Non-steroidal anti-inflammatory drugs (NSAIDs) represent one of the most commonly used classes of drugs in both human and veterinary medicine. However, many clinical side effects have been observed, especially when treatment has been prolonged. While the anti-inflammatory efficacy and safety of repeated administration of firocoxib (Previcox®), which is a selective NSAID COX-2 inhibitor, has been evaluated for short-term use (one to fourteen days), its clinical relevance for longer-term use is not known. As a preliminary study, healthy, adult male and female horses (n = 7) were treated with firocoxib for 40 days concomitant with the collection of blood samples encompassing treatment to assess hematological and biochemical endpoints. Daily oral administration of firocoxib was performed with one 57 mg tablet/animal (0.11-0.14 mg/kg), which was crushed and mixed with feed. Blood samples were collected one day before treatment (D0 or basal sample), during (D10, D20, D30, and D40), and after treatment (D55 and D70). Results indicated some hematological and biochemical effects were significantly reduced (p < 0.05) towards the end of treatment on D40 relative to pre-treatment or baseline values on D0. Post-treatment, all values returned to pre-treatment values within 30 days without any apparent clinical adversities. In conclusion, while these preliminary results are favorable for prolonged use of firocoxib in horses, future studies are required to evaluate the efficacy of prolonged use accompanied with other clinically relevant endpoints in healthy as well as injured or diseased animals.

Ozone therapy acts in the body inducing controlled oxidative stress, thereby improving the antioxidant, immune and circulatory responses. However, very little is known about how this therapy affects oxidative stress indicators in dogs. We aimed to assess the clinical, hematological, biochemical and oxidative stress parameters of healthy dogs subjected to ozone therapy and oxygen therapy by rectal insufflation. Ten healthy dogs were allocated into three experimental groups in a cross-over design: control, without intervention; ozone, which received 100 µg of O3/kg through rectal insufflation; and oxygen, which received an ozone-equivalent volume of medicinal O2 through rectal insufflation. Dogs received four applications weekly and were followed up until the seventh week. Ozone therapy significantly increased the weight, mean corpuscular volume and mean platelet volume and decreased total cholesterol of treated dogs. Regarding oxidative stress, ozone therapy reduced total antioxidant capacity by ferric reduction (TAC-FRAP) in D7 compared with baseline and the control, significantly increased total antioxidant capacity by cupric reduction (TAC-CUPRAC) in D42 and D49 compared with the control group, caused an increase in uric acid compared with the oxygen group and decreased lipid peroxidation on D21 compared with the control group. In conclusion, ozone therapy through rectal insufflation causes transient oxidative stress followed by an antioxidant response and discreetly interferes with a few clinical, hematological and biochemical variables in healthy dogs, although variables still remained within the reference ranges for the species, thus proving the safety of the therapy. Furthermore, oxygen therapy causes oxidative stress without inducing a subsequent antioxidant response.

Obesity, an extremely important factor in feline clinical practice, is estimated to affect up to one third of the feline population. Moreover, it can trigger chronic inflammation, which could predispose to oxidative stress by increasing reactive oxygen species, thereby generating potentially irreversible cellular damage. This study analyzed hematological, biochemical and oxidative stress profiles at various degrees of feline obesity. Forty-five cats were selected and divided into three groups: control (n = 17), overweight (n = 13) and obese (n = 15), after clinical and laboratory evaluation and body condition score. Biochemical and oxidative stress analyses were performed using a photocolorimeter and hematological analyses were performed in a veterinary cell counter. Obese cats showed increased mean corpuscular volume (MCV), red cell distribution width (RDW), HDL cholesterol and triglycerides and decreased activity of gamma-glutamyl transferase (GGT) than control cats, although within the reference ranges for the species. As for oxidative stress, obese cats showed higher total antioxidant capacity (TAC), by the inhibition of 2,2'-Azino-Bis-3-Ethylbenzthiazoline-6-Sulfonic Acid (ABTS), inhibition of ABTS associated with horseradish peroxidase (ABTS + HRP), cupric ion reducing antioxidant capacity (CUPRAC) and ferric reducing antioxidant power (FRAP) methods, while overweight cats had a higher TAC-ABTS + HRP and TAC-FRAP than control cats. We conclude that the conditions of natural obesity and overweight in the feline species alter its hematological, biochemical and oxidative stress parameters.

Neosporosis, an infectious disease caused by the protozoan Neospora caninum, has been associated with economic losses in cattle rearing worldwide. However, previous studies have not presented any evidence regarding the association between serological status of neosporosis and alteration of the reproductive parameters. Thus, this study aimed to determine whether N. caninum is associated with reproductive disorders and to evaluate the possible risk factors of the infection. Blood samples from 202 dairy cows, 51 with a history of reproductive disorders (case group) and 151 without (control group), were collected from different farms in Brazil. Epidemiological questionnaires were conducted with all the farmers. Serum samples were subjected to an indirect fluorescent antibody test to detect antibodies against the parasite. In total, 28.22% (57/202) of the cows were seropositive: 47.06% (24/51) from the case group and 21.85% (33/151) from the control group. By logistic regression, cows aged ≥48 months and cows with history of abortion were 4.8 (95% confidence interval [CI] = 1.91-12.05; p = 0.001) and 2.3 (95% CI = 1.06-5.1; p = 0.034) times more likely to be seropositive, respectively. Furthermore, our results show an association between N. caninum seropositivity and abortion in dairy cows from Brazil with poor management conditions and N. caninum seropositivity risk factors for reproductive disorders.

Propofol is a widely used drug in veterinary medicine to induce anesthesia; as well as the chosen compound for protocols of intravenous anesthesia. The present study aimed to describe the hematological, biochemical and oxidative stress alterations in calves kept under anesthesia by propofol in different dosages. In order to achieve this, eight Holstein calves were induced using propofol in a 5 mg/kg dosage and maintained under continuous propofol infusion for 60 min, having being administered 0.6 mg/kg/h or 0.8 mg/kg/h in crossover design with seven days interval. Blood samples were collected immediately before the anesthesia induction (baseline), and 30 min, 1, 2, 3, 4 and 5 h after the procedure started. Statistically relevant propofol influence was observed both in blood and biochemical parameters, with differences between dosages according to the time of infusion. The drug action over oxidative stress was also observed, causing a raise of the total antioxidant capacity (TAC) with an uric acid increase. Additionally, the increase of triglycerides, induced by the anesthesia maintenance with propofol, caused lipemia in the samples, which was capable of interfering directly in the measurements made by refractometry and spectrophotometry. It was concluded that, in spite of propofol induced alterations in blood and biochemical parameters, such alterations are subtle. In addition to that, the drug presented an antioxidative effect, which reinstates the safety of anesthesia maintenance with propofol in calves.

Metabolic syndrome, or metabolic dysfunction related to obesity in dogs, is a set of factors that may predispose comorbidities secondary to obesity. Adjuvant therapy with an energy-restricted diet, especially with low levels of carbohydrate and fat, is essential for weight loss, in addition to controlling snacks intake. The aim of this study was to evaluate the biochemical profile of obese dogs compared to lean dogs, and also to compare these profiles before and after a 30-day treatment, thus evaluating the possibility of obesity-related metabolic dysfunction and the action of adjuvant dietary therapy in this condition. Cholesterol and its fractions (HDL, LDL and VLDL), triglycerides, systemic blood pressure and glycemia of obese and lean dogs were measured, and seven obese dogs were treated only with a low-calorie diet with low levels of fat and carbohydrates for 30 days; these patients were evaluated before and after treatment. Obese dogs showed higher levels of triglycerides than lean dogs, and dogs treated with low-calorie diet presented weight loss and better outcomes related to biochemical profile, especially triglycerides levels, after treatment.


A síndrome metabólica, ou disfunção metabólica relacionada à obesidade em cães, é um conjunto de fatores que podem predispor à obesidade. A terapia adjuvante com dieta com restrição calórica, especialmente com baixos níveis de carboidratos e gordura, é essencial para a perda de peso, além do controle da ingestão de petiscos. O objetivo do presente trabalho foi avaliar o perfil bioquímicos de cães obesos em comparação aos de escore corporal normal e avaliar esses perfis em um grupo de animais obesos antes e após restrição calórica de 30 dias, verificando a ocorrência da disfunção metabólica relacionada à obesidade e o papel da dieta como tratamento adjuvante desta condição. Colesterol e suas frações (HDL, LDL e VLDL), triglicérides, pressão arterial sistêmica e glicemia de cães obesos e em escore corporal normal foram mensurados, e sete cães obesos foram tratados somente com uma dieta de baixa caloria com baixos níveis de gordura e carboidratos durante 30 dias; estes pacientes foram avaliados antes e após o tratamento. Cães obesos demonstraram valores séricos de triglicérides maiores quando comparados aos cães com escore corporal normal, e cães tratados com a dieta hipocalórica obtiveram perda de peso significativa e melhores resultados relacionados ao perfil bioquímico, especialmente dos níveis de triglicérides após o tratamento.

Corticosteroid therapy has been used for ruminants to allow lung maturation and the birth of premature babies. However, when considering laboratory analyses of these animals, very little data is available regarding hematological and biochemical patterns, especially for premature goats, and the effects of corticotherapy on these parameters are unknown. In this context, the objective of this study was to evaluate the hematological and biochemical parameters during the first hours of life of premature kids from goats subjected to different dexamethasone protocols. For this, the goats were divided into four groups: group I, goats that received 20 mg of dexamethasone at 139 days of gestation; group II, 2 mg of dexamethasone from the 133rd to 136th day of gestation, 4 mg from the 137th to 139th, and 20 mg on the 140th; group III, 16 mg of dexamethasone from the 139th day, with repeated doses every 12 h until elective surgery; and group IV, goats that received 4, 8, 16, and 20 mg of dexamethasone at 137, 138, 139, and 140 days of gestation, respectively. Blood samples were obtained at birth (T0h) and after 1 (T1h), 12 (T12h), 24 (T24h), and 48 h (T48h) of life for hemogram and serum biochemistry assessment of urea, creatinine, total protein (PT), and gamma-glutamyltransferase (GGT). PT levels and GGT activity were lower at birth in all groups and rose after colostrum ingestion. The creatinine values for all the experimental groups did not differ between T0h and T1h; however, they decreased in the subsequent moments. Except for group I, urea concentrations were higher at T48h than at T1h. The red blood cell, hemoglobin, hematocrit, and mean corpuscular hemoglobin counts decreased over time. The total leukocyte count behaved differently in different experimental groups, and was influenced by the levels of dexamethasone, mainly due to the change in the counts of segmented neutrophils and lymphocytes. It was concluded that significant changes in the hematological and biochemical parameters occur in the first hours of life of premature kids, and that the treatment of goats with dexamethasone can affect these parameters in a dose-dependent manner.


A corticoterapia tem sido empregada em ruminantes com o objetivo de permitir a maturação pulmonar e o nascimento de filhotes prematuros. Entretanto, ao se considerar análises laboratoriais desses animais, pouquíssimos dados estão disponíveis quanto aos padrões hematológicos e bioquímicos, especialmente em caprinos prematuros, e tampouco se sabe sobre os efeitos da corticoterapia sobre parâmetros hematológicos e bioquímicos nesses animais. Nesse contexto, objetivou-se avaliar os parâmetros hematológicos e bioquímicos das primeiras horas de vida de cabritos prematuros provenientes de cabras que utilizaram diferentes protocolos de dexametasona. Para tal, as cabras foram divididas em três grupos: grupo I, cabras que receberam 20 mg de dexametasona aos 139 dias de gestação; grupo II, 2 mg de dexametasona do 133° ao 136° dia de gestação, 4 mg do 137° ao 139° e 20 mg no 140° dia; grupo III, 16 mg de dexametasona a partir do 139° dia, com doses repetidas a cada 12 horas até a cirurgia eletiva; e grupo IV, cabras que receberam 4, 8, 16 e 20 mg de dexametasona no 137°, 138°, 139° e 140° dias de gestação, respectivamente. As amostras sanguíneas dos cabritos foram obtidas ao nascimento (T0h), 1 (T1h), 12 (T12h), 24 (T24h) e 48 horas (T48h) de vida para avaliação do hemograma e bioquímica sérica de ureia, creatinina, proteína total (PT) e gamaglutamiltransferase (GGT). Os teores de PT e atividade de GGT foram menores ao nascimento em todos os grupos, elevando-se após ingestão do colostro. Os valores de creatinina em todos os grupos experimentais não diferiram entre T0h e T1H, entretanto, diminuíram nos momentos subsequentes. Com exceção do grupo I, as concentrações de ureia foram maiores no T48h em comparação com o T1h. Os valores de hemácias, hemoglobina, hematócrito e volume corpuscular médio diminuíram ao longo do tempo, enquanto a concentração de hemoglobina corpuscular média aumentou ao longo do tempo. A contagem leucocitária total se comportou de maneira distinta nos diferentes grupos experimentais, demonstrado ser influenciada pelos teores de dexametasona principalmente em decorrência da alteração nas contagens de neutrófilos segmentados e linfócitos. Conclui-se que alterações significativas dos parâmetros hematológicos e bioquímicos ocorrem nas primeiras horas de vida de cabritos prematuros e o tratamento de cabras com dexametasona também pode afetar tais parâmetros de forma dependente da dose.

Visceral leishmaniasis (VL) is a neglected and endemic zoonosis that occurs throughout Brazil; nevertheless, few studies have focused on the early detection of the disease. The municipality of Ourinhos is a non-receptive, silent and vulnerable area for VL, where the seroprevalence of this disease has so far not been investigated. The present study aimed to determine the seroprevalence of canine VL in Ourinhos-SP, and to identify the presence of risk factors. Blood samples were obtained from 604 dogs during a rabies vaccination campaign together with application of a socioeconomic questionnaire, environmental and animal characteristics and tutor's knowledge about the disease. The samples were subjected to indirect ELISA and new samples were collected from reactive and suspect animals, including whole blood and lymph node aspiration evaluated by parasitological method, complete blood count and PCR. No animal was diagnosed as positive based on the combination of direct and indirect tests and the tutors' answers indicated little knowledge about leishmaniasis, being often confused with other diseases transmitted by arthropods; hence, according to the proposed methods, the presence of canine leishmaniasis in the city of Ourinhos was not confirmed and health education campaigns about the disease should be carried out.

Oxidative stress (OS) has been strongly associated with postprandial lipemia (PPL) in humans, and still requires further investigation in dogs. However, since lipemia interferes with spectrophotometric determinations such as those used to assess OS, the present study investigated the effect of PPL on OS parameters of healthy dogs. Twenty dogs had lipemic postprandial samples compared to the average of two non-lipemic moments. Subsequently, PPL was simulated in vitro using a commercial lipid emulsion and twelve pools of non-lipemic serum of these dogs were used to simulate the minimum, median and maximum concentrations of triglycerides obtained during the lipemic state. Serum OS parameters were assessed using the antioxidants uric acid, albumin and total bilirubin; total antioxidant capacity (TAC); total oxidant capacity (TOC); and lipid peroxidation. In vivo PPL caused an increase in albumin, TAC-CUPRAC, TAC-FRAP, uric acid (p < 0.0001), TOC (p = 0.0012) and total bilirubin (p = 0.0245); reduction of TAC-ABTS (p = 0.0008); and did not alter the lipid peroxidation (p = 0.8983). In vitro, levels of albumin increased at the three lipemic concentrations (p < 0.0001), uric acid increased in the median and maximum levels (p < 0.0001), and total bilirubin concentration increased only at the maximum lipemic level (p = 0.0012). All lipemic levels tested increased TAC-ABTS (p = 0.0011) and TAC-FRAP (p < 0.0001). TAC-CUPRAC (p = 0.5002), TOC (p = 0.5938) and lipid peroxidation (p = 0.4235) were not affected by in vitro lipemia. In conclusion, both the in vivo postprandial state and in vitro simulated lipemia affect oxidative stress markers in dogs depending on the oxidative stress marker, and thus the postprandial state and/or lipemic samples should be avoided.

Resumo A leishmaniose visceral (LV) é uma zoonose negligenciada e endêmica presente em todas as regiões do Brasil, mas mesmo assim poucos estudos têm objetivado a detecção inicial da doença. O município de Ourinhos - SP é uma área não receptiva, silenciosa e vulnerável à LV, não havendo até o momento estudos que tenham investigado a soroprevalência no município. Nesse sentido, o presente estudo objetivou determinar a soroprevalência da LV canina em Ourinhos-SP, bem como associar a presença de fatores de risco. Amostras sanguíneas de 604 cães foram obtidas juntamente com a aplicação de questionário socioeconômico, características ambientais e dos animais e conhecimento sobre a doença. As amostras foram submetidas à sorologia por ELISA e novas amostras coletadas de cães reagentes ou suspeitos foram analisadas por método parasitológico direto, hemograma e PCR. Nenhum animal foi considerado positivo na combinação de testes direto e indireto, e as respostas dos tutores indicaram pouco conhecimento sobre a leishmaniose, sendo muitas vezes confundida com outras doenças transmitidas por artrópodes. Dessa forma, de acordo com os métodos propostos, a presença de leishmaniose canina, na cidade de Ourinhos, não foi confirmada. Por isso campanhas de educação em saúde sobre a doença deveriam ser realizadas.

Complete blood count (CBC) is the evaluation of blood cells, which provides resourceful information.Considering that inadequate fasting time is the most common pre-analytical error in laboratory diagnosisand is associated with lipemia in blood samples, which has not yet been adequately investigated in dogs,this study aimed to assess diurnal postprandial changes in the CBC of healthy dogs fed with industrializedfeed. Eighteen clinically healthy dogs aged 2-6 years and weighing over 6 kg were enrolled in the study.All dogs received “Premium” industrialized feed every 12 hours. Blood was collected after a 12-hourfasting period at 6:00 am (baseline), followed by feeding and new blood samples collected hourly forthe next 11 consecutive hours. Red blood cell (RBC) counts, red cell distribution width (RDW), whiteblood cell (WBC) counts, platelets, mean platelet volume (MPV) and hemoglobin were obtained usingan automated veterinary cell counter. Hematocrit (HTC) was determined by Strumia’s microcapillarymethod, differential leukocyte count was performed on hematological dye-stained blood smears and total plasma protein (TPP) was determined using refractometry. Variables were tested for normality anddifferences were considered significant when p < 0.05. A statistically significant decrease was observedin the erythrogram from 2 h for RBC and hemoglobin, from 3 h for HTC and from 4 h for MCV, persistinguntil the end of 11 hours. There was no change in MCHC and RDW. Regarding the leukogram, asignificant increase in WBC was observed from 2 to 7 h, due to the increase in segmented neutrophils 2to 8 h following feeding. Neospora caninum is considered as one of the main causes of reproductive failure in cattle. Vertical transmission is the main route of infection in the bovine host and plays an important role in maintaining the parasite in the herd. Molecular detection of N. caninum is important to determine the occurrence of the disease and to evaluate the genetic diversity of the parasite. The present study aimed at assessing the vertical transmission of N. caninum using molecular techniques to detect the parasite in tissue samples from bovine fetuses collected in a slaughterhouse in the state of São Paulo, Brazil. Seventy fetuses and 70 blood samples from pregnant cows were collected in a slaughtering line. Fresh samples of heart and brain tissue from fetuses were analyzed using molecular assays. Serum samples from fetuses and cows were subjected to an indirect fluorescent antibody test (IFAT) to detect antibodies against N. caninum. Nested PCR targeting the internal transcriber 1 (ITS1) region of the protozoan organism was used in the molecular testing. From the total of fetuses examined, 71.42% were positive for N. caninum by PCR. A higher number of heart samples (47.1%) were positive for the parasite using this technique. Antibodies against the protozoa were detected in 12.9% of serum samples of cows; 2.8% of fetuses were seropositive for this pathogen. Our results show that vertical transmission of N. caninum occurs in cattle from this region of Brazil, and that the use of different diagnostic techniques contributes to successful diagnosis of congenital transmission of the parasite in cattle.

The mechanisms responsible for the imbalance between oxidants and antioxidants in sheep infected with Haemonchus contortus are not well established. This study aimed to prove the hypothesis that oxidative stress occurring during infection by H. contortus varies according to breed, and that the parasite burden correlates with hypoalbuminaemia and anaemia. Thus, after deworming and confirming the absence of infection, two different sheep breeds, Suffolk (n = 15) and Santa Ines (n = 22), were orally inoculated with a single dose of 5,000 L3 of H. contortus. The egg counts per gram of faeces (EPG), packed cell volume (PCV) and concentrations of several plasma markers of oxidative stress (lipid peroxidation, albumin, uric acid, total bilirubin, total antioxidant capacity [TAC], total oxidant concentration [TOC] and the oxidative stress index [OSI]) were quantified before (control group) and during the experimental infection (28, 34 and 42 days post-inoculation). In both breeds, TOC increased at 28 days and TAC increased at 42 days. In Suffolk sheep, there was a positive correlation of EPG with oxidant components (28 days) and a negative correlation of EPG with PCV (42 days). In Santa Ines sheep, there was a positive correlation of EPG with bilirubin (r = 0.492; p = 0.020). H. contortus infection caused oxidative stress, which varied according to the breed. Parasite burden was not associated with hypoalbuminaemia, whereas there was a negative correlation with PCV. This research provides the first evidence that the antioxidant status contributes more to the resilience to H. contortus in Santa Ines sheep compared to Suffolk sheep.

Dogs infected with Leishmania infantum have a reduced number of T lymphocytes. PD-1 (Programmed cell death 1) a new member of the B7-CD28 family that is expressed by immune cells, and its binding to PD-L1 (CD274) or PD-L2 (CD273) induces the deactivation or apoptosis of T cells. This study aimed to evaluate the expression of PD-1 and its ligands, as well as blocking in the induction of apoptosis in T lymphocytes, TNF-α, IL-4 and nitric oxide production by leucokocytes from PBMC and spleen and the parasite load in dogs with visceral leishmaniasis (VL). Our results showed that the expression of PD1 and its ligands was increased in CD3(+) T cells and CD21(+) B lymphocytes within the peripheral blood and splenic mononuclear cells of dogs with VL. In peripheral blood monocytes, only PD-1 ligands exhibited increased expression; however, in spleen macrophages, increased expression of both PD-1 and its ligands was observed. Levels of apoptosis in peripheral blood and splenic T lymphocytes were higher in dogs with VL compared to healthy dogs. Blocking monoclonal antibodies to PD-1 and its ligands in the culture of mononuclear cells from the peripheral blood and spleen decreased the amount of CD3(+) T lymphocyte apoptosis. The concentration of nitric oxide, TNF-α and IL-4 increased in the culture supernatants of peripheral blood mononuclear cells treated with a blocking monoclonal antibody against PD-1. The TNF-α concentration increased in the culture supernatants of splenic cells following all treatments with antibodies blocking PD-1 and its ligands; however, the amount of IL-4 increased only in the presence of a PD-1 blocking agent. Treatment with a PD-1 blocking monoclonal antibody in the mononuclear peripheral blood of dogs with VL reduced the parasite burden while increased TNF-α. We conclude that in canine visceral leishmaniasis, PD-1 and its ligands are involved in the induction of T lymphocyte apoptosis and in regulating the production of nitric oxide, TNF-α, and IL-4, as well as the parasitic load.

Prevalence of hypertension was 28.8%, comprising 22.2% of the dogs from G1 and 58.3% from G2 (P=0.011). The mean arterial blood pressure (BP) of dogs from G1 (135.7 ± 20.5) was lower than from G2 (170.0 ± 26.3) (P <0.001). Urine protein-creatinine ratio (UP/C) revealed values above 0.5 in 75.7% of the dogs, with 34% presenting hypertension. All dogs with hypertension had histopathological and laboratory evidence of glomerular disease. Although there was no statistically significant correlation between elevated BP and the severity of glomerular lesions (P=0.408), there was a statistically significant correlation between elevated BP and increased UP/C in the studied population (P=0.002). Thus, dogs with leishmaniosis and renal disease must be screened for the presence of hypertension so that treatment may be instituted as early as possible, in countries where treatment is allowed, to prevent the progression of renal damage.

O grupo 1 (G1), composto por 54 cães em estágio 1 de doença renal crônica (DRC), e o grupo 2 (G2) por 12 cães em estágios 2 e 3 de DRC. A prevalência de hipertensão foi de 28,8%, compreendendo 22,2% dos cães de G1 e 58,3% dos cães de G2 (p = 0,011). A pressão arterial média (PA) de G1 (135,7 ± 20,5) foi inferior a de G2 (170,0 ± 26,3) (P <0,001). A relação proteína creatinina urinária (P/C U) foi maior que 0,5 em 75,7% dos cães, dos quais 34% possuíam hipertensão. Todos os cães com hipertensão apresentavam doença glomerular. Embora não tenha sido observada correlação estatisticamente significativa entre elevação da PA e severidade das lesões glomerulares (P =0,408), houve uma correlação significativa entre PA elevada e aumento da UP/C (P = 0,002). Portanto, cães com leishmaniose e doença renal devem ser pesquisados ​​quanto à presença de hipertensão, para que o tratamento possa ser instituído o mais precocemente possível em países onde ele é permitido, para evitar a progressão da lesão renal.

Among the uremic toxins proven to affect the neutrophil function in humans with chronic kidney disease(CKD), guanidine compounds stand out. To achieve a clearer understanding of the mechanisms thataffect the immunity of uremic patients, the hypothesis that guanidine acetic acid (GAA) contributesto the inhibition of oxidative metabolism and an increase in neutrophil apoptosis in healthy dogswas investigated in vitro. To this end, neutrophils isolated from ten healthy dogs were incubated inpure RPMI 1640 (control) and enriched with 5 mg/L of GAA. Capillary flow cytometry was usedto quantify superoxide production in neutrophils with the probe (hydroethidine), in the presenceand absence of phorbol-12-myristate-13-acetate (PMA), in order to assess oxidative metabolism.Apoptotic indices were quantified using the Annexin V-PE system, with and without the inductiveeffect of camptothecin. Neutrophils isolated and incubated in a GAA-enriched medium producedsmaller amounts of superoxide (p 0.001) when activated with PMA, however, this inhibition ofoxidative metabolism occurred without significantly altering their viability or rate of apoptosis. Thus, the results show guanidine compounds contribute to immunosuppression in dogs with CKD.

A fim de melhor entenderos mecanismos que afetam a imunidade de pacientes urêmicos, no presente estudo foi investigadain vitro a hipótese de que o composto guanidínico ácido guanidinicoacético (AGA) contribui parainibição do metabolismo oxidativo, aumentando a apoptose dos neutrófilos de cães saudáveis. Paratal, neutrófilos isolados de dez cães saudáveis foram incubados em meio de cultura RPMI 1640puro (controle) e enriquecido com 5 mg/L de AGA. Utilizando-se citometria de fluxo capilar paraa avaliação do metabolismo oxidativo, quantificou-se a produção de superóxido dos neutrófilosempregando-se a sonda hidroetidina, com e sem a presença do estímulo com acetato miristato deforbol (PMA). O índice apoptótico foi quantificado utilizando-se o sistema Anexina V-PE, com e semo efeito indutor da camptotecina. Os neutrófilos isolados e incubados em meio enriquecido com AGA,quando ativados com PMA, produziram uma menor quantidade de superóxido (p 0,001), porém talinibição do metabolismo oxidativo ocorreu sem alterar significativamente a viabilidade e a taxa deapoptose. Assim, os resultados evidenciam que os compostos guanidínicos podem contribuir paraimunossupressão de cães com DRC.
