RESUMEN

BACKGROUND: Although maternal childhood maltreatment has been associated with offspring externalizing symptoms, little is known about the potential mechanisms that contribute to breaking the intergenerational effect of maternal childhood maltreatment. OBJECTIVE: The current study aimed to (a) investigate the intergenerational effect between maternal childhood maltreatment and offspring externalizing symptoms in the Chinese family; (b) examine maternal supportive and harsh parenting as potential mediators of this intergenerational effect; and (c) explore the moderating roles of paternal support parenting, as well as paternal harsh parenting, in this mediation process of maternal supportive and harsh parenting. PARTICIPANTS AND SETTING: The sample consisted of 1111 mother-father-child triads from Beijing, recruited when the children were one and three years old. METHODS: Mothers completed the Childhood Trauma Questionnaire, and both parents completed the Infant-Toddler Social and Emotional Assessment and Comprehensive Early Childhood Parenting Scale. RESULTS: Our results showed that maternal childhood maltreatment was a risk factor for offspring externalizing symptoms at T2 (ß = 0.24, t = 6.51, p < .001), and this effect was mediated by maternal supportive (indirect effect = 0.03, 95%CI = [0.02, 0.05]) and harsh parenting (indirect effect = 0.03, 95%CI = [0.02, 0.07]) at T1. Furthermore, paternal harsh parenting moderated the indirect effect of maternal childhood maltreatment on child externalizing symptoms through maternal supportive parenting. CONCLUSIONS: These findings contribute to our understanding and provide valuable information for disrupting the intergenerational effect of maternal childhood maltreatment.

This study examined the effects of infant negative affectivity (NA) and maternal and paternal depressive symptoms on fathers' and mothers' perceptions of coparenting across the first 2 years following an infant's birth. A total of 147 two-parent families (most couples were White, married, and living together) with healthy, full-term infants were recruited. At each time point, fathers and mothers separately reported their coparenting perceptions via the Coparenting Relationship Scale and their depressive symptoms using the depression subscale of Symptom Checklist-90-Revised. Mothers also reported their children's NA via the Infant Behavior Questionnaire-Revised at 3 to 12 months and the Early Child Behavior Questionnaire at 18 and 24 months. Findings from growth curve models in an actor-partner interdependence model framework suggested that among parents with higher depression, there were steeper declines in coparenting quality reported by parents and their spouses across 3-24 months. In addition, three separate two-way interactions between variables including higher-than-usual parental and spousal depression, as well as higher-than-usual infant NA predicted poorer-than-usual coparenting experiences. Findings indicate that coparenting is a dynamically unfolding construct that is impacted by ongoing changes in the parents' social-ecological niche and suggest the need to consider both parent and child characteristics, and to include spousal influences, to get a comprehensive, whole-family understanding of levels and changes in coparenting relationships. The findings also confirm that coparenting dynamics may benefit from interventions engaging both couples and addressing multiple risk factors from both parents (e.g., depression) and children (e.g., NA).

DNA damage is a modification in the structure of DNA under the influence of endogenous or exogenous factors. DNA damage can cause different types of diseases and is closely related to genetic mutations, cancer, and aging. The cause of the corresponding reaction process is essential for the study of related cancers and other genetically related diseases. Therefore, it is essential to gain a deeper understanding of the various types of DNA damage. This paper provides a comprehensive review of recent advances in the types of DNA damage and associated reaction processes, including damage to DNA bases, nucleotides, and strands, as well as the biological implications of the damage.

This study examined the effects of hypoxemia caused by acute high-altitude hypoxia (AHAH) exposure on the human intestinal flora and its metabolites. The changes in the intestinal flora, metabolism, and erythropoietin content in the AHAH population under altitude hypoxia conditions were comprehensively analyzed using 16S rRNA sequencing, metabonomics, and erythropoietin content. The results showed that compared with those in the control group (C group), the flora and metabolites in the hypoxemia group (D group) were altered. We found alterations in the flora according to the metabolic marker tyrosine through random forest and ROC analyses. Fecal and serum metabonomics analyses revealed that microbial metabolites could be absorbed into the blood and participate in human metabolism. Finally, a significant correlation between tyrosine and erythropoietin (EPO) content was found, which shows that human intestinal flora and its metabolites can help to confront altitude stress by regulating EPO levels. Our findings provide new insights into the adaptive mechanism and prevention of AHAH.

Background: Changes in rainfall induced by climate change will likely influence the utilization of water resources and affect the nutrient cycle in plants in the water-limited desert steppe. In order to understand the response of nitrogen and phosphorus resorption characteristics of plant leaves to precipitation changes, this study compared the nitrogen (N) resorption efficiency, phosphorus (P) resorption efficiency and influencing factors of plants in a desert steppe through water treatment experiments. Methods: A 4-year field experiment was performed to examine the response and influencing factors of nitrogen (N) and phosphorus resorption efficiency of five dominant plants in Stipa breviflora desert steppe to simulated precipitation change in Inner Mongolia, with four simulated precipitation gradients including reducing water by 50%, natural precipitation, increasing water by 50%, increasing water by 100%. Results: Compared with natural precipitation, increasing water by 100% significantly increased soil moisture, and significantly increased the aboveground biomass of S. breviflora, C. songorica, A. frigida, decreased the N concentrations in green leaves of S. breviflora, Cleistogenes songorica, Artemisia frigida, Kochia prostrata, decreased the N concentrations in senesced leaves of C. songorica, decreased the P concentrations in green leaves of K. prostrata and Convolvulus ammannii, decreased the NRE of S. breviflora. NRE was significantly negatively correlated with N concentration in senesced leaves, and PRE was significantly negatively correlated with P concentration in senesced leaves. Conclusions: Increasing water indirectly reduces NRE by reducing plant leaf green leaves nitrogen concentration, and decreasing water indirectly reduces PRE by reducing soil moisture.

Methods The model of heart failure after myocardial infarction was established by left coronary artery liga-tion in rats. Two weeks after modeling, all rats were randomly divided into model group, LGZGD group, and captopril group. Meanwhile sham operation group was set up. The rats were given continuous intragastric administration with drug or distilled water for 28 days, once a day. The behavioral signs of rats in each group were observed. The cardiac function of rats in each group was examined by echocardiography. Serum BNP and NT-ProBNP content were detected by enzyme-linked immunoassay; The changes of myocardial his-topathological and collagen fibers in rats were detected using sirius staining. The contents of oxidative stress index including ROS, SOD in myocardial tissue of rats in each group were observed by DCFH-DA fluorescent probe and Enzyme-linked immunoassay. The ultra-structure of mitochondria was observed by transmission electron microscopy. Expressions of apoptotic proteins ( mitochondrial CytC, cytoplasmic CytC) were detec- ted by Western blot. Expression of proteins related to the Nrf2/BNIP3 pathway were examined by immunoflu-orescence and Western blot. Results LGZGD could significantly improve the cardiac function of rats, reduce the contents of BNP and NT-ProBNP, inhibit the excessive deposition of collagen in myocardial interstiti-um, reduce ROS, increase the content of SOD, improve mitochondrial structure damage, up-regulate the expression of Nrf2 and nuclear translocation, and reduce the expression of BNIP3. Conclusions LGZGD can inhibit the ventricular remodeling and prevent the occurrence of heart failure after myocardial infarction. Its pharmacological effects are mainly related to regulating the Nrf2/BNIP3 pathway, activating Nrf2, promoting its nuclear transfer, and further down-regulating BNIP3 , protecting mitochondrial function, and reducing cardiomyocyte apoptosis.

The intestinal microbial community is the largest ecosystem in the human body, in which the intestinal flora plays a dominant role and has a wide range of biological functions. However, it is vulnerable to a variety of factors, and exposure to extreme environments at high altitudes, as seen on the Qinghai-Tibet plateau, may cause changes in the structure and function of the host intestinal flora. Conversely, the intestinal flora can help the host adapt to the plateau environment through a variety of ways. Herein, we review the relationship and underlying mechanism between the host intestinal flora and the plateau environment by discussing the characteristics of the plateau environment, its influence on the intestinal flora, and the important role of the intestinal flora in host adaptation to the plateau environment. This review aimed to provide a reference for maintaining the health of the plateau population.

The present study examined mothers' emotional availability (EA) during daytime free play and bedtime as a mediator of linkages between maternal nighttime sleep and infant-mother attachment. Participants included 153 mothers (85% White) with infants (53% female). When infants were 1, 3, 6, 9, and 12 months, maternal sleep was assessed using actigraphy and daily sleep diaries for 7 consecutive days. At each time point, mothers' EA was scored from one observation of daytime free play and from one evening observation of infant bedtime by trained observers who were blind to all other participant information. Average scores were created for maternal sleep and EA across the five occasions in the first year. At 12 and 18 months, infant-mother attachment security in the home was scored by blind observers using the Attachment Q-Set, averaged across the two age points, and used in analyses. Mediational analyses revealed that mothers who experienced highly variable sleep and had poor sleep quality were less emotionally available with infants at bedtime during infants' first year of life, which in turn was predictive of lower infant-mother attachment security in the second year, supporting mediation. Linkages between maternal sleep characteristics and daytime EA were less evident. Later maternal sleep timing was also directly predictive of low attachment security, after accounting for maternal EA. Findings emphasize that poor parental sleep places both parenting and infant socioemotional development at risk, and that parental sleep hygiene and sleep habits should be a salient focus of parenting intervention. The second year of life is a time of formative developmental change as basic behavioral systems undergo rapid integration and expansion. This study examined the developmental trajectories of social-emotional (SoE) outcomes and the effects of infant sex and household chaos (HC) on the development of SoE outcomes across the second year of life. The participating families (N = 143) were ethnically homogenous (88% Caucasian) but economically diverse (31% low-to-very-low income). Mothers reported on their children's SoE outcomes including externalizing, internalizing, dysregulating problem behaviors as well as SoE competence when infants (54% girls) were 12, 18, and 24 months old. At each age point, HC was assessed through observations during home visits and compliance to the study protocols. Multilevel modeling revealed increasing developmental trajectories in all of the domains of SoE outcomes across the second year of life, reflecting the premise that these behavioral systems continue to form and become increasingly part of the infant's behavioral repertoire as development unfolds. However, compared to infants in less chaotic homes, infants in more chaotic households experienced steeper increases in both externalizing and internalizing problem behaviors across the second year, and girls showed higher levels of internalizing problem behaviors compared to boys. Results emphasize the increasing trajectories of problem behaviors in relation to ongoing chaotic caregiving environment among infants as young as 2 years of age. This study examined socioeconomic status (SES) and attachment security as predictors of infant nighttime sleep during the second year. Participants included 128 mothers (86% White) with infants (48.4% boys). Data collection took place between April, 2009 and February, 2014. At 12, 18, and 24 months, infant sleep was assessed via actigraphy and daily diaries, and attachment with the Attachment Q-Set. SES indicators included income-to-needs ratios and education. Lower SES predicted greater variability in sleep duration and later sleep timing only for less secure infants. Less secure attachment was associated with poorer sleep at both between- and within-person levels, especially when infants were 12 months of age. Results emphasize the complex interactive effects of environmental and relational factors on infant sleep.

@# Cardiovascular diseases cause significant morbidity and mortality worldwide, incurring a major public health burden. Gastrodia elata Blume is a traditional Chinese herbal medicine that has been widely used to treat central nervous system and cardiovascular diseases. Gastrodin, as the major active component in Gastrodia elata Blume, can confer protection against cardiovascular diseases. In this review, we summarize the anti-inflammatory actions, anti-cardiac hypertrophy, anti-hypertension, anti-atherosclerosis, and angiogenic effects of gastrodin, as well as its protective effects on vascular cells and against myocardial ischemia-reperfusion injury. The medical potential of gastrodin in diabetes-related cardiovascular diseases is also discussed.

OBJECTIVE@#To study the effects of FLT3-ITD length on 32D cell proliferation, apoptosis and sensitivity to FLT3 inhibitor, so as to provide references for stepwise therapy of FLT3-ITD mutated acute myeloid leukemia patients.@*METHODS@#Three different FLT3-ITD mutants with same or adjacent insert sites were selected and constructed in an eukaryotic expression vector. FLT3-ITD mutants stably expressed 32D cell strains were selected with the help of lentivirus system and IL3 free cell culture medium. The proliferation and apoptosis of 32D cell strains after AC220 treatment were detected.@*RESULTS@#FLT3-ITD mutants (ITD1, ITD2 and ITD3) stably expressed 32D cell strains were constructed successfully. In the absence of IL3 factor, the proliferation number of ITD1, ITD2 and ITD3 cell strains were mounted up to 2.3 folds, 3.7 folds, and 4.3 folds after 48 hours, respectively. Under the exposure of FLT3 inhibitor AC220, the IC@*CONCLUSION@#FLT3-ITD mutant expressed cell strains with longer ITD show higher capacity of proliferation and higher tolerance to AC220 treatment.

The present study examined both between- and within-person effects of maternal sleep patterns on quality of mothering at bedtime during infants' first 6 months. Participants included 142 mothers who reported on their daily fall asleep and wake times across seven consecutive days with a daily sleep diary when infants were 1, 3, and 6 months old. At each age point, maternal emotional availability during one night of infant bedtime was observed and scored by trained observers who were blind to maternal sleep patterns. Multilevel modeling revealed that mothers with irregular sleep patterns, especially later average fall asleep times and greater average variability in sleep period across three age points, showed poorer parenting quality with infants at bedtime than other mothers. In addition, both between- and within-person effects of maternal sleep on bedtime parenting quality changed with infant age. Compared to mothers' individual averages across 1, 3, and 6 months, maternal short average sleep period, increased variability in sleep period, and later fall asleep times predicted poorer bedtime parenting quality at 6 months, but not at 1 or 3 months. Results emphasize the importance of maternal sleep regulation and sleep hygiene for maternal parenting quality, especially as infants get older. Using a sample of young adults, the present study investigated how the participants' attachment to romantic partners was related to that with their parents and peers and how this specific attachment experience was associated with their physiological stress response. We examined 121 pairs of young Chinese (N = 242) heterosexual couples (men's age: 22.26 ± 2.40; women's age: 21.62 ± 2.22) and their attachment to parents, peers, and romantic partners as well as their cortisol recovery from romantic conflict. Robust actor-partner interdependence mediation models showed that women's insecure parental and peer attachment was associated with blunted cortisol recovery from romantic stress through their fearful attachment with romantic partners, whereas men's insecure parental and peer attachment was associated with their partners' adaptive cortisol recovery from romantic stress through their fearful romantic attachment. These findings suggested that women's hypothalamic-pituitary-adrenal-axis functioning seemed to be more strongly associated with their own and their partners' attachment relationships compared to men.

Objective:To investigate the difference of isokinetic angle-specific moment curves between anterior cruciate ligament (ACL)-injured patients with and without patellofemoral cartilage injury (PFCI). Methods:A retrospective analysis was performed on patients underwent knee arthroscopy and isokinetic muscle strength testing before surgery from September, 2018 to September, 2019. Seventeen ACL-injured patients with PFCI and 17 ACL-injured patients without PFCI who matched in age, sex and meniscus injury were selected. Before arthroscopy, isometric and isokinetic strength of knee flexion and extension at velocity of 180°/s and 60°/s was tested by isokinetic dynamometer. Normalized torque-angle curves (torque/body mass) were generated in steps of 1° and the differences in angle-specific moment curves between two groups were compared. Results:At 180°/s, there was no significant difference in flexion isokinetic torque both healthy side and affected side between two groups (P >0.05); and no difference in extension torque of the healthy side (P >0.05), however, there was significant difference in extension torque of the affected side at 88° to 90° between two groups (t > 2.102, P <0.05). At 60°/s, there was significant difference in flexion torque of the healthy side at 62° to 82° between two groups (|t| >2.056, P <0.05), and no significant difference was found in flexion torque of the affected side (P >0.05), nor in extension torque of both sides between two groups (P > 0.05). A curve change was found at the beginning of the flexion and extension isokinetic moment curves at the velocity of 180°/s. The isometric knee extension torque was significantly different in the affected side between two groups (t = 2.858, P < 0.01), and no difference was found in isometric knee flexion torque in the affected side as well as both extension and flexion torques in the healthy side between two groups (t < 1.905, P > 0.05). Conclusion:The lower the isokinetic speed, the more significant the difference of strength is between ACL-injury patients with and without PFCI. High speed exercise is recommended for ACL-injured patients with PFCI.

Little is known regarding the dynamic interactions between fathers and mothers in families of children with autism spectrum disorder (ASD) during the parenting process. This study used an actor-partner interdependence mediation (APIMeM) model to investigate the intrapersonal and interpersonal effects of emotion dysregulation and parenting stress on parenting behaviors among 211 pairs (total N = 422) of Chinese parents of children with ASD. The results indicated that for both fathers and mothers, there were significant indirect actor effects of parental emotion dysregulation on parents' own parenting behaviors through their own parenting stress. However, no significant direct or indirect partner effect was found in the analyses. These findings suggest that the emotional parenting dynamics occurred on the individual rather than the dyadic level in these families.

OBJECTIVE@#To detect and analyze the mutation status of FANCJ gene in adult AML patients, so as to provide the basis for studying the mechanism of FANCJ driven AML and guiding the preventim and treatment of deseese.@*METHODS@#The cDNAs were extracted and transeripted from bone marrow cells and normal skin cells in 222 newly diagnosed AML patients. The primers were designed for FANCJ gene coding region, the mutations of FANCJ gene coding region in AML patients as well as the mutations of FANCJ gene in mucous membrane epethelia in patients were detected by PCR and sanger seguencing; the evolutionary conservation of FANCJ mutation in different organisms was analyzed by NCBI Blast online bioinformaties software.@*RESULTS@#The sequencing analysis showed that the mutations of FANCJ gene happened in 11 sites of FANCJ gene coding region, which were as followed: exon5:c.G430A:p.A144T, exon6:c.A587G:pN196S, exon9:c.C1255T:p.R419W, exon10:c.G1442A:p.G481D, exon11:c.C1609G:p.L537V, exon16:c.C2360T:p.P787L, exon17:c.C2440T:p.R814C, exon19:c.C2608T:pH870Y, exon19:c.A2686G:p.I896V, exon19:c.C2830G:p.Q944E, exon20:c.G3412A:p.D1138N. Among them, the repeatability existed in mutations of A144T, N196S, R814C, I896V and Q944E. Beside, the mutation sites of A144, R419, G381, L537, P787, H870, Q944 and D1138 were highly conserved in different organisms.@*CONCLUSION@#Among 222 adult AML patients, the mutations of FANCJ gene have been found in 26 patients, moreover, the mutation sites are relatively conserved in different organisms, and possess important fanction. The results of this study provide the basis for exploring the mexhanism of FANCJ gene driven AML and for guiding the prevantion and treatment of AML.

The aim of this study was to investigate whether uric acid (UA) might exert neuroprotection via activating the nuclear factor erythroid 2-related factor 2 (Nrf2) pathway and regulating neurotrophic factors in the cerebral cortices after transient focal cerebral ischemia/reperfusion (FCI/R) in rats. UA was intravenously injected through the tail vein (16 mg/kg) 30 min after the onset of reperfusion in rats subjected to middle cerebral artery occlusion for 2 h. Neurological deficit score was performed to analyze neurological function at 24 h after reperfusion. Terminal deoxynucleotidyl transferase-mediated dNTP nick end labeling (TUNEL) staining and hematoxylin and eosin (HE) staining were used to detect histological injury of the cerebral cortex. Malondialdehyde (MDA), the carbonyl groups, and 8-hydroxyl-2'-deoxyguanosine (8-OHdG) levels were employed to evaluate oxidative stress. Nrf2 and its downstream antioxidant protein, heme oxygenase- (HO-) 1,were detected by western blot. Nrf2 DNA-binding activity was observed using an ELISA-based measurement. Expressions of BDNF and NGF were analyzed by immunohistochemistry. Our results showed that UA treatment significantly suppressed FCI/R-induced oxidative stress, accompanied by attenuating neuronal damage, which subsequently decreased the infarct volume and neurological deficit. Further, the treatment of UA activated Nrf2 signaling pathway and upregulated BDNF and NGF expression levels. Interestingly, the aforementioned effects of UA were markedly inhibited by administration of brusatol, an inhibitor of Nrf2. Taken together, the antioxidant and neuroprotective effects afforded by UA treatment involved the modulation of Nrf2-mediated oxidative stress and regulation of BDNF and NGF expression levels. Thus, UA treatment could be of interest to prevent FCI/R injury.

Recent studies have shown 5-hydroxymethyl-2-furfural (5-HMF) has favorable biological effects, and its neuroprotection in a variety of neurological diseases has been noted. Our previous study showed that treatment of 5-HMF led to protection against permanent global cerebral ischemia. However, the underlying mechanisms in cerebral ischemic injury are not fully understood. This study was conducted to investigate the neuroprotective effect of 5-HMF and elucidate the nuclear factor erythroid 2-related factor 2 (Nrf2)/antioxidant response element (ARE) signaling pathway mechanism in the striatum after transient global cerebral ischemia. C57BL/6 mice were subjected to bilateral common carotid artery occlusion for 20 min and sacrificed 24 h after reperfusion. 5-HMF (12 mg/kg) or an equal volume of vehicle was intraperitoneally injected 30 min before ischemia and 5 min after the onset of reperfusion. At 24 h after reperfusion, neurological function was evaluated by neurological disability status scale, locomotor activity test and inclined beam walking test. Histological injury of the striatum was observed by cresyl violet staining and terminal deoxynucleotidyl transferase (TdT)-mediated dNTP nick end labeling (TUNEL) staining. Oxidative stress was evaluated by the carbonyl groups introduced into proteins, and malondialdehyde (MDA) levels. An enzyme-linked immunosorbent assay (ELISA)-based measurement was used to detect Nrf2 DNA binding activity. Nrf2 and its downstream ARE pathway protein expression such as heme oxygenase-1, NAD (P)H:quinone oxidoreductase 1, glutamate-cysteine ligase catalytic subunit and glutamate-cysteine ligase modulatory subunit were detected by western blot. Our results showed that 5-HMF treatment significantly ameliorated neurological deficits, reduced brain water content, attenuated striatum neuronal damage, decreased the carbonyl groups and MDA levels, and activated Nrf2/ARE signaling pathway. Taken together, these results demonstrated that 5-HMF exerted significant antioxidant and neuroprotective effects following transient cerebral ischemia, possibly through the activation of the Nrf2/ARE signaling pathway.
