, gene expression profiles were created in duplicate using the Platinum Pig 13K oligonucleotide microarray, which contains 13,610 probes of 70 bp in length and is capable of interrogating 13,297 targets with up to one probe per target. Probe sets were selected according to a 2-fold or greater increase or decrease of gene expression in scNT-CUCs compared to scNT-Ns. Most genes expressed in scNT-Ns were also expressed by scNT-CUCs. However, most genes involved in transcriptional regulation, such as JUN, JUNB, and FOSL2, showed a significant decrease in expression in the scNT-CUCs, which may produce a ripple effect capable of altering the transcriptomes of many other cellular processes, including angiogenesis, antioxidation, and apoptosis. The scNT-CUCs with thrombosis showed extensive apoptosis leading to placental insufficiency and related pathology. Considering that the umbilical cord plays a role in the transportation of metabolites to the fetus, placental insufficiency in scNT-CUCs may be caused by an increase in apoptotic protein expression from scNT-derived umbilical cords with hypoplastic arteries, and our results provide evidence that porcine oligonucleotide microarray analysis is a useful tool for screening scNT-derived abnormalities in pigs.

ABSTRACT

O objetivo deste trabalho foi avaliar e classificar as principais causas de morte de leitões com até seis dias de idade, em umsistema intensivo de suínos confinados (SISCO), localizado na região médio-norte do estado de Mato Grosso durante o anode 2006. Observaram-se 48.416 leitões provenientes de 2.054 matrizes. Verificou-se taxa de mortalidade de 7,43% do totalde nascidos vivos. As causas mais frequentes de mortalidade, em relação ao total de nascidos vivos, foram esmagamento(24,14%), debilitação (8,01%), síndrome diarreica (6,0%), defeitos genéticos (2,41%) e outras causas sem diagnósticoconfirmado durante a necrópsia (15,20%). A taxa de natimortalidade em relação ao total de nascidos foi de 5,59%. Comrelação à sazonalidade das mortes, observou-se maior índice de mortalidade por síndrome diarreica durante a primavera,que coincide com os meses mais quentes do ano, não observando tendência para morte por esmagamento em nenhumaépoca do ano.


The objective of this research was to evaluate and to classify (by means of postmortem findings), the occurrence of the preweaning losses of piglets until the six days of age, in a intensive system of bordered pigs, at the middle north of Mato Grosso State during the year 2006. They had been observed a total of 48.416 piglets proceeding from 2.054 wombs. Was verified the tax of mortality of 7,43% of that livings piglets had been born. The most frequent causes of losses were overlaying (24.14 %), diverse causes (15.20), weakness (8.01 %), diarrhea syndrome (6.0 %) and genetic defects (2.41 %). Stillbirth rate, in relation to total born, was 5.59 %. In relation of periodicity deaths, it was observed a higher number of diarrhea syndrome during the spring, that coincides with the hottest months of the year, wasn’t observed trends of overlaying deaths none time during the year.

ABSTRACT

Sperm 'tail stump' defect was found in ejaculates of a wild boar maintained in captivity. It was in good physical condition, the testes and genital tract were found to be of normal size and consistency. There was no evidence of macroscopic abnormalities at the clinical analysis and at necropsy. The volume and concentration of the semen samples obtained by electroejaculation were lower than normal. The slides examined contained a high level of abnormal spermatozoa (52.7%). The most frequent morphological finding was a droplet-like form attached to the base of the head or a very short stump. The non-stumped spermatozoa had no normal tail but a shortened one. Analysing the histological structure with light microscopy, no ring of spermatozoa was observed lining the lumen of the seminiferous tubules and the characteristically cellular structure was not conserved. The ultrastructural examination evidenced a disorganisation of the normal tubular structure of the flagellum, with lost of regular pattern of the axial bundle of fibrils and the mitochondrial helix. The origin of this abnormality is unknown.

ABSTRACT

Congenital defects like myofibrillar dysplasia (splayleg), umbilical and inguinal hernias, cryptorchism, intersexes, and anal atresia occur relatively frequently in swine. On the other hand, some developmental anomalies like double monsters are very rare. The present paper reports a rare case of a congenital complex malformation including polymelia, duplicitas coli partialis et recti, atresia ani et fistula rectogenitalis, duplicitas corpori uteri, cervicis, vaginae et vulvae and duplicitas vesicae, urethrae et renalis. A plausible interpretation concerning the etiology is that the anomalies arose from unequal partial twinning. The pig has been healthy and inconspicuous. Although no anus was formed defecation took place via a fistula to one of the vaginas. Posture and behaviour of the pig were normal. Cytogenetic analysis of blood lymphocytes revealed no numerical or gross structural anomalies. There have been no further piglets with developmental disorders in the same litter, in a second litter of the same parents and in other twelve litters by the same boar.

Sow longevity influences farm economy and can be considered an important indicator of animal welfare. Body features such as leg conformation can play a key role in sow longevity, although little is known about its effect on culling decisions. Within this context, longevity data from 587 Duroc, 239 Landrace, and 217 Large White sows were analyzed with special emphasis on the effect of leg conformation. Sow longevity was analyzed twice for each breed, testing the effect of a subjective overall score for leg conformation, or the presence or absence of 6 specific leg conformation defects. Each preliminary model also included a teat conformation score with 3 levels, farm or origin, backfat thickness at 6 mo of age, and 2 continuous sources of variation, namely the age at the first farrowing and the number of piglets born alive at each farrowing. Overall leg conformation score influenced (P < 0.01) sow longevity in Duroc, Landrace, and Large White sows, with a greater hazard ratio (HR) for poorly conformed sows (1.56, 2.16, and 1.79, respectively) than for well-conformed sows (0.32, 0.66, and 0.68, respectively). Abnormal hoof growth reduced survivability in Duroc (HR = 2.78; P < 0.001) and Landrace sows (HR = 1.88; P < 0.01); the presence of splayed feet (P < 0.05) or bumps and injuries (P < 0.001) increased the risk of culling in Duroc sows (HR = 2.08 and 3.57, respectively), whereas the incidence of straight pastern increased the HR in Large White sows (HR = 2.49; P < 0.01). In all 3 breeds, longevity decreased for plantigrade sows, with a greater HR in Duroc (HR = 3.38; P < 0.001) than in Landrace (HR = 1.53; P < 0.10) and Large White sows (HR = 1.73; P < 0.05). Teat conformation did not influence sow longevity (P > 0.10). Estimates of heritability for longevity in Duroc sows ranged from 0.05 to 0.07 depending on the algorithm applied. Leg conformation had a substantial effect on sow longevity, where an accurate removal of poorly leg-conformed candidate gilts before first mating could improve sow survival and reduce culling costs. These moderate estimates of heritability indicated that survivability of Duroc sows could be genetically improved by direct selection for leg conformation.
