BACKGROUND: Unsupervised online cognitive assessments have demonstrated promise as an efficient and scalable approach for evaluating cognition in aging, and Alzheimer's disease and related dementias. OBJECTIVES: The aim of this study was to evaluate the feasibility, usability, and construct validity of the Paired Associates Learning task from the Cambridge Neuropsychological Test Automated Battery® in adults enrolled in the Brain Health Registry. DESIGN, SETTING, PARTICIPANTS, MEASUREMENTS: The Paired Associates Learning task was administered to Brain Health Registry participants in a remote, unsupervised, online setting. In this cross-sectional analysis, we 1) evaluated construct validity by analyzing associations between Paired Associates Learning performance and additional participant registry data, including demographics, self- and study partner-reported subjective cognitive change (Everyday Cognition scale), self-reported memory concern, and depressive symptom severity (Patient Health Questionnaire-9) using multivariable linear regression models; 2) determined the predictive value of Paired Associates Learning and other registry variables for identifying participants who self-report Mild Cognitive Impairment by employing multivariable binomial logistic regressions and calculating the area under the receiver operator curve; 3) investigated feasibility by looking at task completion rates and statistically comparing characteristics of task completers and non-completers; and 4) evaluated usability in terms of participant requests for support from BHR related to the assessment. RESULTS: In terms of construct validity, in participants who took the Paired Associates Learning for the first time (N=14,528), worse performance was associated with being older, being male, lower educational attainment, higher levels of self- and study partner-reported decline, more self-reported memory concerns, greater depressive symptom severity, and self-report of Mild Cognitive Impairment. Paired Associates Learning performance and Brain Health Registry variables together identified those with self-reported Mild Cognitive Impairment with moderate accuracy (areas under the curve: 0.66-0.68). In terms of feasibility, in a sub-sample of 29,176 participants who had the opportunity to complete Paired Associates Learning for the first time in the registry, 14,417 started the task. 11,647 (80.9% of those who started) completed the task. Compared to those who did not complete the task at their first opportunity, those who completed were older, had more years of education, more likely to self-identify as White, less likely to self-identify as Latino, less likely to have a subjective memory concern, and more likely to report a family history of Alzheimer's disease. In terms of usability, out of 8,395 received requests for support from BHR staff via email, 4.4% (n=374) were related to PAL. Of those, 82% were related to technical difficulties. CONCLUSIONS: Our findings support moderate feasibility, good usability, and construct validity of cross-sectional Paired Associates Learning in an unsupervised online registry, but also highlight the need to make the assessment more inclusive and accessible to individuals from ethnoculturally and socioeconomically diverse communities. A future, improved version could be a scalable, efficient method to assess cognition in many different settings, including clinical trials, observational studies, healthcare, and public health.

BACKGROUND: Failure of Alzheimer's disease and related diseases (ADRD) research studies to include and engage Black participants is a major issue, which limits the impact and generalizability of research findings. Little is known about participation of Black adults in online ADRD-related research registries. OBJECTIVES: As part of the Community Engaged Digital Alzheimer's Research (CEDAR) Study, this study aims to increase our understanding of facilitators and barriers of Black adults to participating in ADRD-related online registries, as well as to understand their preferences for communication channels. DESIGN, SETTING, PARTICIPANTS, MEASUREMENTS: We invited all Black participants enrolled in the Brain Health Registry (BHR) to complete a cross-sectional online survey. The survey consisted of rating scales and open-text questions asking about their attitudes towards brain health research, reasons for joining and continuing to participate in BHR, difficulties with participating, and preferences for modes of contact and website usage. RESULTS: Of all invited Black BHR participants (N=3,636), 198 (5.5%) completed the survey. The mean age was 58.4 (SD=11.3), mean years of education were 16.3 (SD=2.4), and 85.5% identified as female. Reported facilitators for joining and continuing to participate in BHR were personal interest (e.g., learning more about own brain health) and altruism (e.g., helping research). Among additional registry features which could encourage return, receiving feedback or scores about BHR tasks was rated the highest. Of those who found BHR participation difficult (21%), the most frequent reason was time burden. The most preferred way of receiving study information was via email. Participants reported that the websites that they used the most were YouTube and Facebook. DISCUSSION: The results of our study can inform the development of culturally-responsive registry features and engagement efforts to improve inclusion and participation of Black adults in online ADRD research. Providing participants with feedback about their registry performance and reducing the number of registry tasks are among the recommended strategies.

BACKGROUND: This study aims to understand whether and how participant characteristics (age, gender, education, ethnocultural identity) are related to their feedback about taking a remote, unsupervised, online cognitive assessment. METHODS: The Brain Health Registry is a public online registry which includes cognitive assessments. Multivariable ordinal regressions assessed associations between participant characteristics and feedback responses of older (55+) participants (N=11,553) regarding their Cogstate Brief Battery assessment experience. RESULTS: Higher age, secondary education or less, Latino identity, and female gender were associated with a poorer assessment experience; higher age and a non-White identity were associated with experiencing the assessment instructions as less clear; and higher age, non-White identity, and secondary education or less were associated with rating additional human support with the assessment as more useful. DISCUSSION: Our findings highlight the importance of improving the design and instructions of unsupervised, remote, online cognitive assessments to better suit the needs of diverse communities.

Objetivo. El manual pretende ser una guía, accesible y fácil de consultar, que oriente a los profesionales en la elaboración o adaptación de documentos basados en la evidencia que ayuden a la estandarización de la práctica clínica y a la toma de decisiones velando siempre por la calidad de la misma de forma que siga unos referentes establecidos. Material y métodos. La Comisión de Atención Sanitaria Basada en la Evidencia (CASBE), perteneciente a la estructura de calidad del Hospital Universitario Virgen del Rocío, propuso elaborar un manual para la elaboración de documentos basados en la evidencia que incorpore la relación de productos, la descripción de sus características, atributos, utilidad, metodología de elaboración y ámbito de aplicación de cada uno de ellos. Resultados. El manual contiene 7 herramientas basadas en la evidencia, un capítulo sobre metodología de análisis crítico de la literatura científica, un capítulo con recursos en Internet y anexos con herramientas de evaluación. Conclusiones. Este manual ofrece grandes oportunidades para la mejora de la calidad a los clínicos como guía de estandarización de la práctica asistencial, a los gestores como estrategia para promover y fomentar la elaboración de documentos que ayuden a reducir la variabilidad de la práctica clínica y a los pacientes la oportunidad de participar en la planificación de sus cuidados(AU)

The handbook consists of seven Evidence-Based tools, one chapter on critical analysis methodology of scientific literature, one chapter with internet resources, and some appendices with different assessment tools. Conclusions. This Handbook provides general practitioners with a great opportunity to improve quality and as a guideline to standardize clinical healthcare, and managers with a strategy to promote and encourage the development of documents in an effort to reduce clinical practice variability, as well as giving patients the opportunity of taking part in planning their own care(AU)

Red seal inks from Korea (6), Japan (1) and China (6) were studied to investigate the feasibility of micro-attenuated total reflectance (ATR) FTIR spectroscopy as a tool in the forensic study of questioned documents involving seal inks. The technique was able to differentiate red seal inks of similar colors and different manufacturers. Blind testing has shown that micro-ATR FTIR can identify the origin of the red seal inks with accuracy. Data gathered were converted to a database for future reference. Also, the technique was also successful in determining the sequence of heterogeneous line intersection from a personal seal and a ballpoint pen. The results show that micro-ATR FTIR can be a valuable non-destructive tool for the objective analysis of questioned documents involving different red seal inks.

Determination of simazine and diuron by high-performance liquid chromatography-ultraviolet detection (HPLC-UV) in moss-control agents, seawater and fish in aquariums was investigated and validated. The detection limits are 0.2 (simazine) and 0.4 µg l(-1) (diuron) in blank seawater, and 0.20 (simazine) and 0.30 µg kg(-1) (diuron) in blank fish homogenate, while the recoveries ranged from 93.9% to 102.4% with a relative standard deviation ≤ 9.8% for simazine and diuron. The method was successfully used in the study of simazine and diuron contamination in live fish stored in seafood restaurant aquaria in Korea. It was found that 0.4-2.3% of simazine and <0.10-3.8% of diuron were included the in moss-control agents tested. Of the 66 sampled aquarium seawaters, simazine was found to be present in four samples (3.8-42 µg l(-1)) while diuron was detected in two samples (1.3-1.6 µg l(-1)). For fish homogenates used in a bioconcentration study, simazine content ranges from 0.17 to 1.8 mg kg(-1).

Pressurized liquid extraction (PLE) system was optimized to simultaneously determine polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) in soil samples by gas chromatography with time-of-flight mass spectrometry. PLE parameters (temperature, pressure, static time, and flush volume) and packing materials (activated copper and sorbents such as: Florisil, silica gel, and a combination of Florisil and silica gel) were studied to achieve a one-step extraction and cleanup that could be analytically rapid and reliable. Method detection limit was found to be in the range of 0.1-0.4 mg/kg for PCBs and 0.1-0.6 mg/kg for PBDEs with a relative standard deviation of 1.7-7.3% for PCBs and 2.6-6.3% for PBDEs. A standard reference material for PCBs, NIST-SRM 1939a, spiked with PBDEs standard, was analyzed to substantiate the validity of the optimized method. Experimental values agreed well with the certified values with recoveries of 71.6-117%, and the optimized PLE system has been proven to be useful for the simultaneous determination of PCBs and PBDEs with various congeners in soil samples.

Bioactivity-directed fractionation of the CHCl3-MeOH extract of the leaves of Celaenodendron mexicanum by means of the brine shrimp lethality test and chromatographic techniques led to the isolation of three carboxylic acid triterpenes, the new tirucalla-type triterpene, 3 alpha-hydroxytirucalla-7,24Z-dien-26-oic acid, 3-oxotirucalla-7,24Z-dien-26-oic acid, and epi-oleanolic acid, and three biflavonoids amentoflavone, podocarpusflavone A, and podocarpusflavone B. Four non-active compounds friedelin, maytensifolin B, 3 beta-hydroxyfriedelan-16-one, and celaenodendrolide were also obtained. epi-Oleanolic acid was the most active against brine shrimps with LC50 value of 23.3 microM. In addition, all isolates were tested for in vitro antiprotozoal and cytotoxic activities. 3-Oxotirucalla-7,24Z-dien-26-oic acid and epi-oleanolic acid showed the highest activity against Leishmania donovani promastigotes with IC50 values of 13.7 and 18.8 microM, respectively. Only 3-oxotirucalla-7,24Z-dien-26-oic acid showed activity against Trypanosoma brucei brucei bloodstream forms with IC50 value of 16.8 microM.

Bioactivity-guided fractionation of Stephania dinklagei yielded six compounds including, two zwitterionic oxoaporphine alkaloids, N-methylliriodendronine, and 2-O,N-dimethylliriodendronine, two oxoaporphine alkaloids, liriodenine, and dicentrinone, one aporphine alkaloid, corydine, and one anthraquinone, aloe-emodin. Apart from corydine, the isolates have not been reported as constituents of S. dinklagei. N-Methylliriodendronine, and 2-O,N-dimethylliriodendronine are reported for the first time as natural products. All isolated compound were tested for antiprotozoal activity and cytotoxic activities in vitro. N-Methylliriodendronine was the most active against L. donovani amastigotes (IC50 = 36.1 microM). Liriodenine showed the highest activity against Leishmania donovani, and Plasmodium falciparum with IC50 values of 26.16 and 15 microM, respectively. Aloe-emodin was the only compound active (IC50 = 14 microM) against T. b. brucei.
